Phylogenetic Diversity of Bacteria in an Earth-Cave in Guizhou Province, Southwest of China

  • The objective of this study was to analyze the phylogenetic composition of bacterial community in the soil of an earth-cave (Niu Cave) using a culture-independent molecular approach. 16S rRNA genes were amplified directly from soil DNA with universally conserved and Bacteria-specific rRNA gene primers and cloned. The clone library was screened by restriction fragment length polymorphism (RFLP), and representative rRNA gene sequences were determined. A total of 115 bacterial sequence types were found in 190 analyzed clones. Phylogenetic sequence analyses revealed novel 16S rRNA gene sequence types and a high diversity of putative bacterial community. Members of these bacteria included Proteobacteria (42.6%), Acidobacteria (18.6%), Planctomycetes (9.0 %), Chloroflexi (Green nonsulfur bacteria, 7.5%), Bacteroidetes (2.1%), Gemmatimonadetes (2.7%), Nitrospirae (8.0%), Actinobacteria (High G+C Gram-positive bacteria, 6.4%) and candidate divisions (including the OP3, GN08, and SBR1093, 3.2%). Thirty-five clones were affiliated with bacteria that were related to nitrogen, sulfur, iron or manganese cycles. The comparison of the present data with the data obtained previously from caves based on 16S rRNA gene analysis revealed similarities in the bacterial community components, especially in the high abundance of Proteobacteria and Acidobacteria. Furthermore, this study provided the novel evidence for presence of Gemmatimonadetes, Nitrosomonadales, Oceanospirillales, and Rubrobacterales in a karstic hypogean environment.

Effect of Cell-Wall Broken Spores of Ganoderma lucidum(Leyss. ex.Fr.) Karst on the Lipid Accumulation and Body Weight Reduction in C57BL/6J Mice fed High-Fat Diet (파벽(破壁) 영지포자가 고지방식이 섭취 마우스의 지방축적 및 체중감량에 미치는 영향)

  • This study was performed to investigate the effects of cell-wall broken spores of Ganoderma lucidum on the lipid accumulation and body weight reduction in C57BL/6J mice. Six-week-old C57BL/6J mice were randomly divided into 5 groups and assigned to one of these groups; normal chew diet(Nor) group, high-fat diet(HFD) group, HFD plus spores of Ganoderma lucidum 800 mg/kg/day (HFD + GS/B) group, HFD plus cell-wall broken spores of Ganoderma lucidum 400 mg/kg/day (HFD + BGS/A) group and finally HFD plus cell-wall broken spores of Ganoderma lucidum 800mg/kg/day (HFD + BGS/B). The experimental groups which were treated oral co-administration with cell-wall broken(or original) spores of Ganoderma lucidum and HFD significantly attenuated accumulative body weight gain, compared with HFD group. Administration of these experimental materials also resulted in significant reduction not only the serum levels of total cholesterol, homocysteine but also the lipid accumulation in liver tissue. But in the almost of results the cell-wall broken spores of Ganoderma lucidum were evaluated superior than the original one. These results indicate that cell-wall broken spores of Ganoderma lucidum may inhibit the lipid accumulation in blood as well as liver tissue. Therefore it may be a valuable candidate for the therapy preventing obese induced hyperlipidemic symptoms.

Antiviral Effect of Water Soluble Substance from Elfvingia applanata Alone and in Combinations with Interferons Against Vesicular Stomatitis Virus (New Jersey Serotype) (잔나비걸상 수용성물질의 Vesicular Stomatitis Virus(New Jersey Serotype)에 대한 항바이러스작용과 Interferon과의 병용효과)

  • In order to find less toxic antiviral agents from Basidiomycetes, EA, the water soluble substance, was prepared from the carpophores of Elfvingia applanata (Pers.) Karst. Antiviral activity of EA against vesicular stomatitis virus [New Jersey serotype, VSV(NJ)] was examined in Vero cells using plaque reduction assay in vitro. And the combined antiviral effects of EA with interferon (IFN) alpha and gamma were examined on the multiplication of VSV(NJ). EA caused a concentration-dependent reduction in the plaque formation of VSV(NJ) with 50% effective concentration $(EC_{50})$ of 2.10 mg/ml. The results of combination assay were evaluated by the combination index (CI) that was analysed by the multiple drug effect analysis. The combination of EA with IFN alpha showed more potent effect with CI values of $0.87{\sim}1.59$ for 50%, 70% and 90% effective levels than that with INF gamma with CI values of $1.05{\sim}2.03$.

  • Isozyme patterns of leucine aminopeptidase, esterase and protein from 16 isolates of Ganoderma lucidum were observed by electrophoresis for characterization of the isolates. Even in the same isolate, the patterns of the isozymes from mycelium and cap were different. The esterase patterns from the mycelium could differentiate the isolates. Of 16 isolates, four isolates showed identical patterns. It was assumed that these had the same genetic background.

  • More than one-hundred-twenty specimens of Basidiomycetes, Ascomycetes and slime molds were collected in Seoul National Polytechnic University Campus during the priod from June 1984 to November 1987 and examined for identity. They were classified into 25 families, 53 genera and 81 species. Among them, one genus and 2 species were confirmed as unrecorded taxa in Korea. The unrecorded genus was genus Hapalopilus and was named 'Band Dal Busut' in vernacular. The unrecorded 2 species were Hapalopilus rutilans and Hygrophorus nitratus. They were named 'No Lan Ban Dal Busut' and 'Jil San Beot Geot Busut,' respectively and their characteristics were here reported.

  • Transmissivity is often estimated from specific capacity data because of the expense of conducting standard aquifer test to obtain transmissivity and the relative availability of specific capacity data. Most often, analytic expression relating specific capacity to transmissivity derived by Theis (1963). Brown (1963). and Logan (1964) are used in this analysis. The analytic solution typically used to predict transmissivity from specific capacity in alluvial aquifers assuming influence radius and/or storage coefficient of the aquifers. But those do not agree well with the measured transmissivity in fractured rock aquifers and in heterogeneous aquifers. Razack-Huntely (199l). Huntely-Steffey (1992). and Mace (1997) proposed emphirical rotations between specific capacity and transmissivity in heterogeneous alluvial aquifers. fractured rock aquifers, and karst aquifers. This study focuses on comparison between transmissivity and specific capacity data in volcanic rock aquifers of Jeju Island. Emphirical relation between the log of transmissivity and the log of specific capacity suggests they no linearly related (correlation coefficient 0.951) and the width of $\pm$0.25 log cycles in transmissivity includes 96.6% of data.

Mode of Antiviral Activity of Water Soluble Components Isolated from Elfvingia applanata on Vesicular Stomatitis Virus

  • A preparation of water soluble components (EA) was made from carpophores of Elfvingia applanata (Pers.) Karst and its in vitro antiviral activity on vesicular stomatitis virus [(Indiana serotype, VSV(IND)] was investigated by plaque reduction assay. EA exhibited potent antiviral activity on VSV(IND) growth and negligible cytotoxicity on Vero cells, 50% effective concentration ($EC_{50}C$/) of 104$ug\textrm\/ml$ and 50% cytotoxic concentration ($CC_{50}C$) of 3,793$ug\textrm\/ml$, respectively. Selectivity index (Sl $CC_{50}C$/$EC_{50}C$) of EA on Vero cell and VSV(IND) was about 36.5. EA did not display either a direct virucidal effect on V5V(IND) or induction of antiviral substance by Vero cells upon its treatment. Thus, the mode of antiviral activity of EA was studied at steps of viral adsorption onto cell. When both EA and virus were added to cell monolayers, titer of cell-free virus in culture supernatant increased in ca. 30-40% compared with that of control group and titer of cell-associated virus was 60-100% higher than that of control group. These results suggested that antiviral activity of EA on VSV(IND) might be due to the hindrance of viral entry to cells at eITher endocytosis or loss of envelope.

Mortar Characterization using Electrical Resistivity Method (모르타르의 전기비저항 특성)

  • Cement based mortars are widely used to improve the soft ground of a dam site, highway construction, and karst voids. The mechanical properties of the mortar are well documented in literature, however very limited work is done on their physical properties such as electrical resistivity which is considered as one of the most important physical property known while improving the soft grounds. In this paper, electrical resistivity of the Portland cement mortars is examined by employing the Wenner technique. Cylindrical specimens with various water/cement ratios (w/c) ranges from 0.35, 0.45, 0.50 and 0.65 were cast and tested. The test results showed that the electrical resistivity of the mortar increases with increasing curing time and decreases with increasing water content and w/c. A reasonable, good relation was found between electrical resistivity and compressive strength of mortar.

  • To search germination inhibitors in higher plants, first of all, some experiments containing selection of test weed seeds and effects of solvents and surfactants for bioassay establishment were conducted. Then MeOH-extracts of 45 plants were assayed for germination inhibition activities against Echinochloa crus-galli (L.) Beauv, Digitaria sanguinalis (L.) Scop., Cyperus iria L., Portulaca oleracea L. and Oenothera lamarckiana Ser. seeds. Among them extracts from Rhathiolepis ovata Briat and Picea abies (L.) Karst showed strong inhibitory effect (60-90% inhibition) on the germination of tested weed seeds at 5000 ppm. On the other hand, the extract from Youngia sonchifolia Max stimulated the germination and growth of tested weeds.

The Geomorphic Development of River Terraces along the Middle and the Lower Parts of the Osip-cheon River in Samchok City (三陟 五十川 중.하류부의 河岸段丘 지형발달)

  • The Osip-cheon river flowing on the east side of the Taeback mts. has formed river terraces on the several heights along the middle- and downstream. The river terraces are classified into 5 climatic ones and 7 thalassostatic ones. The thalassostatic ones are found to the height of 145-l50m level at 20-30m intervals. These vertical distribution is caused by the continuous uplift and periodical rise and fall of the sea-level. The high higher surfaces among the thalassostatic ones are the highest among those of Korea. The chronologies of the terraces are correlated to the marine oxygen isotope stages : The thalassostatic terraces on the level of 40 m.a.s.l. are to the stage 7, 70 m.a.s.l.. to the stage 9, 90 m.a.s.l. to the stage 9, 110 m.a.s.1. to the stage 11 and those of 150 m.a.s.1. to the stage 15 among the Interglacial Ages. The landuses and geomorphic landscapes of the Samchok area are chracteristic, because the karst landforms, such as doline and uvala, are developed on the surfaces of the middle-, the higher- and the high higher surfaces of river terrace.
