소설 "토지"에 나타난 경상남도 향토 음식문화 (A Study on the Kyungsangnamdo Native Local Food Culture in the Novel "Toji")

  • This study was intended to outline the characteristics of the food culture in the area of Kyungsangnamdo and its modernization by interpretation and analysis of the novel Toji, which was set in Hadong, Jinjoo in the area of Kyungsangnamdo in the early 20th century. The characteristics of the Kyungsangnamdo area's native dish during the Japanese ruling era in the latter half of the Choson dynasty are as follows. In the first part of the novel, which spans from 1897 to 1908, vegetable and grain food development can be seen in the area of Hadong, the interior plains of Kyungsangnamdo, where there is a typical farming village in the mountains. The second part of the novel, which spans from 1911 through 1917, includes some mentions of the properties of Kyungsangnamdo area's native dishes through the lens of emigrated Koreans living on Gando island. Gando island is in China, and is where Seohee, the heroine, escapes from her homeland and remains for a period of years. There is a unique type of seafood in the Gando area using fresh marine products, exactly the same as in the Kyungsangnamdo area. The third part of the novel spans 1919 through 1929, after Seohee returns to her own country and regains her house. There is a noticeable description of food culture in the area of Jinjoo in Kyungsangnamdo through the description of Seohee focusing on the education of her children. The well-described features of Jinjoo are boiled rice with soup of beef leg bones and Jinjoo bibimbob, with vegetables and a variety of foods using cod. Cod are caught in large quantities in Kyunjgsangnamdo, and cities in the area grow to medium size as the area became traffic-based. The fourth part of the novel spans from 1929 through 1938, and includes very detailed descriptions of characters and background locations. Salted fish combined with the wild ingredients of Mt. Jiri feature prominently in the Kyungsangnamdo's area descriptions. The fifth part spans from 1940 through 1945, and as the Japanese colonization era ends, the foods described in Kyungsangnamdo seem to develop the usage of soybean paste. With abundant fish and shellfish Kyungsangnamdo, the dishes that evolve to use soybean paste include mussel soybean paste soup, picked bean leaves in soybean paste, chaitgook - cold soup from soybean paste, and seolchigook used with seaweed and sea laver.

  • 1. 소규모접종시험을 위해서는 wire brush 접종 또는 다침접종법이 가장 효과적이었으며 포장에서의 대규모시험을 위해서는 전착제를 첨가한 세균현탁액의 살포가 좋았다. 2. 조사지역내에서의 발견된 병은 세균성 점무늬병과 세균성 불마름병 뿐이었고 담배 불마름병균에 의한 피해는 발견할 수 없었다. 3. 병원균은 이병조직속에서 겨울철인 경우 실내에서 6개월까지 생존할 수 있었고 냉장고 속에서는 10개월이상 존재했으며 토양속에서는 불마름병균이 4일, 점무늬병균이 30일간 생존하였다. 4. 종자표면에 부착된 세균은 전염원으로서 중요하지 않았으나 실험실내의 배양기 상에서는 종자의 발아를 억제했다.

한국 근로자의 식품섭취 기호도에 관한 연구 (A Study on the Preferences for Food Intake of Korean Industrial Workers)

  • This study was carried on the preferences for food intake pattern of Korean industrial workers. The survey was conducted from Feb 17, to March 31, 1992 by questionnaires which was composed of 21 items. The subjects of this study were composed of 426 clerical workers, and 439 physical workers. The results were obtained as follows. 1. In the educational background, clerical workers was almost of highschool or collage graduates, and physical workers was almost of highschool graduates. Marginal food cost of per person for lunch a day was from 500 Won to 1,000 Won. They replied that amount of staple food and side dishes were appropriate, and the type of preferable foods was Korean style cooked rice. Also it was known that physical workers thought their nutritive condition more badly than that of clerical workers. So the physical workers considered their health very much when they had meal. It was represented that unbalanced diet, without diet and drinking alcohol effected badly to food intake and taste of food was the most important point in food intake. 2. Preferences for staple foods; Cooked rice was preferred the most by the subjects. Cooked rice with redbean was preferred by clerical workers and cooked rice with soybean was preferred by physical workers and cooked rice with soybean was preferred by physical workers. Also clerical and physical workers liked Kalkooksoo (hot noodle), Nangmyun (cold noodle), bread, vegetable bread and disliked Sphagetti, Ramyun (Instant noodle) in same rate. In the next, cooked rice covered with raw fish pieces (Sushi) was preferred by clerical workers and Mandu-kook was preferred by physical workers. Also they disliked Curriedrice and Hashrice in same pattern. 3. Preference for side-dishes; The Acceptances for side-dishes were high soybean paste soup (clerical workers:43.7%, physical workers:34.4%), Kim chi stew, fried potatoes, crab steaming (37.8%, 36.9%), broiled fish (34.6%, 33.9%), fried green pepper, Ham, Chinese cabbage Kim chi, and pickled garlic by clerical and physical workers all together. But in clerical workers, Wooghugi beef-rib tang, Deodeok muchim, spinach namul, beef jangjeorim, fried shrimp were shown to have high preference in physical workers, beef-rib tang, wild onion and shepherd's purses muchim, soybean sprout, fish jeorim, fried sweet potatoes were high preference. 4. Preference of Snacks; The preferences for snacks were high in strawberry (clerical:36.3%, physical:34.3%), milk (49.9%, 44.4%), waxy rice Doek (30.7%, 30.2%), coffee (40.1%, 38.9%) by two groups in same order.

한국 재래 및 야생종 콩의 Leucine Aminopeptidase 변이 (Variation of Leucine Aminoeptidase Isozyme in Korean Land Races and Wild Soybeans)

  • 한국 재래종 대두(Glycine max) 943계통과 강원도 지성 수집종 야생종(G.soja) 50계통을 polyacrylamide gel electrophoresis와 isoelectric focusing 전기영동으로 LAP 동립효소를 분석한 결과를 요약하면 다음과 같다. PAGE에 의한 Lap1*b는 재래종과 야생종에서 가장 흔한 대립인자였다. Lap1*b의 대립인자 빈도는 한국 야생종(0.94)에서보다 재래종(1.00)에서 더 높았다. 이러한 결과는 한국 재래종 대두는 Lap1 유부자좌에서 Lap1*b 대립인자로 고정되어 있음을 나타내는 것이다. IEF 전기영동법을 이용하여 pH 4∼6.5의 gel에서 LAP 동립효소의 band type을 확인한 결과 재래종과 야생종에서 I type과 II type의 band type을 확인하였다. 재래종과 야생종에서 II type이 6계통(7.2%)과 4계통(8.0%)으로 각각 나타났고 I type은 79계통(92.8%)과 46계통(92.0%)으로 각각 나타났다. 이러한 결과는 다양한 band type이 나타날 수 있는 가능성을 제시하고 있다.

경남지역을 중심으로 한 한국인의 식품 기호도에 관한 조사연구 (Survey on Food Preference in Gyeongnam Area)

  • This research was carried to investigate the food preference of students, from elementary school, middle school, high school, university and industrial workers on Jinju, Sacheon and Jinyang by age and sex. The results were obtained as follows. In case of rice as the stationary food, plain white boiled rice showed the highest preference among all the groups. Most of groups liked most of one-dish meals such as kimbab, fried rice with kimchi, dumplings and bibimbab, especially the elementary school students and middle school students. On the other hand, high percentage of industrial workers disliked the western food such as pizza, hamburger and sphagetti. Most of subjects liked jajangmun, nangmun and bibimgooksu as noodles. As the side dishes generally they prefer the soup to stew. Male prefer the soup with beef and female prefer soup with vegetables. Stew with kimchi and stew with soybean paste showed high preference among most of groups. Most of broil food showed high preference, and students prefer meat to fish as broil cooking materials especially younger students. As a general they liked soybean sprout, spinach and wild sesame leaf as namul cooking method and they liked korean cabbage kimchi, chonggak kimchi, kackdoogi and dongchimi as kimchi. As a dessert subjects liked most of fruits and they liked yoghurt, fruit juice, milk, sikhae and soda as drinks generally. On the other hand snacks such as cooky, candy, cake, corn, rice cake, sweet potato have the lowest percentage preferance as a dessert, but students from elementary schools showed the highest preferance to sweet such as cooky, cake and candy than any other group. These food preferance results showed some nutritional problems especially young age students. They should eat more green-yellow vegetables, liver food and dried small sardine and they should reduce snacks such as candy and cooky and soda drinks. Therefore it is necessary to conduct nutrition education by parents, teachers and dietician together to improve their food habits and their health.

콩에서 흡즙해충 싸리수염진딧물 저항성 자원의 발굴 (Resistance Resources for the Foxglove Aphid in Soybeans)

  • 작물의 병해충 발생은 환경의 여러 가지 요인으로 발생하며 현재 기후의 급격한 변화는 해충류의 급격한 발생빈도 증가를 야기하여 궁극적으로 안정적인 식량 공급의 위협요인이 된다. 본 연구에서는 국내 콩 재배에 있어 많은 피해를 입힐 수 있는 싸리수염진딧물에 대한 저항성 검정지표설정과 저항성 자원발굴을 실시하였다. 기존 콩진딧물에서 사용하는 검정지표는 이동성이 높은 싸리수염진딧물에서 조사의 어려움 있어 비가림온실과 생장상에서의 진딧물 피해 정도를 분석 한 결과를 토대로 싸리수염진딧물 검정지표를 검정한 결과 싸리수염 진딧물은 식물의 피해정도가 안정적으로 나타났고, 간편하게는 접종 14일 후 최초 접종엽의 피해 정도가 싸리수염진딧물 검정지표로 가장 우수하였다. 국내 콩 유전자원의 재배종과 야생종 1,115점의 자원에 대한 Choice test 결과 재배종 23점과 야생종 44점으로 총 67개의 저항성 자원을 선발하였으며 No-Choice test를 통해 재배종에서 8점, 야생종에서 23점으로 항생성을 가진 31개의 유전자원을 선발 할 수 있었다. 선발된 저항성 자원은 생존성 검정결과 선발된 저항성자원에서 진딧물의 생존성이 매우 낮았으며 항생성과 비선호성간에 저항성 반응도 상이함을 확인할 수 있었다. 이상에서 밝혀진 싸리수염저항성 검정지표 및 선발된 자원은 금후 해충저항성 품종개발과 기작구명에 유용하게 활용 가능할 것으로 판단된다.

The Role of Residues 103, 104, and 278 in the Activity of SMG1 Lipase from Malassezia globosa: A Site-Directed Mutagenesis Study

  • The SMG1 lipase from Malassezia globosa is a newly found mono- and diacylglycerol (DAG) lipase that has a unique lid in the loop conformation that differs from the common alpha-helix lid. In the present study, we characterized the contribution of three residues, L103 and F104 in the lid and F278 in the rim of the binding site groove, on the function of SMG1 lipase. Site-directed mutagenesis was conducted at these sites, and each of the mutants was expressed in the yeast Pichia pastoris, purified, and characterized for their activity toward DAG and p-nitrophenol (pNP) ester. Compared with wild-type SMG1, F278A retained approximately 78% of its activity toward DAG, but only 11% activity toward pNP octanoate (pNP-C8). L103G increased its activity on pNP-C8 by approximately 2-fold, whereas F104G showed an approximate 40% decrease in pNP-C8 activity, and they both showed decreased activity on the DAG emulsion. The deletion of 103-104 retained approximately 30% of its activity toward the DAG emulsion, with an almost complete loss of pNP-C8 activity. The deletion of 103-104 showed a weaker penetration ability to a soybean phosphocholine monolayer than wild-type SMG1. Based on the modulation of the specificity and activity observed, a pNP-C8 binding model for the ester (pNP-C8, N102, and F278 form a flexible bridge) and a specific lipid-anchoring mechanism for DAG (L103 and F104 serve as "anchors" to the lipid interface) were proposed.

Gibberellin Production and Plant Growth Enhancement by Newly Isolated Strain of Scolecobasidium tshawytschae

  • We isolated nine endophytic fungi from the roots of salt-stressed soybean cultivar Daewonkong and screened them for growth-promoting secondary metabolites. Of all fungal isolates, P-4-3 induced maximum growth promotion of waito-c rice and soybean. Analysis of the culture filtrate of P-4-3 showed the presence of physiologically active gibberellins $GA_1$, $GA_3$, $GA_4$, and $GA_7$, along with physiologically inactive $GA_{15}$ and $GA_{24}$. The plant growth promotion and gibberellin-producing capacity of P-4-3 was much higher than wild-type Gibberella fujikuroi, which was taken as the control during the present study. The fungal isolate P-4-3 was identified as a new strain of Scolecobasidium tshawytschae through the morphological characteristics and phylogenetic analysis of 18S rDNA sequence. Gibberellins production and plant growth promoting ability of genus Scolecobasidium was reported for the first time in the present study.

종자 특이 프로모터와 대두 Ferritin 유전자에 의한 벼 종실의 철분강화 (Iron fortification of grains by introducing a recombinant gene of ferritin with seed promoters in rice)

  • Ferritin 유전자를 벼의 저장기관인 배유에 특이적으로 발현시킬 수 있는 glutelin, gGlobulin 및 zein 프로모터를 활용하여 쌀알에 최대로 발현시켜, 고부가가치를 가진 가공용 벼 품종을 육성하여 천연의 철 성분이 강화된 유아용 이유식 생산에 이용할 수 있으므로 유아들에게 천연의 철분을 안정적으로 공급할 수 있는 형질전환체를 육성하였다. 종자 저장단백질인 glutelin, globulin 및 zein의 프로모터와 ferritin 유전자를 pMJ21 vector에 pGBF, pGTF 및 pZ4F 등의 Ti-plasmid를 Agrobacterium에 도입하여 동안벼와 화신벼에 형질전환 하였다. 동안벼 종자를 사용하였을 때 pGBF 재조합 유전자는 19.2%, pGTF는 15.0%, pZ4F는 18.4%가 재분화되었고, 화신벼 종자를 사용하였을 때에는 pGBF 재조합 유전자는 6.7%, pGTF는 11.7%, pZ4F는 3.4%가 재분화되었다. 형질전환 벼의 ferritin 유전자의 도입여부는 PCR 분석과 Southern 분석으로 확인하였으며 ferritin 유전자의 유전자 발현은 Norihern 및 Western 분석에 의해 확인하였다. Southern blot 분석 결과로부터 각각의 배유특이 프로모터 유래 형질전환체 중에서 single copy로 도입된 개체를 선발 할 수 있었다. 또한 이들 형질전환 계통들에서 도입유전자의 발현량은 wild type 벼에 비하여 매우 높게 나타났다. 또한 철 단백질의 철분 축적 정도를 분석한 결과 Zein 프로모터를 사용한 형질전환 계통 (T1-2)에서 171.4 ppm으로 wild type과 비교하여 6.4 배의 철분함량 증가를 보였다. 그러나 globulin 및 glutelin 프로모터 유래 형질전환체에서는 wild type과 비교하여 $2.1{\sim}3.0$ 배의 철분함량 증가를 보였다. 벼 형질전환체들의 생육상황을 조사한 결과 초장은 변이 폭이 매우 크게 나타났으며, 대조품종과 비교하여 50%정도 감소한 왜성 및 이형 식물체도 출현되었다. 따라서 본 연구에서는 형질전환체 중에서 표현형 적으로 대조품종과 거의 같은 식물체를 선발하여 후대를 육성하였다. 육성한 T1 세대에서 형질전환체의 초장, 간장, 수장, 분얼수 및 등숙률을 조사한 결과 초장, 간장, 수장, 분얼수에 있어서는 대조 품종과 큰 변이를 보이지 않았으나 등숙률에 있어서는 $53.3{\sim}82.2%$의 비교적 큰 변이를 나타내었다.
