MECHANISM IN ANTIBACTERIAL ACTIVITY OF POLYPHOSPHATES AGAINST PORPHYROMONAS ENDODONTALIS (Porphyromonas endodontalis에 대한 Polyphosphate의 항균기전에 관한 연구)

  • Poly-P has been used to prevent decomposition of foods and has been shown to have inhibitory effect on the growth of gram positive bacteria. The purpose of this study was to evaluate the effect of poly-P on the growth of Porphyromonas endodontalis, a gram negative obligate anaerobic rod, endodontopathic bacterium. P. endodontalis ATCC 35406 was in BHI broth containing hemin and vitamin K with or without poly-P. Inhibitory effect of each poly-P which was added at the beginning(lag phase) or during(exponential phase) the culture, MIC(minimum inhibitory concentration) was determined by measuring the optical density of the bacterial cell at 540nm. Viable cell counts were measured to determined whether poly-P has a bactericidal effect. Leakage of intracellular nucleotides from P. endodontalis was determined at 260nm and morphological change of P. endodontalis was observed under the TEM(transmission electron microscope). Binding of 32P-labeled poly-P to P. endodontalis was examined. SDS-polyacrylamide gel electrophoresis and zymography were performed to observe the changes in protein and enzyme profiles of P. endodontalis, respectively. The results from this study were as follows : 1. The minimal inhibitory concentration(MIC) of poly-P to P. endodontalis appeared to be 0.04~0.05%. 2. Poly-P added to the P. endodontalis culture during the exponential phase of P. endodontalis was as much effective as poly-P added at the begining of the culture, suggesting that the antibacterial effect of poly-P is not much dependent on the initial inoculum size of P. endodontalis. 3. Poly-P are bactericidal to P. endodontalis, demonstrating the decrease of the viable cell counts. 4. Intracellular nucleotide release from the P. endodontalis, was not increased in the presence of poly-P and was not reversed by the addition of divalent cations like $Ca^{2+}$ and $Mg^{2-}$. 5. Under the TEM, it was observed that fine electro-dense materials were prominent in the poly-P grown P. endodontalis, appearing locally in the cell, and the materials were more abundant and more dispersed in the cell as the incubation time with poly-P increased. In addition, highly electron dense granules accumulated in many poly-P grown cells, most of which were atypical in their shape. 6. Binding of 32P-labeled poly-P to P. endodontalis appeared to be 32.8 and 45.5 and 53.4% at 30 minutes, 1 hours and 2 hours, respectively. 7. In the presence of poly-P. the synthesis of proteins with apparent molecular masses of 25, 27, 35, 45 was lost or drastically decreased whereas expression of a protein with an apparent molecular mass of 75 was elevated. 8. Proteolytic activity of P. endodontalis was decreased by poly-P. The overall results suggest that use of poly-P may affect the growth of P. endodontalis, and the anti-bacterial activity of poly-P seems largely bactericidal. Changes in shape, protein expression, and proteolytic activity of P. endodontalis by poly-P may be directly and indirectly attributed to the antibacterial effect of poly-P. Further studies will be needed to confirm the effect of poly-P.

The Study on the Marine Eco-toxicity and Ecological Risk of Treated Discharge Water from Ballast Water Management System Using Electrolysis (전기분해원리를 이용한 선박평형수관리장치의 배출수에 대한 해양생태독성 및 해양환경위해성에 관한 연구)

  • The International Convention for the Control and Management of Ship's Ballast Water and Sediments was adopted at 2004 and then various BWMS (ballast water management system) have been developed. In this study, WET (whole effluent toxicity) test with algae (diatom) Skeletonema costatum as primary producer, invertebrate (rotifera) Brachionus plicatilis as 1st consumer and fish (olive flounder) Paralichthys olivaceus as predator, chemical analysis and ERA (environmental risk assessment) were conducted to assess the unacceptable effect on marine ecosystem by emitting the discharge water treated with AquaStar$^{TM}$ BWMS using electrolysis as main treatment equipment for removing the marine organisms in the ship's ballast water. The most sensitive test organism on discharge water treated with AquaStar$^{TM}$ BWMS was S. costatum that gave the NOEC value of 25.00%, LOEC value of 50.00% and 72hr-$EC_{50}$ value of 69.97% from WET test result for 20 psu salinity treated discharge water. NOEC and LOEC value of B. plicatilis and P. olivaceus exposed at 20 psu salinity treated discharge water were 50.00% and 100.00%, respectively. In the chemical analysis results, total number of substances produced by AquaStar$^{TM}$ BWMS was 18 which were bromate, 7 volatile halogenated organic compounds, 7 halogenated acetic acids, 3 halogenated acetonitriles and chloropicrin. Eighteen substances did not consider as persistence and bioaccumulative chemicals. Uncertainty of toxic property of 18 substances was high. PECs of 18 substances calculated by MAMPEC model were ranged from $4.58{\times}10^{-4}$ to $4.87{\mu}g\;L^{-1}$, PNECs of them were ranged from $1.6{\times}10^{-2}$ to $3.2{\times}10^2{\mu}g\;L^{-1}$. And, the PEC/PNEC ratio of 18 substances did not exceed 1. Therefore, ERA for produced substances indicate that the discharge water treated with AquaStar$^{TM}$ BWMS does not pose unacceptable effect on marine life. And $EC_{50}$ value of S. costatum on discharge water treated by BWMS using the electrolysis had positive correlation with initial TRO concentration, concentration and kind & level of HAAs.

Characteristics of Elements Extraction in Waste Rocks on the Abandoned Jangpoong Cn Mine (장풍 동광산 폐광석 내 원소의 용출 특성)

  • In order to evaluate the geochemical behaviors of elements with waste rocks in the abandoned Jangpoong Cu mine area, total concentration analysis and leaching experiments were performed. The content of elements within waste rocks compared with background values decreased in order of As>>Cu>Pb>Cd>Co. Leaching experiments were carried out at various extraction environments, considering the acid rain ($0.00001{\sim}0.001N\;HNO_3$) and the acid mine drainage ($0.001{\sim}0.1N$ HNO3). After 24 hours of reaction with different acidic solution, the leaching characteristics of waste rocks were classified into three types according to final pH of leaching solution. Type I refers to the case that the final pH of leaching solution was lower than that of the reaction solution due to the dissolution of acidic minerals from rocks, while type 2 and 3 refer to the case that the final pH maintained higher than that of the reaction solution. Theses types include in acid buffering minerals such as clay minerals and carbonate minerals. The leaching characteristics of the elements after the reaction could be categorized into As-Co-Fe, Cu-Mn-Cd-Zn, and Pb. As-Co-Fe started to get leached under 2.5 of pH regardless of changes in the final pH, and Cu-Mn-Cd-Zn showed different initial leaching pH according to the types of final pH changes. Based on the pH value where leaching started regardless of leaching concentration, the relative mobility of each element was in the order of Mn Zn>Cd>Cu>>Fe Co>As>Pb. Thus, more higher mobility elements(Zn, Mn and Cu) were leached by reacting with acid rain water. Acid mine drainage may result in distributions of elements having relatively less mobility(As, Fe, Co and Pb).

Effects of Plant Protein Source Containing Multienzyme on Performance and Milk Characteristics in Sow (포유모돈에 있어 복합효소제 함유 식물성 단백질 공급원이 생산성과 돈유성상에 미치는 영향)

  • A total of thirty sows(Landrace×Yorkshire) were used to determine the effects of plant protein source containing multienzyme on performance, nutrients digestibility and milk characteristics. A feeding trial was conducted for 21 days from parturition to weaning. Experimental diets were supplied for 1 week before the parturition day and throughout the experimental period. Dietary treatments included: 1) Control(CON; basal diet), 2) CGLT(included corn gluten) and 3) FSPM(included fermented soy protein containing multienzyme). Through the entire experimental period, backfat loss and return-to-estrus intervals were not affected by the treatments(P>0.05). Nitrogen digestibility was increased significantly(P<0.05) in FSPM treatment compared to CON treatment. Blood urea nitrogen(BUN) concentration was increased significantly (P<0.05) in FSPM treatment compared to CON treatment. At the initial  period, total protein content of milk was higher significantly(P<0.05) in FSPM treatment compared to CGLT treatment and at the final period, total fat content of milk was higher significantly(P<0.05) in FM treatment compared to CON treatment. Rectal temperature showed similar tendency of change among treatments. The final piglet body weight, weight gain and ADG were higher significantly in FSPM treatment compared to CON treatment. On diarrhea rate in piglet, just one piglet occurred in CGLT treatement. In conclusion, 2.5% dietary plant protein source containing multienzyme suppelmentation improved N digestibility, BUN concentration, fat and protein contents in milk and weight gain in piglet.

The Amino-Carbonyl Reaction in the Fructose-Glycine Mixture System (Fructose-Glycine 혼합계에 있어서 Amino-Carbonyl 반응)

  • This study was conducted to observe the physico-chemical exchange and effect of amino-carbonyl reaction between fructose and glycine . When various buffer solutions were added to equimolar mixture of fructose and glycine at pH 6.0 and $100^{\circ}C$, the browning effect was markedly observed by Mcllvaine buffer. Among the combinations of temperature and reaction time, the deep browning effect was obtained above $100^{\circ}C$, 3hr A marked browning effect obtained above pH 7.0 but little observed below pH 7.0. The browning effect was markedly increased at high fructose concentration. It required 4.0hrs and 32.9hrs to decrease 50% of initial concentration of fructose and glycine at $100^{\circ}C$ and pH 7 but 0.9hrs and 3.8hrs at $120^{\circ}C$, pH 7.0, respectively. The rate constant of fructose and glycine at $100^{\circ}C\;and\;120^{\circ}C$ were $1.78{\times}10^{-1},\;2.11{\times}10^{-2}\;and\;7.74{\times}10^{-1},\;1.83{\times}10^{-1}$, respectively. The formation of HMF was likely to follow the first order kinetics. The addition of 0.1M sodium sulfite, 0.1M sodium bisulfite and 0.1M calcium chloride to equimolar mixture (0.05M) surpressed the reaction up to 76.8%, 76.8% and 96.4%, respectively.

Effect of Paddy Drying by Solar Energy Concentration Blast-Grain Circulation Dryer (태양열집열송풍(太陽熱集熱送風), 곡물순환식(穀物循環式) 건조기(乾燥機)의 벼 건조효과(乾燥效果))

  • A 2.5 ton scale of solar energy concentration blast-grain circulation dryer (SECD) was developed in order to shorten the drying time without damaged paddy. Comparative experiments were carried out on performance, drying efficiency, consistency in moisture content, milling recovery, grade of milled rice, and energy requirement and cost against all that of in-bin drying and storage (IBDS) method. The experiments were performed using mixture of several rice varieties of Tongil type(Japonica-Indica breeding type) under the autumn weather in Korea. The circulating air temperature inside SECD was $4{\sim}5^{\circ}C$ higher than that of IBDS. The moisture content of the paddy during the drying period in SECD was uniform while substantially varied in upper, middle or bottom layer in IBDS. By SECD, 24% initial moisture content of paddy was reduced to 15% after only 3 days of drying as compared to 14 days at IBDS. The percentage of cracked kernels in upper, middle and bottom layers in IBDS was 6, 6 and 12%, respectively, whereas 7% in all layers in SECD. Both types of dryers did not significantly affect the milling recovery of dried paddy and grade of milled rice. Energy requirement of SECD(28.8Kw/2.5ton) for paddy drying was much less than that of IBDS(108Kw/2.5ton).

Effect of Aeration Rates on Emissions of Oxygen and Sulfur compound gases during Composting of Dairy Manure (우분(牛糞) 퇴비화시(堆肥花時) 공기주입률(空氣注入率)이 산소 및 황화합물 가스 배출(排出)에 미치는 영향(影響))

  • This experiment used the enclosed bench-scale reactors was conducted to find out optimal aeration rate for reducing the emission of odors and producing the good-quality compost with the mixture of dairy manure and rice straw. The reactors with gas sampler were aerated at four different rates of 0.09, 0.18, 0.90 and $1.79l\;min^{-1}kg^{-1}$dry solids for 574 hours. The oxygen content within composting pile instantly decreased after aeration. Oxygen limitation(below 15%) in the treatments of $0.90l\;min^{-1}kg^{-1}$ and less was exponentially negative relationship with aeration rates and in the range of 35 to 300 hours after aeration. However, the treatment of $1.79l\;min^{-1}kg^{-1}$ didn't show the oxygen limitation. The oxygen consumption rate and the cumulative amount of oxygen consumed by different aeration rates was ranged in $0.80{\sim}1.57O_2g\;h^{-1}\;kg^{-1}VS^{-1}$, $460{\sim}900O_2g\;kg^{-1}VS^{-1}$, respectively, and they were high in the order of 0.90, 1.79, 0.18, $0.09l\;min^{-1}kg^{-1}$. The maximum oxygen consumption rate was estimated in the range of $1.2{\sim}1.3lmin^{-1}kg^{-1}$. The emission concentrations of sulfur compounds such as hydrogen sulfide, sulfur dioxide and methylmercaptan were remarkably high in the initial composting time. Then they were rapidly decreased with the passing of composting time and clearly with increasing aeration rates. Their average concentrations were in the range of 0.03~2.18, 0~0.50, $0.07{\sim}3.38mg\;kg^{-1}$, respectively and high in the order of methylmercaptan, hydrogen sulfide, and sulfur dioxide. Concentrations of sulfur compounds emitted from composting showed exponentially negative relationship at 1% statistically with the oxygen concentration. It was estimated that hydrogen sulfide and methylmercaptan suddenly increased in the level of 5% oxygen concentration and below, that they were little emitted in 15% and over but sulfur dioxide was emitted in the level of 20% oxygen.

The Effect of Soybean Oil, Dried Palm Oil Powder and Monoglyceride Supplementation on Growth Performance and Serum Lipid Changes in Weaned Pigs (이유자돈에 있어 대두유, 건조팜유분말과 Monoglyceride의 첨가가 성장 및 혈청 지질변화에 미치는 영향)

  • This experiment was conducted to determine the Effect of soybean oil, dried palm oil powder and monoglyceride supplementation on growth performance and serum lipid changes in weaned pigs. One hundred twenty five cross-bred pigs((Y×L)×D, 6.00±0.79kg average initial BW) were used in a 21 d growth trial. Dietary treatments included SOY(containing 5% soybean oil), PALM(containing 5.5% dried palm oil powder) and SOPM0, 12.5 and 25(containing 2.5% soybean oil and 2.7% dried palm oil powder containing 0%, 12.5% and 25.0% monoglyceride, respectively). For the whole period, ADFI was increased in dried palm oil powder containing monoglyceride and PALM treatments compared with SOY and SOPM0 treatment(P<0.05). ADG and G/F were not affected by treatments. Digestibility of fat was higher(P<0.05) for pigs fed SOY diet than fed other diets. SOPM diets containing monoglyceride showed increased fat digestibility compared with PALM diet(P<0.05). SOPM12.5 and SOPM25 treatments were no difference with SOY treatment. However, there were no significant differences in digestibilities of DM, N and DE(P>0.05). Backfat thicknesses were not significantly different among treatments. Pigs fed SOY diet was decreased in serum total cholesterol, HDL-cholesterol, total lipid and was increased in free fatty acid than those fed other diet. Also, triglyceride concentration in serum was increased in PALM treatment compared with SOPM treatment(P=0.06). In conclusion, feeding soybean oil in weaned pigs showed higher fat digestibility and lower cholesterol and triglyceride concentration in serum than feeding only dried palm oil powder. Also, pigs fed monoglyceride were improved fat digestibility compared with those fed dried palm oil powder only.

Grazing Rate and Pseudofaeces Production of Native Snail Cipangopaludina chinensis malleata Reeve on Toxic Cyanobacterium Microcystis aeruginosa (한국산 논우렁이의 유해조류 섭식율 및 배설물 생산)

  • Grazing rates (GR) and pseudofaeces production (PFP) of native snail, Chinese mystery snail (Cipangopaludina chinensis malleata Reeve) on natural colonial morphs of Microcystis aeruginosa was measured. C. chinensis was collected from the upstream of the Geum River (Boryeong, Korea), where they co-habituated with Unio douglasiae and Lanceolaria acrorhyncha. The experiments were performed to evaluate the GR and PFP at different conditions such as; incubation time (1, 3, 5, 7, 9 and 11 hr), body size (3 to 6.1 cm, n=28), snail density (0.5, 1, 1.5 and 2.0 ind. $L^{-1}$) and prey concentration (168.3, 336.7, 505.0 and $673.0{\mu}g\;Chl-{\alpha}L^{-1}$). All experiments were triplicated, and conducted in transparent acrylic vessel (3L in volume). Regarding feeding time, a highest GR (0.538L $gAFDW^{-1}h^{-1}$) and PFP $(7.18mgAFDW^{-1})$ appeared at 1hr and 7hr after snail stocking, respectively. Interestingly, the snail, smaller than 4.5cm in body size, showed a wide range of GR ($-4.173{\sim}1.087L\;gAFDW^{-1}h^{-1}$) for the initial period (1 and 4hrs of stocking), compared to those greater than 4.5cm, which showed a stable FR, higher than 0.5L $gAFDW^{-1}h^{-1}$. Upon density effect, the density of 1.5 ind. $L^{-1}$ induced the most effective inhibition on Microcystis biomass with highest PFP. On the prey concentration, highest GR (0.897L $gAFDW^{-1}h^{-1}$) and PFP (3.67 mg $gAFDW^{-1}h^{-1}$) were induced at the level of $168.3{\mu}g\;Chl-{\alpha}L^{-1}$ and $673{\mu}g\;Chl-{\alpha}L^{-1}$, respectively. GR and PFP of this freshwater snail on the cyanobacterial bloom (M. aeruginosa) varied with the feeding conditions, and they were comparatively high for a short period of time less than 7hrs regardless of the stocking condition. Our results suggest that this freshwater snail has a potential to control cyanobacterial bloom when provided with suitable condition.
