Effects of stocking density on the homeostasis of uric acid and related liver and kidney functions in ducks

  • Objective: Stocking density (SD) is an important issue in the poultry industry, which is related to the production performance, intestinal health and immune status. In the present study, the effects of SD on the metabolism and homeostasis of uric acid as well as the related functions of the liver and kidney in ducks were examined. Methods: A total of 360 healthy 56-day-old Shan-ma ducks were randomly divided into the low stocking density (n = 60, density = 5 birds/m2), medium stocking density (n = 120, density = 10 birds/m2) and high stocking density groups (HSD; n = 180, density = 15 birds/m2). Samples were collected in the 3rd, 6th, and 9th weeks of the experiment for analysis. Results: The serum levels of uric acid, lipopolysaccharide and inflammatory cytokines (interleukin-1β [IL-1β], IL-8, and tumor necrosis factor-α [TNF-α]) were increased significantly in the HSD group. Serious histopathological lesions could be seen in both the livers and kidneys in the HSD group in the 9th week. The mRNA expression levels of inflammatory cytokines (IL-8 and TNF-α) and related pathway components (toll-like receptor 4, myeloid differentiation primary response gene 88, and nuclear factor-κB) were increased significantly in both the livers and kidneys in the HSD group. The mRNA expression levels of enzymes (adenosine deaminase, xanthine oxidase, phosphoribosyl pyrophosphate amidotransferase, and phosphoribosyl pyrophosphate synthetase 1) related to the synthesis of uric acid increased significantly in the livers in the HSD group. However, the mRNA expression level of solute carrier family 2 member 9, which plays an important role in the excretion of uric acid by the kidney, was decreased significantly in the kidneys in the HSD group. Conclusion: These results indicated that a higher SD could cause tissue inflammatory lesions in the liver and kidney and subsequently affect the metabolism and homeostasis of uric acid, and is helpful for guiding decisions related to the breeding and production of ducks.

Inhibition of JAK2/STAT3 and activation of caspase-9/3 are involved in KYS05090S-induced apoptosis in ovarian cancer cells

  • To overcome the poor prognosis of patients with ovarian cancer, attempting to target ovarian cancer with effective antitumor compounds has been conducted for numerous years. Although the 3,4-dihydroquinazoline derivative KYS05090S was known to exert antitumor effects in A549 and ovarian cancer cells by inhibition of T-type Ca2+ channels, the complete underlying antitumor mechanism of this compound remains unclear. Thus, in the present study, the potential apoptotic mechanism of KYS05090S was elucidated in SKOV3 and OVCAR3 ovarian cancer cells. KYS05090S exerted significant cytotoxicity in SKOV3 and OVCAR3 ovarian cancer cells, and also increased the number of apoptotic bodies, and the number of terminal deoxynucleotidyl transferase dUTP nick end labeling positive cells and the sub-G1 population as a feature of apoptosis. Consistently, KYS05090S induced cleavage of poly(ADP-ribose) polymerase and caspase-9/3 in ovarian cancer cells. Notably, KYS05090S attenuated the expression of anti-apoptotic proteins, including cyclin D1 and B-cell lymphoma-2 (Bcl-2), and reduced the phosphorylation of Janus kinase 2 (JAK2) and signal transducer and activator of transcription 3 (STAT3) in ovarian cancer cells. Additionally, KYS05090S blocked the nuclear translocation of STAT3 and suppressed the signaling of JAK2/STAT3 in interleukin-6-treated SKOV3 cells, as a STAT3 activator. Overall, these observations indicated that inhibition of JAK2/STAT3 signaling and activation of caspase-9/3 are critically involved in the effects of KYS05090S on apoptosis in ovarian cancer types, and the compound may be beneficial as a potent antitumor agent.

E-ray를 조사한 쥐의 피부에서 증식된 keratinocyte에 의한 TGF-β1 발현 (TGF-β1 Expression by Proliferated Keratinocytes in the Skin of E-Irradiated Mice)

  • 우리는 방사선피부염 동물 모델을 확립하여, 이차 면역 기관에서의 면역 세포 비율 변화를 관찰하였다. 또한, 방사선 조사에 의한 병소에서 transforming growth factor-${\beta}1$ (TGF-${\beta}1$)과 interlukin-10 (IL-10)의 발현을 증가시킨 세포를 분석하였다. Hairless-1 (HR-1) 쥐의 posterior dorsal 부위에 6 일간 매일 10 Gy 씩 electron (E)-ray를 국부 조사하여 방사선피부염 모델을 만들었다. FACS를 이용하여 면역 세포 비율의 변화를 분석한 결과 비장과 림프절에 존재하는 항원제시세포와 T 세포 및 B 세포들의 비율이 E-irradiation에 의해 영향을 받았다. 피부에서 세포 특이적인 마커와 사이토카인들의 발현 양상은 면역형광염색법으로 확인하였다. 방사선 조사 후, TGF-${\beta}1$과 interlukin-17 (IL-17)은 regulatory T 세포(Treg)보다 keratin-14 (K-14)를 발현하는 진피의 끝부분에서 높게 발현되었다. Interlukin-10 (IL-10)는 Treg 뿐만 아니라 T helper 17 (Th17) 세포, dendritic 세포, macrophage 중 어느 것과도 같은 위치에서 검출되지 않았다. 우리의 데이터는 방사선피부염 동물 모델의 병소 안에서, TGF-${\beta}1$이 증식된 keratinocyte에 과발현된다는 것을 나타낸다.

수삼추출물 첨가 혼합배지에서 조제된 상황 균사체 심부배양물 다당획분의 면역활성 증진 (Immune Enhancement of Polysaccharide from Submerged Culture with Phellinus linteus in the Medium Supplemented with Ginseng Extract)

  • 면역활성을 증진시키기 위하여 수삼추출물(GE, $65^{\circ}$Bx)이 첨가된 액체배지(mushroom complete medium, MCM)에서 상황(Phellinus linteus, PL) 균사체를 배양하여 배양액과 균사체가 모두 포함된 심부배양물을 조제하고 조다당획분(CP)으로 분획하였다. MCM 부피에 대하여 GE가 15% 첨가된 혼합 액체배지에서 배양된 수삼추출물-상황 심부배양물로부터 분획된 PL-GE-15-CP는 $100{\mu}g$/mL의 시료농도에서 GE-5와 10%가 첨가된 PL-GE-5와 10-CP 및 GE가 첨가되지 않은 일반-상황 심부배양물의 PL-CP보다 유의적으로 높은 마크로파지(saline 대조군의 1.45배) 및 Peyer's patch를 경유한 장관면역 활성(1.46배)을 확인할 수 있었다. 활성획분인 PL-GE-15-CP는 DEAE-Sepharose CL-6B를 이용한 분획을 통하여 PL-GE-15-CP의 다른 획분 또는 시료대조군인 PL-CP로부터 분획된 모든 획분보다 유의적으로 높은 마크로파지 활성, IL-12 생산능 및 장관면역 활성(각각 1.54, 3.96과 1.56배)을 갖는 다당획분인 PL-GE-15-CP-II를 분리할 수 있었다. 또한, PL-GE-15-CP-II는 동일 NaCl 농도에서 분획된 시료대조군인 PL-CP-II보다 시료농도 $200{\mu}g$/mouse에서 colon 26-M3.1 carcinoma cell에 대하여 유의적으로 높은 암전이 억제활성도 보여주었다(대조군의 57.3% 억제활성). 한편, 활성 다당획분인 PL-GE-15-CP-II는 주로 중성당(82.45%)과 산성당(12.99%)으로 구성되어 있었으며 구성당 분석결과에서 산성당과 함께 Ara, Man, Gal과 Glc의 중성당으로 구성되어져 있음을 확인할수 있었다(molar ratio; 0.52:0.97:0.63:1.00:0.54). 결론적으로 GE가 첨가된 상황 심부배양물은 일반배지에서의 상황 심부배양물보다 면역활성이 높아 GE가 균사체 활성의 증진에 영향을 주고 있는 것으로 생각되며, 특히 활성성분으로 다당류가 중요하게 관여하고 있는 것으로 사료된다.

산소 장력이 치주인대에서 유래한 세포의 활성과 기능에 미치는 영향 (EFFECT OF OXYGEN TENSION ON THE ACTIVITY AND FUNCTION OF THE CELLS DERIVED FROM HUMAN PERIODONTAL LIGAMENT)

  • 본 연구는 산소 장력이 사람의 치주인대에서 유래된 세포의 활성과 세포외 기질 및 cytokine에 미치는 영향을 알아 보고자 저산소 및 과산소 조건에서 배양한 후 세포의 활성, 총 단백질 합성, 교원질 합성, 그리고 $IL-1{\beta}$, IL-6, $TNF-{\alpha}$를 측정하였다. 교정치료를 위해 발거한 제 1 소구치에서 사람의 치주인대 세포를 채취하여 $37^{\circ}C,\;5\%\;CO_2,\100\%$ 습도의 환경에서 배양한 후 5 내지 6 계대배양하여 실험에 사용하였다. Gaspack system에 $0.2{\mu}m$ Millipore filter를 부착하여 세균 감염을 막았고, 저산소군에는 $10\%\;O_2,\;5\%\;CO_2,\;85\%\;N_2$ 가스를, 과산소군에는 $90\%\;O_2,\;5\%\;CO_2,\;5\%\;N_2$ 가스를 연결하여 산소 장력에 변화를 주었으며 대조군에는 $5\%\;CO_2,\;95\%$ 공기를 공급하였다. $37^{\circ}C$ 에서 2, 4, 6일간 배양한 후에 세포활성을 측정하기 위해 tetrazolium(MTT) assay를 시행하였고 sulforhodamine B(SRB) assay를 이용하여 총 단백질 합성을 알아보았으며, 4-hydroxyproline 측정으로 교원질 합성을 측정해 보았다. 또한 효소면역측정법으로 $IL-1{\beta}$, IL-6, $TNF-{\alpha}$를 측정하여 다음과 같은 결과를 얻었다. 1. 저산소근에저 세포 활성과 총 단백질 합성은 대조군보다 다소 높거나 같은 정도이었다. 2. 과산소군에서 세포 활성은 대조군보다 낮았으며 총 단백질 합성도 다소 감소되었다. 3. 교원질 합성은 저산소군과 과산소군에서 대조군보다 유의하게 감소되었다가 배양기간이 길어짐에 따라 증가되어 대조군과 유사한 정도에 달했다. 4. 효소면역법 측정 결과 IL-6, $TNF-{\alpha},\;IL-1{\beta}$ 순으로 다량 검출되었다. 5. IL-6, $TNF-{\alpha},\;IL-1{\beta}$는 저산소군과 과산소군에서 배양기간이 길어짐에 따라 대조군에 비해 급격히 증가하였다. 5. IL-6, $TNF-{\alpha}$는 과산소 조건에서 배양 6일 후에는 대조군에 비해 유의하게 증가하였으며 이는 배양 2일 또는 4일 후보다도 유의하게 증가한 것이었다.

Skate cartilage extracts containing chondroitin sulfate ameliorates hyperlipidemia-induced inflammation and oxidative stress in high cholesterol diet-fed LDL receptor knockout mice in comparison with shark chondroitin sulfate

  • BACKGROUND/OBJECTIVES: In this study, we investigated the beneficial effects of skate cartilage extracts containing chondroitin sulfate (SCS) on hyperlipidemia-induced inflammation and oxidative stress in high cholesterol diet (HCD)-fed mice in comparison with the effects of shark cartilage-derived chondroitin sulfate (CS). MATERIALS/METHODS: Low-density lipoprotein receptor knockout (LDLR-KO) mice were fed HCD with an oral administration of CS (50 and 100 mg/kg BW/day), SCS (100 and 200 mg/kg BW/day), or water, respectively, for ten weeks. RESULTS: The administration of CS or SCS reduced the levels of serum triglyceride (TG), total cholesterol (TC), and LDL cholesterol and elevated the levels of high-density lipoprotein cholesterol, compared with those of the control group (P < 0.05). Furthermore, CS or SCS significantly attenuated inflammation by reducing the serum levels of interleukin (IL)-1β and hepatic protein expression levels of nuclear factor kappa B, inducible nitric oxide synthase, cyclooxygenase-2, and IL-1beta (P < 0.05). In particular, the serum level of tumor necrosis factor-alpha was reduced only in the 100 mg/kg BW/day of SCS-fed group, whereas the IL-6 level was reduced in the 100 and 200 mg/kg BW/day of SCS-fed groups (P < 0.05). In addition, lipid peroxidation and nitric oxide production were attenuated in the livers of the CS and SCS groups mediated by the upregulation of hepatic proteins of antioxidant enzymes, such as superoxide dismutase, catalase, and glutathione peroxidase (P < 0.05). CONCLUSIONS: These results suggest that the biological effects of SCS, similar to those of CS, are attributed to improved lipid profiles as well as suppressed inflammation and oxidative stress induced by the intake of HCD.

사백산(瀉白散)과 사백산가사삼(瀉白散加沙蔘)이 인간 기관지상피세포의 IL-6, IL-8 및 GM-CSF 발현에 미치는 영향 (The Inhibitory Effects of Sabaek-san and Sabaeksan plus Sasam on the IL-6, IL-8 and GM-CSF mRNA Levels in Human Epithelial Cells)

  • Objective : We aimed to identify the dose-dependent inhibitory effects of Sabaek-san(瀉白散) and Sabaeksan plus Sasam(Adenophorae Radix) 瀉白散加沙蔘) on the mRNA expressions of Interleukin(IL)-6, IL-8 and granulocyte macrophage colony stimulating factor(GM-CSF) involved in the asthma model. Materials and Methods : Through this study, BEAS-2B cell lines, human epithelial cells were used. These cells were stimulated by tumor necrosis factor(TNF)-${\alpha}$, IL-1${\beta}$ and histamine for artificial inflammatory expression. ${\beta}$-action messenger RNA(mRNA) was used for standards. After each 24hours of Sabaeksan and Sabaeksan plus Sasam treatment, total cellular RNAs were collected by treating RNA zol directly on living cells, Then the transcriptional activities of IL-6, 8 and GM-CSF were measured by RT-PCR with electrophoresis, Results : The mRNA expressions of IL-6 are significantly inhibited compared to those of controlled group at 40 and 100ug/ml of Sabaeksan extract and $100{\mu}g/ml$ of Sabaeksan plus Sasam extract (p<0.05). The mRNA expressions of IL-8 are significantly inhibited compared to that of controlled group at 2.40 and 100 ug/ml of Sabaeksan extract and $40.100{\mu}g/ml$ of Sabaeksan plus Sasam extract(p<0.05) THe mRNA expressions of GM-CSF are significantly inhibited compared to those of the controlled group at $100{\mu}g/ml$ of Sabaeksan extract adn $40.100{\mu}g/ml$ of Sabaeksan plus Sasam extract.(p<0.05) Conclusions : This study shows that Sabaeksan and Sabaeksan plus Sasam have dose-dependent inhibitory effects on the mRNA expressions of IL-6, IL-8 and GM-CSF in human epithelial cells. Therefore, these types of herb medicine may inhibit the inflammatory process of asthma. Advanced studies are required to investigate the mechanisms of inhibition by herb medicine in the asthma model.

해조가 2,4,6-trinitrobenzene-sulfonic acid로 유발된 염증성 장질환 동물모델에 미치는 영향 (Effects of Sargassumpallidum on 2,4,6-Trinitrobenzene Sulfonic Acid-Induced Colitis in Mice)

  • Objectives : The aim of the current study was to investigate the effects of Sargassum (Sargassum pallidum (TURN.) C. AG.; SP) on the experimental colitis induced by 2,4,6-trinitrobenzene sulfonic acid (TNBS) in mice. Methods : ICR mice were divided into 7 groups (NOR, CON, $SS50\times5$, $SP20\times3$, $SP50\times3$, $SP20\times5$, $SP50\times5$). TNBS processing was intrarectally applied to all experimental groups on the 3rd experiment day, except the normal group (NOR). For investigating the prophylactic effect, SP at doses of 20 mg/kg ($SP20\times5$) and 50 mg/kg ($SP50\times5$) were orally administered for 5 days. The SP at doses of 20 mg/kg ($SP20\times3$) and 50 mg/kg ($SP50\times3$) were orally administered for 3 days after the colitis induction in order to check the effect of treatment. As a positive control group, sulfasalazine 50 mg/kg ($SS50\times5$) was administrated. Macroscopic findings of epithelial tissue on mice were measured by colon length and macroscopic score. Histologic findings were also checked by crypt cell, epithelial cell, inflammatory cell and edema of submucosa. We measured the ability of SP to inhibit lipid peroxidation and myeloperoxidase activity. We also measured levels of the inflammatory markers, interleukin (IL)-$1\beta$ and cyclooxygenase-2 (COX-2), its transcription factor activation, phospho-NF-${\kappa}B$ (pp65), in the colon by enzyme-linked immunosorbent assay and immunoblot analysis. We measured activation of fecal bacterial enzyme, $\beta$-glucuronidase and degradation activation of fecal glycosaminoglycan (GAG), and hyaluronic acid. Results : Oral administration of SP on mice inhibited TNBS-induced colon shortening and myeloperoxidase activity in the colon of mice as well as IL-$1\beta$ and COX-2 expression. SP also inhibited TNBS-induced lipid peroxidation and pp65 activation in the colon of mice. SP inhibited $\beta$-glucuronidase activation and fecal hyaluronic acid degradation activation as well. Conclusions : SP could be a possible herbal candidate and preventive prebiotic agent for treating inflammatory bowel disease (IBD). Further experiments to differentiate effects of SP on IBD, such as other solutions and extracting times, might be promising.

Modulation of Nrf2/ARE and Inflammatory Signaling Pathways by Hericium erinaceus Mycelia Extract

  • Hericium erinaceus is an edible mushroom used as a medicinal food in Asian countries. In this study, the chemopreventive effects of H. erinaceus mycelia hot water extract (HEW) were evaluated. HEW remarkably induced the luciferase activity of the antioxidant response element (ARE), located in the promoter region of phase 2 and antioxidant genes and regulated by nuclear factor E2-related factor 2 (Nrf2). The up-regulation of ARE activity by HEW corresponded with the induction of Nrf2 and the antioxidant enzyme, hemeoxygenase-1. The inhibition of cyclooxygenase-2 (COX-2) activity is a promising effective approach in cancer chemoprevention, and HEW prominently suppressed COX-2 protein expression in HepG2 cells. Furthermore, HEW showed anti-inflammatory activity by modulating inflammatory mediators such as nitric oxide (NO), inducible NO synthase, tumor necrosis factor-${\alpha}$, interleukin-$1{\beta}$, and the transcription factor, nuclear factor-${\kappa}B$, in lipopolysaccharide-stimulated RAW 264.7 cells. These results suggest that H. erinaceus possessed anti-tumor and anti-inflammatory effects via the modulation of Nrf2/ARE and inflammatory signaling pathways, and may therefore have potential use as a natural chemopreventive agent.

Black ginseng-enriched Chong-Myung-Tang extracts improve spatial learning behavior in rats and elicit anti-inflammatory effects in vitro

  • Background: Chong-Myung-Tang (CMT) extract is widely used in Korea as a traditional herbal tonic for increasing memory capacity in high-school students and also for numerous body ailments since centuries. The use of CMT to improve the learning capacity has been attributed to various plant constituents, especially black ginseng, in it. Therefore, in this study, we have first investigated whether black ginseng-enriched CMT extracts affected spatial learning using the Morris water maze (MWM) test. Their molecular mechanism of action underlying improvement of learning and memory was examined in vitro. Methods: We used two types of black ginseng-enriched CMT extracts, designated as CM-1 and CM-2, and evaluated their efficacy in the MWM test for spatial learning behavior and their anti-inflammatory effects in BV2 microglial cells. Results: Our results show that both black ginseng-enriched CMT extracts improved the learning behavior in scopolamine-induced impairment in the water maze test. Moreover, these extracts also inhibited nitric oxide production in BV2 cells, with significant suppression of expression of proinflammatory cytokines, especially inducible nitric oxide synthase, cyclooxygenase-2, and $interleukin-1{\beta}$. The protein expression of mitogen-activated protein kinase and nuclear $factor-{\kappa}B$ pathway factors was also diminished by black ginseng-enriched CMT extracts, indicating that it not only improves the memory impairment, but also acts a potent anti-inflammatory agent for neuroinflammatory diseases. Conclusion: Our research for the first time provides the scientific evidence that consumption of black ginseng-enriched CMT extract as a brain tonic improves memory impairment. Thus, our study results can be taken as a reference for future neurobehavioral studies.
