The effect of Oligonol intake on cortisol and related cytokines in healthy young men

  • This study investigated the effects of Oligonol intake on cortisol, interleukin (IL)-$1{\beta}$, and IL-6 concentrations in the serum at rest and after physical exercise loading. Nineteen healthy sedentary male volunteers participated in this study. The physical characteristics of the subjects were: a mean height of $174.2{\pm}2.7$ cm, a mean weight of $74.8{\pm}3.6$ kg and a mean age of $22.8{\pm}1.3$ years. Each subject received 0.5 L water with Oligonol (100 mg/day) (n = 10) or a placebo (n = 9) daily for four weeks. The body composition, the white blood cell (WBC) and differential counts as well as the serum cortisol, IL-$1{\beta}$, and IL-6 concentrations were measured before and after Oligonol intake. The cortisol concentration and serum levels of IL-$1{\beta}$ and IL-6 after Oligonol intake were significantly decreased compared to before treatment (P < 0.01, respectively). In addition, the rate of increase of these factors after exercise was decreased compared to the placebo group. There was no change in the WBC and differential cell counts. These results suggest that oral Oligonol intake for four weeks had a significant effect on inhibition of inflammatory markers in healthy young men.

Effects of Interleukin-1${\beta}$ and Tumor Necrosis $Factor-{\alpha}$ on the Release of Collagenase and Gelatinase from Osteoblasts

  • A large number of factors such as osteotropic hormones, cytokines, or growth factors are related to the bone remodeling which is characterized by the coupling of osteoclast-mediated bone resorption and osteoblast-mediated bone formation. Recent investigations have indicated that cytokines such as $interleukin-1{\beta}\;(IL-1{\beta})$ and tumor necrosis $factor-{\alpha}\;(TNF-{\alpha})$ play a potential role in the bone resorption associated with a variety of pathological conditions such as inflammatory osteolytic disease. Collagen is the most abundant protein of the extracellular matrix of bone, and the participation of collagenase in bone resorption has been widely investigated. In this study, effects of $IL-1{\beta}$ and $TNF-{\alpha}$ on the release of collagenase from osteoblastic cells were measured. The gelatinase activity was also measured by gel substrate analysis (zymography) after electrophoresis of conditioned media of osteoblastic cell culture. $IL-1{\beta}$ increased the collagenase activity in ROS17/2.8 and HOS cell culture. $TNF-{\alpha}$ also increased the collagenase activity of osteoblastic cells. When two kinds of cytokines were treated simultaneously in the culture of osteoblastic cells, synergistic increase of collagenase activity was seen in ROS17/2.8 cells. $IL-1{\beta}$ and $TNF-{\alpha}$ significantly increased the collagenase activity after 6 hour treatment in the osteoblastic cell culture, and there was no additional increase according to the culture period. Osteoblastic cells released the gelatinase and molecular weight of this enzyme was measured about 70 KDa as assessed by zymogram. $IL-1{\beta}$ and $TNF-{\alpha}$ showed increase of the gelatinase activity produced by ROS17/2.8 and HOS cells. Taken together, this study suggested that $IL-1{\beta}$ and $TNF-{\alpha}$ can modulate bone metabolism, at least in part, by increased release of collagenase and gelatinase from osteoblasts.

Regulation of Interleukin-1${\beta}$-induced Dedifferentiation and Apoptosis via p38 Mitogen-activated Protein Kinase Pathway in Articular Chondnocytes (연골세포의 탈분화 및 세포고사 억제를 위한 기전연구)

  • Objectives : Interleukin-1 (IL-1)${\beta}$ in articular chondrocytes regulates differentiation, apoptosis, and inflammatory responses. It is still controversial, So, we investigated IL- $1{\beta}$ induces chondrocytes dedifferentiation and death. Also, we studied the role of the mitogen-activated protein kinase (MAPK) subtypes on IL-$1{\beta}$-induced dedifferentiation and apoptosis. Methods : To evaluation of dedifferentiation by chemokines of chondrocytes, we assessed such as proteoglycan, collagen, MMP-3 and MMP-13 by RT-PCR analysis. Also, to assess of apoptosis effect by chemokines, we measured annexin V/propidium iodode (PI) and sub G1 cells in chondrocytes by flowcytometric analysis Results : IL-$1{\beta}$ treatment did not affect activation of ERK-1/2, but stimulation of p38 kinase. Inhibition of phospho ERK-1/2 with PD98059 enhanced IL-1b-induced dedifferentiation, and apoptosis up to 13.5%, whereas inhibition of phospho p38 kinase with SB203580 inhibited dedifferentiation, and apoptosis. Conclusions : Our results indicate that SB203580, p38 kinase inhibitor, inhibits IL-$1{\beta}$-induced dedifferentiation, and apoptosis by the inhibition of type II collagen expression and proteoglycan synthesis of rabbit articular chondrocytes.

The Effect of Jeongshin-tang on Interleukin-1 $\beta$ and $\beta$-Amyloid-Induced Cytokine Production in Human Brain Astrocytes

  • Jeongshin-tang (JST) is a Korean herbal prescription, which has been successfully applied for the various neuronal diseases. However, it's effect remains unknown in experimental models. To investigate the biological effect of JST in Alzheimer's disease (AD) in vitro model, we analized the production of interleukin (IL)-6 and IL-8, and expression of cyclooxygenase (COX)-2 in IL-1β plus β-amyloid [25-35] fragment (A)-stimulated human astrocytoma cell line U373MG. JST alone had no effect on the cell viability. The production of IL-6 and IL-8 was significantly inhibited by pretreatment with JST (1mg/㎖) on IL-1β plus A-stimulated U373MG cells. Maximal inhibition rate of IL-6 and IL-8 production by JST was about 41.22% (P<0.01) and 34.45% (P<0.05), respectively. The expression level of COX-2 protein was up-regulated by IL-1β plus A but the increased level of COX-2 was inhibited by pretreatment with JST (1 mg/㎖). These data indicate that JST has a regulatory effect on cytokine production and COX-2 expression, which might explain it's beneficial effect in the treatment of AD.

Sweating by Exercise Controls Body Temperature through Increase of Interleukin-1beta

  • This study was designed to investigate the expression and production of interleukin-1beta $(IL-1{\beta})$ in human peripheral blood of trained runners and untrained controls after temporary moderate intensity exercise. Male long-distance trained runners (TR) and untrained sedentary control subjects (SED) ran for 1 h at 70% of heart rate reserve (HRR). $IL-1{\beta}$ gene and protein expressions were significantly higher in TR than those with SED at all 3 intervals examined independently. Significant increases in total sweat volume and oral temperature were observed after exercise in both groups, however, there were some differences between the groups. We conclude, therefore, that sweating due to exercise is associated with increase of $IL-1{\beta}$ and it is correlated with decrease of oral temperature.

Effects of gangliosides from deer bone extract on the gene expressions of matrix metalloproteinases and collagen type II in interleukin-1β-induced osteoarthritic chondrocytes

  • BACKGROUND/OBJECTIVES: We investigated the anti-osteoarthritic effects of deer bone extract on the gene expressions of matrix metalloproteinases (MMPs) and collagen type II (COL2) in interleukin-$1{\beta}$-induced osteoarthritis (OA) chondrocytes. MATERIALS/METHODS: Primary rabbit chondrocytes were treated as follows: CON (PBS treatment), NC (IL-$1{\beta}$ treatment), PC (IL-$1{\beta}+100{\mu}g/mL$ glucosamine sulphate/chondroitin sulphate mixture), and DB (IL-$1{\beta}+100{\mu}g/mL$ deer bone extract). RESULTS: The results of the cell viability assay indicated that deer bone extract at doses ranging from 100 to $500{\mu}g/mL$ inhibits cell death in chondrocytes induced by IL-$1{\beta}$. Deer bone extract was able to significantly recover the mRNA expression of COL2 that was down-regulated by IL-$1{\beta}$ (NC: 0.79 vs. DB: 0.87, P < 0.05) and significantly decrease the mRNA expression of MMP-3 (NC: 2.24 vs. DB: 1.75) and -13 (NC: 1.28 vs. DB: 0.89) in OA chondrocytes (P < 0.05).CONCLUSIONS: We concluded that deer bone extract induces accumulation of COL2 through the down-regulation of MMPs in IL-$1{\beta}$-induced OA chondrocytes. Our results suggest that deer bone extract, which contains various components related to OA, including chondroitin sulphate, may possess anti-osteoarthritic properties and be of value in inhibiting the pathogenesis of OA.

  • I) 지금까지 만성골수성백혈병의 치료법으로서 골수이식이나 인터페론의 투여등이 시행되어왔으며 백혈병세포에서 Interleukin-1$\beta$(이하 IL-1$\beta$)이 자율적으로 생성됨이 보고 되어 이러한 질병의 진행에 IL-1$\beta$의 활성증가가 관련될것이라는 견해가 대두되어 왔다. 최근 단핵구성백혈병 환자의 소변에서 분리된 IL-1수용체 길항제(1L-1 receptor antagonist: IL-lRA)가 clonig되었고 인터페론치료에 저항하는 환자들의 치료에 IL-1RA 가 이용될수 있을것이라는 견해가 있다. 따라서 본 연구는 유전공학연구소가 분리 정제한 IL-1RA가 만성골수백혈병 환자로부터 유래한 K562세포주의 증식에 미치는 영향을 평가하고 알파인터페론과의 병용투여시의 상호작용에 관해 검토하였다.

Effect of Foeniculum Vulgare extract on inflammatory response in lipopolysaccharide exposed rats (회향(茴香) 추출물이 LPS를 처리한 Rat의 염증반응에 미치는 영향)

  • Objectives : This study was performed to investigate the effects of Foeniculum Vulgare(FV) extract on the anti-inflammatory of lipopolysaccharide(LPS) exposed rats. Methods : We divided LPS exposed Sprague-Dawley rats into 4 groups. They were control group, feed with 100 mg/kg, 200 mg/kg, 300 mg/kg FV groups. They were administered for 6 weeks. We measured the concentration of plasma interleukin-1${\beta}$(IL-1${\beta}$), plasma interleukin-6(IL-6), plasma tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), plasma interleukin-10(IL-10), the concentration of liver IL-1${\beta}$, IL-6, TNF-${\alpha}$ and IL-10. Results : 1. Plasma IL-1${\beta}$, plasma IL-6 and plasma TNF-${\alpha}$ concentration increased rapidly at 2hours after LPS injection and maintained high levels at 5hours after LPS injection. The concentration of these cytokines in the FV extract groups showed lower values than control group(P<0.05). 2. The concentration of Plasma IL-10 in FV extract groups showed higher values than control group at all times(P<0.05). 3. The concentration of liver IL-1${\beta}$ and IL-6 in FV extract groups showed lower values than control group(P<0.05). The concentration of liver TNF-${\alpha}$ in FV extract groups showed a tendency to decrease and that of liver IL-10 in FV extract groups showed a tendency to increase; however, these values showed no significantly different. Conclusions : In inflammatory response by LPS derivation, the FV gives positive effect.

Anti-inflammatory Effect of Artemisia Capillaris Thunberg in Lipopolysaccharide-exposed Rats (인진호(茵蔯蒿)가 LPS 염증유발 흰쥐의 전염증성 cytokine 생산 및 혈액성상에 미치는 영향)

  • Objectives : The present study investigated anti-inflammatory effect of Artemisia Capilaris Thunberg in lipopolysaccharide-exposed rats. Methods : We divided lipopolysaccharide-exposed Sprague-Dawley rats into 4 groups. They were normal group, feed with 100 mg/kg Artemisia Capillaris Thunberg group, feed with 200 mg/kg Artemisia Capillaris Thunberg group and feed with 300 mg/kg Artemisia capilaris Thunberg group. They were administered for 6 weeks. We measured counts of red blood cell(RBC), the values of hemoglobin(Hb) and packed cell volume(PCV), plasma total protein concentration, albumin concentration, the ratio of albumin/globulin, the activities of plasma glutamic oxaloacetic transaminase(GOT), glutamic pyruvic transaminase(GPT), lactate dehydrogenase(LDH), the counts of white blood cell(WBC), the ratio of neutrophils, lymphocytes, monocytes, basophils, eosinophils, the concentration of plasma interleukin-$1{\beta}$($IL-1{\beta}$), plasma interleukin-6(IL-6), plasma tumor necrosis factor-$\alpha$($TNF-{\alpha}$), plasma interleukin-10(IL-10), the concentration of liver $IL-1{\beta}$ and IL-6, $TNF-{\alpha}$, IL-10. Results : Counts of RBC and the values of Hb and PCV, plasma total protein concentration and albumin concentration, the activities of plasma GOT, GPT and LDH showed no significant difference in the treatment groups. and the ratio of albumin/globulin was increased in Artemisia Capillaris Thunberg groups. The counts of WBC showed lower values in Artemisia Capillaris Thunberg groups than those of control group, In the ratio of neutrophils Thunberg groups. The ration of monocytes, basophils and eosinophils were below 5%, and showed no characteristic trend. The concentration of plasma interleukin-$1{\beta}$($IL-1{\beta}$), plasma inerleukin-6(IL-6) and plasma tumor necrosis factor-$\alpha$($TNF-{\alpha}$) showed a lower values in the Artemisia Capillaris Thunberg groups than those of control group, and the concentration of plasma interleukin-10(IL-10) showed no significant difference in the treatment groups. The concentration of liver $IL-1{\beta}$ and IL-6 showed a lower values in the Artemisia Capillaris Thunberg groups than those of control group, however the concentration of liver $TNF-{\alpha}$ and IL-10 showed no significant difference in the treatment groups. Conclusions : The Artemisia Capillaris Thunberg groups gives positive results of anti-inflammatory response by lipopolysaccharide(LPS) derivation.

  • The author measured IL-8 and $TGF-{\beta}1$ levels of 84 ears - 48 ears of them had treated by antibiotics, 36 of them by Kamihyunggyeyungyotang(KHY) - of pediatric recurrent otitis media with effusion using ELISA assay, and compared them. The results were obtained as follows. 1. The level of IL-8 in KHY group was significantly lower than that in antibiotics group(p<0.05). 2. The level of $TGF-{\beta}1$ in KHY group was lower than that in antibiotics group. According to above results, KHY is considered to be used for treating recurrent otitis media with effusion by controlling the production of interleukin-8 and transforming $growthfactor-{\beta}1$.
