The Induction of Steroid ${\Delta}^1$-dehydrogenase from Arthrobacter simplex IAM 1660 (Arthrobacter simplex의 Steroid ${\Delta}^1$-dehydrogenase의 유도와 유도성 스테로이드의 성질)

  • - Since steroid $\Delta^1$-dehydrogenase synthesis has been known to be inducible, the mechanism of the enzyme induction of Arthrobacter simplex IAM 1660 was investigated. Among various steroids tested for inducers, hydrocortisone was the most effective inducer when hydrocortisone was used as a substrate for steroid $\Delta^1$-dehydrogenase. Steroid $\Delta^1$-dehydrogenase synthesis was effectively induced by progesterone, prednisolone and androstenedione, while the enzyme was less induced by cholesterol and not by phytosterols. The results suggest that the presence of 3-keto group and short side chain of steroids are the favorable factors for the induction of the $\Delta^1$-dehydrogenase synthesis. The enzyme was induced at the highest level when hydrocortisone was added at early log phase to the concentration of 0.01% of the culture and the culture was grown for 15 hours.

Isolation of Alkaline Amylase-Producing Bacillus sp. and Some Properties of Its Crude Enzyme (알칼리성 아밀라아제를 생산하는 Bacillus속 미생물의 분리와 그 조효소의 특성)

  • An alkaline amylase-producing Bacillus sp. GM8901 was isolated and some properties of crude enzyme extract were examined. The microbiological and biochemical characteristics of GM8901 were very similar to those of B. licheniformis. The optimal temperature and pH for the cell growth and amylase production were $50^{\circ}C$ and pH 10.5. The crude amylase extract showed that the optimal temperature and pH were $50{\sim}60^{\circ}C\;and\;pH\;10{\sim}12$, respectively, and that the activity of amylase was stable up to $50^{\circ}C$ and in the range of $pH\;3{\sim}12$.

  • Physiological changes of Escherichia coli during the fed-batch fermentation process were characterized in this study. Overall cellular protein samples prepared at the different stage of fermentation were separated by 2-dimensional gel electrophoresis (2-DE), and differently expressed 15 proteins, Phosphotransferase enzyme I, GroEL, Trigger factor, ${\beta}$ subunit of ATP synthase, Transcriptional regulator KDGR, Phosphoglycerate mutase 1, Inorganic pyrophosphatase, Serine Hydroxymethyl-transferase, ${\alpha}$ subunit of RNA polymerase, Elongation factor Tu, Elongation factor Ts, Tyrosine-tRNA ligase, DnaK suppressor protein, Transcriptional elongation factor, 30S ribosomal protein S6 were identified using matrix-assisted laser desorption / ionization time-of-flight mass spectrometry (MALDI-TOF MS). When bacterial cells grow to high cell density, and IPTG-inducible heterologous protein is produced, expression level of overall cellular proteins was decreased. According to their functions in the cell, identified proteins were classified into three groups, proteins involved in transport process, small-molecule metabolism, and synthesis and modification of macromolecules.

  • Heme oxygenase (HO)-1 is an inducible enzyme that catalyzes the first and rate-limiting step in the oxidative degradation of free heme into ferrous iron, carbon monoxide (CO), and biliverdin (BV), the latter being subsequently converted into bilirubin (BR). HO-1, once expressed during inflammation, forms high concentrations of its enzymatic by-products that can influence various biological events, and this expression is proven to be associated with the resolution of inflammation. The degradation of heme by HO-1 itself, the signaling actions of CO, the antioxidant properties of BV/BR, and the sequestration of ferrous iron by ferritin all concertedly contribute to the anti-inflammatory effects of HO-1. This review focuses on the anti-inflammatory mechanisms of HO-1 actions and its roles in inflammatory diseases.

  • Extracellular capsidiol, sesquiterpenoid phytoalexin, in the medium of pepper (Capsicum annuum L.) suspension cells was not identified from control cells, but highly accumulated in the elicitor-induced cells within 6 hours after the addition of 0.05$\mu\textrm{g}$/$m\ell$ cellulase. Capsidiol production in elicitor-induced cells was markedly suppressed by cytochrome P450 inhibitors, such as ancymidol and ketoconazole demonstrating that biosynthesis of capsidiol is catalyzed by at least on hydroxylation enzyme in the biochemical pathway. Based on protein electrophoresis, two bands, 23.0kDa and 27.5kDa, were identified as newly synthesized polypeptides in the elicitor-induced suspension cells, suggesting that pepper cells which were subjected to elicitor treatment activate specific gene(s) for capsidiol biosynthesis in cultured cells.

  • A strain producing extracellular endo-type inulase was selected from Actinomycetes isolated from soil, and identified as Streptomyces sp. The maximum inulase production was obtained with medium containing inulin 1.0%, yeast extract 1.0%, (NH$_4$)$_2$HPO$_4$ 0.4%, NH$_4$H$_2$PO$_4$0.8%, KCl 0.05%, MgSO$_4$ㆍ7$H_2O$ 0.05%, FeSO$_4$ㆍ7$H_2O$ 0.001% at 96 hours culture in jar fermentor. The endo-type inulase was considered to be an inducible enzyme produced by inulin only.

Isolation of a Pseudomonas aeruginosa Strain Capable of Degrading Acrylamide

  • A new strain of Pseudomonas aeruginosa growing in a rice field contaminated with herbicide and effluents of a factory manufacturing explosives was isolated. This isolate showed excellent growth in unusually high concentration of acrylamide (60 mM). It utilized acrylamide as the sole source of carbon and nitrogen for growth. Other amides such as acetamide, butyramide, isobutyramide, and methacrylamide were also utilized for the growth by this isolate. Acrylamide was degraded into acrylic acid and ammonia by the enzyme amidase. More than $65\%$ of added acrylamide (40 mM) was converted into acrylic acid after 40 h of growth of the culture. Amidase activity was inducible, the highest activity being observed with isobutyramide ($12.5{\mu}M$ ammonia/mg protein/min). These results demonstrate that this bacterium can degrade a variety of amides.

  • Cyclooxygenase-2 (COX-2) is an inducible enzyme expressed in response to a variety of cytokines and other proinflammatory stimuli. It has been known that aberrant up-regulation of COX-2 is associated with resistance to apoptosis. Contrary to the above notion. treatment of MCF10A-ras cells with the anti-tumor agent ET -18-O-$CH_3$ caused increased expression of COX-2 and its mRNA transcript. while inducing apoptosis as revealed by proteolytic cleavage of poly(ADP-ribose)polymerase. caspase-3 activation, and positive TUNEL staining.   • Cyclooxygenase-2 (COX-2) is an inducible enzyme expressed in response to a variety of cytokines. The presence of oncogenic ras has been associated with sustained induction of COX-2, which confers resistance to apoptosis. Contrary to the above notion, we found that MCF10A-ras cells treated with an anti-tumor agent, ET-18-O-$CH_3$, underwent apoptosis as revealed by proteolytic cleavage of poly(ADP-ribose)polymerase, pro-caspase 3 activity, and TUNEL staining, while the same treatment caused an increased expression of COX-2 as well as the elevated production of prostaglandin E$_2$(PGE$_2$).(omitted)

  • In activated macrophages the inducible form of nitric oxide synthase (i-NOS) generates high amounts of toxic mediator, nitric oxide (NO) which contributes to the circulatory failure associated with septic shock. A sesquiterpene lactone compound (yomogin) isolated from medicinal plant Artemisia princeps Pampan inhibited the production of NO in LPS-activated RAW 264.7 cells by suppressing i-NOS enzyme expression. Thus, yomogin may be a useful candidate for the development of new drugs to treat endotoxemia and inflammation accompanied by the overproduction of NO.
