Cloning and Expression of an Acidophilic $\alpha$-Amylase Gene from Bacillus circulans in Escherichia coli (Bacillus circulans의 호산성 $\alpha$-amylase 유전자의 클로닝 및 발현)

  • A new gene encoding an acidophilic TEX>$\alpha$-amylase of Bacillus cil-culans KCTC3004 was cloned into Eschericlzia coli using pUC19 as a vector. The gene localized in the 5.8 kb PstI DNA fragment was expressed independently of its orientation in the cloning vector showing enzyme activity about 40 times greater than that produced by the original B, circulans The optimum pH and temperature of the cloned enzyme were pH 3.6 and 45^{\circ}C.$ respectively. The enzyme hydrolyzed starch to produce maltotriose and maltooligosaccharides. The SDS-PAGE and zymopram of the enzyme produced in E coli(p.4L850) indicated a molecular weight of 55,000.

  • The purpose of this study is to confirm the MKT(Mathematical Knowledge for Teaching) of the in-service mathematics teachers on the statistics(Representative value, Degree of scattering) through the comparative analysis between the sub-elements of the MKT. In addition, it is to examine the factors that cause the difference of the subjects' MKT. To accomplish this, by the subject of 12 secondary in-service mathematics teachers, in this study the test items of the MKT on the statistics were developed and data were collected and analyzed. As a result of the analysis of the MKT test sheet, the CCK(Common Content Knowledge) and SCK(Specialized Content Knowledge) of the mathematics teacher was confirmed as a high score, whereas the and KCS(Knowledge of Content and Students) and KCT(Knowldge of Curriculum and teaching) were confirmed as low scores. In addition, through these results, it was shown that the difference in MKT's elements the middle school and high school teachers obtain occurred slightly.

  • Field measurement units and a system were constructed and installed in a waste-dump slope at the Imgi mine to investigate and analyze the variations in the unsaturated characteristics of the soil. The field instrumentation system was composed of a data acquisition system (DAS), a solar system, and measuring sensors. The rainfall, matric suction, and volumetric water contents were continuously measured from the units in the instrumented site. The variations in matric suction and volumetric water content were primarily affected by the rainfall intensity. At the surface of the slope, the largest increase and decrease in the changes in matric suction and volumetric water content were observed during the wetting and drying processes, respectively. Also, the matric suction and volumetric water content were 5-35 kPa and 0.12-0.24, respectively. However, the ground water level was not suddenly increased just after rainfall but gradually increased after 2 or 3 days later.

  • In this study non-stoichiometric $(K_{0.5}Na_{0.5})_{0.97}(Nb_{0.96-x}Ta_xSb_{0.04})O_3$ ceramics were prepared by the conventional soild-state teaction method. The effect of Ta-substitution on the dielectric and piezoelectric properties were investigated. X-ray diffraction analysis of all the specimens less than x= 15 mol% indicated orthorhombic phase. Thereafter, the specimens showed orthorhombic phase near to pseudo-cubic. Sinterablity of all the specimens was improved due to secondary products such as KCT and KCN. The ceramics with x= 5 mol% showed the optimum velues of pizoelectric constant($d_{33}$)= 150 pC/N, electromechanical coupling factor (kp)= 0.45, electromechanical quality factor (Qm)= 418.9 and dielectric constant(${\varepsilon}_r$)= 567. Accordingly, These results indicate that the composition ceramics is a promising candidate for lead-free material.

Effects of a Herbal Medicine, Yukgunja-Tang Extract Granule, on Functional Dyspepsia Patients by Sasang Constitution: Placebo-Controlled, Double-Blind, Randomized Trial (기능성 소화불량에 대한 육군자탕 엑스과립의 사상체질별 치료효과: 위약 대조, 이중 맹검, 무작위 시험)

  • Objectives This study was aimed to compare effects of Yukgunja-tang (YGJT) extract granule on functional dyspepsia (FD) by sasang constitution. Methods A placebo-controlled, double-blind, randomized, two-center trial was performed. We obtained 3D face images of FD patients participating in this study using face-only scanner and classified them into two types as narrow face and wide face by 3D facial shape diagnosis system (3-FSDS). 96 subjects (48 subjects per type) were enrolled and were randomly allocated into treatment or control groups in a 2:1 ratio. YGJT extract granule or placebo were administered to each group during the 8 week treatment period. One of sasang constitutional specialist diagnosed their constitutions by referring to questionnaires and 3D face images. The primary outcome was total dyspepsia symptom scale (TDS scale), the secondary outcomes were single dyspepsia symptom scale (SDS scale), visual analog scale (VAS) and so on. This trial was registered with clinical research information service identifier: KCT0001920, 15 May, 2016. Results In the Taeeumin treatment group, the TDS scale was significantly decreased after 8 weeks compared to the control group (t=2.331, p=0.025) and the SDS scale also significantly decreased. (t=2.042, p=0.048). The specific effects of the YGJT extract granule without the placebo effect on the Taeeumin with functional dyspepsia were 30.61% in TDS scale and 28.33% in SDS scale. Conclusions The effects of Yukgunja-tang extract granule on functional dyspepsia was different according to Sasang constitution. So the constitution should be considered in randomized controlled trials using herbal extract granules.

Efficacy of non-surgical treatment accompanied by professional toothbrushing in the treatment of chronic periodontitis in patients with type 2 diabetes mellitus: a randomized controlled clinical trial

  • Purpose: The present study aimed to evaluate the clinical benefit of additional toothbrushing accompanying non-surgical periodontal treatment on oral and general health in patients with type 2 diabetes mellitus (T2DM). Methods: We conducted a doubled-blind randomized controlled trial in 60 T2DM patients between June 2013 and June 2014. The patients were randomly assigned to the scaling and root planing (SRP) group; the scaling and root planing with additional toothbrushing (SRPAT) group, in which additional toothbrushing was performed by toothpick methods; or the control group. Microbiological and oral examinations were performed for up to 12 weeks following treatment. Non-surgical treatment was conducted in the experimental groups. The SRP group received scaling and root planing and the SRPAT group received additional toothbrushing with the Watanabe method once a week from the first visit through the fifth visit. The primary outcomes were changes in haemoglobin A1c (or glycated haemoglobin; HbA1c) levels, serum endotoxin levels, and interleukin-1 beta levels. Periodontal health status was measured by periodontal pocket depth, the calculus index, and bleeding on probing (BOP). Results: Both the SRP and SRPAT groups showed improvements in periodontal health and HbA1c, but the SRPAT group showed significantly less BOP than the SRP group. Furthermore, only the SRPAT group showed a statistically significant decrease in serum endotoxin levels. Conclusions: Non-surgical periodontal treatment was effective in improving HbA1c and serum endotoxin levels in T2DM patients. Furthermore, non-surgical treatment with additional tooth brushing had a more favourable effect on gingival bleeding management. Study on the Distribution of Fluorides in Plants and the Estimation of Ambient Concentration of Hydrogen Fluoride Around the Area of the Accidental Release of Hydrogen Fluoride in Gumi (구미 불산 누출사고 지점 주변 식물의 불소화합물 농도 분포 및 공기 중 불화수소 농도 추정에 관한 연구)

  • Objectives: The goal of this study is to identify the distribution of the foliar fluorine content of vegetation surrounding the area where hydrofluoric acid was accidently released in Gumi, Gyeongsangbuk-do on September 27, 2012. In addition, it also aims to estimate the concentration of hydrogen fluoride in the air on the day of the accident. Methods: Samples of plant leaves were collected on October 7, 2012 within 1 km from the site where the accident occurred. These samples were analyzed for soluble fluorine ion with an ion selective electrode. The ambient concentration of hydrogen fluoride was calculated using the fluoride content in the plant via the dose-rate equation (${\Delta}F$=KCT). Results: The arithmetic and geometric means of the concentrations were 2158.2 and 1183.7mg F $kg^{-1}$ for leaves and, 2.4 and 1.1 ppm HF for the air, respectively. The highest concentration of hydrogen fluoride in the air was 14.7 ppm, which is higher than the maximum concentration reported by the government (1 ppm) and the exposure limit (ceiling, 3 ppm). The concentrations of both fluorine and hydrogen fluoride decreased with increasing distance from the accident site and showed a significant decrease outside of a 500m radius from the site (p <0.05). Conclusions: The area around the accident site was highly polluted with hydrogen fluoride according to the results of this study. Considering the persistency of hydrogen fluoride in the environment, long-term monitoring and environmental impact assessment should be pursued.

Fission Yeast-based Screening to Identify Putative HDAC Inhibitors Using a Telomeric Reporter Strain

  • Transcriptional silencing is regulated by promoter methylation and histone modifications such as methylation and acetylation. We constructed a Schizosaccaromyces pombe reporter strain, KCT120a, to identify modifiers of transcriptional silencing, by inserting the $ura4^+$ gene into a heterochromatic telomere region. Two compounds inhibited the activity of histone deacetylases, induced acetylation of histone H3 and caused apoptotic cell death in HeLa cells. Expression of gelsolin and $p21^{waf1/cip1}$ also increased, as it does in response to HDAC inhibitors such as TSA. Therefore, these compounds appear to be potent inhibitors of HDACs, and hence potential anti-cancer drugs. Our observations suggest that a yeast cell-based assay system for transcriptional silencing may be useful for identifying histone deacetylase inhibitors and other agents affecting chromatin remodeling.

Piezoelectric and Dielectric Properties of 0.97[(K0.5Na0.5)(Nb0.97Sb0.03)O3]-0.03[(Bi0.5K0.5)TiO3] Ceramics Modified with K5.4Cu1.3Ta10O29 (K5.4Cu1.3Ta10O29 첨가에 따른 0.97[(K0.5Na0.5)(Nb0.97Sb0.03)O3]-0.03[(Bi0.5K0.5)TiO3] 세라믹스의 압전 및 유전 특성)

  • In this study, piezoelectric and dielectric properties of Lead-free $0.97[(K_{0.5}Na_{0.5})(Nb_{0.97}Sb_{0.03})O_3]+0.03[(Bi_{0.5}K_{0.5})TiO_3]$ (abbreviated as 0.97NKNS-0.03BKT)ceramics synthesized by conventional solid-state reaction process were investigated as a function of $K_{5.4}Cu_{1.3}Ta_{10}O_{29}$ addition. The results indicated that the $K_{5.4}Cu_{1.3}Ta_{10}O_{29}$ addition significantly improved the sinterability, grain growth and piezoelctric properties of 0.97NKNS-0.03BKT ceramics. The optimum values as planar piezoelectric coupling coefficient ($k_p$= 0.355), piezoelectric constant ($d_{33}$= 207 pC/N) and mechanical quality factor ($Q_m$= 128) were obtained when 0.009KCT was added. The electromechanical coupling factor($k_p$) was slightly decreased according to the increasing temperature.

Harmonization of laboratory results by data adjustment in multicenter clinical trials

  • Background/Aims: In multicenter clinical trials, laboratory tests are performed in the laboratory of each center, mostly using different measuring methodologies. The purpose of this study was to evaluate coefficients of variation (CVs) of laboratory results produced by various measuring methods and to determine whether mathematical data adjustment could achieve harmonization between the methods. Methods: We chose 10 clinical laboratories, including Green Cross Laboratories (GC Labs), the central laboratory, for the measurement of total cholesterol, high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), serum triglycerides, creatinine, and glucose. The serum panels made with patient samples referred to GC Labs were sent to the other laboratories. Twenty serum samples for each analyte were prepared, sent frozen, and analyzed by each participating laboratory. Results: All methods used by participating laboratories for the six analytes had traceability by reference materials and methods. When the results from the nine laboratories were compared with those from GC Labs, the mean CVs for total cholesterol, HDL-C, LDL-C, and glucose analyzed using the same method were 1.7%, 3.7%, 4.3%, and 1.7%, respectively; and those for triglycerides and creatinine analyzed using two different methods were 4.5% and 4.48%, respectively. After adjusting data using Deming regression, the mean CV were 0.7%, 1.4%, 1.8%, 1.4%, 1.6%, and 0.8% for total cholesterol, HDL-C, LDL-C, triglyceride, creatinine, and glucose, respectively. Conclusions: We found that more comparable results can be produced by laboratory data harmonization using commutable samples. Therefore, harmonization efforts should be undertaken in multicenter trials for accurate data analysis (CRIS number; KCT0001235).
