Control Efficacy of Serenade Formulation against Rhizoctonia and Pythium Damping-off Diseases (Rhizoctonia 및 Pythium 모잘록병에 대한 Serenade 제제의 방제효과)

  • Damping-off, caused by Rhizoctonia solani and Pythium ultimum, is a very important plant disease and affects seeds and seedlings of many plant species worldwide. To investigate control efficacy of Serenade formulation (1.34%, SC) against Rhizoctonia and Pythium damping-off diseases, seeds of chili pepper and cucumber were sown in soils inoculated with R. solani and P. ultimum and Serenade formulation was applied by soil-drenching with 9-, 19-, and 39-fold dilutions. Control values of Serenade formulation on Rhizoctonia damping-off of chili pepper was 58% and 29% for 9- and 19-fold dilutions, respectively. In the case of cucumber Rhizoctonia damping-off, Serenade treatments showed similar control efficacy with damping-off of chili pepper. On the other hand, control efficacy of Serenade formulation on Pythium damping-off of cucumber was less than control effects on Rhizoctonia damping-off. Only Pythium damping-off of chili pepper treated with 9-fold dilution Serenade was statistically different with untreated control. This result suggest that Serenade formulation could be effectively used for controlling Rhizoctonia and Pythium damping-off diseases.

Isolation and Utilization of Antagonistic Pseudomonas fluorescens from Soils for the Protection of Soybean Sprouts Rot (콩나물 부패병 방제를 위해 토양으로부터 분리한 길항균 Pseudomonas fluorescens의 이용)

  • Thirty-three bacterial and fungal strains were isolated from the rotten soybeans and soybean sprouts to isolate pathogenic microorganisms which cause soybean sprouts rot during soybean sprouts cultivation. In pathogenicity tests of the isolates on soybean sprouts, two isolates(K-17 and K-28) caused soybean sprouts rot and were identified as Erwinia carotovora and Fusarium sp., respectively. To isolate antagonists aganist K-17 and K-28 pathogens, bacteria were isolated from various soybean-cultivated soils and screened by the inhibition zone method. A bacterial isolate(J-232) which inhibited growth of both pathogens was identified as Pseudomonas fluorescens and further examined. The culture filtrate of P. fluorescens J-232 (dilution rate of 500 times) inhibited the growth of Erwinia carotovora K-17 and Fusarium sp. K-28 both on potato dextrose agar medium and on soybean sprouts cultivated in vessel. The development of soybean sprouts rots was observed during cultivation by inoculation of soybean seeds with culture filtrate of both pathogens. The combined inoculation of soybean seeds with culture filtrate of antagonistic bacterium and that of pathogens prevented soybean sprouts rot, and the growth of soybean sprouts was similar to that of control. The soybean sprouts inoculated with antagonists culture filtrate alone did not develop soybean sprouts rot, and the growth of the seedlings was shown to be slightly promoted as compared with that of control.

Virulence and Relative Density of Three Root Rotting Organisms; Pythium ultimum, Pythium echinocarpum and Rhizoctonia solani in Alpine Soils in Chinese Cabbage (고냉지 배추 재배지역 토양에서 배추에 뿌리 썩음을 일으키는 Pythium ultimum, Pythium echinocarpum, Rhizoctonia solani균의 병원력 및 토양내 상대밀도)

  • Fifty three diseased root samples of Chinese cabbage and 22 soil samples were taken from six alpine areas in Kangwon province to examine the organisms causing root rot of Chinese cabbage. Among thirteen microorganisms detected from diseased roots samples, Pythium ultimum, Pythium echinocarpum and Rhizoctonia solani were detected frequently and were pathogenic to Chinese cabbage. Five to 100% of young Chinese cabbage plants grown in the sample soils taken from alpine cabbage fields were infected either alone or together with P. ultimum, P. echinocarpum and R. solani, depending upon the or gin of sample soils. When relative density of the three organsims in the sample soils was estimated based on the infection frequency on the plants grown in those sample soils, P. ultimum was prevalent only in Chahang-i-li area. P. echinocarpum was prevalent in Yongsan-il-li, Hoengwe-li, and Munmaek areas. R. solani was solely found in Maebongsan area where both P. ultimum and P. echinocarpum were not detected. In the three areas where both P. ultimum and P. echinocarpum were present, density of P. echinocarpum was generally greater than that of P. ultimum. Seedlings of thirty major Chinese cabbage varieties were inoculated independently with the three organisms in the greenhouse. Six varieties were resistant to P. ultimum. Three and two varieties were moderately resistant to P. ultimum and P. echinocarpum, respectively. All varieties were highly susceptible to R. solani.

Economic Injury Level of Bemisia tabaci (Gennadius) (Homoptera: Aleyrodidae) on Oriental Melon (Cucumis melo L.) in Greenhouse (참외에서 담배가루이의 경제적 피해수준 설정)

  • This study was conducted to develop economic injury level (EIL) of sweet potato whitefly, Bemisia tabaci, on oriental melon. In greenhouse, seedlings of oriental melon were transplanted at June 8, 2010 and we inoculated adult B. tabaci with the density of 0, 1, 5, 10, or 20 per ten leaves at July 14. Adult of B. tabaci increased approximately twenty five-fold at 60 days after inoculation in the plot of 20 adults per ten leaves. The damages on leaves and fruits by B. tabaci were started to appear at 20 days after inoculation, and the damage rates of leaves or fruits were 28.5 or 31.5 percent at 60 days after inoculation, respectively, in the plot of inoculation with 20 adults per ten leaves. The yield of oriental melon was reduced as the inoculation density of B. tabaci increased, and the relationship between inoculation density of B. tabaci and the rate of damaged fruit could be described by a linear regression Y = 0.961x + 0.0562 ($R^2$ = 0.976). Based on the relationship, the economic injury level was 5.1 adults of B. tabaci per leaf and the control threshold estimated by 80% level of economic injury level was 4.1 adults per leaf for control of sweet potato whitefly.

Gibberellin Production by Newly Isolated Strain Leifsonia soli SE134 and Its Potential to Promote Plant Growth

  • Very few plant growth-promoting rhizobacteria (PGPR) are known to produce gibberellins (GAs). The current study aimed to isolate a phytohormone-producing PGP rhizobacterium from soil and assess its potential to enhance plant growth. The newly isolated bacterium was identified as Leifsonia soli sp. SE134 on the basis of partial 16S ribosomal RNA gene sequence. Application of L. soli culture filtrate significantly increased the biomass, hypocotyl, and root lengths of cucumber seeds as compared with non-inoculated sole medium and distilled water treated controls. Furthermore, the PGPR culture was applied to the GA-deficient mutant rice cultivar Waito-C. Treatment with L. soli SE134 significantly increased the growth of Waito-C rice seedlings as compared with controls. Upon chromatographic analysis of L. soli culture, we isolated, detected and quantified different GAs; namely, $GA_1$ ($0.61{\pm}0.15$), $GA_4$ ($1.58{\pm}0.26$), $GA_7$ ($0.54{\pm}0.18$), $GA_8$ ($0.98{\pm}0.15$), $GA_9$ ($0.45{\pm}0.17$), $GA_{12}$ ($0.64{\pm}0.21$), $GA_{19}$ ($0.18{\pm}0.09$), $GA_{20}$ ($0.78{\pm}0.15$), $GA_{24}$ ($0.38{\pm}0.09$), $GA_{34}$ ($0.35{\pm}0.10$), and $GA_{53}$ ($0.17{\pm}0.05$). Plant growth promotion in cucumber, tomato, and young radish plants further evidenced the potential of this strain as a PGP bacterium. The results suggest that GA secretion by L. soli SE134 might prove advantageous for its ameliorative role in crop growth. These findings can be extended for improving the productivity of different crops under diverse environmental conditions.

Effects of Field Application of Spodoptera litura Nucleopolyhedrovirus to Control S. litura in Chrysanthemum (국화 포장에서 담배거세미나방 방제를 위한 핵다각체병바이러스 살포효과)

  • This experiment was conducted to investigate control effect of Spodoptera litura using nucleopolyhedrovirus in chrysanthemum seedlings of open field and plastic house. Values of LT$_{50}$ of treatment 1.0$\times$10$^{6.8}$ PIBs/ml were 6.2-5.1 days in open field and 6.9-5.4 days in plastic house. Values of LT$_{50}$ and LT$_{95}$ were shorter in open field than in plastic house. Cumulative mortality was 100% in 1.0$\times$10$^{7.8}$ PIBs/ml and also higher in open field than in plastic house. In chrysanthemum field, organic synthetic insecticide (endosulfan EC) killed S. litura larvae in 2 days after application. Motality of S. litura larva inoculated with S. litura nucleopolyhedrovirus (SINPV) 1.0$\times$10$^{6.8}$ PIBs/ml was found out from 4 days after application and maintained during 14 days. Protection values of with SINPV 1.0$\times$10$^{8}$ PIBs/ml after 16 days were 94.0% and 89.4% in open field and plastic house, respectively, and those of endosulfan 350 ppm were 91.4% and 88.6%, respectively.

Characteristic of Progeny in Pepper Transformants (고추 유전자변형체 후대 생육특성 검정)

  • For the resistance test for Phytophthora blight of $T_1$ and $T_2$ transformants in pepper, Phytophthora blight fungus was inoculated to seedlings of the $T_1$ and $T_2$ transformants by concentration (density: zoospore $10^3/ml$). Occurrence rate of blight at 5days after inoculation was 4.0 % in T1-1 line and 10.0% in $T_1-2$ line, and its rate for 12 days after inoculation was 52.0% in $T_1-1$ line, 64.0% in $T_1-2$ line, respectively. Therefore, the lower occurrence rate to blight was enable to select resistant transformants in the some inoculation density (zoospore $10^3/ml$), meanwhile 'Kumtap' and 'Subicho' were 100% in highest occurrence rate to blight. For field test, in which blight was commonly occurred, of the Youngyang Pepper Experiment Station, the acquired transformant resisting to blight was similar to characteristics of domestic varieties, 'Subic ho' for fruit shape, but there are some differences in growth, days to flowering, fruit characteristics. Occurrence of blight in $T_2-1-6$, and $T_2-4-9$ lines was smaller approxmately 30% than commercial varieties, 'Kumtap', although occurrence of blight in field was showed higher difference among tested lines. In this study, we concluded that the transformants showing blight resistance selected from habitual field could be fixed at every generation, and the developed transformation system was also considered to develop transformants in pepper.

Suppression of Powdery Mildew Development in Oriental Melon by Silicate Fertilizer (규산질 비료의 참외 흰가루병 발생 억제 효과)

  • Silicon is known to accumulate in plants and results in greater resistance to diseases and insect pests. In this study, we investigated the effect of silicate fertilizer applied in soil on the development of powdery mildew of oriental melon. Oriental melon seedlings of four-leaf stage were transplanted and grown in a plastic film house. Silicate fertilizer was applied to maintain soil available $SiO_2$ level of 200 mg/kg one week before transplanting. Fungicide triflumizol was sprayed three times; one, two, and three weeks after transplanting. Sphaerotheca fuliginea was inoculated 2 weeks after transplanting. The number of infected leaf and the number of fungal colony in leaves were measured one, two, and three weeks after the inoculation. Three weeks after the fungal inoculation, in the treatment of fungicide triflumizol. infected leaf numbers and number of colony per leaf were reduced by 10 and 58%, respectively. In the silicate fertilizer treatment, infected leaf numbers and numbers of colony per infected leaf were suppressed only by 6 and 16%, respectively, and the efficacy was lower than that of the fungicide triflumizol. The combined treatment of silicate fertilizer and the fungicide suppressed powdery mildew more effectively, and infected leaf numbers and numbers of colony per leaf were reduced by 31 and 80%, respectively. These results indicate that although silicate fertilizer itself is not much effective in the suppression of powdery mildew, it can significantly enhance the efficacy of the fungicide.

Mycorrhizae Formation, Growth and Mineral Nutrient Uptake of Poncirus trifoliata in Response to P Application in Volcanic Ash Soil (화산회토양에서 인산 시용 수준별 탱자 유묘의 공생균근 형성과 생육 및 무기양분 흡수)

  • Mycorrhizae is well known to increase the uptake of P and other mineral nutrients of plants. But if available P levels in soil is too low or high, mycorrhizae formation is limited. This study was carried out to determine the optimum level of available P for mycorrhizae formation of Poncirus trifoliata (trifoliate orange) seedling in volcanic ash soil. Eight levels of P in the range $0-1050mg\;kg^{-1}$ were applied with double superphosphate, and in each P level mycorrhizal fungi inoculated and uninnoculated treatments were included. The seedlings were grown in a greenhouse for 5 months and mycorrhizae formation, growth, and nutrient uptake were measured. As P application level increased, mycorrhizae formation increased at lower range of P application and the highest formation ratio of 43% was found at $100mg\;kg^{-1}$ P level ($2.6mg\;kg^{-1}$ available P in soil). At further higher levels of P application, mycorrhizae formation was rather suppressed. Seedling growth was increased by the inoculation of mycorrhizal fungi, and maximum growth was found at $100mg\;kg^{-1}$ P level where mycorrhizae formation was highest. The growth and mineral nutrient uptake of Poncirus trifoliata seedling represented a significant positive correlation with mycorrhizae formation at all P treatments.

Evaluation of Garlic Germplasm for Resistance to Leaf Blight Caused by Stemphylium vesicarium (마늘 유전자원의 Stemphylium vesicarium에 의한 잎마름병 저항성 평가)

  • Leaf blight caused by Stemphylium vesicarium is one of the most important fungal diseases of garlic (Allium sativum L.) worldwide, which results in a reduction of quality and yield. The breeding of resistant cultivars is an efficient approach to decrease the use of chemical fungicides and minimize crop losses. In this study, to find the resistant garlic resources against S. vesicarium, we evaluated the resistance degree of 20 garlic germplasms. To do this, garlic seedlings at four-leaf stage were rubbed with nonabsorbent cotton and then inoculated with spore suspension (3.0×105 spores/ml of potato dextrose broth) of S. vesicarium by spray method. Three to seven days after inoculation, the infected leaf area (%) of garlic seedling was measured. 'Daeseo' and 'Namdo' were included as susceptible and resistant control cultivars, respectively. After 3 to 7 days of incubation, the infected leaf area (%) of garlic seedling was measured. Our results showed that IT245512, IT245528, and IT244068 lines exhibited the highest resistance against S. vesicarium, whereas IT257134 and IT253043 lines were more susceptible than the susceptible cultivar 'Daeseo'. Based on the results, the resistant genetic resources selected in this study can be used a basic material for resistant garlic breeding system against leaf blight.
