Characterization of Paecilomyces variotii and Talaromyces amestolkiae in Korea Based on the Morphological Characteristics and Multigene Phylogenetic Analyses

  • During fungal diversity surveys of the order Eurotiales in Korea, two fungal strains, EML-DG33-1 and EML-NCP50, were isolated from samples of rat dung and fig tree leaf collected at a garden located in Gwangju in 2014. To complete the National Species List of Korea, it is a prerequisite to verify whether many questionable species, which were previously recorded but not confirmed, indeed present in Korea. Herein, the isolates were confirmed as undescribed species, Paecilomyces variotii and Talaromyces amestolkiae based on the combination of morphological and phylogenetic analyses of multigenes including the rDNA internal transcribed spacer, ${\beta}-tubulin$, and RNA polymerase II subunit 2.

Isolation and Characterization of Three Unrecorded Zygomycete Fungi in Korea: Cunninghamella bertholletiae, Cunninghamella echinulata, and Cunninghamella elegans

  • In a survey of undiscovered taxa in Korea, three zygomycete fungal strains-EML-W31, EML-HGD1-1, and EML-RUS1-1-were isolated from freshwater, grasshopper fecal, and soil samples in Korea. On the basis of the morphological characteristics and phylogenetic analysis of internal transcribed spacer and 28S rDNA, the isolates of EML-W31, EML-HGD1-1, and EML-RUS1-1 were confirmed to be Cunninghamella bertholletiae, Cunninghamella echinulata, and Cunninghamella elegans, respectively. These species have not been previously described in Korea.

Two Endophytic Diaporthe Species Isolated from the Leaves of Astragalus membranaceus in Korea

  • We characterized two endophyte fungi from the leaves of Astragalus membranaceus in Korea. The isolated strains were identified on the basis of the morphological characters and sequences analysis of the internal transcribed spacer and large subunit regions of the rDNA and ${\beta}$-tubulin gene. To the best of our knowledge, this is the first report of Diaporthe oncostoma and Diaporthe infecunda in Korea, and we have provided descriptions and figures.

First Report of Albifimbria verrucaria and Deconica coprophila (Syn: Psylocybe coprophila) from Field Soil in Korea

  • During a survey of fungal diversity in Korea, two fungal strains, KNU17-1 and KNU17-199, were isolated from paddy field soil in Yangpyeong and Sancheong, respectively, in Korea. These fungal isolates were analyzed based on their morphological characteristics and the molecular phylogenetic analysis of the internal transcribed spacer (ITS) rDNA sequences. On the basis of their morphology and phylogeny, KNU17-1 and KNU17-199 isolates were identified as Albifimbria verrucaria and Deconica coprophila, respectively. To the best of our knowledge, A. verrucaria and D. coprophila have not yet been reported in Korea. Thus, this is the first report of these species in Korea.

Seven New Records of Penicillium Species Belonging to Section Lanata-Divaricata in Korea

  • Penicillium species are known to be ubiquitous environmental saprophytes. In the survey of diversity of genus Penicillium, seven new records of Penicillium species belonging to section Lanata-Divaricata were isolated from freshwater and soil samples collected from different locations in Korea. Based on morphological characteristics and multilocus phylogenetic analysis of the rDNA internal transcribed spacer region (ITS), β-tubulin (BenA), and calmodulin (CaM) genes, the isolated strains were identified as P. annulatum, P. camponotum, P. echinulonalgiovense, P. globosum, P. limosum, P. onobense, and P. yunnanense, respectively. This study presents detailed phylogenetic analyses and morphological descriptions of these species that contribute to section Lanata-Divaricata in Korea.

  • While surveying undiscovered fungal taxa in Korea, three rare zygomycetous fungal strains, CNUFC-PTF2-1, CNUFC-TF3-1, and CNUFC-ESAF3-1, were isolated from soil, leaf, and freshwater samples, respectively. The strains were analyzed morphologically as well as phylogenetically based on the internal transcribed spacer region and 28S rDNA sequences. Sequence analysis of the two loci revealed that the isolates, CNUFC-PTF2-1, CNUFC-TF3-1, and CNUFC-ESAF3-1, were identified as Backusella circina, Circinella muscae, and Mucor ramosissimus, respectively. These species have not yet been previously described in Korea.

Diversity of Endophytic Fungi Isolated from Pinus densiflora and Juniperus rigida Distributed in Mt. Baekryeonsan and Mt. Johangsan, Korea

  • We investigated the biodiversity of endophytic fungi in Pinus densiflora and Junipreus rigida from Mt. Baekryeonsan and Mt. Johangsan in Korea. In total, 59 isolates were discovered from 26 host plants and identified using sequences from the rDNA internal transcribed spacer (ITS) region. Twenty taxa of endophytic fungi were identified in this study; 62% of isolates belonged to Leotiomycetes, 34% belonged to Sordariomyetes, and 4% belonged to Agaricomycetes. Results confirmed three previously unreported endophytic fungi in Korea: Phanerochaete concrescens, Creosphaeria sassafras, and Scopuloides rimosa. Community analysis showed that the fungal diversity patterns of the samples from Mt Baekryeonsan and Mt. Johangsan are similar to those from previous studies and varied depending upon host and locality.

First Records of Rare Ascomycete Fungi, Acrostalagmus luteoalbus, Bartalinia robillardoides, and Collariella carteri from Freshwater Samples in Korea

  • The distribution and occurrence of rare ascomycete fungi within freshwater samples in Korea was investigated. Three rare fungal strains, CNUFC-YR537-1, CNUFC-CNUP1-1, and CNUFC-NDR3-1, were isolated using serial dilution method. On the basis of their morphological characteristics and phylogenetic analysis of their internal transcribed spacer regions and 28S rDNA sequences, the three isolates were identified as Acrostalagmus luteoalbus, Bartalinia robillardoides, and Collariella carteri, respectively. To our knowledge, these are the first records of rare genera Acrostalagmus, Bartalinia, and Collariella from Korea, and the first reports of A. luteoalbus, B. robillardoides, and C. carteri from freshwater samples.

Four Unreported Endophytic Fungi Isolated from Roots of Quercus spp. in Korea

  • We isolated endophytic fungi from the roots of Quercus spp. in Korea. The isolates were identified based on morphological and molecular characteristics from the nucleotide sequences of the internal transcribed spacer and large subunit of rDNA regions. We confirmed that the four species we isolated - Oidiodendron citrinum, Acremonium crotocinigenum, Mammaria echinobotryoides, and Pleotrichocladium opacum - have not been recorded previously in Korea. Herein, we present their morphological characteristics and the result of phylogenetic analysis.

Morphology and Phylogeny of Two Novel Species within the Class Dothideomycetes Collected from Soil in Korea

  • Two fungal strains (KNU-JJ-1827 and KNU-JJ-1829) belonging to the class Dothideomycetes were discovered from Jeju Island, Korea during this investigation of soil microfungi. Strain KNU-JJ-1827 showed fewer conidial septations, larger conidiogenous cells, and smaller conidia compared to the previously identified closest species of Didymocyrtis. Strain KNU-JJ-1829 revealed the similar characteristics of the nearest certain species of the genus Parathyridaria with the production of conidiogenous cells and conidia, because no asexual morphs were detected from the closest type strain Parathyridaria rosae. The novelty of the strains was also confirmed by analyzing molecular data using internal transcribed spacer regions and 28S rDNA. The molecular phylogeny also strongly support the detailed description and illustration for each proposed species as Didymocyrtis septata sp. nov. (KNU-JJ-1827) and Parathyridaria ellipsoidea sp. nov. (KNU-JJ-1829) isolated from soil in Korea.
