Productivity and Performance Test of Egg-Type Commercial Korean Native Chickens (산란형 토종닭 실용계의 생산 및 산란 능력 검정)

  • The study was conducted to evaluate the production and the performance test of egg-type commercial Korean Native Chickens. A total of 360 female layers were produced from CY${\times}$C (A), CL${\times}$C (B), CG${\times}$C (C) and CW${\times}$C (D) crossbreds kept at National Institute of Animal Science, Korea, and breed during 20~64 weeks to investigate the productivity and the performance. Layers were divided into 4 crossbreds (6 replications/crossbred, 15 heads/replication). Age and egg weight at first egg was no significantly different among crossbreds. Body weight (BW) at first egg of D crossbred was significantly lower compared to that of other crossbreds (P<0.05). Weekly BW of B crossbred was the highest at 20, 30 and 40 weeks of age, but BW of D crossbred was the lowest among crossbreds. Feed intake (FI) of B crossbred was significantly higher compared to the other crossbreds at 20~24, 32~44 and 52~60 weeks of age. FI of D crossbred was significantly lower compared to other crossbreds after the age of 44 weeks (P<0.05). Average egg weight of B crossbred was significantly higher than the other crossbreds at the age of 24~60 weeks (P<0.05). Egg production ratio (%) of C crossbred was significantly higher compared to the other crossbreds for whole test periods (P<0.05). Hen-housed egg production number at 64 weeks of age for A, B, C and D crossbreds resulted in 241.4, 235.6, 232.3 and 227.0, respectively. Feed conversion ratio of A and C crossbreds was lower compared to the other crossbreds at the age of 20~24 weeks and C crossbred was the lowest among the crossbreds at the age of 60~64 weeks. These results provide the basic data which will be helpful to develope the new strains of commercial Korean Native Chickens.

Changes in the Antigenicity of Chicken Egg White by the Treatments of Protease, Trifluoromethanesulfonic Acid, Heat, and NaOH (효소, Trifluoromethanesulfonic Acid, 열, 및 NaOH 처리에 의한 계란 난백의 항원성 변화)

  • To reduce the antigenicity of egg white (EW), EW was treated with several proteolytic enzymes, trifluoromethanesulfonic acid (TFMS), heat, and NaOH. Competitive indirect enzyme-linked immunosorbent assay (ciELISA), using rabbit anti-EW antibody, was performed to examine the antigenicity of the treated EW's. Enzymatic hydrolysis gave no good effect on the reduction of the antigenicity of EW. Neither did the pretreatment with ${\gamma}-irradiation$ before the hydrolysis reduce the antigenicity. TFMS treatment removed the antigenicity of EW. The antigenicity of EW heated at $120^{\circ}C$ for 30 min or treated with NaOH at 0.3% (w/v) and more, decreased to less than 1/10,000 as compared with that of native EW. The combinatory treatment with NaOH, followed by heat at $70^{\circ}C$ for 15 min had a synergic effect on the reduction of the antigenicity of EW.

Estimation of Genetic Parameters for Growth and Egg Production Traits in Black Korean Native Chicken and Korean White Leghorn Populations (흑색한국재래닭, 한국화이트레그혼 집단의 산육 및 산란 형질 유전모수 추정)

  • This study was conducted to estimate genetic parameters for growth and egg production traits in Black Korean native chicken (L strain) and Korean White Leghorn (F, K strains) using a multi-traits animal model BLUP. Traits used for this study were body weight at 150 days (BW150) and 270 days (BW270), age at first egg (DAY1st), egg weight at first egg (EW1st) and 270 days (EW270), and number of eggs laid by 270 days (EP270), and included 68,688 pedigree and 123,905 performance records collected from 2001 to 2013. In L, F, K strains, heritability estimates of BW150 were high (0.48, 0.52 and 0.50, respectively); of BW270 were high (0.56, 0.57 and 0.56); of DAY1st were medium to high (0.45, 0.39 and 0.31); of EW1st were low (0.15, 0.16 and 0.15); of EW270 were high (0.58, 0.55 and 0.59) and of EP270 were moderate (0.22, 0.21 and 0.20). The genetic and phenotypic correlation of DAY1st with EP270 were highly negative (-0.73 to -0.63 and -0.48 to -0.42). The genetic and phenotypic correlation of EP270 with BW150 and BW270, respectively were low negative (-0.16 to 0.01 and -0.14 to -0.03) and low to moderate positive (-0.08 to 0.07 and -0.13 to 0.04). The genetic and phenotypic correlation of EW270 with BW150 and BW270, respectively were moderate to high positive (0.39 to 0.49 and 0.36 to 0.46) and (0.29 to 0.33 and 0.34 to 0.37). The study showed that there is a potential for genetic improvement of Korean Indigenous chicken through selection program.

Production Traits and Stress Responses of Five Korean Native Chicken Breeds (한국토종닭 5품종의 생산능력 및 스트레스 반응 정도)

  • This study presents the production characteristics and physiological characteristics of five Korean native chicken (KNC) breeds consisting of Hwanggalsaek Jaeraejong (HJ), Korean Rhode Island Red (KR), Korean White Leghorn (KL), Korean Brown Cornish (KC), and Korean Ogye (KO). We investigated their production performances, vitalities, and stress responses. We measured the survival rate, body weight, age at first egg-laying, hen-day egg production, egg weight, amount of telomeric DNA, heterophil-lymphocyte ratio (H/L ratio), and heat shock protein (HSP)-70, HSP-90α and HSP-90β gene expression levels for 493 KNCs. The survival rate was highest in KR, and lowest in KO. Body weights were steadily high in the order of KC, KR, HJ, KO and KL. Average hen-day egg production was highest in KL, and lowest in KC. While the amount of telomeric DNA was highest in KR, and lowest in KC. Furthermore, both the H/L ratio and the HSP-90β gene expression level were highest in KC, and lowest in KR. These results indicated that the KR breed was highly resistant to stress, whereas KC was more susceptible to stress. Taken together, it is considered that with improvements the KC breed would be more suited to be used as a Korean broiler breed while KL would be more appropriately used as a Korean layer breed. In addition, it is considered that the KR breed is appropriate to be used as a maternal chicken breeder based on good production capacity and excellent robustness, while the HJ breed is desirable to be improved as a high-quality Korean meat breed based on its excellent meat quality.

Production Performance of 12 Korean Domestic Chicken Varieties Preserved as National Genetic Resources (국가 보존 유전자원 한국토종닭 12종의 생산능력 고찰)

  • In this study, viability, growth performance and egg production performance were investigated to determine the productive characteristics of 12 Korean domestic chicken varieties which have been collected and conserved for over 25 years in National Institute of Animal Science, Rural Development Administration, Korea. The 1,134 hens were tested and their production traits including survival rate, body weight, age at first egg laying, hen-day and hen-housed egg production, egg weight, and egg quality were measured. Survival rate was the highest in Korean Rhode-D and Korean Native Chicken (KNC) White and the lowest in Korean Cornish Brown (92.2% and 54.3%, respectively). Body weights from 4 to 50 wks were consistently high in the order of Korean Cornish, Korean Rhode, KNC, Korean Ogye, and Korean Leghorn breeds. There was no significant difference in weight between varieties within a breed. Age at first egg laying was the shortest in Korean Leghorn, while it was the longest in Korean Cornish. The hen-day egg production from $1^{st}$ egg to 57 wks was the highest in Korean Leghorn-K, and the lowest in Korean Cornish Brown. Egg weight was the heaviest in Korean Leghorn-F and the lightest in KNC White. The Haugh unit was the highest in Korean Rhode-C and the lowest in Korean Ogye. Taken together, these results suggest that it is desirable that the Korean Cornish variety is improved as the Korean meat breed because of its excellent growth ability, the Korean Leghorn variety is improved as the Korean layer breed because of its good laying performance, and the Korean Rhode and KNC varieties are improved as strong viable breeds because of their good survival rate.

Potential Allelic Association of Microsatellite Markers on Chromosome 1 with Economic Traits in Korean Native Chicken (한국재래닭 1번 염색체내 초위성체 유전표지를 이용한 경제형질 연관 지역 탐색)

  • A total of 17 polymorphic microsatellite markers on chromosome 1 were used for allelic association tests with phenotypic traits in Korean native chicken. Chi-square tests were performed to compare the frequencies of individual alleles between the high and the low trait groups. The frequency of allele 123 of MCW0160 showed a significant difference between the high and the low groups in the trait of egg weight (EW). Three markers, namely ADL0234, UMA1.125 and ADL0101, were found to show significant differences in allelic distribution for the trait of the first lay day (FLD). UMA1.117, ADL0020, UMA 1.019, LMA1 and ADL0238 were found to show significant differences in allelic distribution for the trait of body weight (BW). ADL0101 and ADL0238 were found to show significant differences in allelic distribution for the trait of number of egg production(EP). In this study, we identified the QTL for economic traits at around 94 (MCW0160), 151 (ADL0234), 170 (UMA1.125), 225 (UMA1.117), 285 (ADL0020), 387 (UMA1.019), 418 (LMA1), 500 (ADL0101) and 520 (ADL0238) cM on chromosome 1 in Korean native chicken. The results provided a useful guideline for identification of positional candidate gene and marker-assisted selection for economic traits in Korean native chicken.

Effects of Dietary Supplementation of Macsumsuk and Herb Resources on Egg Performance and Quality in Korean Native Crssbred Chicken (맥섬석과 한방제재 첨가급여가 재래닭 교잡종의 계란생산 및 품질에 미치는 영향)

  • The study was conducted to investigate the effects of dietary supplementation of Macsumsuk and herb resources on the egg performance and quality in Korean native crossbred chickens. A total of three hundred sixty improved Korean native crossbred chickens in primary laying period were randomly allocated to four treatments (30 birds/treatment ${\times}$ 3 replications) for eight weeks. Dietary treatments included CON (basal diets), T1 (basal diets + 0.3% Macsumsuk), T2 (basal diets + 0.3% herb resources) and T3 (basal diets + 0.3% Macsumsuk + 0.3% herb resources). No significant differences were found for feed intake, egg weight and average egg production (P>0.05), however, cracked egg rate was higher in CON than in T1-T3, especially T3 was the lowest (P<0.05). The crude fat content of whole eggs, CON (11.20%) was higher (P<0.05) than the experimental treatments (9.67%-10.01%). Haugh unit, yolk color, eggshell thickness, and eggshell breaking strength tended to be higher in T1 and T2, but there were no significant differences (P>0.05). However, the experimental treatments were significantly higher than CON (P<0.05) for eggshell breaking strength, and T3 (14.91 mg/g) contained lower (P<0.05) cholesterol in egg yolk than other treatments. For fatty acid composition in egg yolk, CON group showed higher saturated fatty acid contents than the experimental treatments, but T1 (67.00%), T2 (66.78%) and T3 (66.82%) were higher than CON for unsaturated fatty acids (P<0.05). In conclusion, supplementating Macsumsuk and herb resources together in diets of Korean native crossbred chickens could decrease egg yolk cholesterol and improve egg quality.

Development of a New Synthetic Korean Native Chicken Breed using the Diallel Cross-Mating Test (토종닭의 이면교배조합 시험을 이용한 신품종 종계 개발)

  • We conducted a 4 × 4 diallel cross-mating test using 934 chickens from four grandparent stock lines to develop a new synthetic breed of Korean native chicken. The mean values, combining ability, and reciprocal effects on survival rate, body weight, and hen-day egg production were analyzed. In phenotypes, most chickens have yellowish-brown, reddish-brown and mixed color feathers. The average survival rate was 86.8±12.3%, with the highest in YH combination. Specific combining ability (SCA) had a greater effect on survival rate than general combining ability (GCA), and the SCA of HY combination was the highest. The 16 cross-combinations were distinctly divided into three weight groups according to their genetic characteristics. At 12 weeks of age, GCA showed a greater effect on weight than SCA, and the SCA of FH combination was the highest. The age at first egg laying was 157 days, and the crosses reached sexual maturity faster than the pure lines. The egg production rate was highest in SY at 79.5±2.1%. The GCA and SCA for hen-day egg production were similar, and the SCA was highest in the HS and FY combinations. The reciprocal effect showed that the offspring's egg production rate was high when S and Y were maternal parents in almost all combinations. In conclusion, FH and HF combinations, which have excellent growth performance, are the most desirable paternal parent stock strains, and FY, FS, HY, and SY combinations, which have excellent laying performance with moderate weight, are the preferred maternal strains.

Analysis of antibody titer of transovarian transmitted diseases in Korean native breeder chickens (토종닭 종계의 난계대 전염병 항체 양성률 분석)

  • This study was carried out to identify the occurrence of transovarian transmitted diseases and antibody positive rates among Korean native breeder chickens. The infection rates with Salmonella pullorum and Salmonella gallinarum among 16-week-old, 36-week-old, and 56-week-old breeder chickens and the antibody positive rates to Egg Drop Syndrome '76, Mycoplasma gallisepticum and Mycoplasma synoviae among 16-week-old, 18-week-old, and 56-week-old breeder chickens were identified, and the antibody positive rates to seven major transovarian transmitted diseases among 1-day-old chicks were investigated. As a result, no infection with Salmonella pullorum and Salmonella gallinarum was found among the investigated subjects of all ages. Vaccinated breeder chickens showed the 100% antibody positive rate to Egg Drop Syndrome '76, and unvaccinated breeder chickens showed the 100% antibody negative rate to Mycoplasma gallisepticum and Mycoplasma synoviae, confirming that there was no infection with Mycoplasma gallisepticum and Mycoplasma synoviae. As a result of the antibody tests of the 1-day-old chicks for transovarian transmitted diseases, it was found that vaccinated chicks showed good antibody positive rates to avian encephalomyelitis, chicken infectious anemia, and avian reovirus, confirming that they had power of defense against the relevant infectious diseases, and that unvaccinated chicks showed the 100% antibody negative rates to avian leukosis, chicken reticuloendotheliosis, and Mycoplasma synoviae, confirming that there was no infection with the relevant diseases. Given that the results of this study showed that among the transovarian transmitted diseases of chickens, there was no history of infection with diseases against which vaccination was not administered and high antibody positive rates were found with diseases against which vaccination was administered, it is judged that chickens with good power of defense against diseases were bred, and it is deemed that constant monitoring and vaccination against transovarian transmitted diseases will be necessary for the control and prevention of the diseases.

Allergenicity of Treated Chicken Egg Whites as Determined by a Passive Cutaneous Anaphylaxis Test, Immunoblot Analysis, and a Mouse Model of Food Allergy (수동피부아나필락시스 시험, immunoblot, 식품알레르기 생쥐모델에 의한 난백 처리물의 알레르기성 평가)

  • The allergenicity of treated chicken egg whites (EW) was evaluated by a passive cutaneous anaphylaxis (PCA) test, immunoblot analysis, and a mouse model of food anaphylaxis. The results of the PCA test revealed that treatment with 0.3% NaOH (w/v) decreased the antigenicity of native EW to 1/4. In addition, treatment with heat ($121^{\circ}C$, 30 min) or 1% NaOH (w/v) decreased the antigenicity to 1/8 and combined treatment with 1% NaOH (w/v) and heat ($70^{\circ}C$, 15 min) decreased the antigenicity to 1/32 of that of the native EW. Immunoblot analysis revealed that the density of EW protein bands decreased in response to heat treatment, and were almost not detectable following the combined treatment. Finally, the murine model of EW anaphylaxis revealed that the mean score of systemic anaphylactic symptoms in EW challenged mice was 1.85, while the mean score in mice challenged with EW that that had been subjected to the combined treatment was only 0.20. The results of this study indicate that the most effective method of reducing EW allergenicity is combined treatment with 1% NaOH (w/v) and heat ($70^{\circ}C$, 15 min).
