Changes of Hemodynamics and Renal Function due to Acute Cadmium Exposure in Rats

  • The systolic and diastolic pressures in anesthetized Sprague-Dawley male rats were greatly decreased after single-dose of Cd treatment without significant changes in heart rate. There was a fluid-shift into the third space and/or -loss through the kidney, since plasma $Na^+$ concentration and hematocrit ratio were significantly increased by acute Cd exposure. The present study showed that the sustained hypotensive effect of single-dose Cd on the cardiovascular system might have resulted from the systemic hypovolemia. Furthermore, renal excretion of electrolytes, including $Na^+$ and $K^+$, and urine flow rate were increased by Cd intoxication. Interestingly, the ratio of $Na^+/K^+$ excretion was increased and reached the maximum level 3 hours after Cd injection and returned to the normal level after 7 hours. Nevertheless, there was no difference in the regression analysis of $Na^+$ excretion and urine flow rate in both groups. Therefore, the increase in the urine volume seemed to enhance the excretion of $Na^+$. This study strongly suggest that the hypotensive effect of Cd is mediated by systemic $Na^+$ loss through the kidney and/or hypovolemia via fluid-shift.

Effect of Bromocriptine on 6-Hydroxydopamine-induced Lipid Peroxidation and Cytotoxicity in vitro and in vivo

  • The present study was to evaluate the protective effects of bromocriptine, which is known as $D_2$ dopamine receptor agonist and used for the treatment of patients with Parkinson's disease (PD), on 6-hydroxydopamine (6-OHDA)-induced neurotoxicity in vitro and in vivo. Lipid peroxidation product (malondialdehyde; MDA) produced by the administration of 6-OHDA was profoundly reduced following the treatment of bromocriptine in a dose-dependent manner in rabbit brain homogenate. Quinone formation by 6-OHDA autoxidation was also attenuated, and its effect was as potent as other antioxidants. Pretreatment of bromocriptine reduced the cytotoxicity of 6-OHDA on SH-SY5Y neuroblastoma cell lines dose-dependently. The loss of striatal dopamine and its metabolite, DOPAC (dihydroxyphenylacetic acid) as well as increase of MDA production caused by intrastriatal injection of 6-OHDA was significantly recovered following the treatment of bromocriptine. The present study clearly showed that bromocriptine had a protective action against 6-OHDA-induced neurotoxicity. These results suggest that bromocriptine has the antioxidant properties, which could be another advantage for delaying the progress of Parkinson's disease.

Antimetastatic Effect of Proteoglycan Isolated from the Mycelium of Ganoderma lucidum IY009 in vitro and in vivo (영지버섯 균사체 (Ganoderma lucidum IY009)로부터 추출한 단백다당체의 전이암 억제 효과)

  • $\beta$-Immunan, a proteoglycan, was isolated from the mycelium of Canoderma lucidum which belongs to a medicinal mushroom. The effects of $\beta$-Immunan on cell-cell and cell-matrix interactions mediated by carbohydrate-recognition and the mechanism responsible for the inhibition of experimental metastasis of Bl6-F10 and B16/BL6 murine melanoma were studied. The results showed that $\beta$-Immunan inhibited Bl6-Fl melanoma cell's adhesion to laminin and asialofetuin-induced homotypic aggregation and reduced invasion against Bl6-F10 murine melanoma cells through matrigel in vivo assay. When $\beta$-Immunan was intraperitoneally administrated to C57B/6 mice bearing B16/BL6 murine melanoma cells, it was decreased the number of pulmonary metastatic colony by the dose dependent manner ranging from 20 to 100 mg/kg/day. The results indirectly indicate that clinical treatment with $\beta$-Immunan might be expected to exhibit anti-metastatic effect. In the pulmonary metastasis, the number of pulmonary metastatic colony of melanoma when $\beta$-Immunan was intraperitoneally administrated to C57BL/6 mice bearing B16/BL6 murine melanoma cells by intravenous injection were decreased by the dose dependent manner ranging from 20 to 100 mg/kg/day.

Survey of the Expression Pattern and Immuno Stimulatory Effect of DNA Vaccine Using β-Galactosidase Reporter System in Olive Flounder (Paralichthys olivaceus)

  • The CMV promoter driven lacZ reporter gene (pcDNA-lacZ) was constructed and used for DNA immunization study. The expression of the lacZ gene was confirmed in vitro using RTG-2 cell line before using for in vivo study in olive flounder (Paralichthys olivaceus). In the dose response study, the maximum expression of the lacZ gene was found in the group injected with 5 ${\mu} g$ of the plasmid DNA. Kinetic study showed a significantly increased expression of $\beta-galactosidase$ gene at 7 days after injection. Effects of DNA vaccine on specific and nonspecific immune responses such as antibody and NO production were studied and the significant effect was found in olive flounder injected with 10 and 15 ${\mu} g$ DNA (sub optimal dose for lacZ gene expression). Two pro inflammatory cytokine genes, $IL-l\beta$ and $TNF-\alpha$, were also found to be up regulated in the muscle injected with the plasmid, suggesting an induction of local inflammatory response.

Characteristics of Aragonite From Underwater and The Cytotoxicity, Cell Division Disturbance and Induction of Morphological Transformation on BALB/3T3 Cells (일부 지하수에서 얻은 Aragonite의 특성과 BALB/3T3 세포에 대한 세포독성, 세포분열장애 및 형태학적 변이유발)

  • Aragonite is one of polymorphs of calcium carbonate of which main form is calcite. We found that white precipitate is formed in much amount by boiling underwater of Inchon, Korea and confirmed that it is aragonite. This study is to evaluate the dimensional characteristics, solubility, acid resistance of aragonite and the cytotoxicity, cell division disturbance and cell transforming ability of it on BALB/3T3 cells. The results are as follows: Lengths of the aragonite were reduced to the 72.7% and 22.7% respectively after 5 months and 7 months of intrapleurai injection to the Sprague-Dawley rat. Strong acid such as 1M HCl dissolved the aragonite instantly but weaker acid pH 2.0 or more could not dissolved aragonite easily. The result of cell growth inhibition showed that cell numbers were decreased as log-doses of treatment of the aragonite were increased 24 hours, 48 hours, and 72 hours later. Cell plating efficiency after the aragonite treatment also showed dose-dependent decrease. Multinuclear giant cell formation was increased in the aragonite treated cells until ID$_{50}$ and after the dose the multinucleate cells were decreased, but remained much higher than negative control cells. Morphological transformation assay showed that the aragonite did not induce transformation in all treated doses.

Effect of Dansamchunghyul-tang on Hyperlipidemia in Rat (단삼청혈탕이 백서의 고지혈증에 미치는 영향)

  • Dansamchunghyul-tang(丹參淸血湯) is a new formula consist of Salviae Miltiorrhizae Radix and some oriental herbs to treatment of hyperlipidemia. In this study, we investigated the effects of the water extract of Dansamchunghyul-tang(DCT) on hyperlipidemia of the rat. We here studied the influence of DCT on hyperlipidemia in Sprague-Dawley rats. The hyperlipidemia was induced by Triton WR-1339 (tyloxapol) intravenous injection at 200mg/kg body weight. After 18 h, blood was collected for the determination of plasma total cholesterol, triglyceride, phospholipid, HDL-cholesterol, and LDL-cholesterol concentrations. In the model of hyperlipidemia induced by triton WR-1339 in rats, DCT showed decreasing effects on total cholesterol, triglyceride, phospholipid level significantly, in a dose-dependent fashion. DCT showed dose-dependent effects on HDL-cholesterol and LDL-cholesterol level insignificantly. In conclusion, DCT showed significant effects on hyperlipidemia, and the present findings indicate that DCT may prove potentially useful for treatment of hyperlipidemia.

Luteolin, a Bioflavonoid, Attenuates Azoxymethane-Induced Effects on Mitochondrial Enzymes in Balb/c Mice

  • Colon cancer (CRC) is a serious health problem throughout the world. Development of novel drugs without side effects for this cancer is crucial. Luteolin (LUT), a bioflavonoid, has many beneficial effects such as antioxidant, anti-inflammatory and anti-proliferative potential. was a potent chemical carcinogen used for the induction of colon cancer. Colon carcinogenesis was initiated by intraperitoneal injection of azoxymethane (AOM) to mice at the dose of 15 mg/body kg weight in Balb/C mice for 3 weeks. Mice were treated with LUT at the dose of 1.2 mg/body kg weight orally. Mitochondrial enzymes such as isocitrate dehydrogenase (ICDH), ${\alpha}$-keto dehydrogenase (${\alpha}$-KDH), succinate dehydrogenase (SDH) and the activities of respiratory chain enzymes NADH dehydrogenase and cytochrome c oxidase were found to be elevated in AOM-treated animals. Treatment with LUT decreased the activities of all the parameters significantly. Hence, LUT might be a potent anticancer agent against colorectal cancer.

Central noradrenergic mechanism in the regulation of blood pressure in SHR

  • The purpose of the present study was to address whether the in vivo noradrenergic neural activities in the locus coeruleus are involved in the regulation of blood pressure. Two groups of the animals were prepared, 1) SHR and 2) age-matched normotensive control, WKY. At the age of 6 and 16 weeks, blood pressure and the releases of NE from the locus coeruleus in SHR and KWY were measured by in vivo microdialysis at three different conditions: 1) normal, 2) elevated state of blood pressure by systemic injected phenylephrine and 3) increased state of neural activity by perfused phenylephrine into the locus coeruleus. The basal release of NE of SHR were significantly higher than that of WKY, Phenylephrine treatment caused elevation of blood pressure in both SHR and WKY in dose-dependent manner. Following phenylephrine injection, the releases of NE from the locus coeruleus of SHR were significantly decreased, whereas the significant change of NE in WKY was observed in the highest dose of phenylephrine. Phenylephrine perfusion into the locus coeruleus through microdialysis probe caused pressor responses and the pressor response in SHR was greater compared with that in WKY. The results from the present study suggests that the noradrenergic nervous system in the locus coeruleus may contribute as one of the development and maintenance factors for hypertension in SHR.

Antidiabetic Metabolism Effect on the water Extract of $Cordyceps$ $Militalis$ in Streptozotocin-Induced Diabetic Rats (밀리타리스 동충하초($Cordyceps$ $Militalis$)열수 추출물이 스트렙토 조토신으로 유발된 당뇨 흰쥐의 항 당뇨 대사에 미치는 영향)

  • This study was carried to investigate the antidiabetic metabolism effect of water extract $Cordyceps$ $Militalis$(C.M.) in Streptozotocin(STZ) induced diabetic rats. Diabetes were induced by intravenous injection of STZ at a dose of 42mg/kg,b.w. dissolved in citrate buffer. The water extract of C.M. was orally administrated once a day for 7 days at a dose of 500mg/kg,b.w(body weight). or 1,000mg/kg.b.w. The content serum glucose was significantly decreased in C.M. treated group compared to the those of STZ-control group. The content of hepatic glycogen and activities of glucose-6-phosphate dehydrogenase(G-6-PDH), glucokinase(GK) were significantly increased, but activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in C.M. treated group compared to the those of STZ-control group. These results indicated that water extract of C.M. would have antidiabetic metabolism effect in STZ-induced diabetic rats.

Antidiabetic Effect of Yacon(Smallanthus sonchifolius) of Root and Leaf in Streptozotocin-Induced Diabetic Rats (Yacon의 뿌리와 잎의 추출물이 streptozotocin으로 유발된 흰쥐의 항당뇨 효과)

  • This study was carried to investigate the antidiabetic effect of ethanol extract of Smallanthus sonchifolius(S.S) in Streptozotocin(STZ)- induced diabetic rats. Diabetes was induced by intravenous injection of STZ at a dose of 43mg/kg dissolved in citrate buffer. The ethanol extract of S,S in leaves and root were orally administrated once a day for 7 days at a dose of 1,000mg/kg. The contents of serum glucose, triglyceride(TG), total cholesterol were significantly decreased in S.S treated group compared to the those of STZ-control group. The content of hepatic glycogen and activity of glucokinase(GK) were significantly increased,and activity of glucose-6-phoshatase(G-6-Pase) was significantly decreased in S.S treated group compared to the those of STZ-control group, but activity of glucose-6-phosphate dehydrogenase(G-6-PDH) was not significantly increased, These results indicated that ethanol extract of S.S would have antidiabetic effect in STZ-induced diabetic rats.
