황기, 작약 및 산수유의 독성평가를 위한 성분분석 및 안정성 시험 (Analysis and Stability Test of the Extracts from Astragali Radix, Paeoniae Radix, and Corni Fructus for Toxicity Study)

  • A simple and reliable reverse phase HPLC method was developed to determine pharmacologically active marker compounds of Astragali Radix, Paeoniae Radix, and Corni Fructus. The stability test of water-extract from three natural medicines were examined for six months. However, no significant changes in the content of the marker compounds of each extract were observed during the investigation.

Clostridium difficile에 대한 항균 한약재 탐색 연구 (A Study of Antimicrobial Activity of Herbal Extracts on Clostridium difficile)

  • Objectives This study was conducted to confirm the possibility of Clostridium difficile infection (CDI) treatment through natural herbal medicines. Methods After screening a total of 77 herbal medicines through the paper disc agar diffusion method, we selected the herbal medicines that showed a effectiveness compared to the positive control vancomycin. Afterwards, drugs that showed inhibitory effects compared to C. difficile without inhibition of Bifidobacterium bifidum and Lactobacillus plantarum, known as beneficial bacteria, were selected and minimal inhibitory concentration (MIC) was confirmed by applying the Broth microdilution method. Results The Coptidis Rhizoma, well known for its antimicrobial effect, was found to have antimicrobial effects on C. difficile, but also had inhibitory effects on the beneficial bacterium B. bifidum. 30% ethanol extraction Crataegi fructus, Corni fructus and Mume fructus had antimicrobial effects on C. difficile without inhibiting the beneficial bacteria B. bifidum and L. plantarum. The MIC values of 30% ethanol extraction Crataegi fructus, Corni fructus and Mume fructus were found to be 10 mg/mL, 20 mg/mL and 5 mg/mL, respectively. Conclusions Crataegi fructus, Corni fructus and Mume fructus were identified as candidate medicines for C. difficile. Further researchs will need to be done in vivo, and to find an optimal extraction method accompanied by economic evaluation.

가시오가피, 당귀와 산수유 성분이 함유된 메주로 제조한 전통 된장의 이화학적 특성 및 관능적 특성 (Physicochemical and Sensory Characteristics of Traditional Doenjang Prepared using a Meju Containing Components of Acanthopanax senticosus, Angelica gigas, and Corni fructus)

  • Changes in the quality characteristics of doenjang prepared with a meju containing components of Acanthopanax senticosus, Angelica gigas and Corni fructus during storage were evaluated. Three types of doenjang produced using each of these components showed lower Hunter's L and a values than the values of the control during the initial stage of storage: however, these values were similar to those of the control after 60 days of storage. Doenjang containing A. gigas showed a similar degree of browning to that of the control during the early stage of storage, while the degree of browning observed in the other samples was much higher than that og the control during this period. Despite these initial differences, the browning in all samples became similar after extended storage. The level of acidity increased gradually for up to 60 days, after which it decreased slightly. The pH of all treatments decreased with storage time. The amino-N contents of all types of doenjang increased gradually, reaching peak values after 60 days. Doenjang with A. senticosus and A. gigas had a strong characteristic flavor that reduced the native flavor of doenjang. Doenjang with C. fructus showed a similar overall palatability to that of the control.

Inhibitory Effects of Corni Fructus Extract on Angiogenesis and Adipogenesis

  • Natural products in Chonnam, Korea were screened via anti-angiogenesis experiments, and 1 candidate product was identified, Corni fructus, which exerted dose-dependent inhibitory effects against angiogenesis, adipogenesis, and cell adhesion. C. fructus extract (CFE) exhibits an angiogenesis inhibitory effect superior to that of the EGCG from green tea leaves. The expression level of angiogenesis and adipogenesis-related signal molecules in the western blotting was reduced by increasing the amount of added CFE. Moreover, a diet supplemented with CFE was deemed more effective in inducing weight loss in LB mice than a representative synthetic diet drug, orlistat, which incidently caused the side effect of denuding the mice of their hair. These results indicate that C. fructus may prove to be a useful anti-adipogenic compound, and these in vitro results may be reflected later under in vivo conditions.

한국산 산수유가 난소적출로 유발된 흰쥐의 골다공증 치료효과에 미치는 영향 (Effects of Korean Corni Fructus On treatment of Osteoporosis In Ovariectomized Rats)

  • Objectives : The present study had been undertaken to investigate the effects of Korean Corni Fructus(KCF) on treatment of osteoporosis in ovariectomized rats. Method : In this experiment, the rats were ovariectomized. Rats were administered by KCF. The levels of bone mineral density, osteocalcin, ALP, calcium, phosphorus in serum, calcium, phosphorus, deoxypyridinoline in urine, calcium, phosphorus, ash weight of bone, body weight and uterus weight were measured. Results : The levels of femoral and fibula-tibial bone mineral density were significantly increased in comparison with OVX group at 4, 8 weeks in KCF group. The levels of serum osteoclacin showed significant decrease in comparison with OVX group at 4, 8 weeks in KCF group. The levels of serum ALP showed significant decrease in comparison with OVX group at 4 week in KCF group. The levels of serum calcium showed significant increase in comparison with OVX group at 8 week in KCF group. The levels of urine calcium and phosphoruls showed significant decrease in comparison with OVX group in KCF group. The levels of femoral and fibula-tibial calcium didn't show significant changes in comparison with OVX group in KCF group. The levels of femoral and fibula-tibial phosphorus didn't show significant changes in comparison with OVX group in KCF group. The levels of femoral and fibula-tibial ash weight didn't show significant changes in comparison with OVX group in KCF group. The levels of body weight were significantly decreased in comparison with OVX group at 4, 8 weeks in KCF group. The levels of uterus weight were significantly increased in comparison with OVX group in KCF group. Conclusion : Reviewing these experimetal results, it appeared that KCF had efficacy on treatment of osteoporosis.

Fructus Corni Officinalis water extract Ameliorates Memory Impairment and Beta amyloid (Aβ) clearance by LRP-1 Expression in the Hippocampus of a Rat model of Alzheimer’s Disease

  • This study evaluated the effects of Fructus Corni Officinalis water extract (FCE) on congnitive impairment and Aβ clearance induced by beta amyloid Aβ (1-42) injection in the hippocampus of rat. Aβ (1-42) was injected into the hippocampus using a Hamilton syringe and micropump (5 ㎍/5 ㎕, 1 ㎕/min, each hippocampus bilaterally). FCE was administered orally once a day (100, 250, 500 mg/kg) for 4 weeks after the Aβ (1-42) injection. The acquisition of learning and retention of memory were tested using the Morris water maze. Aβ accumulation and Aβ clearance in the hippocampus were observed using immunostaining. Aβ (1-42) level in plasma was confirmed using enzyme-linked immunosorbent assay (ELISA). FCE significantly shortened the escape latencies during acquisition training trials. FCE significantly increased the number of target heading to the platform site and significantly shortened the time for the 1sttargetheadingduringtheretentiontesttrial.FCEsignificantlyattenuatedtheAβ accumulation in the hippocampus produced by Aβ (1-42) injection. FCE significantly increased LRP-1 expression around vessels in the hippocampus and Aβ (1-42) levels in plasma. The results suggest that FCE improved cognitive impairment by ameliorate Aβ clearance and Aβ accumulation in the hippocampus. FCE may be a beneficial herbal formulation in treating cognitive impairment including Alzheimer's disease.

산수유의 유리자유기에 의한 간손상 보호효과 및 기전에 대한 연구 (Study on the Protective Effect of Corni Fructus Against Free Radical Mediated Liver Damage)

  • We evaluated the potential protective activity of the traditional Korean medicinal herb, Corni fructus (CF), in an experimental model of hepatotoxicity induced by carbontetrachloride $(CCl_4)$. The CF exhibited a hepatoprotective activity against Chang cell. And The expression of cytochrome P450 2E1 (CYP2E1), measured by RT-PCR and western blot, was significantly decreased in the CF treated Chang cell. But $CCl_4$ and CF has no significant effect on 1A1 and 3A1 isoform of cytochrome P450. Based on these findings, it is suggested that hepatoprotective effects of CF possibly related to antioxidative effects and downregulation of CYP2E1 expression.

육미지황탕 구성약물이 PC12 세포의 산화억제에 미치는 영향 (Antioxidant effects of the Herbs Composing Yukmijihwang-tang on PC12 Cell)

  • Antioxidant effects of Rehmannia vaporata, Discorea Radix, Corni Fructus, Hoolen, Alismatis Radix, and Mountain Cortex Radicis composing Yukmijihwang-tang were studied. The results are as follows; 1. As a result of detecting the defensive effect of each component on cell damage, only the survival rate of cells with 10 ㎎/㎖ Mountain Cortex Radicis was significantly increased. 2. Next, we examined the inhibitory effects of them on ROS occurrence. The result showed significant inhibition of ROS occurrence in cells; with 10 ㎎/㎖ Rehmannia vaporata, cells with 10 ㎎/㎖ Corni Fructus, and cells with 10 ㎎/㎖ Mountain Cortex Radicis. Since the cells with 10 ㎎/㎖ Rehmannia vaporata, however, showed significant cypotoxicity, its result is not meaningful. 3. Finally, the investigation of ROS occurrence / cell found that Corni Fructus and Mountain Cortex Radicis had significant inhibitory effect on ROS occurrence.

자외선이 조사된 무모생쥐에서 홍삼, 사상자, 산수유 혼합 추출물 섭취가 표피 세라마이드 함량 및 관련 효소의 발현에 미치는 영향 (Dietary effect of red ginseng extracts mixed with torilis fructus and corni fructus on the epidermal levels of ceramides and ceramide related enzyme proteins in uv-induced hairless mice)

  • UV-irradiation is a major factor of photo-aged skin, by which pigmentation, wrinkles and laxity are increased. In addition, the epidermal barrier is disrupted, ultimately causing dryness in photo-aged skin. As an effort to search dietary sources for improving the dryness of UV irradiated skin, the dietary effect of red ginseng based functional foods on the epidermal level of ceramides, a major lipid maintaining epidermal barrier, was determined in this study. Albino hairless mice were fed either a control diet [group UV (UV-irradiated control)] or diets with 0.5% (group M0.5) or 1% (group M1.0) of red ginseng extracts mixed with Torilis fructus and Corni fructus (66.7% red ginseng) in parallel with UV irradiation for 5 wks. A normal control group (group C) was fed a control diet without UV irradiation for 5 wks. The epidermal level of ceramides in group UV was significantly lower than that in group C, in which ceramidase, an enzyme involved in ceramide degradation, was highly expressed. In group M0.5, the epidermal level of ceramide was significantly increased to the level even higher than in group C. In addition, protein expression of serine palmitoyl transferase (SPT), a key enzyme involved in de novo ceramide synthesis, was increased in group M0.5. However the epidermal levels of ceramides as well as of ceramidase protein expression in group M1.0 did not differ from those in group UV. In conclusion, we demonstrate that dietary supplementation of red-ginseng extracts mixed with Torilis fructus and Corni fructus at a level of 0.5% level in diet increased the epidermal level of ceramides coupled with the elevated expression of SPT protein.

항산화 천연산물의 조합구성에 의한 Bifidobacterium bifidum의 성장 촉진 효과 (The Growth Promoting Effect of Bifidobacterium bifidum by Combination of Natural Products Bearing Antioxidative Capacity)

  • B. bifidum의 성장은 천연산물의 한 종류 단독으로서도 촉진되었으며, 또한 두 종류, 세 종류, 네 종류의 조합에 의하여 특징적으로 성장이 촉진되었으며, 이것은 항산화력이 강한 천연산물들을 free radical의 제거와 동시에 B. bifidum의 성장 촉진 물질로 사용할 수 있고 이를 이용하여 기능성 항산화 제제의 제조가 가능하리라 사료된다. 이 균주는 연자육 산수유 및 사간 등의 한 종류의 천연산물에 의하여 성장이 촉진되었으며, 두 종류 천연산물의 조합인 연자육과 녹차, 연자육과 산수유 등의 조합, 세 종류의 천연산물 조합인 연자육과 산수유 및 녹차, 연자육과 녹차 및 사간 등의 조합, 네 종류의 천연산물 조합인 녹차, 사간과 산수유 및 감초의 조합, 연자육, 산수유와 녹차 및 감초 등의 조합에서 잘 성장함을 보였으며, 녹차, 사간과 산수유 및 감초의 조합이 대조군보다 2.6배 이상의 성장을 보였다. B. bifidum의 성장을 촉진시킨 4차 배합 조성물의 항산화력은 연자육, 녹차와 사간 및 산수유 조합의 AUC가 694.43로써 대조군의 AUC(116.61)보다 6배이상 높게 나타났으며, 이 조합의 세균성장은 대조군보다 2.6배의 성장 촉진 효과를 보였다. 다음으로는 연자육, 산수유와 녹차 및 감초의 조합(672.14), 녹차, 사간과 산수유 및 감초(633.03)의 조합, 그리고 연자육, 녹차와 사간 및 감초의 조합(560.87)의 순으로 나타났으며, 이들의 항산화력은 대조군의 AUC 116.61보다 5-6배 정도 강한 것으로 나타났고, 항산화력이 강한 조합에서 좋은 세균의 성장 촉진을 보였다. 네 종류의 4차 조합된 천연산물의 hydroxyl radical의 소거활성은 항산화 활성이 강하게 나타난 연자육, 산수유와 녹차 및 감초의 조합에서 84%의 소거활성을 보였으며, 당귀, 음양곽과 백작약 및 녹차의 조합이 100배의 희석액에서 87.99%의 소거 활성을 보였고, 연자육, 녹차와 사간 및 산수유의 조합, 녹차, 사간과 산수유 및 감초의 조합 그리고 연자육, 녹차와 사간 및 감초의 조합에서도 희석 배수에 무관하게 모두 80% 이상의 hydroxyl radical의 소거 활성을 보였으며, 또한 네 종류 조합의 총phenol량은 녹차중의 증제차나 볶음차의 비발효 차 보다 30-32배 이상 높은 것으로 나타났으며, 반 발효차인 오룡차보다는 32-35배, 발효차인 홍차보다는 55-60 정도 높은 것으로 나타났다. 이상의 4종류 조합은 B. bifidum의 성장 촉진 및 free radical 소거 활성이 좋은 것으로 나타나 장의활성을 도울 수 있는 다기능성 제제로서의 가능성을 보였다.
