  • Dehydroquinate dehydratase (DHQD) catalyzes the conversion of 3-dehydroquinic acid (DHQ) into 3-dehydroshikimic acid in the mid stage of the shikimate pathway, which is essential for the biosynthesis of aromatic amino acids and folates. Here, we report two the crystal structures of type II DHQD (CgDHQD) derived from Corynebacterium glutamicum, which is a widely used industrial platform organism. We determined the structures for CgDHQDWT with the citrate at a resolution of 1.80Å and CgDHQDR19A with DHQ complexed forms at a resolution of 2.00 Å, respectively. The enzyme forms a homododecamer consisting of four trimers with three interfacial active sites. We identified the DHQ-binding site of CgDHQD and observed an unusual binding mode of citrate inhibitor in the site with a half-opened lid loop. A structural comparison of CgDHQD with a homolog derived from Streptomyces coelicolor revealed differences in the terminal regions, lid loop, and active site. Particularly, CgDHQD, including some Corynebacterium species, possesses a distinctive residue P105, which is not conserved in other DHQDs at the position near the 5-hydroxyl group of DHQ. Replacements of P105 with isoleucine and valine, conserved in other DHQDs, caused an approximately 70% decrease in the activity, but replacement of S103 with threonine (CgDHQDS103T) caused a 10% increase in the activity. Our biochemical studies revealed the importance of key residues and enzyme kinetics for wild type and CgDHQDS103T, explaining the effect of the variation. This structural and biochemical study provides valuable information for understanding the reaction efficiency that varies due to structural differences caused by the unique sequences of CgDHQD.

  • The presence of traditional media content on online platforms is one of the critical issues nowadays, and Article 17 of the European Media Freedom Act (EMFA) seeks to regulate this. However, it can be seen that the current version of the text is not yet free of flaws: both its harmonisation with the Digital Services Regulation, its use of definitions and the media fast track mechanism it contains would require careful legislative scrutiny before the final text is adopted. The article examines if the self-declaration procedure envisaged by the EMFA would create a loophole for rogue media actors and bring confusion at both the European and horizontal levels or if it would fit the original goal of the EMFA, which is to improve the functioning of the internal European media market and to reinforce the independent media.

  • In various fields such as art, design, music, film, sports, games, and fashion, NFTs (Non-Fungible Tokens) are creating new economic value through trading platforms dedicated to NFT art and content. In this article, I analyze the current state of blockchain technology and NFT art content in the context of an expanding market for blockchain-based NFT art content in the metaverse. I also propose several tasks based on the economic and industrial logic of technological innovation. The first task proposed is to integrate cultural arts on blockchain, metaverse, and NFT platforms through digital innovation, instead of separating or distinguishing between creative production and consumption. Before the COVID-19 pandemic, there was a clear separation between creators and consumers. However, with the rise of Web 3.0 platforms, any user can now create and own their own content. Therefore, it is important to promote a collaborative and integrated approach to cultural arts production and consumption in the blockchain and metaverse ecosystem. The second task proposed is to align the legal framework with blockchain-based technological innovation. The enactment and revision of relevant laws should focus on promoting the development of the NFT trading platform ecosystem, rather than merely regulating it for user protection. As blockchain-based technology continues to evolve, it is important that legal systems adapt to support and promote innovation in the space. This shift in focus can help create a more conducive environment for the growth of blockchain-based NFT platforms. The third task proposed is to integrate education on digital arts, including metaverse and NFT art contents, into the current curriculum. This education should focus on convergence and consilience, rather than merely mixing together humanities, technology, and arts. By integrating digital arts education into the curriculum, students can gain a more comprehensive understanding of the potential of blockchain-based technologies and NFT art. This article examines the digital technological innovation such as blockchain, metaverse, and NFT from an economic and industrial point of view. As a limitation of this research, the critical mind such as philosophical thinking or social criticism on technological innovation is left as a future task.

  • It has been reported that the administration of Limosilactobacillus fermentum alleviates diseases such as osteoporosis and colitis. In this study, we report the complete genome sequence of Limosilactobacillus fermentum KUFM407, a probiotic strain of LAB isolated from Korean traditional fermented food, Kimchi. Whole genome sequencing of L. fermentum KUFM407 was performed on the Illumina MiSeq and Oxford Nanopore MinION platform. The genome consisted of one circular chromosome (2,077,616 base pair [bp]) with a guanine cytosine (GC) content of 51.5% and one circular plasmid sequence (13,931 bp). Genome annotation identified 1,932 protein-coding genes, 15 rRNAs, and 58 tRNAs in the assembly. The function annotation of the predicted proteins revealed genes involved in the biosynthesis of bacteriocin and fatty acids. The complete genome of L. fermentum KUFM407 could provide valuable information for the development of new probiotic food and health supplements.

  • Xiaohongshu, a community-centric social media platform, has pioneered a unique e-commerce model known as 'buyer commerce,' leveraging user-generated content (UGC). Distinctively, Xiaohongshu Live Commerce focuses on fostering deep user relationships and providing superior product and information services, crucial for sustained consumer engagement. This study investigates consumer behavior in purchasing health functional foods via Xiaohongshu Live Commerce, aiming to understand the determinants of continuous usage intention. A novel theoretical framework was devised by integrating the Expectation Confirmation Model (ECM) and the Task-Technology Fit (TTF) model. The research model scrutinizes the impact of Xiaohongshu Live Commerce characteristics, such as perceived usefulness and perceived online intimacy, on task-technology fit. Additionally, it examines the moderating role of perceived risk specific to health functional foods and the influence of expectation confirmation on perceived usefulness, online intimacy, and task-technology fit, alongside their effects on satisfaction and continuous usage intention. The findings reveal that expectation confirmation positively influences perceived usefulness, online intimacy, and task-technology fit. Perceived usefulness significantly enhances task-technology fit, while perceived online intimacy and risk do not significantly affect task-technology fit. Moreover, perceived usefulness and intimacy positively impact consumer satisfaction and continuous usage intention, with task-technology fit playing a pivotal role. Perceived risk moderates the relationship between perceived usefulness and task-technology fit. These insights suggest that companies can augment consumer satisfaction and continuous usage intentions by enhancing the perceived usefulness of technology, effectively managing perceived risks, and continually improving user experience
