Amyloodinium sp. Infestation in Mullet (Mugil cephalus) cultured in a pond on land (육상 수조에서 사육 중인 숭어 (Mugil cephalus)의 Amyloodinium sp. 감염)

  • Amyloodinium sp. was found on the gills of mullet (Mugil cephalus) cultured on land. No external symptoms in the diseased fish were found except decoloration of the gills. In fresh preparations of the gills the parasites were opaque round or oval shape with a bright nucleus and 43.5 ㎛ (18.2~72.7, n=20) in size. In preparations added a drop of Lugol solution, they were black with the same shapes in fresh preparations and 43.5 ㎛ (n=20) in size. The parasites were stained black and blue in a droplet of Lugol solution and Diff-Quick III solution, respectively and their sizes were a little larger than in wet preparations. After stained with May-Grunwald Giemsa, the parasites appeared granular eosinophlic in the peripheral cytoplasm and granular strong basophilic in the center. In silver impregnated specimens the peripheral granules were negative and the central ones positive. The granules appeared brown in purplish cytoplasm after staining with Lugol solution. The parasites developed by binary division when they were cultivated in filtered seawater at 20℃. Histopathologically severe epithelial hyperplasia and fusion in the gill filaments resulted in clubbing, especially the proximal region of the filament. Epithelial hyperplasia was also found in the basal regions of the gill filaments and some epithelial cells were occasionally detached from the filaments. Some pear-shaped trophonts of the parasites with rhizoid attached on the gill filaments showing hyperplasia of the epithelial cells and mucous cells.

Effects of Application Time of GA Paste on Tree and Fruit Growth and Fruit Quality of 'Kamcheonbae' and 'Whangkeumbae' Pears (GA 도포제의 처리시기가 '감천배'와 '황금배'의 생육과 과실 품질에 미치는 영향)

  • To clarify the effects of application time of $GA_{4+7}$ paste (GA paste) on tree and fruit growth, fruit quality, and maturity of 'Kamcheonbae' and 'Whangkuembae' pears, GA paste of 25 mg each was applied directly to fruit stalk 25, 35, and 45 days after full bloom (DAFB). Later application of GA paste tended to increase the fruit weight of 'Kamcheonbae' pear. GA paste applied 45 DAFB increased the titratable acidity of the two cultivars. Fruit firmness of 'Kamcheonbae' was not affected by the GA paste treatment, while that of 'Whangkuembae' was increased by more than 20%. Fruit maturity of the two cultivars was hastened for 4 days by the GA paste treatment at any application time. Sharp increase in the fruit weight of the GA paste-treated 'Kamcheonbae' was noted from early July while that of control exhibited slow increase from early September. The initial growth pattern of 'Whangkeumbae' was similar to that of 'Kamcheonbae', but the growth slowed down from early September, regardless of the treatments.

Development of Sequence Characterized Amplified Region Markers for Cultivar Identification in Persimmon (감 품종 판별용 SCAR 마커 개발)

  • Precise, fast, and cost-effective identification of crop cultivars is essential for plant breeder's rights. Traditional methods for identification of persimmon cultivars are based on the evaluation of sets of morphological characteristics. However, it is difficult to distinguish closely related cultivars using only morphological traits. This study was conducted to develop DNA markers for identification of the 32 persimmon cultivars in Korea and Japan. A total of 309 randomly amplified polymorphic DNA (RAPD) markers were identified using 40 different random primers. Various number of polymorphic bands ranged from 4 (OPP-08) to 14 (UBC159) were detected with an average of 7.7. Resulting 57 RAPD fragments were selected, and their sequences were determined for developing sequence characterized amplified region (SCAR) markers. As a result, 15 of 57 RAPD fragments were successfully converted to SCAR markers. Single polymorphic bands of the same size as or smaller than the RAPD fragments were amplified depending on SCAR markers. Among these markers, a combination of eight SCAR markers (PS225_200, PSN05_420, PSF13_523, PSN11_540, PS372_567, PS485_569, PSP08_635, and PS631_735) provided sufficient polymorphisms to identify 32 persimmon cultivars. These newly developed markers will be a fast and reliable tool to identify persimmon cultivars.

  • In 2004, average temperature was higher, and rainfall was less than those of previous year, The diseases of rice, barley, pepper, chinese melon, apple and oriental pear were surveyed. Seedling diseases, leaf blast, sheath blight and bacterial blight of rice, phytophthova blight, virus diseases and anthracnose of pepper, and sudden wilt syndrome and powdery mildew of chinese melon grown in plastic greenhouse were severe. Especially, sheath blight and bacterial blight of rice occurred two times higher than those of previous year, Panicle blight of rice decreased about 4 times, compared with the previous year, presumed that the lower rainy days, rainfall and RH suppressed spread of the fungal pathogens to panicles of rice. Lower rainfall during mid- and late Aug caused three-times less occurrence of phytophtora blight of red-pepper than that of the previous year, Another diseases surveyed occurred similar or less than those of the previous year.

  • Climate in the year of 2001 could be summarized as severe drought from March to May, unusually high temperature in whole plant growth periods and clear weather condition especially in harvesting time of September and October without any typoons. In rice, major diseases such as leaf and panicle blast, bacterial blight, sheath blight and bacterial grain rot developed little due to unfavorable weather conditions, however, brown spot occurred severely due partly to the reduced use of N-fertilizer, Rice stripe virus infection was unusually high mainly in the west coast areas with four times more diseaseD area campared to the previous year, In pepper phytophthora blight was extremely severe in Cheonnam and Cheonbuk provinces, where had frequent rainfalls during growing period. Incidence of major diseases of tomato and cucumber in 2001 was relatively mild. In watermelon, penicillium fruit rot, one of the peculiar disease, spread over major production areas, whereas CGMMV, usually severe in every years was much reduced. Watermelon plants growing in open-fields were more severely diseased than those in plastic houses. Major diseases of chinese melon and strawberry occurred slightly and in particular, sudden wilt syndrome of chinese melon which was severe in 2000 showed mild infections in 2001. Incidence of white rot of garlic and onion was much lower unlike other years due mainly to spring drought and high temperature in the growing period. Infected area of potato bacterial wilt tended to increase in 2001 while fusarium wilt of sweetpotato was decreased. Apple diseases were generally mild, but powdery mildew of pear increased sharply. Barley scab was not severe as seen in other years.

Effects of Opuntia ficus-indica Pigment and Sodium Lactate on Nitrite-reduced Sausages (선인장색소 및 유산나트륨이 저 아질산염 소시지에 미치는 효과)

  • This study was carried out to investigate the effect of Opuntia ficus indica(prickly pear) pigment and sodium lactate on nitrite-reduced sausages. The a* value, total bacterial counts, pH, water holding capacity, texture analysis, sensory evaluation of sausages, body weight gain and blood glucose of rat for 4 weeks were not significantly different between the control and treatments. However, 2-thiobarbituric acid(TBA) values and cholesterol(total, LDL, HDL) and neutral fat of blood were significantly different between the control and treatments(P<0.05). TBA value was lower in control and cholesterol and neutral fat were lower in treatments. Among the treatments T2(30ppm of nitrite+2% of sodium lactate+0.2% Opuntia ficus indica pigment) was the most effective. In conclusions, this study suggested that T2 can reduce adding level of nitrite from 100 ppm to 30 ppm without any changes in color, shelf life and flavor of sausages. In addition, it had the effect on the reduction of cholesterol and neutral fat in blood.

Effects of Some Chemials on Ethylene Evolution and Abscission of Fruits and Leaves in Oriental Pera (수종(數種)의 약제(藥劑)가 배나무의 낙과(落果) 및 에틸렌 발생(發生)에 미치는 영향(影響))

  • In order to examine the relationship of fruit abscission and ethylene evolution in 'Jojuro' and 'Imamuraaki' pear trees, the thinning chemicals, ${\beta}$-naphthalene acetic acid (NAA), O, O-dimethyl-O-(3-methyl-4 nitrophenyl) posphorothioate (MEP), 1-naphthyl N-methyl carbamate (carbaryl), and 2-chloroethyl phosphonic acid (ethephon) were applied. Concurrently the effect of calcium acetate on the control of fruit abscission were examined when calcium acetate was added to ethephon and carbaryl. 1. All the applied chemicals (MEP, NAA, carbaryl and ethephon) was effective to abscise fruits in 'Jojuro', and MEP and carbaryl in 'Imamuraaki's. 2. The application of ethephon increased the ethylene evolution but other chemicals did not increase it. 3. There were no significant differences in total sugar contents of fruits by MEP, NAA, carbaryl and ethephon treatments. 4. When the calcium acetate was added to the ethephon and carbaryl, the thinning effects were offsetted. 5. The rate of defoliation due to ethephon treatment was controlled by addition of calcium acetate at 0.1~0.25 M. It was clarified that fruit abscission induced by thinning chemicals except ethephon was not directly related to ethylene production and that degree of fruit thinning can be regulated by addition of calcium acetate to thinning chemicals.

Effect of Gibberellin Paste on the Fruit Growth and Maturation in 'Hosui' Pears (Gibberellin 처리(處理)가 배 '풍수(豊水)' 과실(果實)의 생장(生長) 및 성숙현상(成熟現想)에 미치는 영향(影響))

  • This experiment was conducted to find the effect of gibberellin paste on the growth and maturation of 'Hosui' pear fruit and to develop a cultural technique in the middle area of Korea, where fruit quality is often poor due to the short growing season. The application of $GA_{3+4+7}$ paste on the petioles at the young stage of development resulted in the early increase of fruit diameter and weight. Although fruit firmness was slightly low by the $GA_{3+4+7}$ paste treatment, compared to that of control, there were no differences in other quality actors such as soluble solid content and acidity. However, $GA_3$ alone did not affect the fruit growth and quality. Are increase of ethylene evolution of fruit was not confirmed hut ground color development was hastened, resulting in the stimulation of maturity by 3~5 days. Thus, these results may be applicable to the production of high quality 'Hosui' pears in the area where growing season is relatively short.

Analysis of Vibration Suspension Device for Trailer in Agricultural Products (농산물 수송 트레일러의 현가장치 진동 분석)

  • This study was aimed to minimized the impact force and vibration transmitted to transporting agricultural product from the power tiller trailer by installing vibration absorption device. The vibration absorbable trailer (I) mounted with leaf spring suspension and shock absorber was developed and compared on vibration absorption performance with the existing trailer (E) equipped no vibration absorption device. In order to identify the vibration absorption effect of the trailer developed in this study, the vibration accelerations, occurred during driving on paved road with loading 360 kg of pear, were measured and analyzed using FFT analyzer. The magnitude of average vibration acceleration was decreased highly for the improved trailer mounted with vibration absorption device in comparing with existing trailer in the frequency range under 60 Hz and under 80 Hz. And similar vibration absorption effect was represented for the improved trailer in all frequency range. Especially, in the frequency range between 40 Hz and 80 Hz, the magnitude of vibration acceleration for the improved trailer was decreased with 1/3 times in comparing with existing trailer. So, the transporting loss including damage of agricultural product could be decreased highly by using the improved vibration absorbable trailer mounted with leaf spring suspension and shock absorber simultaneously, designed in this study.

Estimation of Allowable Drop Height for Oriental Pears by Impact Tests (충격시험에 따른 배의 허용낙하높이 추정)

  • Impact between fruits and other materials is a major cause of product damage in harvesting and handling systems. The oriental pears are more susceptible to bruising than other fruits such as European pears and apples, and are required more careful handling. The interest in the handling of the pears for the processing systems has raised the question of the allowable drop height to which pears can be dropped without causing objectionable damage. Drop tests on pears were conducted using an impact device developed by authors to estimate the allowable drop height without bruising. The impact device was constructed to hold in a selected orientation and to release a fruit by vacuum for dropping on to a force transducer. The drop height was adjustable for zero to 60 cm to achieve the desired distance between the bottom of the fruits and the top of the impact force transducer. The transducer was secured to 150 kg$\sub$f/ concrete block. The transducer signal was sampled every 0.17 ms with a strain gage measurement board in the micro computer where it was digitaly stored for later analysis. The selected sample fruit was Niitaka cultivar of pears which is one of the most promising fruit for export in Korea. The pears were harvested during the 1998 harvest season from an orchard in Daejeon. The sample fruit was selected from two groups which were stored for 3 months and 5 months respectively by the method of current commercial practice. The pears were allowed to stabilize at environmental condition(18$^{\circ}C$, 65% rh) of the experimental room. One hundred fifty six pears were tested from the heights of 5, 7.5. 10 and 12.5 cm while measurement were made of impact peak force, contact time, time to peak force, dwell time, pear diameter and mass. The bioyield strength and modulus of elasticity were measured using UTM immediately after each drop test. The allowable drop height was estimated on the base of bioyield strength of the pears in two ways. One was assumed the peak force during impact test increasing linearly with time, and the other was based on the actual drop test results. The computer program was developed for measuring the impact characteristics of the pears and analyzing the data obtained in the study. The peak force increased while contact times decreased with increasing drop height and contact times of the sample from the hard tissue group. The allowable drop height increased with increasing bioyield strength and contact times, and also varied with Poisson\`s ratio, mass and equilibrium radius of the pears. The allowable drop height calculated by a theoretical method was in the range from 1 to 4 cm, meanwhile, the estimated drop height considering the result of the impact test was in the range from 1 to 6 cm. Since the physical properties of fruits affected significantly the allowable drop height, the physical properties of the fruits should be considered when estimating the allowable drop height.
