Supplementary Effect by Harvest period of Lentinus edodes on the levels of Blood glucose and Serum lipid in Diabetic KK mice (채취 시기가 다른 표고버섯의 급여가 KK 당뇨 마우스의 혈당과 혈청 지질 수준에 미치는 영향)

  • This study was carried out to investigate the effect of Lentinus edodes which were harvested at different times of maturity on blood glucose and lipid levels in diabetic mice. The diabetic KK mice were fed diets supplemented with Lentinus edodes harvested early (LE) or late (LL) for eight weeks, and control (CO) mice were fed regular diet without Lentinus edodes. Diet intake, body weight, organ weight, blood glucose and serum lipid levels were measured. The mice which were fed diets supplemented with LE and LL showed significantly lower body weights compared to the CO group. There were no significant differences in the diet intake and the weights of different organs including liver, heart, kidneys, and epididymal fat pad among three different groups. Furthermore, the concentrations of serum triglycerides and total cholesterol were significantly lower in LE and LL groups compared to the CO group. These results clearly demonstrate that Lentinus edodes, regardless of their harvested period reduced body weight and serum lipid without affecting blood glucose level in diabetic mice.

Glucose-lowering Effect of Powder Formulation of African Black Tea Extract in $KK-A^y/TaJcl$ Diabetic Mouse

  • We observed the suppressive effect of a powder formulation of African black tea extract prepared from the leaves of Camellia sinensis on type 2 non-insulin dependent diabetic mice, $KK-A^y/TaJcl$. Black tea extract significantly showed suppressive effect of the elevation of blood glucose on oral glucose tolerance test of 8 week-old $KK-A^y/TaJcl$ mice (p<0.05). Long-term treatment with black tea extract showed significant suppression of post-prandial blood glucose and obesity (p<0.05). The weight of the intestine of mice treated with black tea extract was significantly reduced (p<0.05). From these results, African black tea used in this study showed a suppressive effect on the elevation of blood glucose during food intake and the body weight.

  • This study was carried out to investigate the supplementary effects of the rice germ oil compared with soy bean oil on blood glucose level of non-insulin dependent diabetic mice. Forty diabetic KK mice were fed two kinds of experimental diets with 20% lipid from soy bean oil as a control(CO) and rice germ oil(RG) for 8 weeks, respectively. Diet intake, body weight, organs weights and lipids levels of serum, liver and feces were measured. There was no significant difference in food and water intake, body weight gain and organs weights between experimental groups. The concentrations of fasting and random blood glucose were similar between CO and RG groups. There was no significant difference in blood glucose levels after glucose treatment during the glucose tolerance test between two groups. The levels of $HbA_{1c}$ as the index of blood glucose status, and insulin were similar in two groups. These results suggested that rice germ oil can't reduce blood glucose concentration of non-insulin dependent diabetic mice compared with soybean oil. But we need to investigate the hypoglycemic effect of rice germ oil by changing supplementary level and period.

The hepatic total lipid and total cholesterol levels of RG group were significantly lower than those of CO group. The contents of total lipid, triglyceride and total cholesterol excreted in feces of RG group were higher than those of CO group. These results suggested that rice germ oil can reduce serum total cholesterol and LDL-cholesterol and hepatic total lipid concentration of non-insulin dependent diabetic mice compared with soybean oil due to increasing fecal lipid excretion. But we need to investigate the hypolipidemic effect of this oil by supplementary level and period.

  • This study was carried out to investigate the supplementary effects of ${\gamma}$-oryzanol on the blood glucose level in diabetic mice. We supplied 2 experimental diets (CO without and GO with 0.2% ${\gamma}$-oryzanol) to diabetic mice for 8 weeks. Diet intake, body weight and levels of blood glucose, hemoglobin $A_{lc}$ and insulin were measured. Though there was no significant difference in diet intake between experimental groups, the concentration of fasting blood glucose and blood glucose area from glucose tolerance test in diabetic mice was lower in GO group than CO group during the supplementary period of experimental diets. Hemoglobin Ai, was lower and serum insulin level was higher in GO group than CO group without significance. These results suggest that r-oryzanol decrease the blood glucose level, and ${\gamma}$ -oryzanol produced from residual product of rice may be developed with high value.

  • The anti-diabetic effect of green tea fermented by cheonggukjang was evaluated using KK-$A^y$ mice, an animal model of type 2 diabetes mellitus. Over a 90 day testing period, food and water intake decreased significantly in the group fed fermented green tea (FGT) and a group fed commercially available health functional food (PC), when compared with a diabetic control group (DC). The blood glucose levels of FGT mice were lower than in DC mice throughout the test period and were similar to the levels in PC after 60 days. Levels of Hemoglobin A1c (HbA1c) levels and insulin resistance were lower in mice of the FGT group than in mice of the DC group. DNA microarray analysis showed that administration of FGT increased the abundance of 12 mRNA transcripts related to diabetes. Whereas FGT increased hexokinase transcripts related to glycolysis more than 37 fold, levels of Pdx1 (pancreatic and duodenal homeobox1) and Cacna1e (calcium channel) transcripts increased more than 1.8 fold.

  • An experimental study was undertaken to observe the difference in susceptibility of mouse strains to Metagonimus yokogawai infection by estimating it from worm recovery rate and dimension of worms. It was also studied the effects of prednisolone injection on the chronological pattern of worm recovery in ICR mice. The metacercariae were obtained from sweetfish and 300 in each number were given to 5 strains (CBH, A, DBA, $C^{57}BL$ and KK) of mice, and after 7 days period, the worms were collected from their intestine. Prednisolone at the dose of 10 mg/kg was injected to ICR mice every other day from 7 days prior to infection until sacrificed at 6 hours to 35th post-infection day. ICR mice infected with M. yokogawai but untreated were used for controls. The success rate in infection of mice ranged 25.0-83. 3% by strains, the worm recovery rate 1. 2-18. 9%, and the average size of worms O. 554-0. 683 mm long and 0.214-0.244 mm wide. The higher rates and larger size of worms were observed in KK and $C^{57}BL$ strains than others and the difference was statistically significant. In ICR mice for control, the worm recovery rate until 1 day after infection was relatively high (38-66%) but it became much lower (less than 0.7%) during 1-35 days. However, prednisolone injection brought about persistently high recovery rates (16-80%) until 21 days. It was concluded that the susceptibility to M. yokogawai infection is different by strains of mice but it can be elevated by prednisolone injection probably due to suppression of Immune respon3e3 in ICR mice.

Supplementary Effect of the High Dietary Fiber Rice on Blood Glucose in Diabetic KK Mice (고식이섬유쌀의 급여가 KK 당뇨 마우스의 혈당에 미치는 영향)

  • This study was carried out to investigate the supplementary effects of Suwon 464, which has over two times of dietary fiber content compared with normal rice (Ilpum), on blood glucose in diabetic mice. We supplied 5 kinds of experimental diets (corn starch diet as a control (CO), Ilpum polished rice diet (IP), Ilpum brown rice diet (IB), polished rice diet (SP) and brown rice diet (SB) of Suwon 464) to diabetic mice for 8 weeks, after analyzing dietary fiber contents of 5 experimental diets. Diet intake, body weight and contents of blood glucose, hemoglobin $A_{lc}$ and insulin were measured. The dietary fiber contents in CO, IP, IB, SP, and SB diets were 1.0, 1.2, l.4, l.4, and 2.0% respectively. Body weight was lower in SB group than the other groups though there was no significant difference in diet intake among experimental groups. The concentration of blood glucose in diabetic mice was lower in SB group than the other groups during the supplementary period of experimental diets. The hemoglobin Ale and serum insulin levels were lower in SP and SB groups. These results suggested that the brown rice of Suwon 464 with high dietary fiber can control diabetes in diabetic mice by reducing the blood glucose and hemoglobin Ale. Studies on genetic monitoring of inbred mice in conventional breeding unit (일반사육시설 마우스의 유전적 오염에 대한 실태조사 연구)

  • These studies were carried out to survey the genetic contamination of six inbred mice (A, BALB/c, C3H, C57BL/6, CBA and KK) produced and supplied from the conventional breeding unit for improving the quality of mice as experimental animal. We examined alleles of five loci (Akp-1, Car-2, Hbb, Es-1 and Trf) by the use of biochemical markers with celluose acetate electrophoresis. As the results of test, BALB/c, A, C3H, C57BL/6, CBA and KK showed standard alleles in Akp-1, Car-2 and Hbb. But Es-1 of A and C57BL/6 and Trf of A, C3H, C57BL/6 and CBA did allelic divergence in loci. These results suggest that the colonies of A, C3H, C57BL/6 and CBA were genetically contaminated. Therefore, we recommend to eliminate the genetically contaminated mice thoroughly, to check on genetic monitoring regularly and to consider a counterpaln for improving the quality control as soon as possible.

  • 1. In the present experiment, necrotic myocarditis were produced only in suckling mice but also in weanlings or young adult mice by intraperitoneal inoculation with the Tyzzer's organism KK of cat origin, whereas the organism MSK from the mouse was not capable of producing such lesions. 2. Significant difference in pathogenicity for 1-to 6-week-old mice was demonstrable between the two organisms after intraperitoneal inoculation without cortisone.
