A cDNA clone encoding phytocystatin was isolated from Brassica rapa seedlings, through rapid amplification of cDNA ends (RACE). This gene (name as Brcpi1; GenBank accession no.: EF079953) had a total length of 881 bp with an open reading frame of 609 bp, and encoded predicted polypeptide of 203 amino acid (aa) residues including a putative N-terminal signal peptide. Other relevant regions found its sequence included the G and PW conserved aa motifs, and the consensus LARFAV sequence for phytocystatins and the reactive site QVVAG. The BrCPI1 protein shared 95, 94, 81, 80 and 78% identity with other CPI proterins isolated from Brassica oleracea (BoCPI-1), Arabidopsis thaliana (AtCY SB), Glycine max (GmCPI), Oryza sativa (OsCYS-2) and Zea may (ZmCPI) at amino acid level, respectively. Southern blot analysis showed that Brcpi1 was a low copy gene. Expression pattern analysis revealed that Brcpi1 was a tissue-specific expressing gene during reproductive growth and strongly expressed at mature seedling stages. Furthermore, overexpression of Brcpi1 in transgenic Arabidopsis was enhanced tolerance to salt and cold stresses. Meanwhile the juvenile seedling of Brcpi1 transgenic plants was not affected by various concentrations ABA in MS medium. Taken together, the results showed that Brcpi1 functioned as a cysteine protease inhibitor and it exhibited a protective agent against diverse types of abiotic stress, which induced this gene in a tissue- and stress-specific manner.

This study was designed to potentiate the vasodilation effect of Banhabackchulchunma-Tang(BCT) prescription by change of mixture. Six different BCT compositions were made according to mixture of herbs. The vascular relaxation effects of 6 different BCT compositions were examined on phenylephrine(PE)-precontracted rat thoracic aorta. The BCT-1 composition exerted significant relaxation on phenylephrine- or KCI- contracted rat thoracic aorta. Its elaxation was endothelium- independent in both PE- and KCl-induced contraction. Treatment of glibenclamide or tetraethylammonium(TEA) did not affect the relaxation of BCT-1. Vasorelaxation efficacy of BCT-1 was also not influenced by low (25mM) or high (50mM, 80mM) KCl-induced contraction. Furthermore, the contraction by increasing $Ca^{2+}$ concentrations (0.3-10.0mM) to a $Ca^{2+}$-free high K+ (60mM) was significantly reduced by pretreatment with BCT-1 In addition, the relaxant effects were not inhibited by pretreatment of rat aorta with L-NAME, MB, indomethacin and atropine. These results confirm that BCT-1 may exerts its vasodilation effect by endothelium-independent manner. According to the above results, we suggest that the relaxation effect of BCT-1 is endothelium-independent and is related with block of $Ca^{2+}$ influx via $Ca^{2+}$ channel.

This study was performed for the investigation of vasodilatory efficacy and its underlying mechanisms of Jagumhuan(JGH), a herbal remedy. JGH produced completely endothelium-dependent relaxation and relaxed phenylephrine(PE)-precontracted aorta in a concentration dependent manner. The magnitude of relaxation was greater in PE induced contraction than that of KCl, suggesting involvement of $K^+$ channel in the relaxant effect. Both glibenclamide$(10^{-5}M)$, a $K_{ATP}$ channel inhibitor and indometacin, a cyclooxygenase inhibitor, completely prevented this relaxation. The relaxation effects of JGH, involve in part the release of nitric oxide from the endothelium as pretreatment with L-NAME, an NOS inhibitor, and methylene blue, a cGMP inhibitor, attenuated the responses by 62% and 58%, respectively. In addition, nitrite was produced by JGH in human aortic smooth muscle cells and human umbilical vein endothelial cells. The relaxant effect of JGH was also inhibited by 55.41% by tetraethylammonium(TEA; 5mM), a $K_{Ca}$ channel inhibitor. In the absence of extracellular $Ca^{2+}$, pre-incubation of the aortic rings with JGH significantly reduced the contraction by PE, suggesting that the relaxant action of the JGH includes inhibition of $Ca^{2+}$ release from intracellular stores. These results indicate that in rat thoracic aorta, JGH may induce vasodilation through ATP sensitive $K^+$ channel activation by prostacyclin production. However, the relaxant effect of JGH may also mediated in part by NO pathways and $Ca^{2+}$ activated $K^+$ channel.
To assess the university health service organization(UHSO) and its school health programs a questionnaire survey for all of 186 four-year colleges in Korea was conducted from December 1. 1998 to March 31. 1999. The response rate was 91.9 percent(171 universities). The UHSO was established in 116 universities(67.8%L The most common name of UHSO. used in 61 universities(52.6%). was school health center. Only 45(38.8%) of the 116 universities surveyed had a committee to support and run the UHSO. The percentage of universities that had committee was higher in national universities and those with a large number of students. The average number of staff working in the UHSO was 3.5; 4.6 in national universities; and 7.1 in universities with more than 15.000 students. There were 43 universities(37.1%) which had a part-time physician and 104 universities (89. 7%) had full-time nurses. Only 4 universities(3.4%) had a independent facility for the UHSO while most of UHSO were housed in other building. The UHSO had an independent budget in 86 universities(74.1%). The average budget per university was 46.890.000 won: private universities had more budget with 59.170.000 won on average than national universities with 36.990.000 won. The average budget allocated per student was 4.362 won. A regular physical examination was performed in 72 universities(62.1%). The percentage of university that performed regular physical examination was higher in private universities than in national universities. Health counseling was performed in 113 universities(97.4%) and vaccinations in 87 universities(75.0%). Medical care services were performed in 87 universities(75.0%). and the private universities and those with over 10.000 students provided the student with more services. Environmental sanitation was executed in 73 universities(62.9%) and health education was conducted in 68 universities(58.6%)' The school health management activities were promoted in 104 universities(89.7%) out of 116 universities with UHSO. Only 12 universities(10.3%) published reports on the achievements and performance of the UHSO. Only 29 universities(25.0%) had a continuing education for employees of the UHSO.
This study was to evaluate the protein profile of seminal plasma using 2-DE in Hanwoo. Seminal plasma was harvested from five mature Hanwoo, and seminal plasma protein was extracted by M-PER Mammalian Protein Extraction Reagent. Proteins were refined by clean-up kit and quantified by Bradford method until total protein was $300\;{\mu}l$. Immobilized pH gradient (IPG) strip was used 18 cm and 3~11 NL. SDS-PAGE was used 12% acrylamide gel. Each gels were visualized by comassie brilliant blue and silver staining. These spots were analyzed by MALDI-TOF MS and searched on NCBInr. The result, 20 proteins of 36 protein spots were searched through peptide sequencing on the NCBInr. 8 proteins profiled by 2-DE were proved through previous bovine studies and the name of each protein was albumin, nucleobindin, clusterin, TIMP-2, spermadhesin Z13, spermadhesin-1 and BSP proteins (BSP 30 kDa and BSP A1/A2). 12 new proteins were ATP synthase, protein MAK16 homolog, Transmembrane protein 214, E3 ubiquitin-protein ligase BRE1A, dual serine/threonine and tyrosine protein kinase, tissue factor pathway inhibitor 2, alpha-actinin-4, RUN domain-containing protein 3B, catenin alpha-1, protein-glutamine gamma-glutamyltransferase 2, plakophilin-1 and inter-alpha-trypsin inhibitor heavy chain H1 has not been previously described in the bovine seminal plasma study. These proteins may be contribute to define the type of proteins affecting fertility of male and improve the fertilizing ability of semen in Hanwoo.

Diabetes mellitus is a chronic disorder which affects millions of population worldwide. Global estimates published in 2010 reported the world diabetic prevalence as 6.4%, affecting 285 million adults. Foot ulceration and wound infection are major forms of disabilities arising from diabetic diseases. This study was aimed to develop a natural antimicrobial finishing on medical grade textile that meets American Association of Textiles Chemists and Colorists (AATCC) standard. The textile samples were finished with the ethanolic extract of Penicillium amestolkiae elv609, an endophytic fungus isolated from Orthosiphon stamineus Benth (common name: cat's whiskers). Endophyte is defined as microorganism that reside in the living plant tissue, without causing apparent disease symptom to the host. The antimicrobial efficacy of the ethanolic extract of P. minioluteum was tested on clinical pathogens isolated from diabetic wound. The extract exhibited significant inhibitory activity against 4 bacteria and 1 yeast with the minimal inhibitory concentration ranged from 6.25 to 12.5 mg/mL. The results indicate different susceptibility levels of the test microorganism to the ethanolic extract. However, the killing activity of the extract was concentration-dependent. The finished medical textile showed excellent antimicrobial efficacy on AATCC test assays. All the microbial cultures treated with the textile sample displayed a growth reduction of 99.9% on Hoheinstein Challenge Test. The wash durability of the finished textile was found good even after 50 washes with commercial detergent. Besides, the gas chromatography mass spectrometry analysis showed that 6-octadecenoic acid and diethyl phthalate were the main bioactive constituents of the extract. In conclusion, the developed medical textile showed good antimicrobial efficacy on laboratory tests. This work can be extended to in vivo trials for developing healthcare textile products for antimicrobial applications.
Objective : The Annals of the Joseon Dynasty is a primary historical record that has provided a great deal of information about what the Joseon Dynasty was like. However, as of yet, we know very little about the medical officers in Joseon dynasty, such as their government posts and official ranks. The purpose of this study is look in to the activities, government posts, and official ranks of the medical personnel by examining Yeongjosillok. Methods : First, I selected historical records containing '醫' in Yeongjosillok. Then, I organized medical officers' name by reading each record. I screened historical records in Yeongjosillok with their names to analyze their activities, government posts, and official ranks. When there was limited information available, I referred to The Daily Records of Royal Secretariat of Joseon Dynasty. Results : I found 262 historical records in Yeongjosillok containing '醫'. Then I found 26 people who served as medical officers in Yeongjosillok. Also, l found that 11 government posts and 7 official ranks were awarded to them throughout the 110 historical records in Yeongjosillok and The Daily Records of Royal Secretariat of Joseon Dynasty. Conclusion : Through this study, I was able to examine the detailed activities of unknown medical officers by studying the historical records in Yeongjosillok and The Daily Records of Royal Secretariat of Joseon Dynasty. Under the Joseon Dynasty's class-based society, the middle class had various restrictions. However, I found that medical officers that belonged to the middle class received exceptional treatment despite their social status.
Nitric oxide (NO), a diffusible gas, is produced in the central nervous system, including the spinal cord dorsal horn and the trigeminal nucleus, the first central areas processing nociceptive information from periphery. In the spinal cord, it has been demonstrated that NO acts as pronociceptive or antinociceptive mediators, apparently in a concentration-dependent manner. However, the central role of NO in the trigeminal nucleus remains uncertain in support of processing the orofacial nociception. Thus, we here investigated the central role of NO in formalin (3%)-induced orofacial pain in rats by administering membrane-permeable or -impermeable inhibitors, relating to the NO signaling pathways, into intracisternal space. The intracisternal pretreatments with the NO synthase inhibitor L-NAME, the NO-sensitive guanylate cyclase inhibitor ODQ, and the protein kinase C inhibitor GF109203X, all of which are permeable to the cell membrane, significantly reduced the formalin-induced pain, whereas the membrane-impermeable NO scavenger PTIO significantly enhanced it, compared to vehicle controls. These data suggest that an overall effect of NO production in the trigeminal nucleus is pronociceptive, but NO extracellularly diffused out of its producing neurons would have an antinociceptive action.
To investigate whether $AT_{1}$ receptor antagonists are acting by increasing endothelium-de-pendent and -independent relaxation of aortas in normotensive rats, $AT_{1}$ receptor antagonists, losartan and KR-30988, and angiotensin converting enzyme inhibitor, captopril, were orally administered for two weeks (50 mg/kg, b.i.d.). THe blood pressure, heart rate and body weight were not significantly changed by losartan, KR-30988 and captopril compared to the control group. In aortic preparations, the $pD_{2}$ of KR-30988 for ACh-induced relaxation was 8.33 $\pm$ 0.16, significantly (p <0.05) lower than that of control group $(7.71 \pm 0.15)$. ACh-induced relaxation was significantly increased on losartan-treated group (p<0.01) at $10^{-6}$ M of ACh, and in captopril-treated group (p<0.05) at the range of $10^{-7}$ -$10^{-5}$ M of ACh. The $pD_{2}$ values for histamine-induced relaxatio of losartan, KR-30988 and captopril were 5.57 $\pm$ 0.10, 5.85 $\pm$ 0.21 and 5.60 $\pm$ 0.01, respectively, with significant differences in all groups (p<0.01) compared to that of control group (5.13 $\pm$ 0.09). ACh-induced relaxations of aortic preparations were not changed by pretreatment of indomethacin ($10_{-5}$ M), and completely bolcked by pretreatment of L-NAME $(10_{-5}M)$ in all groups. Sodium nitroprusside-induced relaxations were not significantly changed by all drugs tested in this experiments. These results suggest that $AT_{1}$ receptor antagonists, losartan and KR-30988, enhance the endothelium-dependent relaxatio on aortic preparations through the release of, or increase sensitivity, to nitric oxide in nor-motensive rats.
We report a newly naturalized species of the genus Bidens L. (Asteraceae) in Korea, specifically Bidens subalternans DC. var. subalternans. This naturalized plant was discovered Geumjung-gu in Busan; Gyeongsan-si, Jillyang-eup in Gyeongbuk-do; and in Buk-gu in Daegu. Bidens subalternans var. subalternans is distinguishable from other Bidens in Korea by the following combination of characteristics: an odd-pinnate leaf, deeply lobed, narrow lanceolate, four lobes in the corolla, an oblong achene shape. This taxon is termed 'Wang-do-kka-bi-ba-neul' as a common Korean name based on its large size (in particular, the size of leaf). The key for all taxa of the genus Bidens in Korea is also provided here.
