Application as a Cosmeceutical Ingredient of Extract from Euphorbia jolkini (암대극 추출물의 화장품 원료로서의 특성)

  • In this study, we evaluate anti-oxidation, whitening and anti-inflammatory efficacies of Euphorbia jolkini extract for application as a cosmeceutical ingredient. We separated 5 fractions from Euphorbia jolkini extract (70 % MeOH) by MPLC. First and 5th fractions showed a supressive effect on Mn-SOD synthesis in the normal human fibroblasts. They inhibited melanogensis in the B16-F10 melanma cells. Furthermore, 1st and 5th fractions reduced the amounts of $IL-1{\alpha}$, IL-6, COX-II and total NO secreted from the normal human fibroblasts. These results suggest that Euphorbia jolkini extract may be used as an active ingredient in cosmetics.

  • Acyranthes bidentata has been used as anti-arrthritic, aphrodisiac, anti-viral, anti-spasmodic, anti-hypertensive, anti-coagulant and anti-tumor agent. In this study, we evaluated anti-inflammatory effect of polysaccharide extract from Acyranthes bidentata. Acyranthes bidentata polysaccaride decreased $IL-1{\alpha}$, IL-6, COX-2, and total NO synthesis. Our results point to the potential use of Acyranthes bidentata polysaccaride as a cosmeceutical agent for inflammatory cutaneous symptoms.

Characterization of the N-glycosylation of Recombinant IL-4 and IL-13 Proteins Using LC-MS/MS Analysis and the I-GPA Platform

  • Interleukin-4 (IL-4) and IL-13 are cytokines secreted by immune cells. Cytokines induce the proliferation of macrophages or promote the differentiation of secretory cells. The initiation and progression of allergic inflammatory diseases, such as asthma, are dependent on cytokines acting through related receptor complexes. IL-4 and IL-13 are N-glycoproteins. Glycan structures in glycoproteins play important roles in protein folding, protein stability, enzymatic function, inflammation, and cancer development. Therefore, the glycan structure of IL-4 and IL-13 needs to be elucidated in detail for the development of effective therapies. We report the first attempt to characterize the site-specific N-glycosylation of recombinant IL-4 and IL-13 via liquid chromatography-tandem mass spectrometry (LC-MS/MS) analysis. The tandem mass spectra of intact N-glycopeptides were identified using the Integrated GlycoProteome Analyzer (I-GPA) platform, which can automatically and rapidly analyze multiple N-glycopeptides, including their glycan composition and amino acid sequences. The recombinant IL-4 and IL-13 were identified with amino acid sequence coverages of 84% and 96%, respectively. For IL-4, 52 glycoforms on one N-glycosylation site were identified and quantified. In IL-13, 232 N-glycopeptides from three N-glycosylation sites were characterized, with the site Asn52 being the most extensively glycosylated (~80%). The complex glycans were the most abundant glycan on IL-4 and IL-13 (~96% and 91%, respectively), and the biantennary glycans were the most abundant in both recombinant IL-4 and IL-13 proteins.

Effects of Water Extract of Glycyrrhiza uralensis on $\beta$-Hexosaminidase Release and Expression of the Cytokines of RBL-2H3 Mast Cells (감초 추출물이 RBL-2H3 비만세포에서 $\beta$-hexosaminidase 분비 및 Th2 cytokine mRNA 발현에 미치는 효과)

  • The present study was conducted to investigate the anti-allergic reaction with Glycyrrhiza uralensis. We examined cell viability, $\beta$-hexosaminidase release, IL-4 and IL-13 mRNA expression from RBL-2H3 cell after pre-treatment with 0, 100, 250, 500, 1000${\mu}g/m{\ell}$ of Glycyrrhiza uralensis water extracts. Effects of Glycyrrhiza uralensis on the degranulation and pro-inflammatory cytokines (IL-4 and IL-13) expression were evaluated with $\beta$-hexosaminidase assay, and RT-PCR analysis. We observed that Glycyrrhiza uralensis concentrations from 100${\mu}g/m{\ell}$ to 1000${\mu}g/m{\ell}$ had no effect on cell survival. The release of $\beta$-hexosaminidase decreased significantly with all concentrations of Glycyrrhiza uralensis extracts. The expression of the IL-4 and IL-13 mRNA were decreased by Glycyrrhiza uralensis in dose-dependent manner. These results that Glycyrrhiza uralensis has an anti-histamin effects and controls IL-4, IL-13 secretion on allergic reaction.

No Evidence of Association of Interleukin 1A (-889) Genetic Polymorphism with Alzheimer's Disease in Koreans

  • To examine whether the IL-1A (-889) polymorphism associates with a risk for Alzheimer's disease (AD) and acts interactively with the apolipoprotein (APOE) $\epsilon$4 in the development of AD, we performed genotype analyses of the IL-1A and the APOE of the 102 Korean AD patients and 200 Korean non-demented controls. We failed to detect a significant difference in genotypic and allelic frequencies of IL-1A between the AD group and control group. No overexpression of the IL-1A C/T genotype and IL-1A T allele was found when we analyzed the late-onset and early-onset patients, separately. There was no significant genetic interaction between IL-1A polymorphism and the APOE polymorphism. I n conclusion, the IL-1A polymorphism did not contribute to the development of AD independently or interactively with the APOE $\epsilon$4 allele in Koreans.
