The objective of this study was to determine the optimum ratio of methionine to methionine plus cystine for growing pigs. A nitrogen balance trial was conducted using a total of 21 barrows (Large WhiteLandrace) over two replicates. The initial body weight was $20.36{\pm}1.22kg$ (mean${\pm}$SD) in the first replicate and $23.54{\pm}1.02kg$ (mean${\pm}$SD) in the second. For each replicate, the 21 pigs were randomly assigned to one of seven dietary treatments with three observations per treatment. The diets included a methionine and cystine-deficient basal diet with all other essential nutrients meeting nutrient requirements and six diets formulated with graded levels of DL-methionine (0.00, 0.03, 0.06, 0.10, 0.13, 0.16%) and $L-Cystine{\cdot}HCl{\cdot}H_2O$ (0.19, 0.15, 0.11, 0.07, 0.04, 0.00%). This resulted in ratios of methionine to methionine plus cystine of 41.3, 29.6, 35.3, 41.2, 46.0, 51.6 and 57.5%. Each experimental period lasted 12 days consisting of a seven-day adaptation period followed by a five-day total collection of urine and feces. During the collection period, pigs were fed 900 g/day for the first replicate and 1,200 g/day for the second replicate. The feed was provided in three equal portions at 0800, 1500, and 2200 h daily. Pigs had ad libitum access to water after feeding. There was a linear (p<0.01) and quadratic (p<0.01) effect on daily gain and feed conversion as the ratio of methionine to methionine plus cystine increased. Pigs receiving the diets providing a methionine to methionine plus cystine ratio of 51.6% had the best daily gain and feed conversion. Plasma urea nitrogen was also lowest for this treatment. Nitrogen retention increased (p<0.01) as the relative proportion of methionine increased up to 51.6% and then a downward trend occurred at 57.5%. The quadratic regression model, as well as one- and two- slope regression line models, were used to determine the optimum ratio of methionine to methionine plus cystine. Eliminating the 35.3% methionine to methionine plus cystine treatment resulted in $R^2$ values in excess of 0.92. The optimal ratio of methionine to methionine plus cystine was estimated to be 54.15% for nitrogen retention and 56.72% for plasma urea nitrogen.

S. natalensis에서 클로닝된 metK는 1,209 bp의 염기를 가진 유전자로써 아미노산서열에서 S. pristinaespiralis ATCC 25486과 S. peucetius ATCC 27952의 MetK와 96%, S. violaceusniger Tu 4113과 95% 일치하는 매우 높은 상동성을 보였다. 또 pSET152ET 벡터를 이용해 구축한 metK 고발현용 재조합 플라스미드 pCD1를 S. lividans TK24의 genomic DNA에 도입하여 actinorhodin 생산 유도를 시도해 본 결과 R5 고체배지에서 pCD1이 도입되지 않은 균주에서는 actinorhodin 생산을 전혀 확인할 수 없었지만 pCD1이 도입된 형질전환체에서는 actinorhodin 생산이 강하게 유도되었으며 R4 액체배지에서는 actinorhodin 생산량이 10배 증가되었다. 따라서 본 연구를 통해 클로닝된 S. natalensis 유래 metK 유전자는 방선균에서 이차대사산물의 생산을 유도할 수 있음을 확인할 수 있었다.
We developed simple and highly efficient synthesis method for [$^{11}C$]methionine using solid-phase extraction method. For synthesis, we used C18 cartridge. [$^{11}C$]methionine was synthesized on C18 cartridge according to the solid-phase [$^{11}C$]methylation of precursor L-homocysteine thiolactone hydrochloride. The radiochemical yields of [$^{11}C$]methionine was $48.9{\pm}7.93%$ decay corrected (results of 30 syntheses, mean$\pm$SD), with average production higher than 180 mCi. This procedure showed high yield and simple synthesis of [$^{11}C$]methionine.
Soybean is an important crop because its seed has very high protein relative to others. The quality of soy protein is limited by the concentration of the sulfur-containing amino acids in the amino acid profile. Among the supply of various forms of 0.4mM sulfur as S nutrition during seed fill. only 0.4mM L-methionine can inhibit ${\beta}$-subunit synthesis completely and produce the highest glycinin-containing seeds. Compared to 0.4mM sulfate control, seeds supplied by 0.4mM L-methionine have lower ${\alpha}$-, no ${\beta}$-subunit, and highly increased glycinin without altering total protein concentration. Supply of 0.2mM cystine (0.4mM S) did not affect the accumulative pattern of seed storage protein (SSP) subunits. In the supply of L-methionine, 0.2mM treatment showed higher glycinin in seeds but 0.05mM resulted in lower glycinin than tile sulfate control. The relative abundance of ${\alpha}^`$-subunit was not altered by any N or S nutrition. Under 5mM nitrogen, protein concentration was increased about 3-5% by substituting ammonia for nitrate during seed fill independent of nutrition. The increase resulted in the only increase of 7S protein, mainly ${\beta}$-subunit. Our data suggest that the regulatory system of SSP genes responds to the balance between N and S assimilates supplied from mother plant. and controls the di fferential synthesis of their subunits for the maximum protein accumulation in developing soybean seed.
A new isolate, Bacillus sp. ALK-7 can synthesize ethylene from l-aminocyclopropane-l-carboxylic acid (ACC) as well as from methionine. The ACC has only been recognized as a key intermediate found in the metabolic pathway leading to ethylene formation in various plants. The efficiency of ethylene formation from the ACC by Bacillus sp. ALK-7 was about 2 times as high as that from the methionine. The reaction from ACC to ethylene formation was also shown to be mediated by the cell-free extracts of Bacillus sp. ALK-7.
Experiments were conducted to evaluate the nutritive values of supplemental L-lysine, liquid and powder type, and DL-methionine in weanling pigs. For feeding trial, 165 weanling pigs were treated in 2 controls; 18 and 16% CP, 6 supplementations of lysine alone to 16% CP diets; 0.1, 0.2 and 0.4% of liquid and powder type each, and 3 supplementations of lysine + methionine to 15% CP diets; 0.05 + 0.025, 0.1 + 0.05 and 0.2 + 0.1%. Pigs were fed for 5 week to investigate the protein sparing effect of supplemental amino acid, and the optimal supplemental level. A metabolic trial included the measurements of digestibilities of dry matter, crude protein, crude fat, crude fiber, energy, phosphorus and amino acids. The liver acinar cell culture was conducted for the protein synthesis activity of the pigs fed each experimental diet. Supplementation of both type of L-lysine in 16% CP diet showed improved daily weight gain and feed efficiency which were compatible with those of pigs fed 18% CP diet. Groups fed liquid lysine did not differ from those fed powder type in growth performance. Supplementation of lysine and methionine to 15% CP diet did not improve growth performance of pigs to the extent that 18% CP diet was fed. In nutrient digestibility, 16% CP control diet showed significantly (p < 0.05) lower crude protein digestibility than any other treatments. Digestibilities of 16% CP diets with lysine supplementation were equal to that of 18% CP control, while digestibilities of 15% CP diets with the supplementation of lysine + methionine was inferior to that of 18% CP control. Supplementation of lysine alone reduced the nitrogen excretion compared to the none supplemented control groups. However, addition of lysine + methionine excreted more nitrogen than controls. Pigs fed diet supplemented with lysine alone, or lysine + methionine excreted less fecal phosphorus than those fed none supplemetation. Retained protein from liver tissue of pigs fed 18% diet was significantly (p < 0.05) greater than those fed 16% CP diet. A significant difference (p < 0.05) was observed in physical type of lysine. Feeding of powder type showed less secreted protein and greater retained protein in the culture of liver acinar cell. It is concluded that supplementation of lysine at the level of 0.1 to 0.2% can spare 2% of dietary protein and reduce nitrogen excretion by 19.3%. Also, no difference in nutritional values was observed between liquid and powder lysine in weanling pigs.
Selenium(Se)은 사람에게 필수성분이지만 독성이 강한 물질이다. 따라서 생체에서 Se(셀레늄)의 농도조절은 정확하고 효율적으로 이루어져야 한다. 사람은 Se을 음식의 형태로 섭취해야 하고 주로 seleno-L-methionine (L-SeMet)의 형태로 섭취한다. 섭취된 L-SeMet은 methionine 대사와 동일한 효소를 이용하여 Se-adenosyl-L-SeMet으로 대사된다고 알려져 있었다. 그러나 최근 쥐의 간 추출물의 실험에서, L-SeMet이 cystathionine ${\gamma}$-lyase (mouse CGL)의 작용으로 methylselenol ($CH_3SeH$)로 직접적으로 대사된다는 보고가 있었다. CGL은 원래 cystathionine을 L-cysteine과 ${\alpha}$-ketobutyrate, 그리고 $NH_3$로 분해하는 효소로 알려져 있었다. 따라서 본 연구에서는 쥐의 간 추출물 대신에 인간의 CGL을 분리 정제하여 L-SeMet에서 methylselenol의 형성을 확인하고자 하였다. Methylselenol의 표준시료는 dimethyldiselenide를 sodium tetrahydroborate로 환원시켜 준비하였다. 그리고 L-SeMet을 기질로 사용한 효소 반응액 중에서 가스상의 생성물은 GC/MS 스펙트럼으로 분석하였다. 분석 결과 methylselenol의 유도체인 dinitrophenyl selenoether와 일치하였다. 또한 인간 CGL이 L-SeMet에서 methylselenol을 형성하는 반응의 kinetic parameter를 mouse CGL과 비교 분석하였다. 결과적으로 human CGL은 섭취된 L-SeMet의 대사를 책임지고 있는 중요한 효소이다.
It is known that overexpression of S-adenosyl-L-methionine (SAM) synthetase or exogenous addition of SAM enhances the production of actinorhodin, one of pigmented antibiotics found from Streptomyces coelicolor. In order to discover a novel compound as a signal molecule to produce actinorhodin instead of SAM, several compounds were synthesized based on the relationships between structures of the SAM analogues and their actinorhodin productivities. Of these, a few compounds showed better productivities of actinorhodin than SAM.
S-Adenosyl-L-methionine (SAM) is a bioactive material used in the treatment of depression, osteoarthritis, and liver disease. To obtain lactic acid bacteria (LAB) producing high concentrations of SAM, LAB were isolated from commercial kimchi and from prepared kimchi products that contained shrimp jeotgal (fermented salty seafood) or sand lance jeotgal or that were fermented at 5 or $10^{\circ}C$, respectively, when pH was 4.2 to 4.8 and titratable acidity 0.6 to 0.9. Among the 179 LAB strains isolated from the fermented kimchi products, the genus Leuconostoc produced the highest intracellular level of SAM (1.58 mM) and Lactobacillus produced the second highest level (up to 1.47 mM) in the strain culture. This is the first study to quantify SAM in LAB isolated from fermented kimchi prepared by a general kimchi recipe. Ultimately, the selected strains (Leuconostoc mesentroides subsp. mesenteroides/dextranicum KSK417, L. mesentroides subsp. mesenteroides/dextranicum KJM401, and Lactobacillus bifermentans QMW327) could be useful as starters to manufacture fermented foods containing high levels of SAM.
Anionic (sodium lauryl sulphate, NaLS) cationic (cetyl ammonium bromide, CTAB) and non-ionic (Tween-80) surfactants have been found to inhibit the rate of oxiadation L-proline and L-methionine by alkaline $KMnO_4$. A first order dependence of rate of oxidation was observed with respect to $MnO_4{^-}$. The order of reaction in substrate and alkali was found to be fractional nearby 0.65 and 0.55 in Aminoacid and $OH^-$, respectively. An aggregation/association between $MnO_4{^-}$ and surfactant has been confirmed spectrophotometrically. A mechanism, involving kinetically inactive [$MnO_4{^-}$ surfactant] aggregate and consistent with kinetic data, has been proposed. The effect of surfactants has been discussed in terms of hydrophobic and electrostatic interactions.
