Molecular Markers and Their Application in Mulberry Breeding

  • Mulberry (Morus spp.) is an economically important tree crop being cultivated in India, China and other sericulturally important countries for its foliage to feed the silk producing insect Bombyx mori L. Genetic improvements of mulberry lag behind to the same in many other economically less important crops due to the complexity of its genetics, the breeding behavior, and the lack of basic information on factors governing important agronomic traits. In this review, the general usage and advantages of different molecular markers including isoenzymes, RFLPs, RAPDs, ISSRs, SSRs, AFLPs and SNPs are described to enlighten their applicability in mulberry genetic improvement programs. Application of DNA markers in germplasm characterization, construction of genetic linkage maps, QTL identification and in marker-assisted selection was also described along with its present status and future prospects.

Identification and Characterization of Ligninolytic Enzyme by Serratia marcescens HY-5 isolated from the Gut of Insect

  • A lignin degradation bacteria, symbiotic bacteria was isolated from the gut of Sympetrum depressiusculum and tested for its lignin degrading activity using lignin model compounds and related aromatic compounds. The strain was identified as Serratia marcescens HY-5 based on the 165 rDNA, cellular fatty acid composition, biochemical and physiological characteristics. S. marcescens showed 40-50% lignin degrading activity in the media that contained vaillin, guaiacol and dealkaline lignin. S. marcescens showed three ligninase activities [Jaccase, lignin peroxidase(LiP) and Manganase peroxidase(MnP)]. Addition of dealkaline lignin to the basal media increased about 6fold of laccase activity. Vanillic acid or vanillin increase 1.3fold of MnP activity and p-coumaric acid increased 12fold of LiP activity which added to the basal medium.

Current Taxonomical Status of Korean Auricularia Species

  • Auricularia (Auriculariaceae, Basidiomycota) is well-known jelly fungi, and most species are cultivated or utilized medicinally around the world. A total of 127 Auricularia specimens were collected during Korean mushroom forays by the Korea National Arboretum from 2008 to 2020. In this investigation, fungal barcode analysis and morphological examinations of 33 specimens were conducted and five Korean Auricularia species were identified. Of the species identified, A. americana, A. heimuer, and A. villosula were new to Korea, while A. auricula-judae which was previously identified in Korea, was not found. Herein, we discuss the Korean Auricularia species and provide a taxonomic key for their morphological identification.

GC-MS and GC-FID Analysis of Citronella Oil Products for Indicator Ingredient Identification

  • Citronella oil, an essential oil extracted through steam distillation from the leaves and stems of Cymbopogon, is a natural complex substance (NCS) regulated by the Korean government for its use in insect repellents. The component ratios of NCSs like citronella oil vary due to differences in manufacturing processes and origins, presenting a challenge in identifying and quantifying these substances in consumer chemical products. This study analysed ten commercially available products of the most commonly used types of citronella oil, specifically Java and Ceylon types, using gas chromatography (GC)-mass spectrometry (MS) and GC with flame ionization detection (FID). Through chromatographic data, we aimed to determine the components that can qualitatively identify citronella oil and the indicator ingredients that can be used for content analysis.

Morphological Differences between Larvae of the Oriental Fruit Moth (Grapholita molesta Busck) and the Peach Fruit Moth (Carposina sasakii Matsumura) in Korea

  • The oriental fruit moth (Grapholita molesta Busck) and the peach fruit moth (Carposina sasakii Matsumura) are the most severe insect pests affecting apple orchards in Korea. To prevent an outbreak of these two species and to control these agricultural insect pests, it is important to identify them accurately. However, it is hard to classify them when they were in the larval stage since they tunnel into the apple fruit. In this study, surface structures of the two species of larvae were observed using stereo microscope and scanning electron microscope. Distinct differences between the two species of larvae were found. The prothorax spiracles of oriental fruit moth larvae were approximately twice as large as those of peach fruit moth larvae. The arrangements of subventral setae, located around the proleg, were different between oriental fruit moth and peach fruit moth larvae. Furthermore, subdorsal setae of oriental fruit moth were located next to the spiracle on the 8th abdominal segment, while that of peach fruit moth was located above the spiracle. The identification of the two species of larvae observed in this study was confirmed using polymerase chain reaction-restriction fragment length polymorphism method. Surface structural differences are intrinsic characteristics for each species of larvae and can easily be identified using stereo microscope. These specificities will be helpful where a large number of field-collected larvae need to be identified routinely in pest control research.

울릉미역취에서 녹병균 Coleosporium solidaginis의 확인 (Confirmation of Coleosporium solidaginis on Solidago virgaurea subsp. gigantea in Korea)

  • 울릉미역취는 울릉도 원산이지만 한국에서 채소로 널리 재배된다. 울릉미역취의 녹병균은 2014년에 Coleosporium asterum으로 처음 보고되었다. 최근에 북미 시료를 중심으로 미역취속(Solidago)의 녹병균(Coleosporium)을 연구한 논문을 통하여 앞서 한국에서 보고된 울릉미역취 녹병균은 C. solidaginis에 포함시켜야 한다는 의견이 있었다. 이 의견을 확인하기 위하여 고려대학교 표본실에 보존된 울릉미역취 녹병균 시료 3점을 연구하여 C. solidaginis임을 확인하였다. 일본과 중국에서 보고된 양미역취의 녹병균은 C. asterum이나 C. solidaginis과는 구분되는 잠재종으로 추정된다. 본 연구를 통하여 그 동안 북미와 유럽에서만 알려졌던 C. solidaginis가 아시아에도 존재한다는 사실을 처음으로 보고한다.

Identification and Characterization of Expansins from Bursaphelenchus xylophilus (Nematoda: Aphelenchoididae)

  • We identified two novel expansin (EXP) genes in the expressed sequence tag database of Bursaphelenchus xylophilus, designated as Bx-EXPB2 and -EXPB3. Novel Bx-EXPBs encoded 150 amino acids and their similarities in coding sequence were 70.7-84.0% to the previously reported EXPB1 of B. xylophilus. Bx-EXPB2 and Bx-EXPB3 were clustered with Bx-EXPB1 and Bm-EXPB1, respectively, forming the independent phylogeny with other nematode EXPs. All identified Bx-EXPBs contained the signal peptide and were only expressed during the propagative stage, suggesting that they are secreted to facilitate nematode migration through hosts by loosening cell walls during infection. Quantitative real-time PCR analysis showed that the relative accumulation of Bx-EXPB3 mRNAs was the highest among the three Bx-EXPs examined and the order of mRNA accumulation was as follows: Bx-EXPB3 > Bx-EXPB2 >> Bx-EXPB1. Homology modeling of Bx-EXPBs showed that the structurally optimum template was EXLX1 protein of Bacillus subtilis, whichshared residues essential for catalytic activity with Bx-EXPB1 and Bx-EXPB2 except for Bx-EXPB3. Taken together, Bx-EXPB1 and Bx-EXPB2 may be involved migration through plant tissues and play a role in pathogenesis.

고온에서 배추좀나방 유충 지방체의 유전자 발현 변화 분석 (Analysis of Gene Expression in Larval Fat Body of Plutella Xylostella Under High Temperature)

  • 곤충은 변온동물로 육지생태계에서 주로 서식하면서, 식물의 생체량 조절, 종다양성 유지에 중요한 역할을 한다. 주변온도는 변온동물인 곤충의 생리적 반응속도, 뿐만 아니라 생존과 분포를 결정하며, 기후변화에 영향을 준다. 본 연구는 높은 온도에서 곤충의 적응성에 관련있는 유전자를 전사체를 이용하여 동정하였다. 고온에서 사육된 배추좀나방 유충의 지방체로부터 차세대 염기서열 분석법을 이용하여 전사체를 확보하였다. 대사중심인 지방체에서 구조단백질, 열충격단백질, 항산화단백질, 해독효소 들이 동정되었다. 이들 중에서 표피단백질(표피단백질, 키틴합성효소, 엑틴, 카이틴 합성), 스트레스관련단백질(시토크롬 P450), 열충격단백질, 한산화단백질은 발현이 증가되었으나, glutathione S transferase 발현은 오히려 감소되었다. 이상의 결과는 기후변화의 주요인인 온난화에 대한 해충의 생리적 대응과 온도적응을 이해하는데 필요한 기초자료를 제시한다.

국내 미기록 등수국 녹병균 Pucciniastrum hydrangeae-petiolaris (Pucciniastrum hydrangeae-petiolaris, a Newly Found Rust Fungus on Hydrangea petiolaris in Korea)

  • 등수국은 수국과에 속하는 덩굴성 식물로 우리나라의 울릉도, 제주도, 남해안 섬에 분포한다. 2017년 10월 제주도에서 등수국에 발생하는 녹병균을 국내 최초로 발견하였다. 등수국 녹병균을 동정하기 위해, 형태적 특성을 조사하고 internal transcribed spacer (ITS) 및 28S large subunit (LSU) rDNA의 염기서열의 분자계통학적 분석을 수행하였다. 그 결과, 한국의 등수국 녹병균은 일본과 러시아에서 보고된 Pucciniastrum hydrangeae-petiolaris의 형태적 및 분자계통학적 특징과 일치하였으며, 북미에서 보고된 P. hydrangeae와는 상당히 먼 계통학적 관계를 보였다. 본 연구는 한국의 등수국에서 녹병균 P. hydrangeae-petiolaris의 첫 보고이다.

온도 차이에 따른 배추좀나방 유충 지방체에서 발현되는 G 단백질 연관 수용체의 동정 (Identification of G Protein Coupled Receptors Expressed in Fat Body of Plutella Xylostella in Different Temperature Conditions)

  • BACKGROUND: G protein-coupled receptors (GPCRs) are widely distributed in various organisms. Insect GPCRs shown as in vertebrate GPCRs are membrane receptors that coordinate or involve in various physiological processes such as learning/memory, development, locomotion, circadian rhythm, reproduction, etc. This study aimed to identify GPCRs expressed in fat body and compare the expression pattern of GPCRs in different temperature conditions. METHODS AND RESULTS: To identify GPCRs genes and compare their expression in different temperature conditions, total RNAs of fat body in Plutella xylostella larva were extracted and the transcriptomes have been analyzed via next generation sequencing method. From the fat body transcriptomes, genes that belong to GPCR Family A, B, and F were identified such as opsin, gonadotropin-releasing hormone receptor, neuropeptide F (NPF) receptor, muthuselah (Mth), diuretic hormone receptor, frizzled, etc. Under low temperature, expressions of GPCRs such as C-C chemokine receptor (CCR), opsin, prolactin-releasing peptide receptor, substance K receptor, Mth-like receptor, diuretic hormone receptor, frizzled and stan were higher than those at 25℃. They are involved in immunity, feeding, movement, odorant recognition, diuresis, and development. In contrast to the control (25℃), at high temperature GPCRs including CCR, gonadotropin-releasing hormone receptor, moody, NPF receptor, neuropeptide B1 receptor, frizzled and stan revealed higher expression whose biological functions are related to immunity, blood-brain barrier formation, feeding, learning, and reproduction. CONCLUSION: Transcriptome of fat body can provide understanding the pools of GPCRs. Identifications of fat body GPCRs may contribute to develop new targets for the control of insect pests.
