Purification and Properties of Homoseine Dehydrogenases in Canavalia Iineata Cotyledons (해녀콩(Canavalia lineata) 자엽에서 Homoserine Dehydrogenase의 정제 및 특성)

  • Two forms of homo serine dehydrogenase have been isolated from 8-day-old cotyledons of Canavalin lineata by a heat denaturation, ammonium sulfate fractionation, DEAE-8ephacel ion exchange and Sephacryl 8-300 gel filtration chromatographies, and Pro cion red dye, Cibacron blue dye and Resource Q column chromatographies. The molecular weights of T -form (threonine-sensitive) and K-form(threonine- insensitive) were estimated to 230 kD and 135 kD, respectively. In the presence of 10 mM threonine, the activity of T-form was inhibited with almost 70%, but that of K-form was not at all. The Km values tor homo serine of T- and Kform were 1.6 mM and 0.3 mM, respectively. The Km values for NAD of T- and K-form were 2.34 mM and 0.03 mM, respectively. And Km values for NADP of two isozymes were the same as 0.01 mM. The activities of T- and K-form were markedly stimulated up to 4.9and 2.8-fold, respectively, by 400 mM KCI. The partial purified(gel filtration) enzymes(Tform and K-form) can be reversibly converted.verted.

Characteristics of Marine Debris collected from the Coastline of Sandbar in the Nakdong River Estuary (낙동강 하구 사주 해안에서 수거한 쓰레기 종류 및 분포량 추정)

  • This study investigated the spatial distribution of the marine debris in Jinudo in the Nakdong river estuary. Types of marine debris in Jinudo are analyzed through sampling around some portion of its sandbar shoreline. The present study and its main results are summarized as follow: 1) Monitoring system, for evaluating the volume of marine debris, was established in the coastal region of $50\;m\;{\times}\;50\;m\;(=0.002\;km^2)$ over the southern part of Jinudo. All the marine debris of conceivable form are collected and their amount was also estimated. 2) During the $1^{st}$ collection in May 2007, the total weight of the marine debris was 1,110 kg in this site, which means the density of marine debris around the shoreline is $444\;ton/km^2$. After one month later, the collected marine debris was 23.75 kg and so we have the average density of $9.5\;ton/km^2$. From these results, it is suggested that the marine debris of 316.67kg was being accumulated per day over unit area($1\;km^2$). 3) The most frequent marine debris collected during monitoring campaigns was wooden material amounting to 85.86% of the whole collection; the rest were fishing gears such as nets and buoys(5.13%), household waste(2.34%), glass(0.94%) and metals(0.27%).

Distribution of Salinity and Temperature due to the Freshwater Discharge in the Yeongsan Estuary in the Summer of 201 (2010년 여름 담수방류에 의한 영산강 하구의 염분 및 수온 분포 변화)

  • The short-term variation of salinity and temperature in a dyked estuarine environment is mainly controlled by the freshwater discharge from the dyke. We examined the distribution of salinity and temperature by the freshwater discharge in the Yeongsan River estuary using the CTD data obtained from 8 stations through three surveys in June (weak discharge) and August (intensive discharge), 2010. During the weak discharge in June, the surface salinity showed 30-32.5 psu and its horizontal gradient was relatively high around Goha-do (0.25~0.32 psu/km). On the other hand, the salinity of the bottom layer was almost constant in the range of 33 psu. Water temperature ranged $19{\sim}21^{\circ}C$ and displayed higher gradient in north-south direction than the gradient of east-west direction. During the intensive freshwater discharge on August 12, the salinity dropped to 9~26 psu. The maximum horizontal gradient of surface salinity reached 3.8 psu/km in the north of Goha-do where the strong salinity front was formed, and the horizontal salinity gradient of bottom layer was 0.28 psu/km. The horizontal gradient of water temperature was $-0.45^{\circ}C/km$ in the surface and $-0.12^{\circ}C/km$ in the bottom with high surface temperature near the dyke and decreasing gradually to the river mouth. After 3 days of the intensive discharge ($3^{rd}$ survey), the surface salinity increased to 22~26 psu. However, there still existed relatively high horizontal gradient around Goha-do. In the mean time, the bottom salinity decreased to 26.5~27.5 psu, but its gradient was not big as much as the surface gradient. According to time series of CTD profile near the dyke, the discharged fresh water jetted down temporarily and then recovered gradually with the recovering speed of 0.4 m/hour for the discharge case of $13{\times}10^6$ ton. Due to the combined effects of freshwater discharge and surface heating during the summer of 2010, the Yeongsan estuary, in general, underwent intensified vertical stratification, which in turn caused the inhibition of vertical mixing, especially inside area of estuary. Based on the spatial distribution of salinity and temperature, the Yeongsan estuary can be divided into three regions: the Goha-do area with strong horizontal gradient of salinity and temperature, inner estuary from Goha-do to the dyke with low salinity, and outer estuary from Goha-do to the coasts with relatively high salinity.

Characteristics and Activity Changes of Polygalacturonase during Maturation and Postharvest of Persimmon Fruits (감과실의 성숙과 추숙중의 Polygalacturonase활성 변화 및 특성)

  • Polygalacturonase activity was not detected at turning stage but were 55.01 and 206.70 units/100g -fr. wt. in mature and soft persimmon, respectively. Polygalacturonase have two isoenzymes and its molecular weight was estimated to be 55,000 doltons by the method of gel filtration. Vmax and Km of polygalacturonase 1 were 0.195$\mu$ mole reducing-sugar/$m\ell$/30 min. and 3.50mg/$m\ell$, respectively. The optimum temperature and pH of the enzyme appeared to 4$0^{\circ}C$ and 3.5, respectively. Polygalacturonase I had Vmax of 0.110m mo1e reducing-sugar/$m\ell$/30min. and Km value of 2.50mg/$m\ell$, The optimum temperature and pH of polygalcturonase II appear 4$0^{\circ}C$ and 4.0, respectively. Polygalacturonase I was fairly stable at 6$0^{\circ}C$while polygalacturonase II appeared to be stable up to 4$0^{\circ}C$.

The Concentration and Input/Output of Nitrogen and Phosphorus in Paddy Fields (논에서의 질소 및 인의 농도와 유출입)

  • For the purpose of evaluating nutrient loadings into rivers and lakes from agricultural land, especially from paddy fields and also nutrient degradation in drainage channels, the Total Kjeldahl Nitrogen(TKN) and the Total Phosphorus(TP) were investigated in 29.5 ha. paddy fields in Hwa-Sung, Kyong-Ki, Korea, during the period from May 8, 1989 to Sep. 27, 1989. The results of the study can be su㎜arized as follows : 1. Annual inputs into paddy fields were 180 N-kg/ha 46 P-kg/ha. by fertilization, and 15.0 TKN-kg/ha. 10.0 TP-kg/ha. by irrigation, 8.0 TKN-kg/ha. 0.34 TP-kg/ha. by rainfall respectively. The amount of nutrient involved in surface runoff from paddies was 39.0 TKN-kg/ha. 9.2 TP-kg/ha. and in seepage 7.5 TKN-kg/ha. 2.1 TP-kg/ha. respectively 2. In WS1 stream(reach length equals 950m), nutrients decreased 0.31 TKN-mg/L/km, 0.01 TP-mg/L/km and in WS2 stream (reach length equals 750m) which are more meandering and undulating than WS1, the nutrients decreased 0.84 TKN-mg/L/km, 0.11 TP-mg/L/km. From these results, it was concluded that low stream velocity due to meandering and undulation promotes more degradation of nutrient concentrations. 3. For the purpose of decreasing nutrient loads from paddy fields, the amount of fertilizer used needs to be controlled, irrigation weirs need to be constructed in the drainage channels to delay the transportation of nutrients by decelerating the stream velocity and plants such as plantain-lily need to be cultivated in the channel to consume nutrients and therefore enlarge chances of self-purification.

Estimation of Energy Expenditure of Walking and Running Based on Triaxial Accelerometer and Physical Information (3축 가속도계와 신체정보를 이용한 보행 및 주행시 에너지 소비량의 예측)

  • The purpose of this study was to estimate the energy expenditure simply and practically during physical activities. The physical activity is quantified by the integration of the accelerometer signals obtained from the triaxial accelerometer attached at the waist level of the human body. To find a relationship between energy expenditure and accelerometer data, 6 male and 5 female subjects walked and ran on the treadmill with speeds of 1.5, 3.0, 4.5, 6.0, 6.5, 7.0, and 8.5 km/hr. Each subject performed walking at the speed lower than 6.0 km/hr and running at the speed higher than 6.5 km/hr. Actual energy expenditure was determined by a continuous direct gas analyzer. Two predictive equations of walking and running mode for energy expenditure which includes gender, body mass index(BMI) and data from accelerometer were developed using multiple regression analysis. The correlation coefficients and coefficients of determination between the estimated and measured energy expenditure were R=0.936, R2=0.876 and R=0.881, R2=0.776 in walking and running mode, respectively. For further study, experiments on a larger scale of test subjects are essential for acquiring more reliable results.

Estimation of Oceanic Total Precipitable Water from HALE UAV (고고도 장기체공무인기 운영고도에서 해양 총가강수량 추정)

  • In this study, the oceanic Total Precipitable Water (TPW) retrieval algorithm at 16 km altitude of High Altitude Long Endurance Unmanned Aerial Vehicle (HALE UAV) is described. Empirical equation based on Wentz method (1995) that uses the 18.7 and 22.235 GHz channels is developed using the simulated brightness temperature and SeeBor training dataset. To do radiative simulation, Satellite Data Simulator Unit (SDSU) Radiative Transfer Model (RTM) is used. The data of 60% (523) and 40% (349) in the SeeBor training dataset are used to develop and validate the TPW retrieval algorithm, respectively. The range of coefficients for the TPW retrieval at the altitude of 3~18 km with 3 km interval were 153.69~199.87 (${\alpha}$), 54.330~58.468 (${\beta}$), and 84.519~93.484 (${\gamma}$). The bias and RMSE at each altitude were found to be about $-0.81kg\;m^{-2}$ and $2.17kg\;m^{-2}$, respectively. Correlation coefficients were more than 0.9. Radiosonde observation has been generally operated over land. To validate the accuracy of the oceanic TPW retrieval algorithm, observation data from the Korea Meteorological Administration (KMA) Gisang 1 research vessel about six clear sky cases representing spring, autumn, and summer season is used. Difference between retrieved and observed TPW at 16 km altitude were in the range of $0.53{\sim}1.87kg\;m^{-2}$, which is reasonable for most applications. Difference in TPW between retrieval and observation at each altitude (3~15 km) is also presented. Differences of TPW at altitudes more than 6 km were $0.3{\sim}1.9kg\;m^{-2}$. Retrieved TPW at 3 km altitude was smaller than upper level with a difference of $-0.25{\sim}0.75kg\;m^{-2}$ compared to the observed TPW.

Assessing the Impact of Sampling Intensity on Land Use and Land Cover Estimation Using High-Resolution Aerial Images and Deep Learning Algorithms (고해상도 항공 영상과 딥러닝 알고리즘을 이용한 표본강도에 따른 토지이용 및 토지피복 면적 추정)

  • This research assessed the feasibility of using high-resolution aerial images and deep learning algorithms for estimating the land-use and land-cover areas at the Approach 3 level, as outlined by the Intergovernmental Panel on Climate Change. The results from different sampling densities of high-resolution (51 cm) aerial images were compared with the land-cover map, provided by the Ministry of Environment, and analyzed to estimate the accuracy of the land-use and land-cover areas. Transfer learning was applied to the VGG16 architecture for the deep learning model, and sampling densities of 4 × 4 km, 2 × 4 km, 2 × 2 km, 1 × 2 km, 1 × 1 km, 500 × 500 m, and 250 × 250 m were used for estimating and evaluating the areas. The overall accuracy and kappa coefficient of the deep learning model were 91.1% and 88.8%, respectively. The F-scores, except for the pasture category, were >90% for all categories, indicating superior accuracy of the model. Chi-square tests of the sampling densities showed no significant difference in the area ratios of the land-cover map provided by the Ministry of Environment among all sampling densities except for 4 × 4 km at a significance level of p = 0.1. As the sampling density increased, the standard error and relative efficiency decreased. The relative standard error decreased to ≤15% for all land-cover categories at 1 × 1 km sampling density. These results indicated that a sampling density more detailed than 1 x 1 km is appropriate for estimating land-cover area at the local level.

A Study on Hydrologic and Geographic-Characteristics and Design-Width-Determination of Small Rivers in Korea (우리나라 소하천의 수문지형학적 특성 및 계획하폭 결정에 관한 연구)

  • To define a small river in Korea. hydrologic- and geographic-characteristics. such as watershed area, channel slope, channel length and time of concentration are investigated and their limits are presented. The current equations of design-width-determination of the small rivers are examined and two equations for the design-width-determination of the small rivers have been suggested in this paper. A small river in Korea can be defined that watershed area is less than $10km^2$, channel length less than 6 km, channel slope over 0.02. time of concentration less than 30 minutes. As Band Q are defined as a design width and a design flood, respectively, the suggested equation to determine the design width is $B=1.235Q^{064}$ for less than $300m^3/sec$ of design flood. and when B and A are defined as a design width and a watershed area, respectively. the recommended equation is $B=8.794A^{0.56}$ for less than $10km^2$ of watershed area.

Perception on the Traditional Korean Medicine According to the Existence of a Chronic Disease

  • Objectives: Chronic diseases (CDs) continue to increase due to advances in medicine and increase in lifespan, affecting quality of life and resulting in economic loss through treatment costs. This study addressed the perception characteristics of the effectiveness of Korean medicine (KM) based on the existence of a CD to determine the national preference for KM treatment. Methods: Using data from the 2017 National Survey of KM Usage, we evaluated the perception on treatment effect of 16 diseases by dividing them into the CD group and the non-CD group. Response reliability was verified by applying the chi-square test (χ2-test) analysis method of the Statistical Package for the Social Sciences (SPSS) statistical program. Results: The analysis of the perception on effectiveness of KM for patients with CD (n = 1,050, 21.0%) and for patients with non-CD (n = 3,950, 79.0%) showed an overall similar trend for all the 16 diseases. The response rates of having some treatment effect were high for nine diseases namely, disc-related disease (CD: 70.7%, non-CD: 73.1%), osteoarthritis (CD: 72.3%, non-CD: 72.4%), frozen shoulder and shoulder pain (CD: 79.6%, non-CD: 81.4%), back pain (CD: 84.6%, non-CD: 85.0%), sprain (CD: 84.8%, non-CD: 84.1%), facial nerve paralysis (CD: 73.5%, non-CD: 71.7%), stroke (CD: 66.2%, non-CD: 62.8%), digestive disease (CD: 53.3%, non-CD: 50.0%), and common cold and rhinitis (CD: 44.7%, non-CD: 44.8%). Conclusion: The present results found that there was little difference in the perception on effectiveness of KM for each of the 16 diseases in patients with CD and non-CD, however, 70% or more of the respondents recognized some treatment effect on musculoskeletal disorders, regardless of the existence of CD. Preferential political support for KM treatment of chronic musculoskeletal disorders is recommended.
