Effect of Nitrogen Application Level and Regrowth Period on Composition of Fatty Acid in Tall fescue and Bermudagrass (질소시비 수준 및 재생기간이 톨페스큐와 버뮤다그라스의 지방산조성에 미치는 영향)

  • Two experiments were conducted to evaluate the effects of nitrogen (N) application level and regrowth period on the fatty acid (FA) concentration and forage production of tall fescue (Festuca arundinacea Schr.) and bermudagrass (Cynodon dactylon (L.) Pers.). N was applied at 0, 50 and 100 kg/ha, and swards were cut after regrowth periods (15d, 30d and 45d). Concentrations of Individual FA were determined by gas chromatography. FA composition of tall fescue contains a high proportion ($87{\sim}88%$) of total FA contents as palmitic (C16:0), linoleic (C18:2) and linolenic acid (C18:3). The FA composition was not affected by N application, but a longer regrowth period significantly (p<0.05) decreased the composition of linolenic acid (C18:3) and increased those of linoleic acid (C18:2). The FA composition of bermudagrass was not affected by N application, but a longer regrowth period significantly (p<0.001) decreased the composition of linoleic (C18:2) and linolenic acid (C18:3). Linoleic and linolenic acfd of tall fescue were higher than those of bermudagrass. Dry matter (DM) yield of bermudagrass was significantly (P<0.05) affected by N application level and regrowth period. DM yield was higher at high N application and was higher at longer regrowth period. The crude protein concentration was higher at high N application and was low at longer regrowth period. Application of nitrogen fertilizer Increases dry matter (DM) yield and crude protein (CP) content of bermudagrass. These studies demonstrate opportunities to affect the FA concentration and composition of FA in forage through management strategies, which could affect milk FA composition.

Improving the efficacy of Lespedeza cuneata ethanol extract on ultraviolet-induced photoaging (야관문 에탄올 추출물의 자외선 노출에 의한 피부 광노화 개선 효과)

  • This study evaluated the improving efficacy of Lespedeza cuneata ethanol extract on skin photoaging induced by ultraviolet (UV) irradiation. The total polyphenol and flavonoid contents of the extract were respectively $134.98{\pm}1.70$ and $16.20{\pm}0.05$ mg/g, respectively. The superoxide anion radical scavenging activity and electron-donating ability of the extract were shown to be dependent on concentration, and the antioxidant ability was shown to be more effective in superoxide anion radical scavenging activity than in electron-donating ability under the same concentration conditions. In the in vivo test conducted using hairless mouse with skin photoaging induced by UVB irradiation, the skin erythema of the groups treated with the extract (AS) reduced to 28% of the control, and the skin moisture content increased to 131%.. The extract treatment of the UV-damaged skin improved the morphological and histopathological state of the skin. Furthermore, the SOD, GST and CAT activities in the skin tissue of the AS group increased, and the XO activity and TBARS generation decreased. With regard to the genes related to the photoaging skin, the expression of PAK, p38, c-Fos, c-Jun, TNF-${\alpha}$ and MMP-3 in the skin of the AS group were found to have decreased. It was therefore concluded that Lespedeza cuneata ethanol extract can reduce wrinkle formation in the skin due to the regulation of the gene expression caused by the exposure to UVB light.

  • The present study aimed to examine the hepatoprotective effects of ethanol extracts from steam-dried ginseng berry (SGBE) in both $\text\tiny{D}$-Galactosamine/Lipopolysaccharide ($\text\tiny{D}$-GalN/LPS)-sensitized mice and in vitro models. Our results clearly demonstrated that SGBE significantly reduced the level of aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase, and lactate dehydrogenase in blood, and $TNF{\alpha}$ was normalized in 8 h after the treatment with $\text\tiny{D}$-GalN/LPS. Coincidently, major organs remained unimpaired when compared to $\text\tiny{D}$-GalN/LPS control group. Moreover, p38, which stimulates expression of NAFLD-associated cytokines, was markedly inhibited when treated with SGBE. In vitro analysis revealed that the main components of SGBE, linoleic acid and ginsenoside Re/Rd, may play a role in protecting liver from $\text\tiny{D}$-GalN/LPS-induced toxicity. Finally, we concluded that SGBE may be a promising therapeutic agent for preventing damage to the liver.

  • Colon cancer is one of the most common malignancies in the western world and the second leading cause of cancer death in Korea. Epidemiology studies have shown a reduced incidence of colon cancer among populations consuming a large quantity of ${\omega}3$-polyunsaturated fatty acids (${\omega}3$-PUFA) of marine origin. Recently, it has been found that ${\omega}3$-PUFA has an antineoplastic effect in several cancers. This study was designed to investigate the mechanism of the anti-invasive effect of ${\omega}3$-PUFA in colon cancer. ${\omega}3$-PUFA, docosahexaenoic acids (DHA) and eicosapentaenoic acid (EPA) treatment resulted in a dose-dependent inhibition of cell growth in SW480 human colon cancer cells. In contrast, arachidonic acid (AA), a ${\omega}6$-PUFA, exhibited no significant effect. This action likely involves apoptosis, given that DHA treatment increased apoptotic cells in TUNEL assay. Moreover, invasiveness of SW480 cells was inhibited following treatment of DHA in a dose-dependent manner; in contrast, AA had no effect. The levels of MMP-9 and MMP-2 mRNA decreased after DHA pretreatment. MMP-9 and MMP-2 promoter activities were also inhibited by DHA treatment. The levels of NF-kB and p-IkB protein were down-regulated by DHA pretreatment in a dose dependent manner. In addition, DHA inhibited NF-kB promoter reporter activities. These findings suggest that ${\omega}3$-PUFA may inhibit cancer cell invasion by inhibition of MMPs via reduction of NF-kB in colon cancer. In conclusion, ${\omega}3$-PUFA could be used for chemoprevention and treatment of human colon cancer.

Biomass and Nutrient Stocks of Tree Components by Stand Density in a Quercus glauca Plantation (종가시나무 조림지의 임분밀도에 따른 임목 바이오매스 및 양분축적량)

  • This study was conducted to evaluate aboveground tree biomass and nutrient (C, N, P, K, Ca, and Mg) response of tree components by high (1,933 trees $ha^{-1}$) and low (1,200 tree $ha^{-1}$) stand densities in a 27-year-old Quercus glauca plantation. The study site was located in Goseong county, Gyeongsangnam-do, southern Korea. Total 12 trees (6 high and 6 low stand densities) were cut to develop allometric equations and to measure nutrient concentration of tree components. Stand density-specific allometric equations in the high and low stand densities were significant (P < 0.05) in tree components with diameter at breast height (DBH). Also, generalized allometric equations could be applied to estimate tree biomass regardless of the difference of stand density because of no significant effect on slope of stand density-specific allometric equations. Aboveground tree biomass estimated by the allometric equations was significantly higher in the high stand density (177 Mg $ha^{-1}$) than in the low stand density (114 Mg $ha^{-1}$). However, nutrient concentration of tree components was not significantly affected by the difference of stand density. Nutrient stocks in tree components were not significantly between the high stand density and the low stand density, except for the N and P stocks of stem wood. These results indicate that aboveground tree biomass could be significantly affected by stand density, but nutrient concentration among the tree components was not affected by the difference of stand density in a Quercus glauca plantation.

Long-term Prognostic Value of Dipyridamole Stress Myocardial SPECT (디피리다몰 부하 심근관류 SPECT의 장기예후 예측능)

  • Purpose: Dipyridamole stress myocardial perfusion SPECT could predict prognosis, however, long-term follow-up showed change of hazard ratio in patients with suspected coronary artery disease. We investigated how long normal SPECT could predict the benign prognosis on the long-term follow-up. Materials and Methods: We followed up 1169 patients and divided these patients into groups in whom coronary angiography were performed and were not. Total cardiac event rate and hard event rate were predicted using clinical, angiographic and SPECT findings. Predictive values of normal and abnormal SPECT were examined using survival analysis with Mantel-Haenszel method, multivariate Cox proportional hazard model analysis and newly developed statistical method to test time-invariance of hazard rate and changing point of this rate. Results: Reversible perfusion decrease on myocardial perfusion SPECT predicted higher total cardiac event rate independently and further to angiographic findings. However, myocardial SPECT showed independent but not incremental prognostic values for hard event rate. Hazard ratio of normal perfusion SPECT was changed significantly (p<0.001) and the changing point of hazard rate was 4.4 years of follows up. However, the ratio of abnormal SPECT was not. Conclusion: Dipyridamole stress myocardial perfusion SPECT provided independent prognostic information in patients with known and suspected coronary artery disease. Normal perfusion SPECT predicted least event rate for 4.4 years.

Evaluation of Susceptibility of Western Flower Thrips (Frankliniella occidentalis) and Garden Thrips (F. intonsa) to 51 Insecticides (꽃노랑총채벌레와 대만총채벌레에 대한 51종의 살충제 감수성 평가)

  • The susceptibility of the western flower thrips, Frankliniella occidentalis and garden thrips, Frankliniella intonsa was evaluated using 51 commercial insecticides. 15 kinds of insecticides which showed more than 90% mortality against both thrips, F. occidentalis and F. intonsa was selected. Many active ingredients were misused and abused in commercial mixture formulation insecticides. Since the F. intonsa was more susceptible than F. occidentalis, it was considered that both thrips can be controlled by insecticides that showed insecticidal activity on the F. occidentalis. Lethal time ($LT_{50}$ and $LT_{95}$), systemic toxicity and residual toxicity of selected insecticides were compared. Both chlorpyrifos WP and chlorpyrifos + diflubenzuron WP revealed the fastest toxicity within 2 h ($LT_{95}$), while spinetoram WG revealed the slowest toxicity as 62.3 h ($LT_{95}$). Chlorfenapyr SC showed toxicity at foliar and drenching application while spinetoram WG was toxic only in foliar application. Chlorfenapyr SC showed residual effect at 3, 5, 7, 10, 15 days after treatment and both benfuracarb WG and chlorpyrifos WP showed residual effect at 3 days after treatment. As a result of treatment of selected insecticides for field population of F. occidentalis, the population collected from horticultural crops showed lower susceptibility than the population collected from vegetable crops.

Development of a Small Animal Positron Emission Tomography Using Dual-layer Phoswich Detector and Position Sensitive Photomultiplier Tube: Preliminary Results (두층 섬광결정과 위치민감형광전자증배관을 이용한 소동물 양전자방출단층촬영기 개발: 기초실험 결과)

  • Purpose: The purpose of this study was to develop a small animal PET using dual layer phoswich detector to minimize parallax error that degrades spatial resolution at the outer part of field-of-view (FOV). Materials and Methods: A simulation tool GATE (Geant4 Application for Tomographic Emission) was used to derive optimal parameters of small PET, and PET was developed employing the parameters. Lutetium Oxyorthosilicate (LSO) and Lutetium-Yttrium Aluminate-Perovskite(LuYAP) was used to construct dual layer phoswitch crystal. $8{\times}8$ arrays of LSO and LuYAP pixels, $2mm{\times}2mm{\times}8mm$ in size, were coupled to a 64-channel position sensitive photomultiplier tube. The system consisted of 16 detector modules arranged to one ring configuration (ring inner diameter 10 cm, FOV of 8 cm). The data from phoswich detector modules were fed into an ADC board in the data acquisition and preprocessing PC via sockets, decoder block, FPGA board, and bus board. These were linked to the master PC that stored the events data on hard disk. Results: In a preliminary test of the system, reconstructed images were obtained by using a pair of detectors and sensitivity and spatial resolution were measured. Spatial resolution was 2.3 mm FWHM and sensitivity was 10.9 $cps/{\mu}Ci$ at the center of FOV. Conclusion: The radioactivity distribution patterns were accurately represented in sinograms and images obtained by PET with a pair of detectors. These preliminary results indicate that it is promising to develop a high performance small animal PET.

An Initiative Study on Relationship between Algal Blooms and Asian Dust for Regulation of Algal Blooms (조류 성장 억제를 위한 녹조 및 적조 발생과 황사의 상관관계 초기적 연구)

  • Although the problems of the algal blooms have been world-widely observed in freshwater, estuary, and marine throughout the year, it is not yet certain what are the basic causes of such blooms. Consequently, it is very difficult to predict when and where algal blooms occur. The constituents of the Asian dust are in a good agreement with the elements required for the algal growth, which suggests some possible relationship between the algal blooms and the Asian dust. There have been frequently algal blooms in drinking water from rivers or lakes. However, there is no any algal blooms in upwelling waters where the Asian dust cannot penetrate into the soil due to its relatively weak settling velocity (size of particles, $4.5{\pm}1.5{\mu}m$), which implies the possible close relationship of the Asian dust with algal blooms. The present initiative study is thus intended firstly in Korea to illustrate such a relationship by reviewing typical previous studies along with 12 years of weekly iron profiles (2001~2012) and two slant culture experiments with the dissolved Asian dust. The result showed bacterial suspected colonies in the slant culture experiment that are qualitatively in a good agreement with the recent Japanese studies. Since the diatoms require cheap energy (8%) compared to other phytoplankton (100%) to synthesize their cell walls by silicate, the present results can be used to predict algal blooms by diatoms if the concentrations of iron and silicate are available during spring and fall. It can be postulated that the algal blooms occur only if the environmental factors such as light, nutrients, calm water surface layer, temperature, and pH are simultaneously satisfied with the requirements of the micronutrients of mineral ions supplied by the Asian dust as enzymatic cofactors for the rapid bio-synthesis of the macromolecules during algal blooms. Simple eco-friendly methods to regulate the algal blooms are suggested for the initial stage of blooming with limited area: 1) to cover up the water surface with black curtain and inhibit photosynthesis during the day time, 2) to blow air (20.9%) or pure oxygen into the bottom of the water and inhibit rubisco for carbon uptake and nitrate reductase for nitrogen uptake activities in algal growth during the night, 3) to eliminate the resting spores or cysts by suction of bottom sediments as deep as 5 cm to prevent the next year germinations.

A Computer Simulation for Small Animal Iodine-125 SPECT Development (소동물 Iodine-125 SPECT 개발을 위한 컴퓨터 시뮬레이션)

  • Purpose: Since I-125 emits low energy (27-35 keV) radiation, thinner crystal and collimator could be employed and, hence, it is favorable to obtain high quality images. The purpose of this study was to derive the optimized parameters of I-125 SPECT using a new simulation tool, GATE (Geant4 Application for Tomographic Emission). Materials and Methods: To validate the simulation method, gamma camera developed by Weisenberger et al. was modeled. Nal(T1) plate crystal was used and its thickness was determined by calculating detection efficiency. Spatial resolution and sensitivity curves were estimated by changing variable parameters for parallel-hole and pinhole collimator. Peformances of I-125 SPECT equipped with the optimal collimator were also estimated. Results: in the validation study, simulations were found to agree well with experimental measurements in spatial resolution (4%) and sensitivity (3%). In order to acquire 98% gamma ray detection efficiency, Nal(T1) thickness was determined to be 1 mm. Hole diameter (mm), length (mm) and shape were chosen to be 0.2:5:square and 0.5:10:hexagonal for high resolution (HR) and general purpose (GP) parallel-hole collimator, respectively. Hole diameter, channel height and acceptance angle of pinhole (PH) collimator were determined to be 0.25 mm, 0.1 mm and 90 degree. The spatial resolutions of reconstructed image of the I-125 SPECT employing HR:GP:PH were 1.2:1.7:0.8 mm. The sensitivities of HR:GP:PH were 39.7:71.9:5.5 cps/MBq. Conclusion: The optimal crystal and collimator parameters for I-125 Imaging were derived by simulation using GATE. The results indicate that excellent resolution and sensitivity imaging is feasible using I-125 SPECT.
