The Effects of Pear Phenolic Compound and Herbal Drugs According to the dose and Duration on the Respiratory System of Asthma Mice Induced by Ovalbumin (배(이(梨)) 추출물과 고경(枯梗), 행인(杏仁) 배합제제의 용량 및 기간별 투여가 Ovalbumin으로 유발된 천식 생쥐에 미치는 영향)

  • Objectives: Oriental pear was used as traditional remedies for respiratory diseases like cough and asthma etc. The purpose of this study was to observe the effects of preventing asthma by the combination of phenolic compound extracted from pear and herbal drugs. Methods: In order to study the effects of preventing asthma by the combination of phenolic compound extracted from pear and herbal drugs(Platycodon grandiflorum, Prunus armeniaca) on allergic asthma, mice were pre-treated by oral administration of the solution before antigen sensitization for 10 days and 20days. And 2 days later, mice were actively sensitized with a subcutaneous injection of ovalbumin and 13 days later, they were provoked with ovalbumin aerosols. The experimental groups were divided 6groups(10d1P, 10d2P, 10d4P, 20d1P, 20d2P and 20d4P) by meditation quantity and period. We measured isometric contractile responses to acetylcholine(ACh) and KCl in the isolated tracheal smooth muscle(TSM), IL-4, eosinophil and lymphocyte in bronchoalveolar lavage fluid(BALF), IgE in serum, WBC, RBC and hemoglobin in blood. Results: Contractile responses of TSM to ACh, the sensitivity of TSM to Ach and the maximal contractile response of TSM to KCl were decreased by direct proportion of meditation quantity. Eosinophil and IL-4 level in BALF were more significantly decreased, and IgE level in serum was more significantly increased in 10d4P and 20d4P group than the control group. Conclusions: Based on the above results, it is assumed that oral administration of the combination of phenolic compound extracted from pear and herbal drugs(Platycodon grandiflorum, Prunus armeniaca) can help the preventing effects of allergic asthma.

The Relationships Between Valgus Collapse Knee Position and Quadriceps Activity During a Single Limb Step Down in Female Subjects (젊은 여성의 한쪽 다리 스텝다운 동작 시 슬관절 외반 정도와 대퇴사두근 근활성도 간의 상관관계)

  • Background: This study was designed to investigate the correlation between electromyography (EMG) activities in the vastus medialis oblique (VMO) vs vastus lateralis (VL) activity ratio and the valgus collapse knee position while stepping down. Methods: Twenty healthy women volunteered to participate in this study. We measured the frontal-plane projections of the knee valgus angle, knee valgus distance, and hip adduction angle by using a digital camcorder. After 3 repetitions of the step down (dominant side) exercise, the findings of the static and dynamic phases were analyzed. EMG activities data of the VMO:VL activity ratio were recorded during the step down exercise and were normalized to the maximal voluntary isometric contraction (MVIC) of the quadriceps. A paired t-test was used to compare the findings of the static and dynamic phases. We analyzed the Spearman's rank order correlation coefficient between the and VMO:VL ratio. Results: Hip adduction angle, knee valgus angle, VMO activity, VL activity, VMO:VL activity ratio were statistically higher in the dynamic phase than in the static phase (p<.05). Frontal-plane projections of knee valgus angle were significantly correlated with hip adduction angle (r=.459, p<.05) and knee valgus distance (r=.505, p<.05). However, the EMG activity ratio of the VMO and the VL did not show a significant change during step down exercise with respect to hip adduction angle (p=.875), knee valgus angle (p=.618), and knee valgus distance (p=.701). Conclusion: The results from this study indicate that frontal-plane projections of knee valgus angle were associated with hip adduction angle and knee valgus distance. On the basis of these results, the knee valgus distance may be used to determine the valgus collapse knee position while stepping down.

Evidence for Adenosine Triphosphate (ATP) as an Excitatory Neurotransmitter in Guinea-Pig Gastric Antrum

  • We explore the question of whether adenosine 5'-triphosphate (ATP) acts as an excitatory neurotransmitter in guinea-pig gastric smooth muscle. In an organ bath system, isometric force of the circular smooth muscle of guinea-pig gastric antrum was measured in the presence of atropine and guanethidine. Under electrical field stimulation (EFS) at high frequencies (>20 Hz), NO-mediated relaxation during EFS was followed by a strong contraction after the cessation of EFS (a 'rebound-contraction'). Exogenous ATP mimicked the rebound-contraction. A known $P_{2Y}-purinoceptor$ antagonist, reactive blue 2 (RB-2), blocked the rebound-contraction while selective desensitization of $P_{2Y}-purinoceptor$ with ${\alpha},{\beta}-MeATP$ did not affect it. ATP and 2-MeSATP induced smooth muscle contraction, which was effectively blocked by RB-2 and suramin, a nonselective $P_2-purinoceptor$ antagonist. Particularly, in the presence of RB-2, exogenous ATP and 2-MeSATP inhibited spontaneous phasic contractions, suggesting the existence of different populations of purinoceptors. Both the rebound-contraction and the agonist-induced contraction were not inhibited by indomethacin. The rank orders of agonists' potency were 2-MeSATP > ATP ${ge}$ UTP for contraction and ${\alpha},{\beta}-MeATP\;{\ge}\;{\beta},{\gamma}-MeATP$ for inhibition of the phasic contraction, that accord with the commonly accepted rank order of the classical $P_{2Y}-purinoceptor$ subtypes. Electrical activities of smooth muscles were only slightly influenced by ATP and 2-MeSATP, whereas ${\alpha},{\beta}-MeATP$ attenuated slow waves with membrane hyperpolarization. From the above results, it is suggested that ATP acts as an excitatory neurotransmitter, which mediates the rebound-contraction via $P_{2Y}-purinoceptor$ in guinea-pig gastric antrum.

Effects of Verapamil and Tetracaine on Acetylcholine-and Oxytocin-induced Uterine Contraction Pattern (Acetylcholine및 Oxytocin에 의하여 야기되는 렛드 자궁수축에 미치는 Verapamil 및 Tetracaine의 영향)

  • The effects of verapamil and tetracaine on acetylcholine-and oxytocin-induced contraction of uterus from estrogen-treated rat were examined. Isometric tensions were recorded on the Physiograph and stored in TriGem 20XT computer as digitized data for off-line analysis of the components, which described the contraction patterns: trought tension (T), peak tension (P), contraction frequency (F), and duration (D). In the acetylcholine-induced contraction, verapamil $(0.25\;{\mu}M)$ significantly decreased P and D. In contrast, tetracaine $(42{\mu}M)$ decreased F, but increased D. In the low oxytocin-induced contraction, verapamil $(0.25\;{\mu}M)$ decreased P and D, and tetracaine $(42{\mu}M)$ decreased F but increased D. In the high oxytocin-induced contraction, verapamil decreased P and D, but tetracaine decreased P without affecting on other components. These results suggest that the analysis of effects of a certain inhibitor on the components of contraction allow to postulate its specific inhibitory mechanism of the smooth muscle contraction.

Characterization of Prunus necrotic ringspot virus Isolate from Peach in Korea (국내 복숭아에서 분리한 Prunus necrotic ringspot virus의 특성)

  • In this paper, we report a characterization of Prunus necrotic ringspot virus (PNRSV) isolate. The virus was identified from 'Yumyeong' peach showing mild mosaic on leaves in commercial orchard of 'Umsung', Chungbuk province in Korea. The virus isolate produced ringspot symptom on the inoculated cotyledons and systemic mosaic and malformation on the upper leaves of Cucumis sativus. Systemic mottles were appeared in Chenopodium quinoa. When the buds of the virus infected stem were grafted on the healthy young Prunus persica GF305 seedlings, line pattern with mosaic appeared within 3 months. Isometric virus-like particles were found in parenchyma cells and plasmodesmata of C. sativus leaves inoculated mechanically with the virus. The cDNA fragments of PNRSV coat protein (CP) region, approximately 675bp, were synthesized from genomic RNA extracted from virus-infected leaves by RT-PCR using specific primer pairs. Partial nucleotide sequences of the CP regions were determined and analyzed with the known PNRSV. The CP gene of PNRSVKorea isolates showed 93.9~94.7% similarity to the 4 known PNRSV isolates.

Effects of Cordyceps militaris Mycelia(CMM) herbal acupuncture at BL13, LU4 on airway smooth muscle, airway inflammation, IgE and Interleukin-4 in mouse model of allergic bronchial asthma (동충하초(冬蟲夏草) 약침(藥鍼)이 백서(白鼠)의 기관지평활근(氣管支平滑筋) 장력(張力)과 염증(炎症) 및 면역(免疫) 반응(反應)에 미치는 영향(影響))

  • Objectives : It has been reported that Cordyceps militaris Mycelia(CMM) has an effect on deficiency allergic asthma(虛喘) clinically. The aim of this study was to determine an appropriate oriental treatment and the doses of CMM to treat asthma. Methods : In order to study the effect of herbal acupuncture solution of the CMM on allergic asthma, the mouse were pretreated by CMM herbal acupuncture at BL13, LU4 before antigen sensitization. 2 days later Mice were actively sensitized with a subcutaneous injection of ovalbumin(OA) and 13 days later they were provoked with OA aerosols. IL-4, lymphocyte, macrophage in bronchoalveolar lavage fluid(BALF), IgE in serum, WBC, RBC, HGB in blood, and in vitro isometric contractile responses of the isolated tracheal smooth muscle(TSM) to acetylcholine$(ACh,\;0.1-1000\;{\mu}M)$, KCl were measured. Results : Contractile responses of TSM to ACh were significantly increased in CMM herbal acupuncture 1 group $(Ach\;1000\;{\mu}M)$, CMM herbal acupuncture 2 group $(ACh\;1,\;10\;{\mu}M)$, CMM herbal acupuncture 3 group $(Ach\;0.3,\;1,\;30,\;300\;{\mu}M)$. The sensitivity of TSM to ACh was significantly decreased in CMM herbal acupuncture 3 group. The maximal contractile response of TSM to ACh was significantly decreased in CMM herbal acupuncture 1, 3 group. The maximal contractile response of TSM to KCl was significantly decreased in CMM herbal acupuncture 1, 2, 3 group. The counts of lymphocytes in BALF was significantly increased in CMM herbal acupuncture 3 group. The counts of macrophages in BALF was significantly decreased in CMM herbal acupuncture 3 group. Interleukin-4 level in BALF was significantly increased in CMM herbal acupuncture 1,3 group. and it was increased in CMM herbal acupuncture 2 group, but there was no significance. Serum IgE level was significantly decreased in CMM herbal acupuncture 1, 2, 3 group. The counts of WBC in blood was significantly increased in CMM herbal acupuncture 1, 3 group Conclusion : Based on the above results it is assumed that CMM herbal acupuncture at BL13, LU4 can help the treatment of deficiency allergic Asthm.

Comparing the Muscle Strength of the Iliopsoas with the Muscle Activity of the Rectus Femoris according to Knee Flexion Angles in Supine and Sitting Positions (바로 누운 자세와 앉은 자세에서 무릎관절 굽힘 각도에 따른 엉덩허리근의 근력과 넙다리곧은근의 근활성도 비교)

  • Purpose : The muscle strength of iliopsoas (IL) was measured commonly in sitting position with hip and knee flexed 90°. However, there is no study to determine the muscle strength of IL in various test positions. Therefore, the purpose of this study was to compare the muscle strength of IL and muscle activity of rectus femoris (RF) according to test position and knee flexion angle. Methods : Twenty healthy subjects were participated for this study. The muscle strength of IL and muscle activity of RF were measured by hand-held dynamometer and surface electromyography during maximum voluntary isometric contraction (MVIC) of IL, respectively. The muscle strength of IL and muscle activity of RF was measured in 4 conditions as follows; 1) knee flexion angles 90 ° in supine, 2) 130 ° in supine position, 3) 90 ° in sitting, 4) 130 ° in sitting. Each condition were performed randomly by three repetitions. Results : The muscle strength of the IL was the main effect on the test position and knee flexion angle (p<.05), and the muscle activity of RF was the main effect only on the knee flexion angle (p<.05). There was also no interaction between the factors (p>.05). In supine position, the muscle strength of IL in knee flexion 130 ° was significantly less than that in knee flexion 90 ° (p<.0125). In knee flexion 90 °, the muscle strength of IL in supine position was significantly greater than that in sitting position (p<.0125). The muscle activity of RF in knee flexion 130 ° was significantly less than that in knee flexion 90 ° in supine and sitting positions (p<.0125). Conclusion : When the muscle strength of IL was measured in clinic and sports fields, the supine position with knee flexion 130 ° was recommended to prevent the muscle activation of RF and to maintain the trunk stability.

호흡기,소화기계 감염환자로부터 전염성 바이러스 분리 및 특성

  • 호흡기계 및 소화기계에 감염된 전염성 바이러스에 대한 역학적 기초자료로 이용하고자 1999년 1월부터 12월까지 부산지역에서 분리된 전염성 바이러스의 특징과 계절적 발생추이, 환자의 성별, 연령별 발생에 대해 조사한 결과는 다음과 같다. 1. 1999년도 바이러스 가검물 2261건에서 분리한 호흡기계감염 바이러스 279건과 소화기계 감염 바이러스 83주를 분리하였으며, 이중 인플루엔자 바이러스 A형이 96주(29.6%), B형이 107주(33.0%)로 대부분을 차지하였다. 2. 1999년의 바이러스 분리의 성별 분포는 총 360명의 환자 중 179명(49.7%)의 남성 및 181명(50.3%)의 여성으로 비슷한 양상을 나타내었다. 이중 호흡기계의 경우 279명의 감염환자 중 남성이 130명(46.6%), 여성이 149명(53.4%)으로 여성의 감염율이 비교적 높았으나, 소화기계의 경우 83명의 감염환자 중 남성이 51명(61.4%), 여성이 32명(38.5%)으로 남성의 감염율이 거의 2배정도 높게 나타났다. 3. 1999년의 연령별 분포는 10세 이하의 어린이가 194명(59.9%)으로 대부분을 차지하였으며, 이중 인플루엔자 바이러스가 99명(30.6%)으로 가장 높은 감염을을 나타내었다 유행성이하선염 바이러스의 감염어린이 중에 $l1{\sim}15$세의 연령층이 15명으로(53.3%)로 가장 높게 나타났다. 4. 1999년 월별 감염율은 호흡기계 감염증 바이러스의 경우 1월부터 4월까지, 그리고 12월에 증가 추세를 보이면서 4월에 가장 높은 감염율을 나타내었다 소화기계 감염증 바이러스의 경우 9, 10, 11월을 제외한 모든 월별에 관찰되었으며, echo와 coxsackie 바이러스는 무균성 수막염 환자에서 하절기에 집중적으로 발생하였다. 동절기에 유행하는 설사 바이러스는 12월에 비교적 높은 양상을 나타내었다. 5. 인플루엔자 바이러스는 MDCK 세포에서, 아데노 바이러스와 유행성 이하선염 바이러스는 HEp-2 세포에서, 파라인플루엔자 바이러스는 Vero 세포에서, 그리고 echo, coxsackie B 바이러스와 장내 바이러스는 HEp-2, Vero, BGM 세포에서 뚜렷한 세포병변 효과를 나타내었다. 6. 분리한 바이러스는 전자현미경으로 관찰한 결과 인플루엔자 바이러스 A 형(HINI, H3N2)은 95nm, B 형은 70nm크기의 구형을 나타내었으며, 바이러스표면의 지질 이중층이 뚜렷하게 관찰되었다. 아데노 바이러스는 외피가 관찰되지 않았으며, nucleocapsid는 symmetry이고 크기는 71nm로서 바이러스 입자 표면에 icosaheral capsomer의 배열이 명확하게 관찰되었고, 파라인플루엔자 바이러스와 유행성 이하선염 바이러스는 외피가 있는 구형의 큰 viron으로180, 170nm 크기이었다. 7. Echo와 coxsackie B group 바이러스는 모두 외피가 없는 isometric 형으로 크기는 $30{\sim}45nm$ 이었고, enteric adeno 바이러스는 84nm 크기로서 외피가 없고, 입자 표면에 capsomer의 배열이 명확하게 관찰되었고, rotavirus는 크기가 70nm이며 외층 capsid 단백질과 내층 capsid 단백질이 두층으로 되어 있는 전형적인 수레바퀴 모양을 나타내었다. 이상의 결과로 보아 호흡기계 및 소화기계에 감염되는 전염성 바이러스는 연중 지속적으로 분리되고 있으며 전염성이 강하여 집단 발생은 일으키는 경우도 많고 최근 들어 유행성 이하선염과 흥역 바이러스의 발생률이 높은 추이를 나타내고 있지만 아직은 특이한 바이러스 치료제가 개발되어 있지 않았으므로 지속적인 대책과 아울러 장기적인 발생 가능에 대한 예방책을 흥보하여야 할 것으로 보이며 계속적인 바이러스성 전염병 유행예측조사 및 역학조사가 적극적으로 이루어져야 할 것으로 사료된다.

Isolation of Enterovirus Causing Aseptic Meningitis in Busan, 2000-2002 Years (2000-2002년 바이러스성 뇌수막염의 발생양상 및 특성)

  • Enteroviruses isolation were attempted from samples obtained from aseptic meningitis-suspected patients in hospitals in Busan during 2000-2002. Enteroviruses were found in 2 of 292 cases in 2000, 4 of 371 cases in 2001, 83 of 703 cases in 2002. In 2000, the isolated viruses were found to be echovirus serotype 11 and coxsackievirus serotype B2. Coxsackievirus serotype B5 was isolated in 2001 and in 2002, echovirus serotypes 2, 3, 6, 7, 9, 13, 25, 30 were isolated in 70 cases while coxsackievirus serotypes B3 and B4 were isolated in 10 cases. Various specimens tended to emerge over the years. The occurrence in 2000 tended to be mostly focus during the cold months, December through January, while in 2001, it occurred in May. In 2002, occurrence was found to be distributed from April to November with the highest rate during June and July. The strains of Vero and HEp-2 of echovirus and coxsackievirus, respectively, are highly infectious. Electron micrograph of echovirus and coxsackievirus show that they are small nonenevolped, isometric-shaped viruses. Isolated RNA from strains of echovirus and coxsackievirus showing cytopathic effects were used to undergo nested PCR which resulted in a 436 bp single band in all the strains. The serotype was sent to the Department of Virology at the Korean National Institute of Health for identification.

The Effect of Kinesio Taping on Muscle Strength Improvement, Blood Fatigue Factors, Muscle Fatigue and Damage Index (Kinesio taping이 근력, 혈중 피로물질 및 근 손상 물질 농도변화에 미치는 영향)

  • The purpose of this study was to examine the effect of kinesio taping on muscle strength and changes of muscle fatigue and damage. 10 male subjects participated in 1-RM and isometric cybex muscle strength tests with and without taping application. Muscle strength (bench press, leg press) and extension (knee, shoulder) strength were significantly increased after taping, but there was no significant difference in flexion (knee, shoulder) strength. The concentration of fatigue factors (ammonia, phosphorous), muscle damage index substances (CK, LDH), IGF-I and creatinine were reduced after taping, but there were no significant differences.
