The in vitro regeneration was established, and the genetic stability among the mother plants (control) and the micropropagated green plants was evaluated using ISSR markers in Imperata cylindrica 'Rubra', Poaceae which containing important bioenergy plants. Green shoots were multiply induced from growing point culture via callus on MS medium supplemented with 0.01 mg/L NAA and 2 mg/L BA, and the shoots were proliferated on the MS medium with rooting. Rooted plantlets were transplanted to the pot with 100% survival rate. Using ISSR markers, somaclonal variation was analyzed in eight mother plants (control), ten green-regenerant cultivated at culture room (ReR) and ten green-regenerant cultivated at field condition (ReF). All ISSRs produced a total of 97 bands, and the scorable bands varied from one to seven with an average of 4.4 bands per primer. The polymorphism rate of ReRs and ReFs was 4.1% and 3.1% respectively, showing higher rate than that of control (0%). The genetic similarity matrix (GSM) among all accessions ranged from 0.919 to 1.0 with a mean of 0.972. According to the clustering analysis, ReFs and mother plants were divided into two independent groups. The results indicate that no clear genetic diversity was detected among regenerated plants, and ISSR markers were useful tool for identification of somaclonal variation of regenerants.

Sunflower (Sunking4505) and Rape (Sunmang) are oil-seeds containing high oleic acid, and these are used for the production materials of bio-diesel and applying for phytoremediation. The effect of cadmium on germination rate and the growth of the plants is evaluated. Object seeds were placed in Cd (0, 1.5, 4, 12, 30, 60, 100, 150, 300, 500 mg/L) solutions for seven days, and germination rate, root length, shoot length, seedling length, and dry weight were observed. $IC_{50}$, seedling vigor index, and tolerance indices were computed, and data were statistically analyzed by Analysis of Variance (ANOVA). Germination rate as well as root, shoot, and seedling length decreased as the cadmium concentration increased except dry weight. The $IC_{50}$ of sunflower and rape are 112 and 10 mg-Cd/L, respectively. Only one of the sunflower seeds is germinated at 500 mg-Cd/L whereas rape seeds are not germinated more than 150 mg-Cd/L solution. Root has higher cadmium sensitivity than shoot, and sunflower has higher germination rate, growth, and seedling vigor index than rape. In case of tolerance indices, sunflower has lower value than rape at relatively low concentration, but has higher value at high concentration.
The author succeeded in rearing the young blue crab from the first stage of zoe ato the true crab shape, and during this time he observed their growth and metamorphosis. The relationships between the number of eggs carried by female crabs (E) and the carapace width (C) and body weight (W) are shown as follows: E= 27.9049C-281.8155, E=0.5682 W-116.4606. There are five zoeal stages and a megalopa in the complete larval development of the blue crab. Water temperature in rearing aquaria ranged from 21.4 to $25.2^{\circ}C$. The duration of each zoeal stage was two days on the average. After the fifth moulting, the zoea becomes megalopa and 5 to 6 days later the megalopa moults and develops into the first stage of adult crab shape. The carapace width of megalopa measured about 1.70 mm and the carapace length, from the tip of the rostrum to the posterior dorsal margin of the carapace, was about 2.78 mm on the average. The carapace width and length of the first crab, 18 days after hatching, measured about 4.48 mm and 2.62 mm respectively. After two days, the first crab moulted and grew into the second crab with about 6.47 mm in carapace width and 4.66 mm in carapace length. The larval rearing in the outdoor tank shelved better results than in the indoor aquarium. The highest mortality occurred when the first stage of zoea moulted into the second stage. Percentage of crabs which survived, from the first crab to the ninth crab stages, was about $55\%$. The relationships between rearing days (D) and the carapace width (C), carapace length (L) and body weight (W) of the crab stages during 40 days of rearing are shown as follows. Carapace width, Indoor: C=1.1250D+1.7227 Outdoor C=1.3465D -0.2449 Carapace length, Indoor: L=0.6654D+1.6712 Outdoor: L=0.7893D+0.6919 Body Weight, Outdoor: $$W=1.15e^{0.12423D}$$ Indoor: $$W=6.759\times10^{-2}D^{1.2598}$$ (9-19 day old crabs) Outdoor: $$W=4.136\times10^{-2}D^{1.6024}$$ (21-40 day old crabs) During the crab stage, the following relationships between the number of moulting times and the carapace width (C), carapace length (L) and body weight (W) were found as follows: $$C=5.2e^{0.28119N}$$$$L=3.65e^{0.26372N}$$$$W= 0.14e^{0.7037N}$$ The relationships between the carapace length (L) and the carapace width (C) and body weight (W) of the crab stages are shown as follows: Carapace length, mm Formula 2.62-27.17 L=1.6864C-1.0387 7.47-18.53 $$W=9.367\times10^{-5}C^{3.5567}$$ 22.11-27.17 $$W=3.406\times10^{-5}C{3.8571}$$
Objectives : High co-morbidity of periodic limb movements during sleep(PLMS) and obstructive sleep apnea syndrome(OSAS) is well known and their incidences tend to increase in the elderly. Previous studies have inconsistently rep0l1ed increase or no change of periodic limb movement index(PLMI) by nasal continuous positive airway pressure(CPAP) in OSAS without analyzing possible variables affecting PLMI. We attempted to examine PLMI change evoked during CPAP titration and also factors affecting it in OSAS. Methods : Twenty-nine OSAS patients(M:F=26:3, mean age: $51.6{\pm}10.6\;yrs$) without other sleep disorders except for PLMS were selected, based on the nocturnal (baseline) polysomnograhy. Another night of noctumal polysomnography was performed for CPAP pressure titration. We compared between those two nights PLMI, mean and lowest $SaO_2$, and sleep variables. We also calculated PLMI differences between baseline and CPAP nights, named as delta PLMI (value of CPAP night PLMI minus value of baseline night PLMI). Correlations were calculated between delta PLMI and factors such as age, body mass index, applied CPAP pressure, baseline night values of respiratory disturbance index, mean and lowest $SaO_2$, and sleep parameter differences between baseline and CPAP nights. Results : Decrease of RDI(p<.01) and increase in mean and lowest $SaO_2$ (p<.05, p<.01) were observed during CPAP night. No sleep parameters showed significant change except for the decrease of total stage 1 sleep%(p<.01) during CPAP night. Ten out of 29 patients showed PLMI increase, while the other 19 patients showed either no change(n=14) or even PLMI decrease(n=5) during CPAP night. The 10 patients showing PLMI increase during CPAP night showed a significant positive correlation between delta PLMI and baseline night RDI(p<.05), which meant that PLMI increase was found to be more prominent in higher RDI patients than in lower RDI ones. There were no significant correlations between delta PLMI and other factors in the other 19 patients. Conclusions : We suggest that during the baseline night PLMS would have been underscored and/or masked due to the overlapping of PLMS and apneas/hypopneas or the arousals induced by apneas/hypopneas. Despite its still unknown mechanism, the CPAP application may unmask PLMS and increase PLMI in a subgroup of OSAS patients. It needs to be evaluated further whether the chronic CPAP use sustains the above finding.
Formerly, adult-tiger puffer, Takifugu rubripes with ova caught in the sea, were used for seedling production. But it was difficult to secure naturally-ripened adults. For the purpose of adult tiger puffer in captivity, this study was carried out. To determine the growth 220 tiger puffers hatched in 1990 (3-year-old) and 1991 (2-year-old) were used. For spawning and egg incubation leading to fry development, eggs were stripped from tiger puffers hatched in 1988 (5-year-old) and 1990 (3-year-old) through human chorionic gonadotropin (BCG) treatments. In May, 1993, mean body length and mean body weight of 2-year-old tiger puffer were $30.72\pm1.35cm\;and\;1,048\pm228 g,$ and that of 3-year-old tiger puffers were $36.02\pm1.17cm$ and $1,402\pm66g$ respectively. The relationship between body length (L) and body weight (W) of 2-year-old the tiger puffers during the experiment period was represented as $W\;=\;1.7892L^{31524}\times10^5$ (r= 0.9436) and that of 3-year-old, $W=\;3.2840L^{36099}\times10^6$ (r= 0.9070) respectively. The GSI in female 2-year-old-fish changed from $0.23\times0.l2\;to\;0.74\pm0.08$, during the experiment period, and in male it didn't change remarkably until November, but thereafter it increased and showed a peak of $8.69\pm5.09$. The GSI of 3-year-old-fish showed a peak of $8.05\pm5.58$ in April in female and $12.65\pm4.60$ in May in male. The change of HSI in 3-year-old-fish was correlative to the change of GSI, but in 2-year-old-fish it was little correlative. In female gonad of 2-year-old tiger puffer, the mature oocytes reached $350{\mu}m$ in April, but thereafter they didn't spawn and became atrophied. But in male gonad, a great number of spermatozoa were crowded in the testicular lobuli in April. Female gonad of 3-year-old tiger puffer had the mature oocytes of 650 pm in March and the ripe oocytes, $900{\mu}m$ in April. Male testis development was similar to that of 2-year-old-fish. Egg-stripping after hormone treatments was possible past 139 hours and 142 hours from each of two 5-year-old-fish (500IU/kg, BW), and after 114 hour from a 3-year-old-fish (1,000 IU/kg, BW) under water temperature $16.3\~17.8^{\circ}C$. Eggs stripped amounted was 650 g and 400 g from two 5-year-old-fish and 610 g from the 3-year-old-fish, and fertilization rates were $98.0\%,\;97.4\%\;and\;96.5\%$ respectively. All the hatched larvae devloped into normal fry.

The midwater pair trawl which is being used at present in Korea have several problems. Firstly, it is difficult to control the net height on high towing speed. Secondly, net breaking often occurs owing to floats and thirdly, the volume of net on the net drum is so large. This study is aiming for examining the possibility of application for the Korean midwater pair trawl through the model experiment of non-float midwater pair trawl. The model of non-float midwater pair trawl was manufactured as 1/100 of the full scale net which is being used in bottom pair trawl for 850ps class considering the Tauti's Similarity law. The model experiment was carried out to analyze the opening efficiency according to the variation of lower warp length and the opening efficiency was investigated between th proto type and non-float type. The results obtained can be summarized as follows ; 1. The hydrodynamic resistance of non-float type was about 10~20% smaller than that of the proto type and it increased about 1ton according to the increase of dL at the condition of the same flow speed. The resistance acting on the lower warp decreased about 5% but that of the upper warp increased according to the increase of lower warp length (dL) at the condition of the same flow speed. 2. The net height of the non-float type decreased almost linearly according as the increased of flow speed and it increased in a logarithmic functional form with the increase of the lower warp length at the condition of the same flow speed. On the decreasing rate of the net height, the non-float type was lower than the proto type and the difference of the decreasing rate was about 12% at 3.0 knot, 25% at 4.0 knot, 25% at 4.0 knot respectively when dL was 30m. 3. The net width of non-float type was not varied so much as only 2m range and was larger than that of proto type. 4. The mouth area of non-float type decreased in a exponential functional form. On the decreasing rate of the mouth area, the non-float type was lower than the proto type. The filtering volume increased in a logarithmic functional form with increasing flow speed and the filtering volume of proto type decreased steeply over 3.0knot, but that of non-float type increased until 4.0knot. 5. The optimal length of lower warp was when the value of dL was about 30m and the optimal position of front weight was at the connection point of four net pendants.
BACKGROUND: Mine drainage from metal mining districts is a well-recognized source of environmental contamination. Oxidation of metal sulfides in mines, mine dumps and tailing impoundments produces acidic, metal-rich waters that can contaminate the local surface water and soil. METHODS AND RESULTS: This experiment was carried out to investigate the pollution assessment of heavy metal on the water quality of mine drainage, paddy soils and sediment in lower watershed affected by mine drainage of the Sambo mine. The average concentrations of dissolved Cd (0.018~0.035 mg/L) in mine drainage discharged from the main waste rock dumps(WRD) was higher than the water quality standards (0.01 mg/L) for agricultural water in Korea. Also, the average concentrations of dissolved Zn, Fe and Mn were higher than those of recommended maximum concentrations (Zn 2.0, Fe 5.0, Mn 0.2 mg/L) of trace metal in irrigation water proposed by FAO (1994). The average contents of Pb and Zn in paddy soils was higher than those of standard level for soil contamination(Pb 200, Zn 300 mg/kg) in agricultural soil by Soil Environmental Conservation Act in Korea. Also, the concentrations of Cd, Pb and Zn in sediment were higher than those of standard level for soil contamination (Cd 10, Pb 400, Zn 600 mg/L) in waterway soil by Soil Environmental Conservation Act in Korea. The enrichment factor (EFc) of heavy metals in stream sediments were in the order as Cd>Pb>Zn> As>Cu>Cr>Ni. Also, the geoaccumulation index (Igeo) of heavy metals in stream sediments were in the order as Zn>Cd>Pb>Cu>As>Cr>Ni, specially, the geoaccumulation index (Igeo) of Zn (Igeo 3.1~6.2) were relatively higher than that of other metals in sediment. CONCLUSION(s): The results indicate that stream water and sediment were affected by mine drainage discharged from the Sambo mine at least to a distance of 1 km downstream (SN-1, SN-2) of the mine water discharge point.
1. On the basis of the samples collected on the eastern coast of Koje-Do from May to September, 1969, studies have been made on the growth and the relationships between the carapace length and the body length, and between the carapace length and the body weight of Penaeus japonicus Bate. 2. The mean carapace length of P. japonicus was 51mm in May, 57mm in June, 47mm in July and 50mm in September respectively. 3. As a result of the present studies two populations of P. japonicus exist in waters around Koje-Do, namely the spring and fall spawning populations. 4. The relationship between the carapace length ($\iota$) and the body length(L) and between the carapace length and the body weight (W) are indicated by the following equations: May $$L=2.6544{\iota}+3.1258$$$$W=1.892{\iota}^{1.9844}$$ June $$L=2.8659{\iota}+2.1796$$$$W=1.082{\iota}^{2.4323}$$ July $$L=2.5840{\iota}+3.3090$$$$W=1.290{\iota}^{2.3094}$$ September $$L=2.4234{\iota}+4.5775$$$$W=1.599{\iota}^{2.1857}$$ 5. With regard to the relationships between the carapace length and the body length and between the carapace length and the body weight there is no significant difference between the populations spawning in June and September. 6. The relationships between the carapace length ($\iota$) and the body length (L) and between the carapace length and the body weight (W) for the samples cultured at three different localities are indicated by the following equations: Koje-do $$L=3.7738{\iota}+0.0805\;(r=0.934)$$$$W=0.4690{\iota}^{3.0713}$$ Oma-do $$L=2.993{\iota}+1.6455\;(r=0.990)$$$$W=0.6328{\iota}^{2.6579}$$ Kumdang-do $$L=3.2749{\iota}+0.9055\;(r=0.983)$$$$W=0.5768{\iota}^{2.8076}$$ 7. During the larval stages the relationship between the body length (L) and the rearing day (D) is indicated by the following equations: Zoeal stages (1-3) L=0.1279D+0.2686 (r=0.979) Mysis (1) - Post larva (6) L=0.1697D+0.5634 (r=0.994) Post-larvs (7) - Post larvs (21) L=0.1344D+1.9501 (r=0.978)
This experiment was conducted to find out the optimum condition of aeration rates for removal of malodor and to improve the compost quality. The aspect of ammonia emission and amounts of volatilization were investigated in the enclosed composting reactor of 242 liters piled with mixed materials of dairy manure and rice straw, which adjusted to 65% of initial moisture content and controlled by four different aeration rates. Mature temperature increased suddenly in initial composting time and decreased with Increasing aeration rates. The treatment of $1.79l\;min^{-1}kg\;dry-solids^{-1}$ results in overcooling and rapid drying of composting materials because of too much aeration. The average concentration of ammonia emitted from composting for 24 days was the range of 25.3 to $239.8mg\;l^{-1}$ and was highest in the treatment of $0.09l\;min^{-1}kg\;dry-solids^{-1}$, followed by 0.90. 0.18 and $1.79l\;min^{-1}kg\;dry-solids^{-1}$. The range of maximum concentration by different aeration rates was $335{\sim}2279mg\;l^{-1}$ and it wan highest in the treatment of $0.09l\;min^{-1}kg\;dry-solids^{-1}$, followed by 0.18, 0.09 and $1.79l\;min^{-1}kg\;dry-solids^{-1}$. Relationship between the ammonia concentration emitted and temperature matured under different aeration rates showed an exponential positive correlation with 1% significance and had a trend of clear increase in ammonia concentration with increasing temperature over $50^{\circ}C$. Most of ammonia volatilized within plays after composting. The volatilization rate of ammonia ranged from 0.056 to 0.453 per dry solids of materials and it was highest in the treatment of $0.09l\;min^{-1}kg\;dry-solids^{-1}$, followed by 0.18, 0.09 and $1.79l\;min^{-1}kg\;dry-solids^{-1}$. Amounts of ammonia volatilized under composting condition of this experiment was estimated to be highest in the aeration range of 0.9 to $1.0l\;min^{-1}kg\;dry-solids^{-1}$.
Strain improvement and bioprocess development were undertaken to enhance hyaluronic acid(HA) production by Streptococcus zooepidemicus cells. Using a high-yielding mutant strain, statistical medium optimization was carried out in shake flask cultures, resulting in 52% increase in HA production (5.38 g/l) at the optimal medium composition relative to the parallel control cultures. For sufficient supply of dissolved oxygen (DO), which turned out to be crucial for enhanced production of HA, agitation system and speed were intensively investigated in 5 L bioreactor cultures. Increase in oxygen mass transfer coefficient (kLa) through increment of agitation speed (rpm) and 35% expansion of diameter of the newly-designed impellers showed significantly positive effects on HA production. By installing an expanded Rushton-turbine impeller for efficient break-down of sparged air, and an extended marine impeller above the Rushton-turbine impeller for efficient mixing of the air-born viscous fermentation broth, maximum amount of HA (9.79 g/l) was obtained at 450 rpm, 1.8 times higher level than that of the corresponding flask culture. Subsequently, the possibility of bioprocess scale-up to a 50 L bioreactor was investigated. Despite almost identical maximum HA production (9.11 vs 9.25 g/l), the average HA volumetric productivity (rp) of the 50 L culture turned out only 74% compared to the corresponding 5 L culture during the exponential phase, possibly caused by shear damages imposed on the producing cells at the high stirring in the 50 L culture. The scale-up process could be successfully achieved if a scale-up criterion of constant oxygen mass transfer coefficient (kLa) is applied to the 50 L pilot-scale bioreactor system.
