Deletion of the oligopeptide transporter Lmo2193 decreases the virulence of Listeria monocytogenes

  • Background: Listeria monocytogenes is a gram-positive bacterium that causes listeriosis mainly in immunocompromised hosts. It can also cause foodborne outbreaks and has the ability to adapt to various environments. Peptide uptake in gram-positive bacteria is enabled by oligopeptide permeases (Opp) in a process that depends on ATP hydrolysis by OppD and F. Previously a putative protein Lmo2193 was predicted to be OppD, but little is known about the role of OppD in major processes of L. monocytogenes, such as growth, virulence, and biofilm formation. Objectives: To determine whether the virulence traits of L. monocytogenes are related to OppD. Methods: In this study, Lmo2193 gene deletion and complementation strains of L. monocytogenes were generated and compared with a wild-type strain for the following: adhesiveness, invasion ability, intracellular survival, proliferation, 50% lethal dose (LD50) to mice, and the amount bacteria in the mouse liver, spleen, and brain. Results: The results showed that virulence of the deletion strain was 1.34 and 0.5 orders of magnitude higher than that of the wild-type and complementation strains, respectively. The function of Lmo2193 was predicted and verified as OppD from the ATPase superfamily. Deletion of lmo2193 affected the normal growth of L. monocytogenes, reduced its virulence in cells and mice, and affected its ability to form biofilms. Conclusions: Deletion of the oligopeptide transporter Lmo2193 decreases the virulence of L. monocytogenes. These effects may be related to OppD's function, which provides a new perspective on the regulation of oligopeptide transporters in L. monocytogenes.

  • A gram(+) bacteria that produced microbial flocculant was isolated from soil and classified as a Bacillus species and named as Bacillus sp. A56. The culture conditions of the strain for fluocculant production were studied in a shake flask. Optimum temperature and initial pH for flcculant production were 30C and 6.5, respectively. The optimized medium has gollowing composition: glucose 20g/l, NH4NO3 0.5g/l, K2HPO4 1.0g/l, KH2PO4 0.8g/l, MgSO4.7H2O 0.2g/l, MnSO4.4-6H2O 0.3g/l, CaCO3 0.5g/l, yeast extract 0.3g/l, tryptone 0.3g/l in tap water.

봉독의 젖소 유방염 유래 그람 양성 및 음성 세균별 항균효과 분석 (Antibacterial effect of bee venom against Gram-positive and negative bacteria isolated from mastitis in dairy cattle)

  • Mastitis is an inflammatory condition of the mammary gland, most often caused by bacterial infections, resulting in significant economic losses to the dairy industry. Antimicrobial resistance has been of great concern because of the extensive clinical use of antibiotics. For this reason, the development of new compounds as an alternative treatment to bovine mastitis is needed. Bee venom has been widely used as an oriental treatment for several inflammatory diseases and bacterial infections. The aim of the present study was to evaluate the antimicrobial activity of bee venom on bacteria isolated from bovine mastitis. A total of 107 isolates from bovine mastitic milk samples collected in 2019 and 2020 in Jeonbuk province. All bacterial isolates were tested for susceptibility to bee venom of the honey bee (Apis mellifera). In order to obtain comprehensive antibacterial activities of the bee venom, we measured the minimal inhibitory concentration (MIC) of the bee venom against bacterial strains. Bee venom showed significant inhibition of bacterial growth of Gram-negative bacteria Citrobacter spp., Escherchia coli, Klebsiella spp., Pseudomonas spp., Serratia spp. and Raoultella with MIC values of 96, 81, 72, 230, and 85 ㎍/mL, respectively, and Gram-positive bacterial Enterococcus spp., Staphylococcus spp. and Streptococcus spp. with MIC values of 29, 21 and 16 ㎍/mL, respectively. The results indicated that the MIC values were different depending on the bacterial strains, and those of Gram-positive bacteria were lower than those of Gram-negative bacteria for bee venom. These findings suggested that bee venom could be an effective antimicrobial treatment for bovine mastitis; however, further research is necessary to evaluate the mechanism underlying the antimicrobial action, its effectiveness/safety in vivo and effective application for therapeutic use.

Bactericidal Effect of Combination of Atmospheric Pressure Plasma and Nisin on Meat Products Inoculated with Escherichia coli O157:H7

  • This study was conducted to investigate the bactericidal effect of nisin (Nisin) only, atmospheric pressure plasma (APP) only, and a combination of APP and nisin (APP+Nisin)(APP+Nisin) on beef jerky and sliced ham inoculated with Escherichia coli O157:H7, gram-negative bacteria. The bactericidal effect against E. coli O157:H7 and Listeria monocytogenes was confirmed using a nisin solution at a concentration of 0-100 ppm, and APP+Nisin was tested on beef jerky and sliced ham using 100 ppm nisin. Beef jerky and sliced ham were treated with APP for 5 min and 9 min, respectively. In the bacterial solution, 100 ppm nisin out of 0-100 ppm nisin exhibited the highest bactericidal activity against L. monocytogenes (gram-positive bacteria; p<0.05); however, it did not exhibit bactericidal effects against E. coli O157:H7 (gram-negative bacteria). The APP+Nisin APP+Nisin exhibited a 100% reduction rate in both E. coli O157:H7 and L. monocytogenes compared to the control group, and was more effective than the Nisin. The APP+Nisin decreased the number of colonies formed by 0.80 and 1.96 Log CFU/g for beef jerky and sliced ham, respectively, compared to the control, and exhibited a higher bactericidal effect compared to the Nisin (p<0.05). These results demonstrate the synergistic bactericidal effect of APP and nisin, providing a possible method to improve the limitations of nisin against gram-negative bacteria. In addition, this technology has the potential to be applied to various meats and meat products to control surface microorganisms.

Kenaf methanol 추출물의 가축 주요 병원성 균에 대한 항균효과 (Antibacterial Effect of Hiscus cannabinus L. Methanol Extract against Pathogenic Bacteria in Domestic Animals)

  • 본 시험은 무궁화과에 속하는 초본 식물인 Kenaf (Hibiscus cannabinus L.)를 경상대학교 시험농장에서 2010년 6월 1일 파종하여 같은 해 11월 18일 수확하여 건조 한 후, 꽃 (HCME-F)과 잎 (HCME-L)에서 추출한 메탄올 추출물을 이용하여, 세포 독성 시험 및 주요 병원균에 대한 항균효과를 규명하였다. 항균효과에 사용된 균주는 가축에서 피부 질환, 유방염 및 소화기질병을 유발하는 그람양성균인 St. aureus 와 Str. epidermidis, 그람음성균인 S. typhimurium 과 E. coli 균을 공시균주로 사용하였다. 추출물의 안전성을 평가하기 위해 두 추출물 HCME-F과 HCME-L을 0, 25, 50, 100 및 $200{\mu}g/ml$ 농도로 첨가한 배지에서 RAW 264.7 세포와 24시간 반응 후 세포 독성을 측정해 본 결과, 세포 독성이 인정되지 않았으며, 항균효과 시험결과 그람양성균인 St. aureus와 Str. epidermidis 균에 대하여 1, 50 및 $100{\mu}g/ml$의 추출물 농도 및 반응시간 경과에 따라 항균 효과가 증가되었으나, 그람음성균인 S. typhimurium 과 E. coli 에서는 항균효과가 인정되지 않았다. 종합적으로 Kenaf의 꽃과 잎에서 추출한 메탄올 추출물이 세포에 대한 안전성이 입증되었고, 가축과 사람의 피부 질환 및 유방염의 대표적 그람양성균인 St. aureus와 Str. epidermidis 균에 대한 항균효과를 보여, Kenaf의 꽃과 잎을 이용한 선택적 그람양성균 치료제 및 사료첨가제 개발이 가능할 것으로 판단된다.

지실 추출물의 세포독성과 항균효과 (Cytotoxity and Antimicrobial Effects of the Extract of Poncirus trifoliata)

  • This study was carried out to evaluate cytotoxic effects of Poncirus trifoliata Raf. extract on lymphocytic leukemia tumor (L1210) cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethyl thiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay The comparison of Ic50 Values of Poncirus trifoliata Raf. extract in L1210 cell lines showed that their susceptibility to these fractons decreased in the following order: adriamycin > Fr.4> Fr. 6> Fr. 5> Fr. 3> Fr. 1> Fr. 2 by the MTT assay. In order to develop an antumicrobial agent, Poncirus trifoliata Raf. was extracted wit ethanol, and then it was fractionated with several mobile phase. The antitumor activities of fractions of the ethanol soluble extract was investigated. The minimal inhibitory concentrations (MIC) of fractions of the ethanol soluble extract of Poncirus trifoliata Raf. against microorganisms were also examined. Antimicrobial activities of ampicillin and ketoconazole as references were compared to those of fractions of the ethanol soluble extract of Poncirus trifoliata Raf. The antimicrobial activities of all fractions from the extract had growth inhibition activities against gram-positive bacteria, gram-negative bacteria and fungi $(MIC\;>\;200{\mu}g/ml)$. These results suggest that fraction 4 of the ethanol soluble extract of Poncirus trifoliata Raf. possessed the most antitumorous agent.

흑마늘박 추출물의 항균 활성과 미생물 제어를 위한 시금치 세척에의 이용 (Antimicrobial Activity of Black Garlic Pomace Extract and Its Application to Cleansing of Fresh Spinach Leaves for Microbial Control)

  • 본 연구에서는 흑마늘 진액 가공 후 발생하는 흑마늘박의 활용 가능성을 검토하고자 흑마늘박 추출물의 항균성과 시금치에 대한 세척 적용 효과를 분석하였다. 흑마늘박 추출물은 주요 병원성 미생물인 L. monocytogenes, S. aureus, E. coli O157:H7, S. Typhimurium에 대해 모두 항균성을 나타냈으며, 특히 그람 양성균인 L. monocytogenes와 S. aureus에 더 효과적으로 작용하였다. 시금치에 0.5% 흑마늘박 추출물을 처리한 후 $4{\pm}1^{\circ}C$에서 9일 동안 저장하면서 미생물 수 변화를 측정하였다. 흑마늘박 추출물 처리는 대조구와 비교하여 저장 9일 동안 시금치의 총 호기성 세균 수를 1.23~1.35 log CFU/g, 효모 및 곰팡이 수는 0.82~1.12 log CFU/g 감소시켰다. 또한, 흑마늘박 추출물 처리는 저장 9일 동안 시금치의 Hunter 색도 값 및 총색차 값을 저장 초기와 유의적인 차이 없이 지속시켰기에 색도 품질 유지 측면에서 단순 물 세척 처리구보다 더 효과적이라고 판단된다. 따라서 본 연구 결과를 통해 흑마늘박 추출물은 병원성 미생물에 대해 높은 항균성을 가지면서 동시에 시금치와 같은 신선 농산물의 미생물 제어를 위한 효과적인 세척 처리 물질로써 활용될 수 있다고 판단된다.

Lactobacillus sp. GM7311이 생산하는 박테리오신의 Gram 양성균에 대한 작용형태 (Mode of Action of the Bacteriocin from Lactobacillus sp. GM7311 against Gram Positive Bacteria)

  • Lactobacillus sp. GM7311이 생산하는 박테리오신의 Gram 양성균에 대한 작용 형태를 알아보기 위하여, 세가지 시험균주의 TSB 배양액에 일정한 시간 간격별로 $100\;Bu/m{\ell}$ 되도록 박테리오신을 첨가한 후 그 균수 변화와 작용 특성을 살펴보았다. 우선, 시험 균주의 증식 시기별로 박테리오신을 첨가한 경우 Listeria monocytogenes는 대수증식기 후기 이후에 억제작용을 나타낸 반면, Bacillus subtilis의 경우는 대수증식기 중기 이전까지의 시험균에 대해서 강한 억제작용을 보였으며, Staphylococcus aureus는 정지기 이전의 시험균에 대해 큰 항균 효과를 나타냈다 또한 박테리오신을 처리한 세 가지 시험 균주를 전자 현미경으로 관찰했을 때, L. monocpogenes와 B. subtilis는 세포벽 파괴에 의한 세포내 물질의 유출이 관찰되었으나, S. aureus는 다른 두 균주와 달리 박테리오신에 의한 세포벽의 파괴 현상은 없었으며 다만 세포벽이 대조구에 비해 엷어진 것이 관찰되었다. 아미노산 및 지방산 조성의 변화를 조사하여 박테리오신을 처리하지 않은 대조구와 비교하였을 때, 아미노산의 경우는 세 균주 모두에서 대부분의 성분이 감소하는 경향을 보였고 지방산은 큰 조성상의 변화나 공통된 특정지방산 성분의 감소 등은 나타나지 않았으며 각 성분들의 함량이 증감하는 경향을 보였다.

식품표시 관련 교육경험에 따른 중학생들의 식품표시에 대한 인식과 활용실태 (Physical and Chemical Composition, Anti-Microbial Activity and Cytotoxicity of Kiwifruit Skin)

  • This study was conducted to examine the chemical composition of kiwifruit skin, and to est its anti-microbial activities and cytotoxicities, thus, exploring ways for the economic utilization of kiwifruit skin. Four varieties of kiwifruit were examined: Daeheung, Bidan, Haegeum No.1 and Hayward. Vitamin C content in the fruit skins of Bidan, Daeheung, Haegeum No.1 and Hayward were 72.44, 67.22, 62.51 and 61.44mg/100g, respectively. Total amino acids content in the fruit skins of Bidan, Haegeum No.1, Hayward and Daeheung ere 808.31, 706.02, 629.07 and 464.83mg/100g dry weight, respectively. K and Ca content ere $17.20-45.70{\mu}g/mL$ and $4.58-10.15{\mu}g/mL$. While, other inorganic matter contents were below $4.89{\mu}g/mL$. Anti-microbial activity of kiwifruit skin extracts, in terms of the diameter of inhibition zone when tested against five gram positive and three gram negative microbial trains (even in the concentration of 2,000mg/L), was less than 14.1mm. The hyperplasia inhibition of lung cancer cells by methanol extracts from Bidan and Haegeum No.1 using concentrations of 800mg/L were 27.7% and 14.5%, however, those from Daeheung and Hayward were below 3% Consequently, it will be useful to know that kiwifruit skin can be added to processed goods which demand for higher concentrations of vitamin C, amino acids, K and Ca.

Biochemical Characterization of $\small{L}$-Asparaginase in NaCl-Tolerant Staphylococcus sp. OJ82 Isolated from Fermented Seafood

  • $\small{L}$-Asparaginase from gram-positive bacteria has been poorly explored. We conducted recombinant overexpression and purification of $\small{L}$-asparaginase from Staphylococcus sp. OJ82 (SoAsn) isolated from Korean fermented seafood to evaluate its biotechnological potential as an antileukemic agent. SoAsn was expressed in Escherichia coli BL21 (DE3) with an estimated molecular mass of 37.5 kDa, determined using sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Consistent with asparaginases in gram-negative bacteria, size-exclusion chromatography determined SoAsn as a homodimer. Interestingly, the optimal temperature of SoAsn was $37^{\circ}C$ and over 90% of activity was retained between $37^{\circ}C$ and $50^{\circ}C$, and its thermal stability range was narrower than that of commercial E. coli $\small{L}$-asparaginase (EcAsn). Both SoAsn and EcAsn were active between pH 9 and 10, although their overall pH-dependent enzyme activities were slightly different. The $K_m$ value of SoAsn was 2.2 mM, which is higher than that of EcAsn. Among eight metals tested for enzyme activity, cobalt and magnesium greatly enhanced the SoAsn and EcAsn activity, respectively. Interestingly, SoAsn retained more than 60% of its activity under 2 M NaCl condition, but the activity of EcAsn was reduced to 48%. Overall, the biochemical characteristics of SoAsn were similar to those of EcAsn, but its kinetics, cofactor requirements, and NaCl tolerance differed from those of EcAsn.
