Determination of Vitamin B12 and Biotin in Foods for Special Dietary Uses with Immunoaffinity Column (면역친화성 컬럼을 이용한 특수용도식품 중 비타민B12와 비오틴 분석 연구)

  • This study was conducted to improve the standard method for vitamin B12 and biotin contained in foods for special dietary uses to ensure the specificity of the complex matrix properties of foods. For the food code, the test method was improved to determine vitamin B12 and biotin by high-performance liquid chromatography (HPLC)-UV using column-switching after concentration using immunoaffinity column. The immunoaffinity columns contain a gel suspension of monoclonal antibody specific to the vitamin of interest so that it can be used to concentrate the vitamin B12 and biotin and remove interferences from the food extracts. Moreover, validation of advanced new methods was carried out to support the suitability of the proposed analytical procedure (specificity, linearity, detection limits (LOD), quantitative limits (LOQ), accuracy, and precision). The improved analytical method is being used to monitor relevant food items on sale. The results of this study showed that the new analytical method is suitable and appropriate for managing food intended for special dietary uses.

Survey of Ochratoxin A in Cereal-based Korean Traditional Foods by HPLC (HPLC를 이용한 전통식품 중 오크라톡신 A 오염도 조사)

  • To determine rapid and reliable analytical method for ochratoxin A detection in cereal-based Korean traditional foods, ochratoxin A content was measured by high-performance liquid chromatography (HPLC) with immunoaffinity column clean-up. Recoveries of ochratoxin A in tested samples ranged from 68.4 to 85.3%. Occurrences of ochratoxin A were 15, 10, and 5% for Kochujang, Deonjang, and Kanjang, respectively. None was detected in Sunsik (mixed cereals). Average levels of ochratoxin A ranged from $0.5\;to\;1.3{\mu}g/kg$, lower than maximum residue level of $5-50{\mu}g/kg$ of ochratoxin A recorded in foreign food code.

Development of a Screening Method and Device for the Detection of Escherichia coli from Agri-Food Production Environments and Fresh Produce

  • This study was conducted to develop a screening method using Colilert-18 and a device for the detection of E. coli from agri-food production environments and fresh vegetables. The specificity and sensitivity of Colilert-18 by temperature ($37^{\circ}C$ and $44^{\circ}C$) were evaluated with 38 E. coli and 78 non-E. coli strains. The false-positive rate was 3.8% (3/78) and 0% (0/78) at $37^{\circ}C$ and $44^{\circ}C$, respectively. The detection limit of E. coli at $37^{\circ}C$ at <1.0 log CFU/250 ml was lower than that at $44^{\circ}C$. The efficiency of the developed device, which comprised an incubator equipped with a UV lamp to detect E. coli in the field, was evaluated by measuring the temperature and UV lamp brightness. The difference between the set temperature and actual temperature of the developed device was about $1.0^{\circ}C$. When applying the developed method and device to various samples, including utensils, gloves, irrigation water, seeds, and vegetables, there were no differences in detection rates of E. coli compared with the Korean Food Code method. For sanitary disposal of culture samples after experiments, the sterilization effect of sodium dichloroisocyanurate (NaDCC) tablets was assessed for use as a substitute for an autoclave. The addition of one tablet of NaDCC per 50 ml was sufficient to kill E. coli cultured in Colilert-18. These results show that the developed protocol and device can efficiently detect E. coli from agri-food production environments and vegetables.

  • While various foodborne pathogenic bacteria can be detected more rapidly via polymerase chain reaction than via conventional plating methods, it is impossible to distinguish between viable and dead cells in DNA-based assays. Hence, propidium monoazide (PMA) treatment has been introduced to detect living cells. The purpose of this study is to evaluate the applicability of the PMA treatment and real-time qPCR method for the detection of Cronobacter sakazakii and to compare it to that of plate counting. Based on our positive results, we suggest the use of PMA treatment and real-time qPCR for the detection of viable Cronobacter sakazakii in various food sources and an update of the Korean Food Code.

Development of Analytical Method and Monitoring of Veterinary Drug Residues in Korean Animal Products

  • This study was conducted to determine the residual amount of veterinary drugs such as meloxicam, flunixin, and tulathromycin in animal products (beef, pork, horsemeat, and milk). Veterinary drugs have been widely used in the rearing of livestock to prevent and treat diseases. A total of 152 samples were purchased from markets located in major Korean cities (Seoul, Busan, Incheon, Daegu, Daejeon, Gwangju, Ulsan and Jeju), including Jeju. Veterinary drugs were analyzed by liquid chromatography-tandem mass spectrometry according to the Korean Food Standards Code. The resulting data, which are located within 70-120% of recovery range and less than 20% of relative standard deviations, are in compliance with the criteria of CODEX. A total of five veterinary drugs were detected in 152 samples, giving a detection rate of approximately 3.3%; and no food source violated the guideline values. Our result indicated that most of the veterinary drug residues in animal products were below the maximum residue limits specified in Korea.

Comparative Study of the Efficiency of GC with Large Volume Injector and SPE Clean-up Process Applied in QuEChERS Method (GC-대용량 주입장치와 SPE를 적용한 QuEChERS 잔류농약 분석법의 효율성 비교)

  • This study was conducted to compare STQ method, multi-residue method in Korean food code and QuEChERS method for validated selected and accuracy, reproducibility and efficiency. A total of 45 selected and targeted pesticides were the analyzed by GC and 5 of them were crops (apple, potato, green pepper, rice, soy bean). $R^2$ values were calculated in the standard calibration curve was over 0.990. Recovery tests were performed by three replications in two levels and the relative standard deviation of the repeated experiments was less than 30%. The average percentage of recoveries in the multi-residue method in Korean food code was 89.13%, QuEChERS method was 92.45% and STQ method was 85.28%. In addition, matrix effects in multi-residue method in Korean food code was 24.61%, QuEChERS method was 23.98% and STQ method showed 11.24%. The STQ method is easy and showed high clean-up effect in extracting the sample solution than the QuEChERS method and clean-up with C18, PLS, PSA cartridge columns. A large volume of the sample was injected in order to compensable for the problem, that occurred due to high detection limit in the analyser. When the STQ method was applied using a large volume injector, the standard calibration curve showed a higher linearity $R^2=0.990$, and method detection limit was 0.01 mg/kg. It showed an average recovery of 91.84% and the relative standard deviations of three replications repeated in two level process was less than 30% and had an average matrix effect of 17.90%.

An Outline of Food Ethics (음식윤리 개요)

  • This review was performed to outline the food ethics to the professionals in the field of food science and industries. Food ethics is the ethics or ethical consideration concerning food; while, the keywords of food ethics are a life, happiness and the wisdom. The ultimate goal as well as the first principle of food ethics is a sustainable living of human race. In order to reach the goal a coexistence of humans and nature, coexistence of humans and humans, and the fulfillment of essential requirements of a food are needed. The needs yield six categorical imperatives such as a respect for life, preservation of nature, distributive justice without deception or fakes, prior consideration of consumer, moderation and balance neither overs nor shorts, and the assurance of safety. The core principles corresponding to the categorical imperatives include a respect for life, environmental preservation, justice, priority of consumer, dynamic equilibrium, and the priority of safety. The core principles approach is a simple, clear, and practical method in solving ethical issues. The code of food ethics is also based on the core principles.

Proficiency test for analyzing illegal compounds in food (식품 중 부정유해물질의 분석 능력평가)

  • To perform proficiency test for determining anti-impotence drug-like compounds in food, interlaboratory test has been done. Targets were 6 such as sildenafil, tadalafil, vardenafil, homosildenafil, hydroxy-homosildenafil and pseudo-vardenafilI. Total 13 institutes were participated and all is in charging of food analysis. To do double blind test, the reference materials were made as 13 different ginseng drink samples and each sample had random 3 targets. By the official method for anti-impotence drug-like compounds in food code, sample was just diluted in water, extracted in organic solvents, determined by HPLC/UV and then confirmed by LC/MS. The analytical duration was 60 days after receiving sample. Ten out of 13 institutes were satisfied by evaluation of Z-score and RSZ according to the regulation for managing analytical quality assurance.

  • Koji is steamed rice that has had koji-kin, or koji mold spores, cultivated onto it. The isolation, culture, and microscopic examination of molds in the koji require the use of the selective media and special microscopic slide techniques. If simple wet mount slides of molds were attempted, it became apparent that wet mount slides made from mold colonies usually don't reveal the arrangement of spores that is so necessary in identification. The process of merely transferring hyphae to a slide breaks up the hyphae and sporangiophores in such a way that identification becomes very difficult. The slide culture method is superior to wet mounts in that the hyphae, sporangiophores, and spores remain more or less intact when transferred. The procedure that will be used to produce a mold culture on a slide that can be observed directly on the slide. We investigated the contamination rate of penicillium spp. on the 21 kinds of koji distributed at Korea. The contamination rate of Penicillium spp. were not detected at 21 products by slide culture method. These results will be used to reestablish a mold determination of koji and food in Food Additives Code.

Analysis of Amoxicillin, Ampicillin, Oxolinic Acid, and Flumequin in Foods (식품 중 아목시실린, 암피실린, 옥소린산 및 플루메퀸의 분석)

  • The present study was performed to validate an analytical method for veterinary drugs, including amoxicillin, ampicillin, oxolinic acid and flumequin, in meat, fish, and their products as established in the 2007 Korea Food Code, and to monitor the respective drugs in 6 kinds of foods, namely beef, pork, flatfish, rockfish, freshwater eel and shrimp. Additionally, an HPLC-ESI-MS/MS method was developed for the qualification of these veterinary drugs. As a result of monitoring, these drugs were detected in 14 (3.6%) out of 393 samples. In particular, amoxicillin was detected in 9 flatfish samples, and oxolinic acid and flumequin were detected in 4 samples and 1 sample of freshwater eel, respectively. The detection levels of amoxicillin ranged from 0.009 to 0.078 mg/kg, in which 1 sample of flatfish contained more than the MRL (0.05 mg/kg). Flumequin detected in 1 sample of freshwater eel, was less than the MRL (0.5 mg/kg). Therefore, the majority of detected levels were less than the MRLs, with the exception of 2 samples showing amounts that were 1-2 times higher than the MRLs. The detected veterinary drugs were identified and confirmed by the HPLC-ESI-MS/MS method created in this study.
