Comparison of Loop-mediated Isothermal Amplification and Korea Standard Food Codex (KFSC) Method for Detection of Salmonella Typhimurium, Listeria monocytogenes Artificially Inoculated in Yuk-hwe and Yuk-sashimi (육회와 육사시미에 접종된 Salmonella Typhimurium와 Listeria monocytogenes 검출을 위한 Loop-mediated isothermal amplification와 식품공전의 배지 시험법, real-time PCR의 검출 성능 비교)

  • The object of this study is to compare the performance of the 3M Molecular Detection Assay 2 (3M MDA 2) and the Korea Standard Food Codex (KSFC) Method (i.e., isolation media and real-time PCR) in detecting Salmonella Typhimurium and Listeria monocytogenes in traditional Korean foods. Yuk-hwe and Yuk-sashimi (types of raw beef dishes) were artificially inoculated with $10^0-10^4CFU/25g$ of L. monocytogenes and S. Typhimurium. Citrobacter freundii and Listeria innocua were used as competitive microflora. After enrichment, the samples were analyzed using 3M MDA 2 and real-time PCR. All samples inoculated at concentrations of $10^0-10^4CFU/25g$ without competitive microflora were positive for S. Typhimurium and L. monocytogenes, as detected by 3M MDA 2 and Korea Standard Food Codex (KFSC) Method. In addition, part of the samples were positive for the presence of C. freundii and L. innocua. The 3M MDA 2 - Salmonella and Korea Standard Food Codex (KFSC) Method showed similar detection performances in Yuk-hwe and Yuk-sashimi. The 3M MDA 2 method for Salmonella and Listeria, which is a LAMP-based technology, can be used for rapid detection of S. Typhimurium and L. monocytogenes in raw beef. LAMP bioluminescence assays provide results on the subsequent day and are simple to use compared with the Korea Standard Food Codex (KFSC) Method, particularly in terms of DNA preparation.

Isolation of Wild Yeasts from Riversides in Geumgang Upstream, Taean and Suncheonman Seashores and Microbiological Characteristics of the Unrecorded Wild Yeasts (금강 상류와 태안, 순천만 해변들로부터 야생효모의 분리 및 국내 미기록 효모들의 균학적 특성)

  • This study aimed to isolate wild yeasts from waters and soils on the riversides of Geumgang upstream in Geumsan, seashores of Cheongpodae in Taean and Suncheonman in Suncheon, Korea, and to investigate their unrecorded microbiological characteristics. Forty-nine species of wild yeasts, including Aureobasidium pullans YH 4-3, were isolated from 40 samples of Cheonnaegang in Geumgang upstream. Fifty-six species and 36 species of wild yeasts were also isolated from 80 samples of Cheongpodae Beach and Suncheonman Mashland, respectively. Species from Candida genus were isolated from all three locations. Among them, Candida michaelii NNIBRFG28278, Sporobolomyces japonicus NNIBRFG28271, Dioszegia buhagiarii NNIBRFG28279, Ustilago spermophora NNIBRFG28273, Nakazawaea pomicola NNIBRFG31590, Candida natalensis NNIBRFG31591, Candida pseudorhagii NNIBRFG31592, Candida santamariae NNIBRFG31593, Cutaneotrichosporon terricola NNIBRFG31594, and Meira nashicola NNIBRFG31595 represent newly recorded yeast strains in Korea. Almost all of these unrecorded yeasts were oval shaped, and Sporobolomyces japonicus NNIBRFG28271 and Candida natalensis NNIBRFG31591 have ascospores. All the strains grew well in yeast extract-peptone-dextrose (YPD) and yeast extract-malt extract (YM) media, and Ustilago spermophora NNIBRFG28273 grew well in vitamin-free medium. Sporobolomyces japonicus NNIBRFG28271 grew well in 15% NaCl-containing YPD medium, and Ustilago spermophora NNIBRFG28273 and Dioszegia buhagiarii NNIBRFG28279 assimilated starch, and Ustilago spermophora NNIBRFG28273 produced ethanol.

Growth factors improve the proliferation of Jeju black pig muscle cells by regulating myogenic differentiation 1 and growth-related genes

  • Objective: The growth rate of pigs is related to differentiation and proliferation of muscle cells, which are regulated by growth factors and expression of growth-related genes. Thus, the objective of this study was to establish optimal culture conditions for Jeju black pig (JBP) muscle cells and determine the relationship of various factors involved in muscle growth with the proliferation of JBP muscle cells. Methods: Muscles were taken from the femur skeletal muscle of JBP embryos. After isolation of the muscle cells, cells were cultured in a 6-well plate under four different culture conditions to optimize culture conditions for JBP muscle cells. To analyze proliferation rate of JBP muscle cells, these muscle cells were seeded into 6-well plates at a density of 1.5×105 cells per well and cultured for 3 days. Western blot and quantitative real-time polymerase chain reaction were applied to verify the myogenic differentiation 1 (MyoD) expression and growth-related gene expression in JBP muscle cells, respectively. Results: We established a muscle cell line from JBP embryos and optimized its culture conditions. These muscle cells were positive for MyoD, but not for paired box 7. The proliferation rate of these muscle cells was significantly higher in a culture medium containing bFGF and epidermal growth factor + basic fibroblast growth factor (EGF+bFGF) than that without a growth factor or containing EGF alone. Treatment with EGF and bFGF significantly induced the expression of MyoD protein, an important transcription factor in muscle cells. Moreover, we checked the changes of expression of growth-related genes in JBP muscle cells by presence or absence of growth factors. Expression level of collagen type XXI alpha 1 gene was changed only when EGF and bFGF were added together to culture media for JBP muscle cells. Conclusion: Concurrent use of EGF and bFGF increased the expression of MyoD protein, thus regulating the proliferation of JBP muscle cells and the expression of growth-related genes.

A Study on the Staging of Scientific Imagination -History and Current Status of SF Theaters (과학적 상상력의 무대화에 대한 시론 -SF연극의 역사와 현재)

  • This paper is an attempt to discuss the history and the current state of 'SF theaters.' SF theater is still an unfamiliar genre to the public, and may surprise some, given that the stage is perceived as an insufficient space for stretching the scientific imagination. Since 2010 works that bring the scientific imagination into the theater have frequently been performed, and a recognition of SF theaters began to be established. Producers came to be absorbed in human psychology, and our isolation amidst the progress in technology, as well as in the absurdities of the world, while giving up the ideal of realistic descriptions. This became the foundation for SF theaters in South Korea today. Starting from the research history and the conceptual change in SF theaters, this study examined the status of SF dramas going back to the colonial period for SF theaters. Through inquiring into the history of SF theaters, we were able to derive the following implications and problems. Firstly, as they are based on future society or technical improvement without consideration of scientific probability or rationality, the scientific imagination is too absent for the work to be named 'SF theater.' Secondly, while being highly evaluated as an attempt to integrate science and stage in an era that emphasizes convergence, when we delve into the creativity of a material it is noticeable that the view of the world is still regressive. Thirdly, there are many cases in which scripts lean on SF classics or Japanese original works. Nevertheless, if young creators' diverse attempts in a genre can breathe with the contemporary audience desiring a new material, the foundation of a Korean-style SF theater may be expanded to include more significant work.

Behavior and perception characteristics on the isolated students and the bystanders in physical education (체육수업 참여 학생들의 교우관계 소외자와 방관자 행동 특성 및 인식 탐색)

  • The purpose of this study is to determine the nature and recognition of the isolated students and bystanders in respect of friendship in Middle School Physical Education. To achieve these objectives, we analyzed statistical material through PQR and inductive category by completing open questionnaires for 360 students bullied in public middle school in a large city. According the results, first, damages defender of participants showed the highest percentage, 70.2%. There were bystanders(12.3), the alienated(7.4) inflictor(5.5), contributor(4.6) in descending order. Second, according to the results of inductive analysis regarding the nature and recognition of bystanders in physical education, the biggest reason for not helping their colleagues in need is as following: ① improper friendship(32.8), ② lack of moral and emotional empathy(28.1), ③ lack of exercise ability(15.6), ④ psychological burden(12.5), ⑤ improper class attitude(10.9). Determination of volatile and residual iodine during the dissolution of spent nuclear fuel (사용 후 핵연료 용해 중 휘발 및 잔류 요오드 분석)

  • The determination of iodine in the spent nuclear fuel and the volatile behavior during its acid dissolution have been studied by NAA(neutron activation analysis) and electron probe microanalysis (EPMA). Simulated spent fuels (SIMFUELs) were dissolved in $HNO_3$(1+1) at $90^{\circ}C$ for 8 hours. The iodine remained in a dissolver solution after dissolution, and that condensed in dissolution apparatus and trapped in the adsorbent by volatilization during the dissolution were determined, respectively. The condensed iodine was recovered by the redistillation with $HNO_3$(1+1) after transfer of the dissolver solution. The iodines in the dissolver and redistilled solution were separated by solvent extraction followed by ion exchange or precipitation method and determined by RNAA (radiochemical neutron activation analysis). The ion exchange column and filtration kit used for the isolation of iodine, which were prepared with a polyethylene tube, were used as an insert in the pneumatic tube for neutron irradiation. The iodine volatilized during the dissolution of SIMFUELs was collected in a trapping tube containing Ag-silica gel (Ag-impregnated silica gel) adsorbent, and the distribution of iodine trapped in the adsorbents were determined by EPMA. The adsorbing characteristics shown with the SIMFUELs were compared with those shown with a real spent fuel from the nuclear power plant.

Relationship between Digital Informatization Self-Efficacy and Life Satisfaction in the Elderly - the Mediating Effect of Social Capital

  • The purpose of this study is to suggest action directions for preventing social isolation and improving life satisfaction of the elderly by verifying the mediating effect of social capital in the relationship between the elderly's digital information self-efficacy and their life satisfaction. For this purpose, the 2022 digital information gap survey data were used to analyze the relationship between digital information self-efficacy, social capital, and the elderly's life satisfaction using SPSS 26.0 and AMOS 24.0. As a result, first, the elderly's digital information self-efficacy was found to have a positive (+) effect on life satisfaction. Second, the elderly's digital information self-efficacy was found to have a positive (+) effect on social capital. Third, the social capital of the elderly was found to have a positive effect on life satisfaction. Fourth, the social capital of the elderly was found to have an indirect mediating effect in the relationship between digital information self-efficacy and life satisfaction. Based on this, practical and policy measures were presented to revitalize digital information education that older people can apply in real life, develop a digital platform for forming online-based social capital, communities suited to the digital information capabilities of older people, and revitalize information groups.

A Study on the Change Trends and Implications of North Korea-Russia Relations: Focusing on the Comprehensive Strategic Partnership Agreement (북·러 관계의 변화 동향과 시사점 고찰 - 포괄적 전략 동반자 협정을 중심으로 -)

  • Relations between North Korea and Russia have recently reached an inflection point in the wake of the Russia-Ukraine war, and are expected to have a significant military and geopolitical impact on the Korean Peninsula and the security environment in East Asia in the future. The 'Comprehensive Strategic Partnership Agreement between North Korea and Russia' was signed on the occasion of Kim Jong-un's visit to Russia in September 2023 and Russian President Putin's visit to North Korea in June 2024. The security environment on the Korean Peninsula and in East Asia is anticipated to be significantly impacted by developmental changes in North Korea-Russia relations. As North Korea and Russia formed a paramilitary alliance, stipulating provisions that can be interpreted as automatic military intervention, North Korea's military support for the ongoing war in Ukraine was secured, and the basis for Russia's military intervention in future Korean Peninsula issues was laid. Additionally, the possibility of transferring Russian military technology to North Korea increased as an institutional mechanism was established to implement joint measures to strengthen the defense capabilities between the two countries. If Russia uses its position as a permanent member of the UN Security Council to neutralize international sanctions against North Korea and provide large-scale economic support, North Korea will break away from international isolation and strengthen its strategic autonomy. Therefore, this study examines the historical process of change in North Korea-Russia relations and examines the main contents and implications of the 'Comprehensive Strategic Partnership Agreement' concluded after the North Korea-Russia summit in June 2024.

Correlation of Effective Dose and BMI in Radioiodine($^{131}I$) Therapy (방사성옥소($^{131}I$) 치료 시 유효선량과 체질량지수의 상관관계)

  • Purpose : The aim of this study was to predict radiation dose at 1 meter with BMI(body mass index) in thyroid cancer patients treated with radio-iodine and provide the efficient guideline in the management of patients. Methods : 140 patients from thyroidectomy for thyroid cancer were enrolled. All subjects under went 150 mCi radio-iodine therapy and performed whole body scan 1 week later. BMI(weight divided by square of height) was calculated to evaluate the amount of fatty tissue indirectly. The radiation dose at 1 meter was measured initially and on 2nd days. the relation of values with BMI were analyzed statically. As for the method of statistical analysis, using Med calc Version 9,2,2,0 Program. Results : (1) The initial effective dose was inversely correlated with the BMI. Significance level was 0.0004. (2) We obtained the following formula from the data of initial effective dose and BMI: Y = -30.91X + 350.4(${\mu}Sv/h$)(Y: initial radiation dose, x: Group). (3) After 21.55 hours, than radiation dose was less than those recommended by ICRP or NRC in 53% of the population. Conclusion : Using BMI, the initial radiation dose and 2nd days dose can be predicted in thyroid cancer patients before radio-iodine therapy. It may be used for predicting the time of discharge and control the isolation room. We were able to predict the radiation exposure after discharge using this calculated value.

Screening and Identification of a Cesium-tolerant Strain of Bacteria for Cesium Biosorption (환경유래의 세슘 저항성 균주 선별 및 세슘 흡착제거 연구)

  • One of the issues currently facing nuclear power plants is how to store spent nuclear waste materials which are contaminated with radionuclides such as $^{134}Cs$, $^{135}Cs$, and $^{137}Cs$. Bioremediation processes may offer a potent method of cleaning up radioactive cesium. However, there have only been limited reports on $Cs^+$ tolerant bacteria. In this study, we report the isolation and identification of $Cs^+$ tolerant bacteria in environmental soil and sediment. The resistant $Cs^+$ isolates were screened from enrichment cultures in R2A medium supplemented with 100 mM CsCl for 72 h, followed by microbial community analysis based on sequencing analysis from 16S rRNA gene clone libraries(NCBI's BlastN). The dominant Bacillus anthracis Roh-1 and B. cereus Roh-2 were successfully isolated from the cesium enrichment culture. Importantly, B. cereus Roh-2 is resistant to 30% more $Cs^+$ than is B. anthracis Roh-1 when treated with 50 mM CsCl. Growth experiments clearly demonstrated that the isolate had a higher tolerance to $Cs^+$. In addition, we investigated the adsorption of $0.2mg\;L^{-1}$ $Cs^+$ using B. anthracis Roh-1. The maximum $Cs^+$ biosorption capacity of B. anthracis Roh-1 was $2.01mg\;g^{-1}$ at pH 10. Thus, we show that $Cs^+$ tolerant bacterial isolates could be used for bioremediation of contaminated environments.
