The Occurrence of Extrafloral Nectaries in Korean Plants (韓國植物의 花外蜜腺分布)

  • Extrafloral nectaries have been shown in many studies to promote mutualistic interactions between plants and insects(usually ants) that visit the glands. The insects gain sugars, water and amino acids secreted by the extrafloral nectaries and benefit the plants by reducing the damage caused by plant's inseet herbivores. Little is known about the occurrence of extrafloral nectaries in plants growing in Asia. To learn about the occurrence of extrafloral nectary bearing plants in Korea, living plants and herbarium material were examined for the glands. In addition, the cover of plants with extrafloral nectaries and the proportion of woody plants with extrafloral nectaries were measured in three forest communities on Kangwha Island. 131 species of plants belonging to 53 genera and 30 families were found to have extrafloral nectaries. These 131 species comprise about 4.0% of Korea's flora, a highet percentage of extrafloral nectary bearing plants than occurs in the studied areas of North America. Extrafloral nectary bearing plants occupied 7, 23 and 55% of the covers and comprised 15, 21 and 15% of the woody plants in the three different forests, a significant level of occurrence. Many important Korean crop plants were found to have extrafloral nectaries including : sesame (Sesamum indicum L.), squash (Cucurbita moschata Duchesne), sweet potato (Ipomoea batatas Lam), persimmon (Diospyros kaki Thunb.) cotton (Gossypium indicum Lam.), mung bean (Phaseolus radiatus L.), red bean (Phaseolus angularis W.F.), peach (Prunus persica (L) Batsch.), plum (Prunus salicina Lindl.). Many of these cultivated and wild plants may receive protection by ants and other beneficial insects that visit their extrafloral nectaries.

Development of Solanum hougasii-specific markers using the complete chloroplast genome sequences of Solanum species (엽록체 전장유전체 정보를 이용한 Solanum hougasii 특이적 분자마커 개발)

  • Solanum hougasii, one of the wild Solanum species, has been widely used in potato breeding since it exhibits excellent resistance to diverse important pathogens. S. hougasii can be directly crossed with the cultivated tetraploid potato (S. tuberosum) owing to its EBN (Endosperm Balanced Number) value of 4, which is same as that of S. tuberosum although it is an allohexaploid. In this study, the complete chloroplast genome sequence of S. hougasii was obtained by next-generation sequencing technology, and compared with that of the chloroplast genome of seven other Solanum species to identify S. hougasii-specific PCR markers. The length of the complete chloroplast genome of S. hougasii was 155,549 bp. The structural organization of the chloroplast genome in S. hougasii was found to be similar to that of seven other Solanum species studied. Phylogenetic analysis of S. hougasii with ten other Solanaceae family members revealed that S. hougasii was most closely related to S. stoloniferum, followed by S. berthaultii, and S. tuberosum. Additional comparison of the chloroplast genome sequence with that of five other Solanum species revealed five InDels and 43 SNPs specific to S. hougasii. Based on these SNPs, four PCR-based markers were developed for the differentiation of S. hougasii from other Solanum species. The results obtained in this study will aid in exploring the evolutionary and breeding aspects of Solanum species.

  • Solanum demissum is one of the wild Solanum species originating from Mexico. It has wildly been used for potato breeding due to its resistance to Phytophthora infestans. S. demissum has an EBN value of four, which is same as that of S. tuberosum, so that it is directly crossable for breeding purposes with the cultivated tetraploid potato (S. tuberosum). In this study, the chloroplast genome sequence of S. demissum obtained by next-generation sequencing technology was described and compared with those of seven other Solanum species to develop S. demissum-specific markers. Thetotal sequence length of the chloroplast genome is 155,558 bp, and its structural organization is similar to those of other Solanum species. Phylogenetic analysis with ten other Solanaceae species revealed that S. demissum is most closely grouped with S. hougasii and S. stoloniferum followed by S. berthaultii and S. tuberosum. Additional comparison of the chloroplast genome sequence with those of seven other Solanum species revealed two InDels specific to S. demissum. Based on these InDels, two PCR-based markers for discriminating S. demissum from other Solanum species were developed. The results obtained in this study will provide an opportunity to investigate more detailed evolutionary and breeding aspects in Solanum species.

  • This study was initiated to build runoff plots, install soil and water quality monitoring systems and collect background data from the plots and neighboring soils as the 1st year study of a 5 year project to assess soil quality and develop the management practices for environmentally sound agriculture in mountainous soils. Eleven $3{\times}15m$ runoff plots and monitoring systems were installed at a field of National Alpine Agricultural Experiment Station to monitor soil quality and discharge of nonpoint source pollutants. Corn and potato were cultivated under different fertilizer, tillage and residue cover treatments. The soil has a single-layered cluster structure that has a relatively good hydrologic properties and can adsorb a large amount of nutrient. Concentrations of T-N, $NH_4$-N, and $NO_3$-N of surface soil sampled in the winter were relatively high. Runoff quality in the winter and thawing season in the spring was largely dependent on surface freezing, snow accumulation, temperature, surface thawing depth and so on. Runoff during the thawing season caused serious soil erosion but runoff quality during the winter was relatively good. Serious wind erosion from unprotected fields after the fall harvest were obserbed and best management practices to reduce the erosion need to be developed.

  • To know a degree of pollution in agricultural products cultivated around 4 unworked mines located in Chollanam-Do, we investigated a concentration of heavy metal and trace metal to provide the basic data for its residual limits. 28 samples of seven kinds of agricultural products(lettuce, onion, potato, radish, pepper, pumpkin, bean) collected from 4 unworked mines located in Chollanam-Do in 2001 were analyzed by Mercury Analyzer for mercury and Atomic Absorption spectrophotometer for Pb, As, Cd, Cu, Mn, and Zn. Hg contents were detected N.D. ∼trace level(0.01 ppm and less) and As content were detected N.D. ∼ 0.029 lpm but most of same)leE were not detected. Cd contents were detected N.D∼0.124 ppm. The results of Hg, As, Pb and Cd content showed that for all the 7 species of agricultural products studied, none have accumulated levels dangerous enough to Pose health problems. The average contents of Cu were 3.070 ∼ 7.825 ppm in bean, the Mn were 3.688 ∼23.935 ppm in lettuce ailed the Zn were 5.690 ∼21.171 ppm in bean, respectively.

Scab of Tea (Thea sinensis) Caused by Cladosporium herbarum in Korea

  • In 2001, a black scab disease was observed in tea plant (Thea sinensis) cultivated in the hillsides of Hwngaemyon and Hadong-gun, Gyeongnam province, Korea. The disease symptoms initially appeared on leaves, green twigs and stems, showing small dark brown spots on the infected areas, which gradually expanded. A fungus was isolated from diseased leaves and green twigs. It grew readily on potato dextrose agar, forming dark green to dark gray colonies. The optimum temperature for mycelial growth was about 20$^{\circ}C$. The diameter of growing hyphae was 3.5-5.8 $\mu\textrm{m}$. Conidia were ellipsoidal, ovoid or subspherical, and mostly one-celled but occasionally septate. The size of one-celled and septate conidia were 3.7-12.4${\times}$3.4-5.2 $\mu\textrm{m}$ and 9.3-18.7${\times}$3.8-7.2 $\mu\textrm{m}$, respectively. Conidia were formed in long branched chains on the erected conidiophores, which were dark brown in color and 28.9-218.3${\times}$3.0-6.1 $\mu\textrm{m}$ in length. The fungus was identified as Cladosporium herbarum on the basis of its morphological characteristics. The black scab disease occurring in tea caused by Cladosporium herbarum has not been previously reported in Korea.

Case studies for estimation nitrogen and phosphorous balance with different cropping systems in upland (현장사례를 통한 밭 작부의 질소와 인산양분 수지 평가)

  • Cropping system is very important for environment conservation and improvement of nutrient recovery rates in agricultural land. This case study was conducted to identify the nutrient balance of major upland cropping types with different districts. Typical cropping systems at MuAn and HaeNam located in the south coast district were cultivated with soybean-chinese cabbage or garlic and garlic-soybean-chinese cabbage-hot pepper. Alpine district, PyeongChang was cultivated with double cropping of chinese cabbage and potato-radish or chinese cabbage for one year. Typical cropping type of AnDong, YeongYang where are well known for hot pepper cultivation was hot pepper's mono cropping and hot pepper-soybean or hot pepper-sesame for one year. But SeoSan and HongSeong, where are located in mid-west coast had variety cropping system compareed to other districts. Double cropping of chinese cabbage, a heavy fertilizing crop, caused higher nutrient balance in the field when it was cultivated consistently. Experiment showed that heavy fertilizing crops, such as chinese cabbage, garlic and onion, need to be rotated with soybean and sesame cultivation which need smaller fertilizer level. Alpine arable land has shown higher nutrient balance than other areas, and the introduction of rotation crops are needed to reduce nutrient balance and environmental protection.

Development of New Strains of Wolfiporia cocos for Sclerotium Formation by 2-Way Cross-Breeding (이원교배에 의해 균핵 형성하는 복령 균주 개발)

  • Wolfiporia cocos is an important medicinal fungus that has been used in regions of Northeast Asia including Korea, Japan, and China. W. cocos is classified in Korea into two types (red bokryeong and white bokryeong) based on the internal colors (yellow orange-pale pink and white) of the sclerotium. Generally, the W. cocos type cultivated on farms produces white sclerotium. In this study, we endeavored to select strains that form sclerotium in sawdust medium using 2-way cross-breeding among two cultivated strains and three wild strains. Monospores were isolated from the fruiting bodies of cultivated and wild strains on potato dextrose agar. Thirty-nine strains of 338 hybrid strains isolated formed sclerotia with white or yellow colors upon culture for 3 months in Pinus densiflora sawdust medium. Selection for sclerotium forming strains using sawdust culture follows a very simple and easy procedure that is presented for the first time in this paper. We plan to test selected strains in the field to aid in developing new varieties for the future.

Studies on the Quality of Processing Potatoes grown at Different Locations (裁培地域에 따른 加工用 감자의 品質에 關한 硏究)

  • Influence of different cultivated areas on the processing-grade tuber yield, specific gravity, reducing sugar content and tuber qualities of five promising varieties was studied to get the basic information for selecting potato varieties with good processing quality under the different cultivated conditions on Korea. The average total tuber yields of 5 tested varieties at the 2nd harvest time was 3,.051kg/10a in Daekwallyoung. The processing-grade tuber yield of the late-maturing variety of Gemchip was over 3.2 tons per 10a, whereas that of late-maturing variety, NS1 was 2.8 tons per 10a. The dry matter content of the tubers produced from Daekwallyoung was the highest, followed by the order of Bosung, atlantic was the highest in dry matter content, followed by the order of NS1, NS2, Superior and Gemchip. Degrees of change in glucose content as affected by change of cultivated areas were different among varieties. Atlantic and NS1 showed less change of glucose content as compared with other varieties.

Vitamin C Quantification of Korean Sweet Potatoes by Cultivar and Cooking Method (국내산 고구마의 품종 및 조리방법별 비타민 C 함량)

  • This study was carried out to investigate the amounts of vitamin C in 22 sweet potato cultivars cultivated in Korea as well as evaluate the effects of cooking methods on vitamin C contents. Methods for determining vitamin C was validated by determining linearity, specificity, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy using HPLC. Results showed high linearity in the calibration curve with a coefficient of correlation ($R^2$) of 0.9999. The LOD and LOQ values for ascorbic acid (AA) were 0.03 and $0.10{\mu}g/mL$, respectively. The relative standard deviations (RSDs) for intra- and inter-day precision of AA were less than 5%. The recovery rates of AA and dehydroascorbic acid (DHA) were in the range from 98.21~98.64 and 98.28~100.68%, respectively. Depending on cultivar, contents of AA, DHA, and total ascorbic acid (TA) in sweet potatoes varied in the range from 37.76 (Sinyulmi)~89.25 (Juhwangmin), 23.37 (Sinjami)~63.94 (Sinyulmi), and 68.52 (Sinjami)~115.95 (Juhwangmin) mg/100 g, respectively, and their average levels were $56.98{\pm}12.53$, $36.46{\pm}9.03$, and $93.44{\pm}12.00mg/100g$, respectively. The average TA levels were also dependent on flesh color, whish was significantly higher in general sweet potato and orange sweet potato than in purple sweet potato. Steaming, baking, and frying processes significantly reduced AA (10.61~58.41%), DHA (2.57~52.81%), and TA (14.54~49.92%) contents in sweet potatoes. The highest reduction of AA, DHA, and TA contents was observed after baking, followed by steaming and frying. We expect that the basic information provided by this study will be useful to plant breeders and food scientists.
