Prognostically Significant Fusion Oncogenes in Pakistani Patients with Adult Acute Lymphoblastic Leukemia and their Association with Disease Biology and Outcome

  • Background and objectives: Chromosomal abnormalities play an important role in genesis of acute lymphoblastic leukemia (ALL) and have prognostic implications. Five major risk stratifying fusion genes in ALL are BCR-ABL, MLL-AF4, ETV6-RUNX11, E2A-PBX1 and SIL-TAL1. This work aimed to detect common chromosomal translocations and associated fusion oncogenes in adult ALL patients and study their relationship with clinical features and treatment outcome. Methods: We studied fusion oncogenes in 104 adult ALL patients using RT-PCR and interphase-FISH at diagnosis and their association with clinical characteristics and treatment outcome. Results: Five most common fusion genes i.e. BCR-ABL (t 9; 22), TCF3-PBX1 (t 1; 19), ETV6-RUNX1 (t 12; 21), MLL-AF4 (t 4; 11) and SIL-TAL1 (Del 1p32) were found in 82/104 (79%) patients. TCF3-PBX1 fusion gene was associated with lymphadenopathy, SIL-TAL1 positive patients had frequent organomegaly and usually presented with a platelets count of less than $50{\times}10^9/l$. Survival of patients with fusion gene ETV6-RUNX1 was better when compared to patients harboring other genes. MLL-AF4 and BCR-ABL positivity characterized a subset of adult ALL patients with aggressive clinical behaviour and a poor outcome. Conclusions: This is the first study from Pakistan which investigated the frequency of5 fusion oncogenes in adult ALL patients, and their association with clinical features, treatment response and outcome. Frequencies of some of the oncogenes were different from those reported elsewhere and they appear to be associated with distinct clinical characteristics and treatment outcome. This information will help in the prognostic stratification and risk adapted management of adult ALL patients.

Development of KD- Propeller Series using a New Blade Section (새로운 날개단면을 이용한 KD-프로펠러 씨리즈 개발)

  • A new propeller series is developed using the newly developed blade section(KH18 section) which behaves better cavitation characteristics and higher lift-drag ratio at wide range of angle-of-attack. The pitch and camber distributions are disigned in order to have the same radial and chordwise loading distribution with the selected circumferentially averaged wake input. Since the geometries of the series propeller, such as chord length, thickness, skew and rate distribations, are selected by regression of the recent full scale propeller geometric data, the performance prediction of a propeller at preliminary design stage can be mure realistic. Number of blades of the series propellers is 4 and the expanded blade area ratios are 0.3, 0.45, 0.6 and 0.75. Mean pitch ratios are selected as 0.5, 0.65, 0.8, 0.75 and 1.1 for each expanded area ratio. The new propeller series is composed of 20 propellers and is named as KD(KRISO-DAEWOO) propeller series. Propeller open water tests are performed at the experimental towing tank, and the cavitation observation tests and fluctuating pressure measurements are carried out at the cavitation tunnel of KRISO. $B_{P}-\delta$ curves, which can be used to select the optimum propeller diameter at the preliminary design stage, are derived from a regression analysis of the propeller often water test results. The KD-cavitation chart is derived from the cavitation observation test results by choosing the local maximum lift coefficient and the local cavitation number as parameters. The caviy extent of a propeller can be predicted more accurately by using the KD-cavitation chart at a preliminary design stage, since it is derived from the results of the cavitation observation tests in the selected ship's wake, whereas the existing cavitation charts, such as the Burrill's cavitation chart, are derived from the test results in uniform flow.

Evaluation for Rock Cleavage Using Distribution of Microcrack Spacings (II) (미세균열의 간격 분포를 이용한 결의 평가(II))

  • The characteristics of the rock cleavage in Jurassic granite from Geochang were analysed. The evaluation for the three directions of rock cleavages was performed using the parameters such as (1) frequency of microcrack spacing(N), (2) total spacing(${\leq}1mm$), (3) mean spacing($S_{mean}$), (4) difference value($S_{mean}-S_{median}$) between mean spacing($S_{mean}$) and median spacing($S_{median}$), (5) density of spacing(${\rho}$), (6) difference value between two exponents for the whole range of the diagrams(${\lambda}_H-{\lambda}_L$), (7) mean value of exponent(${\lambda}_M$), (8) mean value of exponential constant($a_M$), (9) difference value between two exponents for the section under the initial points of intersection(${\lambda}t_H-{\lambda}t_L$), (10) mean value of exponent(${\lambda}t_M$) and (11) mean value of exponential constant($at_M$). The results of correlation analysis between the values of parameters for three rock cleavages and those for three planes are as follows. The values of (I) parameters(1, 2, 7 and 8) and (II) parameters(3, 4 and 5) are in orders of (I) H(hardway, (H1 + H2)/2) < G(grain, (G1 + G2)/2) < R(rift, (R1 + R2)/2) and (II) R < G < H. On the contrary, the values of the above two groups(I~II) of parameters for three planes show reverse orders. Besides, the values of parameter $6({\lambda}_H-{\lambda}_L)$, parameter $9({\lambda}t_H-{\lambda}t_L)$, parameter $10({\lambda}t_M)$ and parameter $11(at_M)$ for three planes are in orders of R(rift plane, (G1 + H2)/2) < H(hardway plane, (R2 + G2)/2) < G(grain plane, (R1 + H2)/2), H < G < R, H < R < G and R < H < G, respectively. The values of the above four parameters for three rock cleavages show the various orders of R < H < G, R < H < G, H < G < R and H < G < R, respectively. Meanwhile, the spacing values equivalent to the initial points of contact and intersection between the two directions of diagrams were derived. The above spacing values for three rock cleavages are in order of rift(R1 and R2) < grain(G1 and G2) < hardway(H1 and H2). The spacing values for three planes are in order of rift plane(G1 and H1) < hardway plane(R2 and G2) < grain plane(R1 and H2). In particular, the intersection angles for three rock cleavages and three planes are in order of rift and rift plane < hardway and hardway plane < grain and grain plane. Consequently, the two diagrams of rift(R1 and R2) and rift plane(G1 and H1) show higher frequency of the point of contact and intersection. These characteristics of change were derived through the general chart for three planes and three rock cleavages. Lastly, the correlation analysis through the values of parameters along with the distribution pattern is useful for discriminating three quarrying planes.

Change of physical properties after diameter increase by electroplating of orthodontic rectangular stainless steel wire (전기도금을 이용한 스테인레스 스틸 각형 선재의 굵기 증가 후 물성 변화)

  • The purpose of this study was to evaluate clinical applications of electroplating method through investigation of the physical properties of orthodontic rectangular wires according to varying their cross section. For the study, it was accomplished to electroplate the 0.016-inched orthodontic rectangular stainless steel wire. The cross section of stainless steel orthodontic rectangular wire increased from $0.016{\times}0.016inch\;to\;0.017{times}0.017inch$ by electroplating. The wire was heat treated to improve an adhesion between the wire and electroplated metal. h three-point bending test and torsion test were conducted in order to compare physical properties among three wire groups; $0.016{\times}0.016wires(group 016),\; electroplated\;0.016{\times}0.016wires(group\;016P)\;and\;0.017{\times}0.017$ wires (group 017). Through the investigations of each wire group, following results were obtained 1. At three-point bending test, the group Ol6P showed higher tendency in the degree of stiffness, yield strength and ultimate tensile strength than the group 016. Stiffness and ultimate tensile strength showed statistically significant differences between two groups at three-point bending test (p<0.05). 2. Stiffness, yield strength, and ultimate tensile strength of the group 016P showed lower tendency than those of the group 017 Stiffness showed statistically significant differences between two groups at three-point bending test (p<0.05). 3. Torque/twist rate, yield torsional moment, and ultimate torsional moment of the group 016P showed higher tendency than those of the group 016. All measurements showed statistically significant differences between two groups alter torsion test (p<0.05). 4. Torque/twist rate, yield torsional moment, and ultimate torsional moment of the group 0166P showed lower tendency than those of the group 017. Yield torsional moment, and ultimate torsional moment showed statistically significant differences between two groups after torsion test (p<0.05).

Studies on the Effects of Amylase Addition to Rice Extrusion on the Rheological Properties of the Extrudate for Weaning Food Base (압출조리를 이용한 쌀 이유식 제조에서 아밀라제 첨가가 물성변화에 미치는 효과)

  • The effects of amylase addition during extrusion cooking of rice on the physico-chemical properties of the extrudates were investigated in order to develop rice-based Korean style weaning food products. A laboratory-built single screw extruder was used, the enzymes used were Termamyl 120LS(amylase from Bacillus licheniformis, NOVO Co.), BAN 240L(amylase from Bacillus amylolichuefaciens, NOVO Co.) and malt powder. By the addition of enzymes, the water soluble index of the extrudates increased by 3 times compared to that of the extrudates without enzyme and the concentration of reducing sugar in the extrudates increased drastically at 28 feed moisture. The gel permeation chromatographic pattern showed that the large molecular starch fractions diminished by the addition of enzyme during extrusion and the low molecular fraction increased. The residual enzyme activity in the extrudate were 27% for the most thermo-resistance enzyme by treating at $140^{\circ}C$ in the metering section of the extruder. The sediment volume of the extrudate dispersion increased as the metering section temperature increased to $140^{\circ}C$ . By the addition of enzymes the viscosity of extrudate dispersion was redused $1/4{\sim}1/200$ of that of the extrudates without enzyme. It allowed to use 1.8 times of solid material to the weaning food formulation to attain the same level of consistency as the commercial products. It proves that the addition of amylase during rice extrusion is effective to increase the energy density of weaning food by starch degradation, which results in the increases of water solubility, reducing sugar content, dispersibility and fluidability.

A Study on the Guidelines on the Insertion of Metal Stiffeners in the Restoration of Stone Cultural Heritages (석조문화재 복원을 위한 금속보강재 매입방법 표준화 연구)

  • Stone cultural heritages are repaired by the use of metal stiffeners. The problem is that this type of repair has been based on the experience of workers without specific guidelines and has caused various problems. This is to suggest the structural reinforcement and behavioral characteristics of metal rods to minimize the secondary damage of materials and have the specimens tested and verified to establish the guidelines on how to insert metal stiffeners. When only epoxy resin is applied to the cut surface, only 70% of the properties of the parent material are regenerated and it is required to structurally reinforce the metal stiffener for the remaining 30%. The metal rod is under the structural behavior after the brittle failure of stone material and the structural behavior does not occur when the metal stiffener is below 0.251%. When it accounts for over 0.5%, it achieves structural reinforcement, but causes secondary damage of parent materials. The appropriate ratio of metal stiffener for the stone material with the strength of $1,500kgf/cm^2$, therefore, should be between 0.283% and 0.377% of the cross section of attached surface to achieve reversible fracture and ductility behavior. In addition, it is more effective to position the stiffeners at close intervals to achieve the peak stress of metal rod against bending load and inserting the stiffener into the upper secions is not structurally supportive, but would rather cause damage of the parent material. Thus, most stiffeners should be inserted into the lower part and some into the central part to work as a stable tensile material under the load stress. The dispersion effect of metal rods was influenced by the area of reinforcing rods and unrelated to their diameter. However, it ensures stability under the load stress to increase the number of stiffeners considering the cross section adhered when working on large-scale structures. The development length is engineered based upon the diameter of stiffener using the following formula: $l_d=\frac{a_tf_y}{u{\Sigma}_0}$. Also, helically-threaded reinforcing rods should be used to perform the behaviors as a structural material.

Taxonomic reconsideration of Lactuca hallaisanensis H. Lév. (한라고들빼기 (Lactuca hallaisanensis H. Lév.)의 분류학적 재고)

  • In order to evaluate taxonomic status of Latcuca hallaisanensis H. $L{\acute{e}}v$., an endemic species of the Jeju-do Island, we investigated fruit wall structure and chromosome morphology. The fruit wall structure had 10-11 obtuse costae in the transverse section. The costa was wholly occupied by libriform fiber cells, and the underlying fibersclereid tissue was only one to three cells layers thick. Also, the intercosta lacked fiber-sclereid layers. Somatic chromosome numbers and karyotype of Latcuca hallaisanensis were recorded for the first time. This diploid species (2n=10) with the same basic number of x=5 has the total chromosome length $23.3{\mu}m$ and the length of each chromosome falls in $1.9{\mu}m-2.9{\mu}m$. It possess the karyotype complement i.e., 3sm+2st and a characteristic chromosome pair (No. 1 and 2) with a secondary constriction at the distal portion of the short arms. The overall similarity in external morphology (involucre, achene etc), chromosome morphology as well as in fruit wall anatomy between Lactuca hallaisanensis and Crepidiastrum s. lat. clearly indicated that this species should be treated as Crepidiastrum, rather than Lactuca.

Photochemical Response Analysis on Drought Stress for Red Pepper (Capsiumannuum L.)

  • The aim of this study is to determine the drought stress index through photochemical analysis in red pepper (Capsiumannuum L.). The photochemical interpretation was performed in the basis of the relation between Kautsky effect and Photosystem II (PSII) following the measurement of chlorophyll, pheophytin contents, and $CO_2$ assimilation in drought stressed 5-week-old red pepper plants. The $CO_2$ assimilation rate was severely lowered with almost 77% reduction of chlorophyll and pheophytin contents at four days after non-irrigation. It was clearly observed that the chlorophyll fluorescence intensity rose from a minimum level (the O level), in less than one second, to a maximum level (the P-level) via two intermediate steps labeled J and I (OJIP process). Drought factor index (DFI) was also calculated using measured OJIP parameters. The DFI was -0.22, meaning not only the initial inhibition of PSII but also sequential inhibition of PSI. In real, most of all photochemical parameters such as quantum yield of the electron transport flux from Quinone A ($Q_A$) to Quinone B ($Q_B$), quantum yield of the electron transport flux until the PSI electron acceptors, quantum yield of the electron transport flux until the PSI electron acceptors, average absorbed photon flux per PSII reaction center, and electron transport flux until PSI acceptors per cross section were profoundly reduced except number of QA reducing reaction centers (RCs) per PSII antenna chlorophyll (RC/ABS). It was illuminated that at least 6 parameters related with quantum yield/efficiency and specific energy fluxes (per active PSII RC) could be applied to be used as the drought stress index. Furthermore, in the combination of parameters, driving forces (DF) for photochemical activity could be deduced from the performance index (PI) for energy conservation from photons absorbed by PSII antenna until the reduction of PSI acceptors. In conclusion, photochemical responses and their related parameters can be used as physiological DFI.

A Study on the Analysis of the Combustion Behavior and Carbonization Pattern of Vinyl Flooring on Which a Real-Scale Combustion Test Was Performed (실물 연소 실험이 진행된 비닐장판의 연소거동 및 탄화 패턴 해석에 관한 연구)

  • A real-scale combustion test was conducted on a vinyl flooring in a divided space, with 50 mL of an inflammable liquid sprayed on it. The combustion behavior of the vinyl flooring was studied in real time, and the carbonization patterns of the surface and cross-sections of the carbonized vinyl floor were analyzed. When the flame ignited by gasoline reached its peak, a continuously flaming region, intermittent flaming region, plume region, etc., were formed. The combustion of 50 mL gasoline on vinyl flooring took 26 s, and a halo pattern was observed. This test involved spraying kerosene evenly on the vinyl flooring and attempting to ignite the flooring using a gas torch, which failed. After the combustion of the vinyl flooring was complete, its carbonized range was measured to be 600 mm in length and 380 mm in width, and the carbonized area was 1000 ㎟. Heat transformed the coated layer of surface of the carbonized vinyl flooring into a carbonized layer, which became harder. The analysis of cross-section of the boundary surface of the carbonized vinyl flooring using a stereoscopic microscope showed that the vinyl flooring was bubbling, and that the white boundary layer at the bottom of the coated layer had disappeared.

Design of Microstrip Array Antenna for Satellite Reception (위성수신용 마이크로스트립 어레이 안테나 설계)

  • In this paper, the microstrip array antenna is studied to replace the parabolic antenna in the direct satellite reception. A microstrip array antenna has been used in extremely limited area, but if it is applied to practical life like a direct satellite reception antenna, we expect that it will be used in various way. First of all, if we use a microstrip array antenna for a direct satellite reception antenna, it should be guaranteed characteristics of broadband frequency. Therefore, the goal of this paper is designing technique an antenna which guarantees broadband frequency band for a direct satellite reception. In this paper, the proposed microstrip antenna is fed by orthogonal two feed lines to a rectangular patch and a sequentially rotated feeding technique is designed proposed for a good axial ratio in broadband frequency band. The rectangular patch is designed to satellite reception band, and the width and length are W=L=8.9 mm ($0.352{\lambda}o$) respectively. The antenna's ground plane has dimensions of $250{\times}250mm$. The experimental results verify that the proposed antenna had the axial ratio of above 1dB broader than that of the conventional feeding antenna. In order to verify the performance, a $8{\times}8$ array having two pairs was fabricated and tested. The maximum gain is 20.8 dB, the sidelobe level confirm less than -10 dB. It is verified by link budget calculation that C/N=6.7 dB can be obtained for domestic use if this proposed antenna is used in Koreasat reception system.
