가상 이상 개념을 이용한 물리법칙의 2D 시각화 시뮬레이션 (Two Dimensional Visualization Simulation of Physical Law using Virtual Anomaly)

  • 경험을 통한 지식의 습득이 단순한 기호나 수식을 통한 지식의 습득과 비교해서 보다 안정적이고 명료하게 학습자에게 정보를 전달한다. 컴퓨터 기반의 교육용 시스템은 가상현실기술, 지능형 교육 시스템, 개념의 시각화기술, 시뮬레이션 기반 시스템, 미세한 세계(MicroWorld)를 이용한 형태로 연구되어지고 있다. 본 논문에서는 교육용 시스템의 여러 형태 중에서 시뮬레이션 기반으로 물리법칙을 2D로 시각화하여 학습자에게 교육할 수 있는 방법을 제안한다. 특히 물리법칙 중 중력의 법칙을 시뮬레이션하여 확인함으로써 간접적 경험을 통하여 보다 명료하게 지식을 습득한다. 시뮬레이션을 통한 명확한 지식 전달을 위해서 본 논문에서는 실시간적인 물리법칙의 시각화, 가상 이상 현상(virtual anomaly), 물리법칙의 시각화를 시뮬레이션한다. 그리고 학습자는 이러한 방법들을 통해서 효율적이고 효과적인 지식습득이 가능함을 보인다.

  • The fashion of late Yi dynasty had explored a possibility of new fashional beauty from its own experiences throughout the history of Korean fashion excluding any interference of foreign fashions, and brought out some characteristics ; first, the aesthetical value focused on the more human fashion resulted in the exposure of waist. This aesthetical change in the fashion of late Yi dynasty produced out results as follows ; the length and width of jacket became short and narrow maintaining the basic form, while the width of skirt became increased to be contrasted exceptionally with the upper coat. This mode of fashion reveals the transitional movement from the emphasis on balance to that on unbalance in the late period of Yi dynasty. But regarding the fact that the proportional change by the ways of putting on skirt became to be similar to the golden ratio, we can see many examples throughout the folk paintings by Shin Yun bok, we find that the fashional beauty of that age pursued ideal harmony and unity all the time being in spite of its tendency toward non-refinement. Second, we see another peculiarity in the mode of woman's fashion from the paintings by Shin Yun-bok who always depicted the hu-man nature frankly. For example, the jacket became shorter to emphasize lady's slim waist, and underwears began to be exposed after the upper part of skirt had been to expose lady's waist while the bottom line pulled up highter than before. Thus, as the function of robe had changed from a mere means of covering into an expression of beauty, the fashion of late Yi dynasty began to express a perfectly new beauty possessing an erotic mood and sensu-ality of a woman, which was its inner aesthetic need. Third, this emergence of new fashional beauty made the mental value of man and the aesthetic views of that time quite different from the previous ones, and finally there came various expressions of a new beauty. There-fore, the woman's fashion of late Yi dynasty is noted for the exposure and emphasis on the body line of a woman after the aesthetic view-point had changed of time toward pragmatism and humanism, while upholding the traditions of Korean clothes and pride for the nation. And our ancestors realized very Korean fashional beauty by pursuing the expressional ways of gentle, indirect emphasis upon the feminine and natural beauty to exclude and ar-tificial, direct and straightforward exposures and images.

Anti-proliferation Effects of Interferon-gamma on Gastric Cancer Cells

  • IFN-${\gamma}$ plays an indirect anti-cancer role through the immune system but may have direct negative effects on cancer cells. It regulates the viability of gastric cancer cells, so we examined whether it affects their proliferation and how that might be brought about. We exposed AGS, HGC-27 and GES-1 gastric cancer cell lines to IFN-${\gamma}$ and found significantly reduced colony formation ability. Flow cytometry revealed no effect of IFN-${\gamma}$ on apoptosis of cell lines and no effect on cell aging as assessed by ${\beta}$-gal staining. Microarray assay revealed that IFN-${\gamma}$ changed the mRNA expression of genes related to the cell cycle and cell proliferation and migration, as well as chemokines and chemokine receptors, and immunity-related genes. Finally, flow cytometry revealed that IFN-${\gamma}$ arrested the cells in the G1/S phase. IFN-${\gamma}$ may slow proliferation of some gastric cancer cells by affecting the cell cycle to play a negative role in the development of gastric cancer.

Defining the N-Linked Glycosylation Site of Hantaan Virus Envelope Glycoproteins Essential for Cell Fusion

  • The Hantaan virus (HTNV) is an enveloped virus that is capable of inducing low pH-dependent cell fusion. We molecularly cloned the viral glycoprotein (GP) and nucleocapsid (NP) cDNA of HTNV and expressed them in Vero E6 cells under the control of a CMV promoter. The viral gene expression was assessed using an indirect immunofluorescence assay and immunoprecipitation. The transfected Vero E6 cells expressing GPs, but not those expressing NP, fused and formed a syncytium following exposure to a low pH. Monoclonal antibodies (MAbs) against envelope GPs inhibited cell fusion, whereas MAbs against NP did not. We also investigated the N-linked glycosylation of HTNV GPs and its role in cell fusion. The envelope GPs of HTNV are modified by N-linked glycosylation at five sites: four sites on G1 (N134, N235, N347, and N399) and one site on G2 (N928). Site-directed mutagenesis was used to construct eight GP gene mutants, including five single N-glycosylation site mutants and three double-site mutants, which were then expressed in Vero E6 cells. The oligosaccharide chain on residue N928 of G2 was found to be crucial for cell fusion after exposure to a low pH. These results suggest that G2 is likely to be the fusion protein of HTNV.

OPTHiS Identifies the Molecular Basis of the Direct Interaction between CSL and SMRT Corepressor

  • Notch signaling is an evolutionarily conserved pathway and involves in the regulation of various cellular and developmental processes. Ligand binding releases the intracellular domain of Notch receptor (NICD), which interacts with DNA-bound CSL [CBF1/Su(H)/Lag-1] to activate transcription of target genes. In the absence of NICD binding, CSL down-regulates target gene expression through the recruitment of various corepressor proteins including SMRT/NCoR (silencing mediator of retinoid and thyroid receptors/nuclear receptor corepressor), SHARP (SMRT/HDAC1-associated repressor protein), and KyoT2. Structural and functional studies revealed the molecular basis of these interactions, in which NICD coactivator and corepressor proteins competitively bind to ${\beta}-trefoil$ domain (BTD) of CSL using a conserved ${\varphi}W{\varphi}P$ motif (${\varphi}$ denotes any hydrophobic residues). To date, there are conflicting ideas regarding the molecular mechanism of SMRT-mediated repression of CSL as to whether CSL-SMRT interaction is direct or indirect (via the bridge factor SHARP). To solve this issue, we mapped the CSL-binding region of SMRT and employed a 'one- plus two-hybrid system' to obtain CSL interaction-defective mutants for this region. We identified the CSL-interaction module of SMRT (CIMS; amino acid 1816-1846) as the molecular determinant of its direct interaction with CSL. Notably, CIMS contains a canonical ${\varphi}W{\varphi}P$ sequence (APIWRP, amino acids 1832-1837) and directly interacts with CSL-BTD in a mode similar to other BTD-binding corepressors. Finally, we showed that CSL-interaction motif, rather than SHARP-interaction motif, of SMRT is involved in transcriptional repression of NICD in a cell-based assay. These results strongly suggest that SMRT participates in CSL-mediated repression via direct binding to CSL.

Antioxidant and Anti-inflammatory Properties of Raw and Processed Fruits and Vegetables

  • Reactive oxygen species (ROS) generated from metabolic reactions cause oxidative DNA damage, which results in oxidative tissue injury. Therefore, there is an increasing demand in the intake of high antioxidant sources in order to maintain a healthy environment in cells. In this study, we investigated the antioxidant and anti-inflammatory activities of Malus domestica (apple), Pyrus communis L. (pear), Daucus carota L. (carrot), Brassica oleracea var. (broccoli), Brassica oleracea var. capitata (cabbage), and Raphanus sativus L. (radish) obtained from the local market. Since these are common fruits and vegetables that are widely consumed, we aimed to investigate their beneficial properties, placing particular emphasis on their antioxidant and anti-inflammatory properties. The samples were processed via an indirect heating method and their properties were compared to their raw forms. Based on DPPH and ABTS assays, processed samples showed better antioxidant activities when compared to raw samples and processed pear samples exhibited the best antioxidant activity. The anti-inflammatory activities of the samples were also investigated in LPS-treated RAW 264.7 cells. mRNA expression of pro-inflammatory mediators and cytokines (iNOS, COX-2, $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6) was assessed using RT-PCR. As expected, processed samples exhibited better iNOS inhibition when compared to their raw forms and processed broccoli and cabbage samples exhibited outstanding anti-inflammatory effects. The samples, up to 1 mg/mL concentration, did not exhibit cytotoxicity against RAW 264.7 cells as demonstrated by cell viability assays. Altogether, processed broccoli and cabbage samples exhibited the strongest anti-inflammatory properties.

Modulation of Glial and Neuronal Migration by Lipocalin-2 in Zebrafish

  • Background: Glial cells are involved in immune and inflammatory responses in the central nervous system (CNS). Glial cells such as microglia and astrocytes also provide structural and functional support for neurons. Migration and morphological changes of CNS cells are associated with their physiological as well as pathological functions. The secreted protein lipocalin-2 (LCN2) has been previously implicated in regulation of diverse cellular processes of glia and neurons, including cell migration and morphology. Methods: Here, we employed a zebrafish model to analyze the role of LCN2 in CNS cell migration and morphology in vivo. In the first part of this study, we examined the indirect effect of LCN2 on cell migration and morphology of microglia, astrocytes, and neurons cultured in vitro. Results: Conditioned media collected from LCN2-treated astrocytes augmented migration of glia and neurons in the Boyden chamber assay. The conditioned media also increased the number of neuronal processes. Next, in order to further understand the role of LCN2 in the CNS in vivo, LCN2 was ectopically expressed in the zebrafish spinal cord. Expression of exogenous LCN2 modulated neuronal cell migration in the spinal cord of zebrafish embryos, supporting the role of LCN2 as a cell migration regulator in the CNS. Conclusion: Thus, LCN2 proteins secreted under diverse conditions may play an important role in CNS immune and inflammatory responses by controlling cell migration and morphology.

도시가로경관의 이미지 동질화를 위한 환경설계적 고찰 - 대구시 동성로를 중심으로 (An Environmental Study on the Image Identification of Urban Streetscape (The Case Study of Tongsung-Ro in Taegu City))

  • A study on the image identification of urban streetscape is valuable for illuminating identity that is not yet fully approached in the field of environmental design. This analysis of urban streetscape for image identification allows us to make a more detailed exploration of an important approaching methods in dealing with the structural characteristics of identity. As a matter of fact, the earlier indirect studies on this image identification were made by environmental designers, such as architectural and urban designer in the field of environmental perception and came to its environmental cognition & environmental pattern research with assistances by such researchers as K. Lynch A. Rapoport & Christopher Alexander. Through its environmental perception research, we can see its structural characteristics that is aesthetic & visual structural contents of physical environmental elements. And we can see its cognitive characteristics through the environmental cognitive research, that is continuity, territoriality, identity of place, uniqueness or individuality, meaning & symbolism. Through its environmental pattern research, we can see its physical, socio - economic, cultural and symbolic pattern identification contents, that is physical form of the city, style of the street, pattern of streetscape, socio- economic & geographical locality, arid life cycle, life style, common style of the behavior, cultural pattern of the activity, socio - cultural expression of the symbol. In these process, we can set up a set of the environmental design criterias from those three integral studies for identity. And for an environmental research, Tongsung-Ro around the CBD (central business district) in Taegu City was selected for a case study, because this streetscape is suitable for that approaching methods in this study.

무인 헬리콥터 사진촬영시스템을 이용한 도로 절개지 붕괴사면 3차원 입체 지형 추출 (3D Stereoscopic Terrain Extraction of Road Cut Failure Slope Using Unmanned Helicopter Photography System)

  • 붕괴된 사면의 정보는 제2차 피해의 발생우려가 있기 때문에 신속 정확하게 지형자료를 획득할 수 있어야 하고, 필요에 따라서 접근하지 않고 간접적인 측량방식으로 보다 효율적으로 표현할 수도 있어야 한다. 따라서 본 연구에서는 무인 헬리콥터 사진촬영시스템을 이용하여 붕괴된 도로 절개지 사면에 근접하여 상공에서 정지비행으로 대상지역의 영상을 촬영하였다. 그리고, 무타켓 Total Station에 의해 관측된 점과 영상해석에 의해 분석된 좌표점 10개와 비교한 결과 절대치 평균값은 X축 방향으로 평균 0.056m, Y축 방향으로 0.082m, Z축 방향으로 0.066m으로 나타났다. 또한, 검사점 10점에 대한 오차의 RMSE는 X축 방향으로 0.015636m, Y축 방향으로 0.021319m, Z축 방향으로 0.018734m로 나타났다. 따라서, 이러한 절개지 붕괴사면의 지형에 대하여 관계형 영상정합방법에 의해 접근하지 못하는 위험지역에서의 사면 범위와 각사면의 종 횡단면을 나타낼 수가 있었다.

메이지·다이쇼 작품의 てくれ·てください의 표현 양상 (Expressions of requests using give and receive verbs in the era of Meizi and Taisyo)

  • Request expressions can be defined as expressions that demand or ask the other person to do certain movements. There are direct request expressions that ask the other person to do certain movements directly and indirect request expressions that ask the other person to do certain movements by describing the speaker's condition. The study analyzed gender and hierarchy of speakers and listeners who used 'tekure' and 'tekudasai' in dialog examples of the Meiji Period and the Taisho Period. In those periods, the modern Tokyo dialect was formed and established. "Toseishoseikatagi"in Meiji 10s,"Ukigumo""Natsukodachi""Tajotakon"in Meiji 20s,"Hakai""Botchan"in Meiji 30s,"Huton""Inakakyoshi" in Meiji 40s and "Aruonna"in the Taisho Period were analyzed for the study. 'kure' was used more by male speakers than female speakers. Examples by female speakers were shown on the novels after Meji 30s. In case of male speakers, they often used it to listeners with an equitable relationship at "Toseishoseikatagi"in Meiji 10s but they often used it to younger listeners at "Hakai"in Meiji 30s. 'okure' was used more by female speakers than male speakers. Listeners were varied from older ones to younger ones. In case of female speakers, 'okure' was used more often at "Aruonna"in the Taisho Period than the other novels. In case of male speakers, 'okure' was used only at "Ukigumo""Natsukodachi"and "Hakai". 'Okurenasai' was used outstandingly by female speakers on the form of 'okun_'. In case of 'kudasai', female speakers used it more than male speakers at "Toseishoseikatagi" and "Aruonna"but male speakers used it more than female speakers at "Tajotakon"and "Hakai". Listeners were varied from older ones to younger ones. 'o~kudasai' was not shown until Meiji 20s but shown after Meiji 30s among the analyzed novels. According to gender, it was used a little bit more often by female speakers than male speakers. According to hierarchy, listeners were usually older than speakers. 'o~nasatekudasai' was used more often by male speakers than female speakers. Listeners were also usually older than speakers.
