Changes in the Growth Characteristics and the Content of Decursin and Decursinol angelate in Angelica gigas Nakai by Mineral and Hormonal Agent Treatment (미네랄 및 호르몬 제제 처리에 따른 당귀의 생육특성 및 Decursin, Decursinol Angelate 함량 변화)

  • Background: Angelica gigas Nakai is a perennial herb belonging to the family Umbelliferae. Its roots are utilized in traditional medicine. The aim of this study was to increase the yield of and the content of two indicator components (decursin and decursinol angelate) in A. gigas Nakai. Methods and Results: The roots of A. gigas Nakai were harvested 4-times from late August to late October in 2017. Two agents (trace element-TE, and plant hormone-HM) were applied 4 times at intervals of 2 weeks. The content of the two indicator components were analyzed by high performance liquid chromatography. The HM treatment showed the greatest increase in underground part yield and root diameter. The content of the two indicator components in the control (non-treatment) group was the highest in the underground part, but was higher in the aerial parts in the agent treatment group. After treatment with the agents, the content of the indicator components tended to decrease in the underground part. However, the total content of the indicator components in the two agent treatment groups exceeded the level of 6% set by the Korean Pharmacopoeia. Conclusions: The highest underground part yield was found in the HM treatment group, while the highest content of decursin and decursinol angelate were found in the control group. This study provides basic information for yield improvement in A. gigas.

Evaluation of Agri-Environmental Performance of Convention on Biological Diversity Using Winter Paddy Field in Korea

  • Winter migratory birds gather in paddy rice fields to feed shed rice grains. The Korean Ministry of Environment has practiced a policy program Contract on Paddy Field Management (CPFM) during winter fallow since 2002. This program starts with a contract between local governments and farmers, and the government pays a differential subsidy to farmers who finish spreading rice straw, cultivating barley, letting the whole rice plant without harvest, and submerging paddy fields for winter migratory birds. As more local governments have operated CPFM program, the total area on the contract and subsidy budget has increased yearly since 2002. This program could have its stable position as a successful policy by giving profits to farmers. With the program extended, the population of winter migratory birds has been greatly. For the evaluation of environmental performance of a policy, we analyzed this CPFM program by introducing some indicators in the form of Driving Force-State-Response Framework. The indicators were composed of 3 categorized indicators; the area of paddy land contracted under this program as 'driving force' indicator, population of birds, the number of bird species and the amount of feces as 'state' indicators, and the size of monetary support, the number of farmers or local governments participating, and public perception as 'response' indicators. The contract area of paddy field under CPFM could be a good biodiversity indicator reflecting potential performance of this policy measure in the light of its linkage to the population of winter migratory birds. And the share of CPFM land of the whole agricultural land might also be used as a useful indicator of policy evaluation for improvement of wildlife diversity. The 'state' indicators such as population of birds and the number of bird species could be matched to 'driving force' indicator, but the total CPFM area of each site could not reflect the effect of areas under different management practices. However, the amount of bird feces could reflect differences in environmental performance with management practices as 'state' indicators. The development of indicators indicating 'response' such as farmers' behavior, public perception, and policy makers' willingness is also needed to support a successful implementation and improvement of this policy measure with the development of 'performance' indicators integrating all these indicators.

Studies on the microbiological assay method for tabtoxin produced in pseudomonas syringae pv. tabaci (Pseudomonas syringae pv. tabaci가 생산하는 tabtoxin의 미생물학적 검색방법에 관한 연구)

  • Tabtoxin produced in Pseudomonas syringae pv. tabace irreversibly inhibits its known physiological target, glutamine synthetase so that causes wildfire disease on leaves of host plant. In this study, we examined a rapid and sensitive microbiological method for tabtoxin assay in several media. In minimal A agar medium nd minimal glucose agar medium, growth inhibition zone of Agrobacterium tumefaciens was larger than that of other indicator strain. However, mostly, growth inhibition zone of indicator strains on the minimal glucose agar medium was smaller than that of on the miniaml A agar medium. In complex agar medium, growth inhibithiton zone was not observed in all the tested indicator strains. Pseudomonas syringae pv. tabaci produced more tabtoxin according to the incubation time. When glutamine was added to the minimal glucose agar medium, growth inhkbition zone of Agrobacterium tumefaciens was reduced.

An inventory of Korean living collections in the Arnold Arboretum of Harvard University, USA

  • As part of a study to understand the current status of Korean plant species propagated in foreign countries, we conducted extensive surveys on Korean living collections in the Arnold Arboretum of Harvard University, the oldest public arboretum in North America. Specifically, we collected and identified the voucher specimens of the living collections and further examined the collection databases to understand the history of how Korean plants were introduced to the Arboretum. We found that a total of 154 Korean taxa (belonging to 49 families and 81 genera) have been planted in the arboretum. Most plants originated from seeds that were directly collected in Korea or obtained via seed exchanges with botanical gardens in Asia, Europe, and U.S.A. Of the 154 taxa, 16 are Korean endemic species, and three are listed as endangered species. In addition, 62 taxa are as floristic regional indicator species of Korea used by the Ministry of Environment.

A New Method on the Measurement of Catalase Activity of Panax ginseng C.A. Meyer Tissues (인삼조직에서 Catalase Activity측정에 관한 새로운 Method)

  • We report a newassay method on the measurement of the catalase activity, whose utilzation value is considered to be remarkable in the field of plant biochemistry. We named this method as a De-Coupling method. The essence of de-coupling method is the separation between the enzyme reaction and the indicator reaction. The optimum condition of the enzyme reaction was found to be following: on addition of 1 ml of substrate (H2O2: 20mM) to the fixture of the crude extract of enzyme (volume: 0.2 ml) and the ammonium phosphate buffer (volume: 1.8 ml; 0.93 M phosphate, 1.6M NHB, 2.5 M methanol, pH 7.0). After 30, 60 and 90 seconds of the enzyme reactions are proceeded, the reactions are terminated by 25% of tai-chloro-acetate (final concentration of 5%), respectively. The precipitated materials by tai-chloro-acetate was removed by the centrifugation (2000g, 10minutes). Formaldehyde produced in the enzymatic reaction was reacted with 2ml of acetylacetone (60mM). The indicator reaction -(HANTSCH REAKT10N)- in which lutidine is formed, was proceeded for 60 minutes at $25^{\circ}C$.

  • Digestibilities of nutrients and energy are among the most important parameters to be determined in feed evaluation research. The apparent digestibility coefficients (ADC) of protein, total carbohydrate (TCHO), fat, energy and amino acids were determined for 14 common feed ingredients using chromic oxide as external indicator with Israeli carp (Cyprinus carpio). The ingredients tested were; corn, corn starch, gelatinized starch, wheat middling, wheat grade inferior, corn gluten meal, rapeseed meal (solvent extracted), soybean meal (solvent extracted), blood meal (drum dried), feather meal(hydrolyzed), file fish meal (flame dried), sardine fish meal (steam dried), sardine fish meal (flame dried) and brewers yeast (dehydrated). The overall ADC values were high in Israeli carp showing high capacity to digest their feed ingredients irrespective of plant or animal sources. In addition the ADC of plant protein was high enough to support the successful supplementation of fish meal with other plant proteins.

Studies on Purification and Serology of Potato Virus X (감자바이러스 X의 순화와 혈청학적 연구)

  • Potato virus X was purified especially for the preparation of antisera for diagnosis and identification. Potato virus X was isolated Iron infected plants by means of indicator plants and identified in electron microscopy. Isolated PVX was multiplied in tomato plants and purified by a modified procedures. The purity of PVX was 0.59mg/m1. Purified PVX was injected into rabbits once a week for 5 weeks. Antiserum was collected 10 days after the last injection. Produced antiserum was determined 1/1024 titers by means of microprecipitin tests and showed sharp reactions in agar gel-diffusion tests.

The Vascular Plants in Is. Gageo (Jeollanam-do) (가거도(전라남도)의 관속식물상)

  • This study was carried out to investigate the flora of Is. Gageo (Jeollanam-do). The vascular plants were collected 13 times (from June 2008 to September 2011), and 465 taxa in total were identified, including 112 families, 301 genera, 402 species, 3 subspecies, 49 varieties, 9 forms and 2 hybrids. 171 taxa were first records for the region. Among them were seven taxa of Korean endemic plants and the Ministry of Environment-designated plants, which should be protected by the wildlife protection law, were identified as three taxa. There were 54 taxa of the flora with the levels III, IV, V in the floristic indicator plants which was designated by Ministry of Environment. The naturalized plants were identified as 29 taxa, and their naturalization ratio and urban index were found to be 6.2% and 9.0% respectively. There existed many kinds of plants resources having conservational value like $Loxogramme$ $salicifolia$, $Sarcandra$ $glabra$, $Osmanthus$ $insularis$, $Saussurea$ $polylepis$, $Carex$ $ligulata$ var. $austrokoreensis$, $Hosta$ $jonesii$, $Cyrtosia$ $septentrionalis$, etc., in this area. As therefore clear that appropriate plans to conserve the natural habitat of Is. Gageo must be made immediately.

Purification and Serology of Potato Virus S (감자 바이러스 S의 순화와 항혈청제조)

  • he study was conducted to produce an antiserum of potato virus S for identification and screening of seed-potatoes. Potato virus S was isolated from infected plants and identified by means of indicator plants and electro microscopy. Isolated potato virus S was multiplied in Nicotiana deebneyii and the virus was purified by a modified method that was developed through this study. The purity of potato virus S was 1.18mg/ml. Purified potato virus S was injected into rabbit intravenously once a week for 5 weeks. Antiserum was collected 10 days after the last injection. The produced antiserum was determined to have a titer of, 1/2048 by means of microprecipitin tests.

Studies on the Distributiion of Vascular Plants in the Islands around the Korean Peninsula (한반도 주변도서의 관속식물 분포에 관한 연구)

  • Distributional pattern of vascular plant species in terms of species-area relationship and distances from their species pool on the 53 islands selected in the South Korea were studied. From the relationship between vascular plant species and area the equation of species-area was established as the follow: S=cA2 c=135.52 z=0.226 S; number of species A; area of island In the equation the Z value 0.226 is lower than those of any other localities reported in the world. Z value increased with the increasing distance from the species pool while C value decreased. The relationships of Z and/or C and distance from species pool, D, were established; Z=0.002D+0.176 C=-39.5logD+178.5 The tendency of common species decrease of them with the increasing islands was fifted to the Fisher's logarithm series. Common species 50% above was 39 species, including the most common species, Pinus thunbergii. Quotient of Similarity value by Sorensen (1948) on common species was relatively low i.e. 0.27∼0.47. It is noticeable for their vegetation conditions that the naturalized plant as a indicator of disturbance, for example Erigeron canadensis, was widely distributed.
