Effects of 3-Amino-1,2,4 Triazole and Diethyldithiocarbamate on Paraquat Toxicity in Rats (흰쥐에서 Aminotriazole과 Diethyldithiocarbamate가 Paraquat의 독성에 미치는 영향)

  • The effects of superoxide dismutase(SOD) and catalase on the toxicity of paraquat(PQ) were studied using diethyldithiocarbamate(DDC), 3-amino-1,2,4-triazole(AT) which are inhibitors of Cu, Zn-SOD and catalase in rats. Sprague Dawley rats were divide into 6 groups: control, DDC, PQ, AT, DDC+PQ, and AT+PQ group. The PQ (50 mg/kg body weight(BW); about half dose of $LD_{50}$) was administered with orally, otherwise AT(1.0g/kg BW) and DDC(1.0g/kg BW) were administered by intrperitoneal(iP) injection. The survival rate of rats in PQ+AT group was significantly decreased compared with PQ group while the difference of survival rate between DDC group and DDC+PQ group was not significant. The SOD activity after administration of DDC was decreased in liver, lung and kidney, but catalase activity was not changed. The catalase activity in liver, lung and kidney of AT treated rats was decreased, while SOD activity was not changed in this group. The effects of DDC and AT to the PQ toxicity was also observed in primary cultured rat Skin fibroblasts. The viable cells that was measured with MTT method, was decreased in AT+PQ treated group compared to PQ treated group, but the difference of cell viability between DDC treat group and DDC+PQ treated group was not observed. This result, AT potentlate PQ toxicity while DDC were not affect, suggested that the decreased catalase activity lead to elevation of hydrogen peroxide levels and PQ toxicity may be correlate with the hydrogen peroxide rather than the superoxides.

  • A study on the antitoxic effects of Semen Mungo extracts against Cadmium Chloride Toxicity in Rats This study was performed to find out the effect of Semen mungo against Cadmium toxicity. The experimental rats were divided into 5 groups such as control group, Cadmium alone treatment group, three simultaneous treatment groups of semen mungo and cadmium. Rats were given pellets administration with three dosage of semen mungo glabrae such as 4mg/kg body weight for four weeks. The results were summarized as follows on: ' 1. The simultaneously administration of semen mungo and cadmium significantly more decreased cadmium concentration in liver and kidney tissues compared to the administration of cadmium alone.(p<0.05) 2. The simultaneously administration of semen mungo and cadmium significantly more decreased metallothionein concentration in liver and kidney tissues compared to administration of cadmium alone(p<0.05) 3. When liver and kidney tissues were observed with optical microscope obvious changes were visible in those tissues.1

  • The use of cisplatin is limited by severe side effects such as renal toxicity. Our platinum-base drug discovery is aimed at developing drugs capable of diminishing toxicity and improving antitumor activity. We synthesized new Pt (II) complex analogue [Pt (cis-DACH)(DPPP)]. 2NO$_3$ (PC) containing cis-1,2-diaminocyclohexane as a carrier ligand and 1,3-bis(diphenylphosphino) propane as a leaving group. Furthermore, nitrate was added to improved the solubility. In this study, its structure was determined and its antitumor activity against SKOV-3 and NIH-OVCAR-3 human ovarian adenocarcinoma, and in vitro cytotoxicity was determined against primary cultured rabbit kidney proximal tubular and renal cortical cells of human kidney using colorimetric MTT assay. PC demonstrated acceptable antitumor activity against SKOV-3 and NIH-OVCAR-3 human ovarian adenocarcinoma and significant activity as compared with that of cisplatin. The toxicity of PC was found quite less than that of cisplatin using MTT and $^3$H-thymidine uptake tests in rabbit proximal tubular cells and human kidney cortical cells. PC was used for human cortical tissue in 7 weeks hitoculture by the glucose-consumption tests. We determined that the new platinum drug has lower nephrotoxicity than cisplatin. Based on these results, this novel platinum (II) complex compound (PC) represent a valuable lead in the development of a new anticancer chemotherapeutic agent capable of improving antitumor activity and low nephrotoxicity.

A Study on the Preventive Effect of Kam Doo Decoction on the Subacute Lead Toxicity in Rats (흰쥐에서 아급성 연독성에 대한 감두탕의 예방효과에 관한 연구(I) - 장기 및 대변의 축적에 미치는 영향을 중심으로 -)

  • This study was performed to investigate the preventive effect of KDD against lead toxicity. KDD of 133, 266, 532 and 1,064 mg/kg were administered twice to the rats of Sprague-Dawley strain and then 300 mg/kg lead acetate was given to times, respectively. 1. The accumulation effects of KDD against to lead showed the changes of lead concentration by time variation. But, no statistical significance were showed on 8 and 10 weeks for kidney, spleen, 8 weeks for liver, and 4, 6 and 8 weeks for duodenum. In the femur, statistical significance existed during the whole experimental period. The relatively high concentration of lead detected in the feces of the experimental group means that KDD facilitated excretion of lead. 2. The histopathological effect of KDD against lead showed cytomegaly, karyomegaly, inclusion body, urinary cast and hemosiderin of kidney in the experimental group I (Pb 300 mg/kg). Recovery of KDD administrated group was inclined to increase by KDD concentration. But, spleen's histopathological recovery of KDD aginst to lead did not show as much as kidney. In conclusion, this study revealed the preventive effect of KDD against lead toxicity and its mechanism inferred to facilitate lead excretion in feces following hinderance of lead absorption in the gastric-intestine and organs.

Effects of Onion Diet on Carbon Tetrachloride Toxicity of Rats (양파식이가 흰쥐에서 사염화탄소 독성에 미치는 영향)

  • This study designs to investigate effects of onion diet on carbon tetrachloride toxicity of rats. Experiments were performed with week's feeding, body weight, food intake, ratio of orgen weight/dody weight, serum lipid levels, superoxide dismutase and catalase activity and malondialdehyde(MDA) content in liver and kidney were determined. The content of serum total cholesterol in each group were lower than those of control group(p<0.05), especially at onion juice treated group. Serum HDL-cholesterol level of CBB and CBJ groups was significantly lower than that of control group (p<0.05). Superoxide dismutase activities of liver and kidney were significantly increased by carbon tetrachloride treatment and decreased by onion feeding. MDA contents in liver and kidney of CCl4 treated rats were significantly decreased by boiled and fresh onion fed group, compared with CCl4 treated control group. This result suggested that onion diet has a protective effect of CCl4 induced hepatotoxicity and nephrotoxicity of rat.

  • The term Butterfly tea refers to decoction of Mariposa christia vespertilionis leaves which is widely consumed by cancer patients throughout Malaysia and has gained a huge popularity among Malaysians, not only cancer patients but also researchers to discover the real potential of this plant. Herein, the study is aimed at evaluating the possible toxicity in 28-day subacute oral toxicity of ethanolic extract M. christia vespertilionis in male Sprague Dawley rats. The 28-day subacute toxicity study was conducted to detect the no-observed adverse effect level (NOAEL). In this study, a total of 30 rats were divided into the control, 5% DMSO (vehicle), low dose (75 mg/kg), medium dose (125 mg/kg) and high dose (250 mg/kg) groups. The extract was administered daily from day 1 until day 28. At the end of the study, the animals were humanely sacrificed and assessed for the effect extract of Mariposa christia vespertilionis leaves on body weight and relative organ weights and haematological, biochemical and histopathological parameters. The haematological and serum biochemical parameters for the assessment of kidney and liver injuries were carried out. Results of haematological and serum biochemistry results showed no changes in the control and treated groups. In the histopathology, evaluation of kidney tissues in all treated groups showed no significant (p > 0.05) lesions. In contrast to kidney, liver tissues showed significant differences (p < 0.05) in lesions observed in low dose (430 mg), medium dose (700 mg) and high dose (1480 mg) groups with very mild, mild and mild to moderate lesion of hepatic necrosis, in the respective groups, and very mild hepatic degeneration and hepatitis were scored in all three groups.

  • Objectives : This study was carried out to investigate the biological activities of methanol extracts of fermented Camellia japonica leaf and flower. Methods : Methanol extracts of fermented Camellia japonica leaf and flower were prepared and a dose of 100 and 400mg/kg/day was administered orally into mice. And after appropriate weeks, changes of serum enzyme activities were investigated to confirm its effects on serum glucose, cholesterol and short term administration safety. Results : Fermented flower extract showed significant decrease of serum level of cholesterol. And showed no toxicity on kidney and liver within the dose of 400mg/kg/day. Conclusion : Thus above result showed no toxicity on kidney and liver in male and female mice.

Effects of Humic Acid on the Cadmium-Induced Fish Toxicity (Humic acid가 카드뮴 어독성에 미치는 영향)

  • Cadmium, a major aquatic pollutant in many parts of the world, is toxic and readily accumulated in aquatic organisms. It mainly exists in water as complexes with organic ligands such as dissolved organic carbon and this complexation is known to have effects on the aquatic toxicities. In this study, acute toxicity, histology, and bioaccumulation were studied to evaluate effects of humic acid on cadmium toxicity and bioavailability to fish using Oryzias lalipes and Cyprinus carpio. 96h-LC$_{50}$ of cadmium was 6.38mg/L using Oryzias latipes. However, the mortality showed the dose-dependent decrease when humic acid was added to the test solution. When fish were exposed to 5 mg/L of cadmium, histological changes in the exposed organs (caudal fins, gills, kidney, liver, and gut) of C~lprinus carpio were decreased by humic acid, especially in kidney and liver. Bioaccumulation of cadmium also decreased by treatment of humic acid. It seems that the formation of complexes between cadmium and humic acid may decrease bioavailability of cadmium to fish, and thus reduce the toxic effects of cadmium.

Effect of Dietary Cysteine Level on Cadmium on Cadmium and Lead Toxicity in Rats (식이내 Cysteine 수준이 흰쥐의 카드뮴과 납중독에 미치는 영향)

  • This study has been investigated the potenial of increased dietary cysteine to alter the effects of cadmium and lead on tissure and bone metal concentrations, excretion and tissue metallothionein(MT) concentrations. Fifty-four male rats of Sprgue-Dawley strain weighing 149$\pm$17g were divided into 9 groups according to body weight. Nine experimental diets with different cadmium (0ppm, 400ppm), lead(0ppm, 710ppm) and cysteine (0.06%, 0.45%, 0.90%) levels were given to rats for 30 days ; Food intake, weight gain, F.E.R, and weights of liver, kidney and femur were decreased in cadmium supplied groups than in cadmium free groups. Urinary and fecal cadmium excretions were increased and MT synthesis we induced in liver, kidney and small intestine in cadmium supplied groups. In lead supplied groups, weight gain and F.E.R were decreased. With cysteine supplementation in cadmium supplied groups, weight gain and F.E.R, and weights of liver, kidney and femur were increased. Cadmium excretion in feces and MT concentrations in liver and kidney were also increased with cysteine supplementation. In lead supplied groups, there was no significant increase in food intake, weight gain and F.E.R with cysteine supplementation. Lead excretion in feces was increased in cysteine supplemented groups. In conclusion, effect of cadmium administration was more toxic than lead adminstration. Cysteine alleviated cadmium and lead toxicity by increasing metallothionein concentration and fecal excretions of heavy metals. Especially, effect of cysteine supplementation was more effective in cadmium groups than in lead groups. Effect of cysteine supplementation was not different with level of cysteine supplementation in both cadmium and lead groups.
