Effects on the Rumen Microbial Fermentation Characteristics of Lignosulfonate Treated Soybean Meal (Lignosulfonate처리 대두박의 반추위 내 미생물 발효특성에 미치는 영향)

  • This study was conducted to investigate the effects on fermentation characteristics of rumen microorganism by different types and levels of lignosulfonate treated soybean meal (LSBM) in in vitro test and rumen simulation continuous culture (RSCC) system in dairy cows. The experiment I was control and 12 treatments (each with 3 replications) in vitro test to demonstrate composition of different types of treatments with lignosulfonate (Desulfonate, Na, Ca and solution) and levels (2, 4 and 8%) of soybean meal in the dairy cow diet. LSBM source treatments in the dairy cow diet showed pH value, $NH_3$-N concentration and total VFA concentration lower than control at all levels and incubation times (p<0.05). Dry matter digestibility of LSBM source treatments showed lower than control (p<0.05). Gas production and rumen microbial synthesis was decreased by rumen microbial fermentation for incubation times. Undegradable protein (UDP) concentration of all LSBM treatments was decreased for incubation times, and significantly higher than control (p<0.05). In the experiment II compared diets of the control, LSBM Na 2%, LSBM Sol 2%, which are high performance to undegradable protein (UDP) concentration experiment I in vitro test, and heated treatment lignosulfonate (LSBM Heat) 2% in the dairy cow diet from four station RSCC system ($4{\times}4$ Latin square). A rumen microbial fermentation characteristic was stability during 12~15 days of experimental period in all treatments. The pH value of LSBM treatments was higher than control treatment (p<0.05). The $NH_3$-N concentration, VFA concentration and rumen microbial synthesis of LSBM treatments were lower than control (p<0.05). The undegradable protein (UDP) showed LSBM Na 2% (45.28%), LSBM Sol 2% (43.52%) and LSBM Heat 2% (43.49%) higher than control (41.55%), respectively (p<0.05). Those experiments were designed to improve by-pass protein of diet and milk protein in the dairy cows. We will conduct those experiments the in vivo test by LSBM treatments in dairy cows diet.

Effects of Dicarboxylic Acid as an Alternative to Antibiotic on in vitro Rumen Parameters, Milk yield and Milk Compositions in Lactating Cows (항생제 대체제로서 Dicarboxylic Acid 급여가 in vitro 반추위 발효성상, 착유우의 유량 및 유성분에 미치는 영향)

  • This study was undertaken to investigate the effects of dicarboxylic acid supplementation, as replacement antibiotics, of on in vitro ruminal parameters and milk yield and milk composition in lactating cows. in vitro treatments were 1) Con (4 g of basal diet), 2) CM (4 g of basal diet + 0.05 ml of monensin), 3) CR (4 g of basal diet + 0.1 ml of dicarboxylic acid) and in vivo treatments were 1) Con (25 kg of basal diet/head/day), and 2) CR (25 kg of basal diet + 5 g of dicarboxylic acid/head/day), respectively. A total 10 lactating dairy cows ($649{\pm}19kg$ average body weight, $99{\pm}65$ average milking days) were divided in to two groups according to mean milk yield and number of days of postpartum. The cows fed a basal diet during adaptation (2 wk) and experimental diets during the treatment periods (4 wk). In the first in vitro experiment, there were no statistical differences between treatments in pH, gas production, and ammonia-N and lactic acid concentration during incubation. However, dry matter digestibility was significantly higher in CR treatment compared to control or CM treatment (P<0.05). Total VFA was tended to higher in CR treatment than those of control and CM treatment (P>0.05). In the second experiment, milk yield was significantly higher in treatment (40.39 kg) compared to control (35.19 kg), (P<0.05). Milk composition and MUN were not changed by dietary supplementing dicarboxylic acid. Therefore the present results reporting that supplementation of dicarboxylic acid might enhance the stabilization of ruminal fermentation and increase the milk yield of lactating cows.

Effects of Different Cutting Height on Nutritional Quality of Whole Crop Barley Silage and Feed Value on Hanwoo Heifers

  • The present study evaluated the effects of different cutting height on nutritive value, fermentation quality, in vitro and in vivo digestibility of whole crop barley silage. Whole crop barley forage (Yuyeon hybrid) was harvested at height of 5, 10, and 15 cm from the ground level. Each cutting height was rolled to make round bale and ensiled for 100 days. After 100 days of ensiling, pH of silage was lower (p<0.05) in 5 cm, but no difference between 10 and 15 cm of cutting height. The content of lactate and lactate to acetate ratio were increased (p<0.05) in 5 cm of cutting height, whereas the acetate content was higher (p<0.05) in 10 and 15 cm than that of 5 cm cutting height. Aerobic stability was greater (p<0.05) in silages of 10 and 15 cm of cutting height. Three total mixed rations (TMR) were formulated with silages from the three different cutting heights (TMR5, TMR10, and TMR15) incorporated as forage at 70:30 ratio with concentrate (dry matter [DM] basis). In vitro dry matter digestibility was higher (p<0.05) in the TMR5 and TMR10 than that in TMR15, whereas in vitro neutral detergent fiber digestibility was higher (p<0.05) in the TMR10 and TMR15 than that in TMR5. Concentration of $NH_3-N$ was highest (p<0.05) in the TMR10 followed by TMR15 and TMR5. Total volatile fatty acid was decreased (p<0.05) with increased cutting height. The digestibility of DM and neutral detergent fiber were highest (p<0.05) in TMR15, than those in TMR5 and TMR10, whereas acid detergent fiber digestibility was higher (p<0.05) in TMR5 than that in TMR10. The results showed that increasing cutting height, at least up to 10 to 15 cm, of whole crop barley forage at harvest (Yuyeon) may be beneficial for making silage for TMR formulation and increasing digestibility of DM and NDF.

Effects of Hyolbuchukeo-tang Extracts on Blood and liver of Hyperlipidemia Rats Induced by High Fat Diet (혈부축어탕(血府逐瘀湯) 추출물(抽出物)이 고지방식이(高脂肪食餌)로 유발(誘潑)시킨 고지혈증(高脂血症) 흰쥐의 혈액(血液) 및 간(肝) 기능(機能)에 미치는 영향(影響))

  • Objectives: The purpose of this study was to investigate the effects of Hyolbuchukeo-tang extracts on the hyperlipidemia rats induced by high fat diet. Methods and Materials: In vitro; The extracts prepared for Hyolbuchukeo-tang by hot water extraction (HH), fermentation(HF) and UMPM extraction(HU) method. The extracts were examined for levels of polyphenol compounds, antioxidant activities, and inhibitory potencies for HMG-CoA reductase. In vivo; Sprague-Dawley male rats of weighing $150{\pm}5g$ were randomly divided into six groups ; normal control diet(NC), and high fat diet(HC), high fat diet with treated lovastatin of 10mg/kg(PC), high fat diet with Hyolbuchukeo-tang extracts; HH, HF and HU treated extracts of 300mg/kg, respectively. Also, we compared the effects of the extracts of HH, HF and HU on rats fed high fat diet for four weeks. Results: 1. The content of polyphenol compounds and electron donating abilities(EDAs) was the HF higher than HH and HU. The superoxide dismutase(SOD)-like activities were proportionate in consistency and they appeared highly from all extracts. The HMG-CoA reductase inhibition activities was highest activities in the HU. 2. The activities of serum GOT and GPT were significantly lower in the HH and HF groups. The level of serum triglyceride was significantly decreased in the HF group. HH and HU groups were significantly decreased in the atherogenic index(AI). The total cholesterol concentration in liver was significantly decreased in the HF group, and HU showed more significantly decreased in the triglyceride than of the lovastatin. Also, photomicrographs of liver tissue showed higher fat accumulation in the HC group than in the HH, HF and HU groups. Conclusions: These result suggest that the hyper-lipidemia caused by a high fat diet was effectively inhibited the administration of HF and HU.

  • We have cloned several E. coli sfs genes which stimulate mal gene expression with $crp^{{\ast}1}$). One the genes (pPVC2) was sequenced and potential CRP binding site is located in the upstream of the putative promoter in the regulatory region. In order to investigate the regulation of the sfs1 gene by the cAMP-CRP complex, we have constructed the sfs-lacZ fusion gene in this research. The overall transcriptional stimulations of sfs1 gene in the presence cAMP were confirmed by ${\beta}-galactosidase$ activity and Western blot analysis of sfs1-lacZ fusion gene. Transcriptional regulation by cAMP-CRP was also confirmed by Northern blot analysis. End-labelled DNA of the DNA fragment in sfs1 regulation region were used for gel retardation assay to examine the CRP-DNA complex in the presence of cAMP. Results here indicate that CRP binding site in the regulatory region of sfs1 gene is positive regulator for the expression of sfs1 gene.

Effects of Dietary DFA IV on Lumen Short Chain Fatty Acids Production and Intestinal Environment in Rats (식이 DFA IV의 흰쥐 장내 단쇄지방산 생성 및 장내환경 개선효과)

  • This study examined the improving effect of DFA IV obtained from bacterial fermentation on the gut health. The effects of the dietary DFA IV on the intestinal mass, short chain fatty acids production and pH were evaluated in vivo. Sprague-Dawley rats were fed the 0% (control) or 1% DFA IV supplemented diets for 3 weeks. Supplementation of DFA IV resulted in a significant increase in cecal tissue and wall weights. Together with the lowering of the cecal and colonic pH, the amount of acetate and butyrate increased by 1.6 and 3.2 fold of the control group in the cecum, respectively, in the rats fed DFA IV diets. The DFA IV diet also significantly increased the cecal lactate 1.5 fold compared to the control diet, indicating that dietary DFA IV stimulated the growth of lactic acid bacteria and bifidobacteria in the intestine. Based on the above results, it is concluded that the dietary DFA IV may be used as a putative prebiotic supplement.

Biological Activities of Ethanol Extracts from Hericium erinaceus Mycelium on Angelica keiskei and Angelica keiskei Pomace (신선초와 신선초박에 배양한 노루궁뎅이버섯 균사체 에탄올 추출물의 생리활성 효과)

  • The purpose of this study was to investigate the antioxidant, nitrite scavenging, angiotensin-converting enzyme (ACE), and tyrosinase inhibition activities of ethanol extracts from Hericium erinaceus (HE) cultured in Angelica keiskei (AK) and Angelica keiskei pomace (AKP). Antioxidant activities of the HE, AK, AKP, HE+AK, and HE+AKP extracts were higher than that of control, but lower than those of BHT and Vit C. Tyrosinase inhibition activity was the highest in AKP+HE extract (90%), whereas those of HE, AK, AKP, and AK+HE extracts were about 57, 61, 82, and 78%, respectively. Nitrite scavenging abilities of various samples were above 80% at pH 1.2, and there was no significant difference between the HE, AK, AK+HE, and AKP+HE extracts at pH 3.0, excepting the AKP extract. ACE inhibition activity was the highest in AKP extract (60%), whereas those of HE, AK, AK+HE and AKP+HE extracts were about 42%, 50%, 35% and 40%, respectively. These results suggest that H. erinaceus cultured in AK and AKP may be used as a raw material for functional foods, and food additives, as well as in the cosmetic industry as a natural source of bioactive compound after further investigation in vivo.

Protective Effect of Ferments of Hot-water Extract Mixture from Rhodiola sachalinensis and Red Ginseng on Oxidative Stress-induced C2C12 Myoblast (C2C12 근육세포의 산화적 손상에 대한 홍경천-홍삼 추출물 혼합액 발효물의 보호효과)

  • Rhodiola spp. and red ginseng have been used for food and medicinal applications in disease chemoprevention in many Asian countries. Increased oxidative stress by reactive oxygen species (ROS) has been proposed to be a major cause of muscle fatigue. The present study was designed to investigate the protective effects of a fermented hot-water extract mixture from Rhodiola sachalinensis and red ginseng (MFR) on cell damage and the antioxidant enzyme system in $H_2O_2$-induced oxidative stress in skeletal muscle cells. C2C12 myoblasts were treated with various concentrations of NFR (non-fermented Rhodiola sachalinensis extract), FR (fermented hot-water extract from Rhodiola sachalinensis) and MFR for up to 5 days after the standard induction of differentiation, followed by semi-quantitative RT-PCR. MFR treatment dose-dependently protected oxidative damage of C2C12 cells. The treatment with MFR also enhanced mRNA expressions of MyoD, Cu/Zn SOD, Mn-SOD and GPX up to 16%. These results indicate that MFR exerts an anti-oxidative effect through a mechanism (s) that may involve the up-regulation of antioxidant enzymes, which may be important for the cellular redox environment in muscle cells.

Effects of the Brown Seaweed Residues Supplementation on In Vitro Fermentation and Milk Production and Composition of Lactating Dairy Cows (미역부산물 첨가가 In Vitro 발효성상과 젖소의 산유량 및 유성분에 미치는 영향)

  • This study was conducted to investigate effects of the brown seaweed residues supplementation on in vitro fermentation, and milk yield and milk composition of dairy cows. Therefore, two experiments consisting of an in vitro and an in vivo growth trial were used. In in vitro experiment, brown seaweed residues(BSR) was supplemented in basal diet with 0, 1, 2 and 4% respectively, and incubated for 3, 6, 9, 12, and 24 h. The pH value, ammonia-N and VFA were investigated. The pH value tended to increase with increasing BSR during the incubation. Particularly, pH was significantly higher in BSR treatments compared with control at 9 h(p < 0.05). While, ammonia-N concentration was not significantly different across treatments during the whole incubation. BSR supplementation did not affect total VFA production, but acetate was linearly increased in BSR treatments compared with control at 12 h(p < 0.05), and its concentration was highest(92.70 mM) in 4% BSR among treatments. The concentration of iso-butyrate tended to increase in BSR treatments in comparison to control during the incubation. In addition, the concentration of iso-valerate was higher in BSR treatments compared with control at 12 and 24 h. In growth trial, BSR was added(800 g/d/animaI) to diets of dairy cow. Dry matter intake was not affected by BSR supplementation, but daily milk yield(kg) significantly increased in BSR treatment compared with control(p < 0.05). However, milk composition(%) and milk yield(kg) were not significantly different between treatments. Milk fat(% and kg/d) tended to slightly decrease in BSR treatment compared with control(3.59% and 1.06 kg/d vs. 3.32% and 1.01 kg/d), The contents of C16:0 and C20:4 in milk significantly increased in BSR treatment compared with control reflecting from dietary fatty acid composition. The content of C18:0 in milk which is end product of biohydrogenation of CI8 unsaturated fatty acids in the rumen significantly increased in BSR treatment compared with control(p < 0.05). C18:2 content in milk tended to decrease, but tended to increase trans-II C18:l and CLA contents in milk in BSR treatment compared with control. In conclusion, it could be summarized that BSR may stabilize rumen pH, and it could improve milk yield and CIA content in milk with more than 4% of diet. Therefore, BSR could be beneficially used in dairy diets as a feed additive.

Evaluation of Biological Activities of Fermented Rice Bran from Novel Black Colored Rice Cultivar SuperC3GHi (슈퍼자미(SuperC3GHi) 미강 발효물의 생리활성)

  • Rice bran is by-product from the dehulling process of rice, an important staple food in Korea. Various studies have been reported immune-enhancing effects of rice bran cultured with Lentinus edodes. In particular black rice bran that contains anthocyanin has the beneficial effects on immune system. SuperC3GHi, a recently released black colored rice cultivar, has higher contents of cyanidine-3-glucoside (C3G), a strong antioxidant than those of currently cultivated black rice cultivars. In this study, we investigate the beneficial effects of fermented SuperC3GHi rice bran (C3G-F) with Lentinus edodes. Using in vivo and in vitro model for inflammation and allgergy, anti-inflammatory and anti-allergic effects of C3G-F were determined. C3G-F induced tumor suppressive effect on Sarcoma-180 cell and B16 melanoma cell injected to mice. These results suggest that the capacity of C3G-F seems to act as a potent immunomodulator and with the absence of notable side-effects, C3G-F could be used as a novel functional food ingredient having possible therapeutic effects against immunological disorders.
