태백산분지 북익부 고품위 석회석의 생성은 크게 동생적(syngenetic)과 후생적(epigenetic) 관점으로 나뉘어 있으며, 산소-탄소 안정동위원소 비 특성을 이용하여 이들의 생성환경을 고찰하였다. 북익부의 4개 광산(GMI, 정선, 백운, 백광)과 남익부의 상동(오미아)광산, 그리고, 삼척지역의 쌍용동해광산을 대상으로 하였으며, 이들과의 비교를 위해 정선지역 대기층 노두시료를 포함하였으며, 이들을 색상에 따라 암회색, 회색, 담회색, 백색으로 구분하였다. 산소 안정동위원소 비는 4.5 ~ 21.6 ‰의 넓은 범위의 변화를 보이지만, 탄소 안정동위원소 비는 -1.1 ~ 0.8 ‰ (방해석 맥 제외)로 매우 좁은 범위를 보여, 열수의 작용이 크지 않았음을 지시한다. 또한, 광산별, 색상별 산소-탄소 안정동위원소 비의 분포범위의 차이는 없으며, 주변 노두에서 채취된 탄산염암과 유사한 범위를 보이는 것으로 보인다. 이와 같은 결과는 고품위 석회석의 생성에 있어서 열수의 영향은 거의 없었다는 것을 지시한다. 산소 안정동위원소 비 값은 암석의 색상변화(백색화)보다는 조직의 변화와 관련되는데, 산소 안정동위원소 비 15 ‰ 전후를 기준으로 탄산염암의 산상을 구분할 수 있다. 이보다 높은 경우 괴상 또는 층리가 잘 보존된 특징을 보이는 반면, 낮은 경우 하나의 시료 내에서 두 가지 이상의 색상이 나타나고 있으며, 층리는 교란되거나 거의 소멸하여 흔적만 확인된다. 두 가지 이상의 색상이 나타나는 경우 밝은 부분이 어두운 부분에 비해 현저히 낮은 산소 안정동위원소 비를 보이는 경우가 다수 관찰되며, 이는 열수와의 반응에 의한 현상으로 해석된다. 다만, 이와 같은 현상은 소규모로 나타나고 있으며, 광상 전체적인 고품위 석회석의 생성에 대한 해석으로는 적당하지 않다. 특히, 재결정화한 백색의 탄산염암에서 높은 산소 안정동위원소 비 값을 보이는 것은 이 지역 탄산염암의 재결정화에 열수의 작용이 매우 적었음을 지시하며, 주변 마그마에 의한 열적 영향만을 받은 결과로 해석된다. 또한, 고품위 석회석광상 내 광체의 분포가 수평으로 연속적이며, 고품위 석회석 상하 주변에 백운석이 층상으로 협재되는 등의 산상을 고려할 때, 연구지역 고품위 석회석의 생성은 동생적(syngenetic) 해석이 보다 더 타당할 것으로 판단된다. 따라서, 태백산분지 전반적으로 순차층서적 관점에 의한 퇴적상의 해석이 필요할 것으로 사료되며, 이는 추가적인 고품위 석회석 광체의 확보에 매우 중요한 요소가 될 것이다.
산소안정동위원소는 추적자로서의 뛰어난 특성에도 불구하고 분석상의 어려움으로 해양수괴 연구에 많이 활용되지 못하고 있다. 물시료에 함유된 산소의 안정동위원소비를 측정하기 위해 가장 많이 사용하는 방법은 $H_2O/CO_2$ 평형기를 이용해 시료인 물을 이산화탄소와 동위원소평형을 이루게 한 뒤, 이산화탄소의 동위원소 비율을 측정하는 것이다. 이러한 자동 $H_2O/CO_2$ 평형기를 이용한 물 시료 분석시의 정밀도는 ${\pm}0.1%o$, 해양에서 이를 적용하기 위해서는 정밀도를 보다 높이는 것이 요구된다. 따라서 본 연구에서는 분석상의 오차를 감소시킬 수 요인들을 파악하여 분석장치의 보완 및 분석방법의 개선으로 정밀도 최소 0.05%o 향상시킴으로서 산소동안정동위원소의 변화폭이 작은 해양에서도 추적자로서 활발히 응용될 수 있도록 기여하고자 하였다. $H_2O/CO_2$ 평형기를 이용하는 경우 가장 큰 문제점은 시료가스가 전처리 장치로부터 질량분석기까지 이동해야 하는 거리가 멀고 평형가스가 팽창해야 하는 부피가 크다는 것이다. 그러므로 단순히 압력 차를 이용해서 가스를 이동시킬 경우, 가벼운 동위원소종이 선택적으로 이동되는 동위원소 분별작용이 일어날 수 있고, 분석 장치 전체의 부피가 플라스크에 담긴 시료가스의 부피에 비해 크기 때문에 시료가스가 충분히 혼합되지 않을 수가 있다는 점이다. 이러한 점을 보완하기 위해 액체질소 트랩과 고진공 균일도를 향상시켰다. 동일한 해수시료를 기존방법과 보완방법을 이용하여 각각 14번씩 분석한 결과 평균 측정값은 0.023, 0.024%o 서로 거의 동일한 값을 보였으나 표준편차는 각각 ${\pm}0.081$, ${\pm}0.021%o$ 네 배가량 개선된 결과를 보이고 있다. 그 결과 해양에서의 동위원소 분석시 발생할 수 있는 중요한 오차 요인을 현저하게 감소시킴으로서 동위원소 변화폭이 작은 해양수괴 연구에 크게 기여할 수 있을 것으로 기대된다.
Stable isotope profiles with fine-scale resolution were constructed from the fossil mollusk shells, Mercernaria mercernaria, obtained from the late Pleistocene transgressive deposits of Gomez Pit, Virginia, USA. Incremental sampling were made along the axis of maximum growth to provide high-resolution ${\delta}^{18}$O and ${\delta}^{13}$C records. The ${\delta}^{18}$O shell profiles exhibit a series of pronounced cycles in the overall amplitude, corresponding to strong seasonal variations in temperature, which is apparently positive environmental variable. Contrasts between the patterns of ${\delta}^{18}$O and ${\delta}^{13}$C profiles reflect the relationship influencing the seasonal carbon cycling in the shallow marine environment. Positive anomalies of the ${\delta}^{13}$C values during the summer were observed to be out of phase with the ${\delta}^{18}$O profile. Such relatively heavier carbon source may be alternated due to seasonal methanogenesis during the summer. A hypothesized methane-based system may be operated in the shallow and marginal marine environment, resulting in a ${\delta}^{13}$C enriched bicarbonate pool, in which the heavier isotope seems to be incorporated to the shell carbonate.
The recent progress on metabolic systems engineering was reviewed based on our recent research results in terms of (1) metabolic signal flow diagram approach, (2) metabolic flux analysis (MFA) in particular with intracellular isotopomer distribution using NMR and/or GC-MS, (3) synthesis and optimization of metabolic flux distribution (MFD), (4) modification of MFD by gene manipulation and by controlling culture environment, (5) metabolic control analysis (MCA), (6) design of metabolic regulation structure, and (7) identification of unknown pathways with isotope tracing by NMR. The main characteristics of metabolic engineering is to treat metabolism as a network or entirety instead of individual reactions. The applications were made for poly-3-hydroxybutyrate (PHB) production using Ralstonia eutropha and recombinant Escherichia coli, lactate production by recombinant Saccharomyces cerevisiae, pyruvate production by vitamin auxotrophic yeast Toluropsis glabrata, lysine production using Corynebacterium glutamicum, and energetic analysis of photosynthesic microorganisms such as Cyanobateria. The characteristics of each approach were reviewed with their applications. The approach based on isotope labeling experiments gives reliable and quantitative results for metabolic flux analysis. It should be recognized that the next stage should be toward the investigation of metabolic flux analysis with gene and protein expressions to uncover the metabolic regulation in relation to genetic modification and/ or the change in the culture condition.

Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS) is one of the powerful methods that enable analysis of small molecules as well as large molecules up to about 500,000 Da without severe fragmentation. MALDI-TOF MS, thus, has been a very useful an analytical tool for the confirmation of synthetic molecules, probing PTMs, and identifying structures of a given protein. In recent nuclear medicine, MALDI-TOF MS liner ion mode helps researcher calculate the average number of chelator(or linkage) per an antibody conjugate, such as DOTA-(or DFO-) trastuzumab for labeling a medical radioisotope. This simple technique can be utilized to improve the labeling method and control the quality at the development of antibody-based radiopharmaceuticals, which is very effected to diagnosis and therapy for in vivo tumor cells, with radioisotopes like $^{89}Zr$, $^{64}Cu$, and 177Lu. To minimize the error, MALDI-TOF MS measurement is repeatedly performed for each sample in this study, and external calibration is carried out after data collection.
As an important contribution to the planed drilling (IODP) in the central part of the Arctic Ocean, we are currently working on a refined chronostratigraphy for Marine Isotope Stage (MIS) 16 to MIS 2 on the exciting material from ODP Site 910A (Leg 151) which has been recovered from the marginal Eastern Arctic Ocean (the Yermak Plateau - the Atlantic/Arctic Ocean Gateway). Several stratigraphic age fix-points support the interpretation of the stable oxygen. isotope record of planktonic foraminifer N, pachyderma sin. that is punctuated by several short-term meltwater events. We believe that our new record will serve as 'the important correlating tool for establishing a basic stratigraphy for the Quaternary Arctic Ocean as well as for generating high-resolution paleoenvironmental reconstructions in the central Arctic Ocean. Furthermore, our study will provide reference stratigraphic data sets for interpreting the micropaleontological, sedimentological and organic / inorganic - geochemical proxies of the new boreholes that will be drilled on the Lomonosov Ridge(Central Arctic Ocean) in the frame of the "Arctic Coring Expedition' (ACEX, IODP) in summer 2004.
Secondary ion mass spectrometry (SIMS) is a promising tool to measure isotope ratios of individual uranium particles in environmental samples for nuclear safeguards. However, the analysis requires prior identification of a small number of uranium particles that coexist with a large number of other particles without uranium. In the present study, this identification was performed by scanning electron microscopy - energy dispersive X-ray analysis with automated particle search mode. The analytical results for an environmental sample taken at a nuclear facility indicated that the observation of backscattered electron images with × 1000 magnification was appropriate to efficiently identify uranium particles. Lower magnification (less than × 500) made it difficult to detect smaller particles of approximately 1 μm diameter. After identification, each particle was manipulated and transferred for subsequent isotope ratio analysis by SIMS. Consequently, the isotope ratios of individual uranium particles were successfully determined without any molecular ion interference. It was demonstrated that the proposed technique provides a powerful tool to measure individual particles not only for nuclear safeguards but also for environmental sciences.
A total of 23 springs distributed in the southern and northern sides of Mt. Hala in Jeju Island were seasonally sampled and analyzed for their major ion chemistry and oxygen and hydrogen isotope compositions to investigate their hydrogeochemical and isotopic characteristics. Dissolved ion concentrations of the south-side springs slightly increase with decreasing altitude. This indicates that dissolved ion concentrations of groundwater recharged at higher altitudes increase by water-rock interaction during the downgradient migration of groundwater through highly permeable volcanic aquifer. Dissolved ion concentrations of the north-side springs also slightly increase with decreasing altitude, but dramatically increase at ~300 m.a.s.l. This may indicate a sudden input of contaminants to the north-side groundwater system around ~300 m.a.s.l. Springs located in areas above ~300 m.a.s.l. have very low concentrations of dissolved ions, showing little seasonal variations. Whereas springs located in areas below ~300 m.a.s.l. show a big seasonal variation in the concentration of dissolved ions. Seasonal variation of oxygen isotope compositions of springs is ~3$\textperthousand$ for high-altitude springs (~1700 m.a.s.l.) and is ~2$\textperthousand$ near shore, indicating an attenuation of the variation through mixing with other groundwater bodies during migration.
The gold-silver deposits of the Nakcheon, Eunchi and Jungbong mine in the northern Taebagsan mining district are composed of fissure fil1ing veins emplaced in Precambrian meta-sediments and Jungbongsan granite. Based on the changes of ore texture and mineralogy, ore mineral chemistry, fluid inclusion and mineralizing condition, a regional zoning is recognized from the Nakcheon to the Eunchi and Jungbong ore deposits, and this trend of zoning is also recognized by stable isotope compositions. Stable isotope compositions show that the source of su1fur and carbon is mainly igneous origin, and the water of ore fluid in the Nakcheon ore deposits is mainly magmatic origin but much of meteoric water is involved in ore fluid of the Eunchi and Jungbong ore deposits. The ore deposits of study area is polymetallic meso to epithermal type genetically related to the acidic igneous pluton. Due to the differntial erosion level and mineralized depth, each ore deposits has a slightly different characteristic of mineralization; The Nakcheon ore deposits belong to meso-epithermal type, but the Eunchi and Jungbong ore deposits belong to epithermal type.
To study the origin of organic matter in meteorite, terrestrial rocks which contain or-ganic compounds similar to the ones found in carbonaceous chondrites are studied and compared with Muchison meteorite. Hydrocarbon molecules were extracted by benzene and methanol from bituminous coal and oil shale and the extracts were partitioned into aliphatic, aromatic, and polar fractions by silica gel column chromatography. Carbon isotopic ratios in each fractions were analysed by GC-C-IRMS. Molec-ular compound identifications were carried by GC-MS Engine. Bituminous coal and oil shale show the organic compound composition similar to that of meteorite. Oil shale has a wide range of ${\delta}^{13}C,-20.1%_0~-54.4%_0$ compared to bituminous coal, $-25.2%_0~34.3%_0$. Delta values of several molecular compounds in two terrestrial samples are different. They show several distinct distributions in isotopic ratios compared to those of meteorite; Murchison meteorite has a range of ${\delta}^13C\;from\;-13%_0\;to\;+30%_0$. These results provide interpretation for the source and the formation condition of each rock, in particular alteration and migration processes of organic matter. Especially, they show an important clue whether some hydrocarbon molecules observed in meteorite are indigenous or not.
