A study on Alkalophilic Bacteria Producing $\beta$-Galactosidase. -Isolations and Cultural Characteristics (I)- ($\beta$-Galactosidase를 생산하는 호알카리성 세균에 관한 연구 -균주의 분리 및 배양조건 (I)-)

  • A strain of alkalophilic Bacillus sp. YS-309 has been isolated from domestic soil. It belongs to genus Bacillus from its morphological and biochemical characteristics. The strain grows better in the alkaline media rather than in the neutral media. The optimum pH and temperature for growth were observed at 8.5 and 4$0^{\circ}C$, respectively. Glucose, lactose and maltose were appeared as good carbon source but soluble starch and fructose were utilized uneffectively for growth. Concentrations of lactose had affected both the cellular growth and the enzyme productions. The maximum growth and the highest enzyme productions were obtained at 0.5%(w/e) of lactose added in the media. B-Galactosidase from Bacillus sp. YS-309 was produced inducibly into the cell and total enzyme activities per ml were gradually decreased when the concentration of glucose increased.

  • One of the most serious problems in KEPCO system operation is higher fault current than the SCC(Short Circuit Capacity) of circuit breaker. There are many alternatives to reduce the increased fault current such as isolations of bus ties, enhancement of SCC of circuit breaker, applications of HVDC-BTB(Back to Back) and FCL(fault current limiter). But, these alternatives have some drawbacks in viewpoints of system stability and cost. As the superconductivity technology has been developed, the resistance type HTS-FCL(High Temperature Superconductor Fault Current Limiter) can be one of the most attractive alternatives to solve the fault current problem. To evaluate the accurate transient performance of resistance type HTS-FCL, it is needed that the dynamic simulation model considering transient characteristics during quenching and recovery state. Under this background, this paper presents the EMTDC model for resistance type HTS-FCL considering the nonlinear characteristic of final resistance value when quenching and recovery phenomena by fault current injection and clearing occurs.

  • A novel Si via structure is suggested and fabricated for 3D MEMS package using the doped silicon as an interconnection material. Oxide isolations which define Si via are formed simultaneously when fabricating the MEMS structure by using DRIE and oxidation. Silicon Direct Bonding Multi-stacking process is used for stacked package, which consists of a substrate, MEMS structure layer and a cover layer. The bonded wafers are thinned by lapping and polishing. A via with the size of $20{\mu}m$ is fabricated and the electrical and mechanical characteristics of via are under testing.

  • In this paper, fully embedded and miniaturized diplexer device has been developed and characterized for dual-band/mode CDMA handset applications. The size of the embedded diplexer is significantly reduced by embedding high Q circular spiral inductors and high DK MIM capacitors into a low cost organic package substrate. The fabricated diplexer has insertion losses and isolations of -0.5 and -23 dB at 824-894 MHz and -0.7 and -22 dB at 1850-1990 MHz, respectively. Its size is $3.9mm{\times}3.9mm{\times}0.77mm$. The fabricated diplexer is the smallest one which is fully embedded into a low cost organic package substrate.

  • In this paper, electrostatically actuated direct contact type RF MEMS switches have been designed and demonstrated. As driving structures of the switch, cantilever, bridge, and torsion spring beam structures are used and the actuation voltage characteristics of the switches have been compared and discussed. The designed RF switches are fabricated with the surface micromachining technology using the electroplated gold and nickel structures. The characteristics of the fabricated switches are measured and analyzed. The switch, which is fabricated using the 510 ${\mu}m$-length bridge structure with the thickness of 1.5 ${\mu}m$, is actuated with 15 V driving voltage. The insertion losses are less than 0.2 dB over the measured frequency ranges from 0 to 20 GHz and the isolations are more than 30 dB.

Studies on Yeasts Isolated from Marine Substrates (I) (해양효모의 분류 1)

  • As a part of taxonomical and ecological studies on the yeasts in marine environments, several kinds of yeasts were isolated from Zostera marina several invertebrates (penaeus, Meretrix and Neptunus) and surface sea water, which are collected at the two established sites of estuarine areas ; Dolsan isaland in Youchun district and Baiksu-ri in Youngkwang district. The obtained results can be summarized as follows. 1. Ascosporgenous Yeasts. Hansenula sp I and Hansenula sp II were isolated from Zotera marina and Hanse nular sp I from penaeus. 2. Asporogenous Yeasts. Trichoporon fermentans, Torulopsis, ernobii and Toruopsis dattia were isolated from Zostera marina, Candida krusei from Meretrix and Neptunus, Torulopsis pinus from surface sea waters, and Phodotorula aurantiaca from Penaeus. 3. More notable isolations of several species from Zostera marina than the other sources could be assumed as related to the higher sugar concentration of this plant.

Studies on the Constituents of Berberis amurensis Ruprecht (매발톱나무의 성분에 관한 연구)

  • Berberis amurensis Ruprecht(Berberidaceae) is a medicinal plant indigenous to the middle and northern part of Korean peninsula. The woody parts of this plant have been used for the ocular, peptic and intestinal disorders. The stems of this plants were extracted with MeOH and the MeOH extract was partitioned between organic phases and water layer, successively to fractionated quarternary alkaloids. The acetone-soluble part of guarternary alkaloidal fraction had antibacterial activities and it contained four protoberberine alkaloids such as palmatine(I), Berberine(II), Jatrorrhizine(III) and coptisine(IV), and one aporphine alkaloid, magnoflorine(V). Although the isolations of the compounds I, II, IIII, IV and V from different sources were reported, this is the first report that Berberis amurensis contained the compounds. When the contents of compound I(palmatine) and II(berberine) were quantified and compared with those of other plant parts, cortex contained higher palmatine and berberine than any other part of the plant.

The Disease Severity and Related Pathogens Caused by Root Rot on 6 Years Old Ginseng Cultivation Fields (인삼 6 년근 수확지의 뿌리썩음병 발생현황 및 관련 병원균 동정)

  • Ginseng (Pnanx ginseng C. A. Meyer) is famous worldwide, and is very important cash crop and medicinal herb in Korea. It takes four to five years to produce harvestable ginseng roots, and ginseng is attacked by several pathogens during cultivation. We investigated the disease rate caused by ginseng root rot from 6 years old ginseng cultivation fields (Chungnam; 9 fields, Chungbuk; 11 fields, Gangwon 5 fields). The highest disease severity was Dangjin D (2.9) and the lowest one was Gaesan C (0.6). Of the 625 isolations, 340 isolations were classified as Ilyonectria radicicola and Fusarium solani. Finally, genetic diversity of I. radicicola and F. solani was confirmed by sequence analysis. Among the I. radicicola group, I. mors-panacis, which is known as highly virulent pathogen, and I. liriodendri, I. robusta and I. cyclamicicola, which are weakly virulent pathogens, were identified. In the case of F. solani, it is divided into two groups, but it is necessary to conduct diversity research through genetic analysis and pathogenetic studies using various markers. Based on these results, it could be used as a basic data for control of ginseng root rot pathogens.
