Increased cryo-survival rate in ejaculated human sperm from infertile men following pre-freeze in vitro myo-inositol supplementation

  • Objective: To investigate the effects of in vitro myo-inositol (Myo-Ins) supplementation of cryopreserved human semen on the cryo-survival rate (CSR). Methods: Semen samples were obtained from 41 infertile men. Following routine semen analysis, each sample was divided into two equal aliquots (0.5 mL each). One aliquot was treated with 1 mg of Myo-Ins dissolved in $10{\mu}L$ of sperm preparation medium. The second aliquot was treated with $10{\mu}L$ of the same medium (control). Both aliquots were incubated for 20 minutes prior to freezing to slow the freezing process. The frozen samples were examined for post-thaw percentages of total motility (TM), progressive motility (PM), and the CSR, defined as the percentage of post-thaw TM divided by the percentage of pre-freeze TM and multiplied in 100. The results were expressed as median and interquartile range (25th and 75th percentiles). Results: The pre-freeze TM (50% [30%-50%]) and PM (35% [20%-35%]) were significantly higher than the post-thaw TM and PM in the MyoIns group (15% [10%-35%] and 10% [5%-20%]; p<0.001 and p<0.001, respectively) and the control group (10% [6%-30%] and 5% [3%-15%]; p<0.001 and p<0.001, respectively). The CSR of the 41 semen aliquots supplemented with Myo-Ins (40% [25%-70%]) was significantly higher than that of the control samples (30% [13%-58%], p=0.041). The CSR of the 26 abnormal semen samples that were supplemented with Myo-Ins (38% [20%-50%]) was significantly higher than that of the control samples (23% [12%-30%], p=0.031). Conclusion: In vitro Myo-Ins supplementation of ejaculated human sperm from infertile men resulted in a significant increase in the CSR in samples with abnormal pre-freeze sperm parameters.

Change of the saccharide content of fruit bodies by the addition of saccharides on oyster mushroom substrate (느타리버섯 배지의 당첨가에 따른 자실체의 당성분함량 변화)

  • Oyster mushroom is one of the popular mushrooms for Korean people and it is thus one of the mushrooms that were mostly cultivated in Korea in addition to winter and king oyster mushrooms. To promote the consumption in terms of its nutritional value and determine the absorption mechanism in its physiological and metabolic aspects, we investigated the sugar composition of its fruit bodies according to the amount of sugar component in the medium. As a result from the treatment of 4 different sugar components to the sawdust medium of oyster mushroom varieties (i.g., Suhan 1 ho, Yeorum, Chunchu 2 ho), we detected fructose, glucose, ribose, xylose as a monosaccharide with ${\alpha}$-lactose and trehalose as a disaccharide. The sugar alcohols were also detected including glycerol, mannitol, myo-inositol, and sorbitol. The sugar components that were detected in all treatments were trehalose, mannitol, ${\alpha}$-lactose, fructose, glucose, ribose, and myo-inositol and the rest of the components were found in some treatments with their different quantity included in the fruit bodies. The amount of monosaccharides included in fruit bodies is comparatively low and Suhan 1 ho contained them more than other varieties. There was no remarkable difference in the amount of sugar components in fruit bodies depending on treated sugars and their amounts in the treatment. With the increase of lactose treatment, the sugar components were markedly increased and the amount of trehalose in the fruit bodies was different according to the varieties. Therefore, it is not possible to detect the trend of sugar components according to the medium used in the cultivation. The sugar alcohols in the fruit bodies were comparatively contained more in Suhan 1 ho and Chunchu 1 ho. The amount of mannitol in the fruit bodies was not possible to be detected in terms of its trend according to the treatment. For myo-inositol, its quantity is gradually increasing in Suhan 1 ho and Chunchu 1 ho and it is not possible to detent the trend in Yeorum. In conclusion, we only detected ${\alpha}$-lactose as a sugar component that showed a correlation between the amount of treatment and the amount of the fruit bodies. Therefore, the sugar components detected in the fruit bodies were transformed rather than direct absorption into fruit bodies and further studies are needed to address this question.

Effect of Germanium Treatment in Culture Medium on Germanium Absorption by Callus Induced from Brown Rice (배지내 게르마늄 처리가 현미 유도 캘러스의 게르마늄 흡수에 미치는 영향)

  • This study was carried out to investigate the effect of Germaniwn(Ge) treatment in the culture media on the Ge absorption by the callus induced from brown rice cv. Dongjinbyeo. MS medium was more effective on the growth ratio of callus and the content of Ge and some inorganic elements except K in callus than N$_{6}$ medium. The more Ge treatment in the N$_{6}$ or MS medium, the more Ge absorption by the callus, but the growth ratio of callus and the content of Ca, Mg, and K in callus were decreased. The content of Fe, Mn, Zn, and Cu was increased under treatment up to 100~200mg /$\ell$ Ge, but tended to be decreased under treatment more than that of Ge concentration. Under treatment less than 150mg /$\ell$ Ge, GeO$_2$(inorganic Ge) was more effective on the Ge absorption by callus than Ge-132(organic Ge), but Ge-132 was more effective on the Ge ab-sorption by callus and the activity of callus in case of treatment more than 150mg /$\ell$ Ge. The lower pH of culture medium, the higher Ge content in the callus. When callus was cultured on medium supplemented with Ge and 0.1~1.0mM of citric acid or myo-inositol, content of Ge and some inorganic elements in callus, as well as growth and dry weight of callus, were tend to increase in comparison to control, but myo-inositol was more effective on them than citric acid.cid.

Inactivation of Brain myo-Inositol Monophosphate Phosphatase by Pyridoxal-5'-Phosphate

  • Myo-inositol monophosphate phosphatase (IMPP) is a key enzyme in the phosphoinositide cell-signaling system. This study found that incubating the IMPP from a porcine brain with pyridoxal-5'-phosphate (PLP) resulted in a time-dependent enzymatic inactivation. Spectral evidence showed that the inactivation proceeds via the formation of a Schiff's base with the amino groups of the enzyme. After the sodium borohydride reduction of the inactivated enzyme, it was observed that 1.8 mol phosphopyridoxyl residues per mole of the enzyme dimer were incorporated. The substrate, myo-inositol-1-phosphate, protected the enzyme against inactivation by PLP. After tryptic digestion of the enzyme modified with PLP, a radioactive peptide absorbing at 210 nm was isolated by reverse-phase HPLC. Amino acid sequencing of the peptide identified a portion of the PLP-binding site as being the region containing the sequence L-Q-V-S-Q-Q-E-D-I-T-X, where X indicates that phenylthiohydantoin amino acid could not be assigned. However, the result of amino acid composition of the peptide indicated that the missing residue could be designated as a phosphopyridoxyl lysine. This suggests that the catalytic function of IMPP is modulated by the binding of PLP to a specific lysyl residue at or near its substrate-binding site of the protein.

Carbohydrate, Amino Acid and Phenolic Contents of Rice Leaves in Relation to Adult-Plant Resistance to Leaf Blast (벼 잎의 탄수화물, 아미노산, 페놀화합물 함량과 잎 도열병에 대한 성체식물 저항성과의 관계)

  • Six rice cultivars showing various types of resistance or susceptibility to Pyricularia oryzae in the field were used to study the relationship between susceptibility and contents of carbohydrates, amino acids and phenolics at different plant maturities and leaf ages under controlled environmental conditions. No consistent differences between the susceptible cultivars and adult-plant-resistant cultivars were found in terms of the contents of individual carbohydrates, total amino acids and phenolics in rice leaves throughout the plant development. Only the adult-plant-resistant cultivar Dobong had lower contents of inositol throughout the plant development than the other cultivars. The amounts of sucrose, inositol, glucose, and fructose increased in all tested cultivars at eight leaf stage than those at five leaf stage, but slightly decreased at twelve leaf stage. In contrast, total amino acids and phenolics gradually decreased during plant development. With increasing age of rice leaves, the amounts of total soluble amino acids and phenolics gradually increased in healthy fifth leaf tissues, although there were no significant differences between the cultivars. In particular, a high level of phenolics existed in old fifth leaves of the cultivar Dobong. These results suggest that increased resistance to blast of matured rice plants and old leaves may be the two different phenomina derived from the physiological changes occurring during plant development and leaf senescence, probably functioning differently in P. oryzae development.

Characteristics and purification of proteoglycan from Phellinus igniarius (Phellinus igniarius로부터 분리한 단백다당류의 분리 및 특성)

  • The proteoglycan, intracellular and extracellular, extracted from the liquid culture of Phellinus igniarius were purified and characterized. The mycelial productivity was proved to be better in shaking culture compared to standing culture. The productivity of intracellular proteoglycan of Phellinus igniarius appeared to be similar in two culturing methods. The standing culture of Phellinus igniarius produced 6 times as much extracellular proteoglycan compared to shaking culture. The proteoglycan were purified to a single peak by ion exchange chromatography(DEAE-cellulose) followed by gel filtration(Sepharose 2B). PIEPDG contained 79.0% total sugar and 7.2% protein. PIEPAG contained 56.7% total sugar and 40.8% protein. PIIPDG contained 64.8% total sugar and 17.4% protein. PIIPAG contained 56.9% total sugar and 41.5%n protein. The molecular weights of all the fractions were estimated to be above 100,000, from 134KDa of PIEPDG to 560 KDa of PIEPAG. The results of sugar analysis by HPLC showed that PIEPDG contains glucose only. The sugar part of PIIPDG and PIIPAG were consisted of glucose and inositol. The PIEPAG contained three kinds of monosaccharides, glucose, fructose and inositol.

  • Inositol 1,4,5-triphosphate receptor type 1 (IP3R1) is a $Ca^{2+}$ release channel that responds to the second messenger IP3 and that modulates diverse cellular functions such as contraction/excitation, secretion, gene expression and cellular growth. We discovered single nucleotide polymorphisms (SNPs) within IP3R1 gene and analyzed associations between gene polymorphisms and carcass traits in Korean cattle (Hanwoo) in order to develop novel DNA markers at genomic level. Three SNPs were detected at the position of g.1428617A>G, g.1418843C>T and g.1414377C>T with 24 unrelated Hanwoo samples by direct sequencing of the PCR products. We found that genotype of g.1414377C>T SNP was associated with live weight (P<0.05) and carcass weight (P<0.01) using the general linear model of SAS package. These results suggest that polymorphism of IP3R1 gene was associated with weight-related traits in Hanwoo.

  • 1-O-oleoyl glycerol, 2-O-oleoyl myo-inositol and methyl 2-O-oleoyl-${\alpha}$-D-glucopyranoside were used as surfactants in this study. The solution properties and solubilization process of those nonionic surfactants were examined by the phase equilibria. As a result of this study, we have found that phase behavior of two component systems of surfactants/$H_2O$/cyclohexane depends on temperature respectively. The three phase regions of three component systems appeared in the temperature range of $27^{\circ}C{\sim}32^{\circ}C$, $36^{\circ}C{\sim}45^{\circ}C$ and $38^{\circ}C{\sim}52^{\circ}C$ and solubilization of water and oil was high in those three phase ranged As the temperature was varied in the two component systems, liquid crystals of hexagonal were observed to in the case of 1-O-oleoyl glycerol, and liquid crystal of lamella types were observed in the case of 2-O-oleoyl myo-inositol and methyl 2-O-oleoyl-${\alpha}$-D-glucopyranoside.

  • The lipid and fatty acid compositions of neutral lipid, glycolipid and phospholipid were identified and quantified by thin-layer chromatography and gas chromatography. Main constituents of the neutral lipid were triglyceride, free sterol and esterified sterol in the free lipid, and free fatty acid and monoglyceride in the bound lipid. In the outer part, there existed 25.7% triglyceride in the free lipid and it was not nearly found in the bound lipid. Main constitutents of the glycolipid were digalactosyl diglyceride and esterified steryl glycoside in the free lipid, and digalactosyl diglyceride in the bound lipid. Free lipid didn't contain trigalactosyl diglyceride but bound lipid contained 2.0% of it. Main constituents of the phospholipid were lysophosphatidyl choline, phosphatidyl inositol, phosphatidyl choline, phosphatidyl glycerol and phosphatidyl ethanolamine in the free lipid, and phosphatidyl choline and phosophatidyl inositol in the bound lipid. The predominant fatty acids of three fractions, neutral lipid, glycolipid and phospholipid showed almost the same pattern as that of the total free and bound lipids. The content of palmitic acid was relatively higher in the polar lipids(glyco and phospholipid). Therefore, saturated fatty acid ratio of polar lipid was higher than that of neutral lipid. found lipid contained more saturated fatty acids as compared with the free lipid.

  • Bacillus subtilis BD0310 isolated from tea leaves was used for the development of a biofungicide against Pestalotiopsis longiseta causing gray blight of tea plants. The optimum growth conditions were investigated for the mass cultivation of the microbial agent. The optimum temperature and cultivation time were determined as $12{\sim}24$ hours at $30^{\circ}C$ and the optimum initial pH was pH 7.0 in nutrient broth. Among the tested carbon sources of fructose, galactose, glucose, glycerol, inositol, lactose, maltose, sorbitol and starch, maltose and inositol were found to highly increase antifungal activity of the microbial agent against P. longiseta. Yeast extract and tryptone apparently increased antifungal activity of the microbial agent among the tested nitrogen sources of casein, tryptone, malt extract, yeast extract and $(NH_4)_2SO_4$. The results will make a contribution to mass production of the antagonistic bacterium Bacillus subtilis BD0310 for development of a microbial agent controlling gray blight of tea plants.
