Design and Implementation of OASIS Considering Web Accessibility (웹 접근성을 고려한 전통의학정보포털 설계 및 구현)

  • This study shows evaluation of how much OASIS meets "the korean web content accessibility guidelines" and analysis of some of the accessibility problems and their solutions in OASIS(Oriental Medicine Advanced Searching Integrated System) which is the only web site that offers papers and project information related to Traditional Medicine in Korea. The evaluation criteria to determine if OASIS is accessible is classified into four sub items; Perceivable - if information and user interface components is presentable to users in ways they can perceive, Operable - if user interface components and navigation are operable, Understandable - if information and the operation of user interface are understandable, Robust - if content is robust enough that it can be interpreted reliably by a wide variety of user agents, including assistive technologies. Based on the measured results, OASIS has just been redesigned and implemented in more accessible and effective way. OASIS that improves web accessibility for the disabled is expected to help them study oriental medicine more easily and conveniently by providing equal access and equal opportunity to use the web.

  • It has been known many pretreatment methods of Pinellia ternata. In this study, we chose one of them, ${\lceil}$Gang-Banha${\rfloor}$ and compared the process step by step in terms of taste an icon of Pinellia ternata. Identification of the major components and understanding the mechanism of pretreatments should be studied near future.

Age-related Changes in blood Chemistry and Thyroid hormones in Senescence Accelerated Mice(SAM R1 and SAM P6) (노화 방지를 위한 한약재의 효능 연구 (I))

  • Aging process can be explained by many factors. In this study, we counted complete Blood Cells (CBC) such as WBC, Lymphocytes, Monocytes, Granulocytes, RBC, HGB, and HCT of both SAM P6 and SAM P1 during the aging process. Plasma albumin, glucose, alkaline phosphatase, calcium, creatinine, inorganic phosphate, urea concentrations were also measured at the same time. In addition to these, plasma concentrations of cortisol, total T3, and total T4 were analyzed by Chemiluminescent Immunoassay. There were no change in CBC counts of SAM R1 and SAM P6 during this study. Plasma concentratins of albumin and glucose decreased significantly in SAM R1. However, plasma alkaline phosphatases and creatinine concentration in SAM P6 decreased significantly at 16 week after birth comparing to the control. Total T4 levels were siginificantly increased although cortisol and total T3 concentrations were the same in SAM R1 groups. Especially, the after birth of creatinine, alkaline phosphatase, T4 of SAM P6 at 16 week were significantly different form those of SAM R1. At 12 week after birth, pilose antler extract was given 5g/kg/day p.o. for 0, 7, 14, 21, and 30 days each in both SAM R1 and SAM P6. The RBC, HGB, and HCT levels started to increase significantly from 7 days after the dose at SAM P6 only. Total T4 conectrations were elevated gradually during the study although the antler extract administration did not prevent or inhibit the increase in total T4 concentration during the study. Therefore, the elevation of erythrocytes after administration of the extract needs to be studied in future.

Screening of Korean Herbal Medicines with Inhibitory Activity on Advanced Glycation End Products (AGEs) Formation (III) (한국약용식물의 최종당화산물 생성저해활성 검색 (III))

  • Enhanced formation and accumulation of advanced glycation end products (AGEs) have been implicated as a major pathogenesis process leading to diabetic complications, normal aging, atherosclerosis and Alzheimer's disease. In our ongoing project to discover novel treatments for diabetic complications from natural sources, we have investigated on the inhibitory activity of 67 ethanol extracts from 57 Korean herbal medicines against the formation of AGEs in vitro. Of these, 22 extracts were found to have a significant AGEs inhibitory activity ($IC_{50}$<50 ${\mu}g$/ml) compared with aminoguanidine ($IC_{50}$=75.98 ${\mu}g$/ml). Particularly, 6 extracts from 3 herbal medicines, Castanea crenata (flower, leaf, bark-twig), Acer tatarium subsp. ginnala (fruit) and Sapium japonicum (leaf, twig) showed (approximately 8-17 fold) stronger inhibitory activity than that of aminoguanidine.

Development of Pulse Analysis System Based on Convenience and Compactness (사용 편의성과 소형화 기반의 맥파 측정장치 개발)

  • To improve and downsize the KIOM-Pulse Analysis System(KIOM-PAS), which has been traditionally used in clinical trials, a miniaturized PAS(M-PAS) with a pneumatic pump has been developed. M-PAS is composed of a measurement module and an arm-mounting module, on which an arm can be placed. The measurement module is equipped with a pressing component and sensor, which is a wearable wristband. The arm-mounting module includes a pneumatic motor, data acquisition board and valves. In addition, the measurement module is divided into a fixing module of band type for attachment to a pulsation site and a sensing module, which includes a sensor and a tube. The fixing module and sensing module are constructed independently, and the detachable fastening method improves the posture convenience of the subject during measurement. M-PAS has been reduced to 1/6 the size of KIOM-PAS, and the measurement time is shortened by 22%. Using a simulator, the difference between the waveforms measured by the two devices exhibited a high degree of similarity of within 3.65%. M-PAS represents improvements in size and convenience compared with KIOM-PAS, and it is expected to be widely used in clinics in the future because it improves the attachment method of the fixing module.

A Study on the Reliability Assessment of Sasang Constitution Questionnaire Developed by KIOM for Japanese (일본인 대상자를 통해 살펴본 KIOM 체질 설문지의 신뢰도 평가)

  • 1. Objectives : This study aimed to assessing the reliability of questionnaire for Japanese as one of method to diagnose Sasang Constitution and evaluating the possibility of applying domestic questionnaire to foreign site. 2. Methods : A cross-sectional survey approved by IRB (Independent Review Board) at Tohoku University was performed for 119 participants with informed consent at a particular city in Japan using self-reported questionnaires developed by KIOM (Korea Institute of Oriental Medicine). Among the entire samples, 112 were retested. The test-retest reliability was assessed by Kappa for normal variables and internal consistency was evaluated by Cronbach's alpha. 3. Results : Four of 77 questions are impossible to calculate consistency owing to unsymmetric matrix between primary reply and secondary one. Seventeen of them (22%) showed 0.4 or below in Kappa, 56 were ranged from 0.4 to 0.8 and no question marked 0.8 or over. Internal consistency was possibly checked for several questions about character, digestion, sweat, excrement, urine, cold-heat pattern. Cronbach's alpha for questions except urine and cold-heat pattern was over 0.6 4. Conclusions : On the basis of Kappa value by two self reported questionnaire, 72.7% of questions showed 0.4 or over so that reliability were highly secured. And other questions except for urine and heat-cold pattern marked 0.6 or over in order that internal consistency was also successfully maintained. Accordingly, when subsequently applying KIOM questionnaire to foreign site, domestic questionnaire should be properly adjusted in social and cultural background, lifestyle, etc.

The studies about cutting, processing and decokting methods of oriental medicinal plants II : Rehmanniae Radix Preparata (한약재의 절단, 수치, 전탕법에 관한 연구II : 숙지황)

  • To improve the quality control of frequently used oriental medicinal plants, the storage, cutting, processing and decokting methods of Rehmannia glutinosa Libosch. var. purpurea Makino(熟地黃) were examined. The contents of 5-hydroxymethyl-2- furaldehyde(5-HMF) of Rehmannia glutinosa Libosch. var. purpurea Makino were analysed by HPLC at various conditions as the standard component. Raw materials were classified into 3 groups with their specific gravity(天 地 人黃) and determined the ding weights and the contents of water, the solid component of Ji-Hwang(地黃) was the most. Rehmannia glutinosa Libosch. var purpurea Makino was steamed and dried nine times and analysed the content of 5-HMF on every time. In the case of Ji-Hwang, the standard component of the ninth sample was produced the most. But which of the tenth samples of Chun-Hwang and Yin-Hwang(天黃 人黃) were the most. The best decokting conditions which were determined by the yield of 5-HMF were as follows : non cutting, soaking time was 0 minutes, decokting time was 75 minutes after boiling. The contents of sugar on each of samples were determined. The contents of standard component increased by steaming number and the contents of sugar decreased.

Assessment of anatomical characteristics of the medicinal plant African cherry (Prunus africana) for its accurate taxonomic identification

  • The genus Prunus (family: Rosaceae) consists of over 400 plant species and exhibits vast biodiversity worldwide. Given the wide distribution of this genus, its taxonomic classification is important. Anatomical characteristics are conserved and stable and can therefore be used as an important tool for the taxonomic characterization of plants. Therefore, this study aimed to assess and document the anatomical characteristics of the leaf, stem, and seed of P. africana using micrographs and photographs for possible use in the identification, quality control, and phylogenetic analysis of the species. The anatomical sections of a young stem revealed a cortex consisting of isodiametric parenchyma cells, druse crystals, primary vascular bundles, and pith. The mature stem bark majorly consisted of the rhytidome, with the periderm densely arranged in multiple layers; a cluster of stone cells; and sclerenchyma. The leaf sections were hypostomatic, with stomata sizes ranging from 18.90-(22.34)-26.90 × 15.41-(18.40)-21.22 ㎛. The leaf sections showed the presence of characteristic druse crystals, vascular bundles, and mesophyll layers. The pericarp contained the epicarp, mesocarp, and endocarp, with their thickness being approximately 350-400, 300-350, and 30-50 ㎛, respectively. In addition, it contained a seed testa with a thickness of approximately 50-60 ㎛. The morphological and anatomical characteristics observed in P. africana leaves, stems, and seeds in this study could serve as useful data for the taxonomic identification of this species.

Effects of Boehmeria platanifolia Extract on Muscle Amelioration in Dexamethasone-Induced Muscle Atrophy Mouse Model (개모시풀추출물의 Dexamethasone 유도 근위축 마우스 모델에서 근개선 효과 연구)

  • Objective : This study was conducted to investigate the muscle-improving and therapeutic effects of Boehmeria platanifolia (BP) in a mouse model of dexamethasone-induced muscle atrophy. Methods : Muscle atrophy was induced in C57BL/6 mice by intraperitoneal administration of dexamethasone for 12 days. BP extract was administered orally at doses of 100 mg/kg and 200 mg/kg for 19 days, starting 7 days before the intraperitoneal administration of dexamethasone. Mice were weighed during the experimental period, and muscle strength and muscle weight were measured at the end of the experiment. The gastrocnemius (GASTROC) muscles of mice were isolated and the cross-sectional area (CSA) of the muscle fibers was measured after H&E staining. Results : Dexamethasone-induced muscle atrophy mice had a decrease in body weight compared to normal mice, and BP-administrated mice did not show significant change in body weight compared with a control group. Muscle strength in mice with induced muscle atrophy was reduced compared to normal and significantly increased with BP administration and positive control. In addition, the weight of the quadriceps (QUAD) muscle and fiber size of the GASTROC muscle, which was reduced in sarcopenia-induced mice, was increased by BP. Conclusion : BP extract increased muscle strength, muscle weight, and muscle fiber size in dexamethasone-induced muscle atrophy mice. This suggests that the efficacy of BP extracts in improving muscle strength and preventing and treating sarcopenia may be beneficial for the development of potential therapeutic or functional products.

Studies on Gene Expression of baicalin treated in HL-60 cell line using High-throughput Gene Expression Analysis Techniques (Baicalin을 처리한 HL-60 백혈병 세포주에서 대규모 유전자 분석 발현 연구)

  • Baicalin, a biologically active flavonoid form the roots of Scutallaria baicalensis (Skullcap), have been reported to not only function as anti-oxidants but also cause anticancer effect. We investigated the mechanism of baicalin-induced cytotoxicity and the macro scale gene expression analysis in leukemia cell line, HL-60 cells. Baicalin (10 μM) were used to treat the cells for 6h, 12h, 24h, 48h and 72h. In a human cDNAchip study of 65,000 genes evaluated 6, 12, 24, 48. 72 hours after treated with Baicalin in HL-60 cells. Hierarchical cluster against the genes which showed expression changes by more than two fold. One hundred one genes were grouped into 6 clusters according to their profile of expression by a hierarchical clustering algorithm. For genes differentially expressed in response to baicalin treatment, we tested functional classes based on Gene Ontology (GO) terms. This study provides the most comprehensive available survey of gene expression changes in response to baicalin treatment in HL-60 cell line.
