Bisphenol A의 급성노출이 마우스의 면역기능에 미치는 영향 (Effects of Acute Oral Administration of Bisphenol A on the Immune Function in Mice)

  • In order to investigate the effects of bisphenol A (BPA) on immune system in mice we examined the various immunological parameters. After single oral administration of BPA to female ICR mice, the weights of bodies and lymphoid organs, splenic cellularity and hematological parameters were examined on day 2 and 7. Among them WBC and splenic cellularity were slightly decreased on day 2. To assess the effects of BPA on humoral immune responses, splenic IgM plaque forming cell (PFC) and serum IgM were assayed. When BPA was administered after immunization with SRBC, but not before immunization, IgM PFC against SRBC was significantly lowered in a dose dependent manner. Serum IgM level was also decreased on day 4 when high dose (2000 mg/kg) of BPA was administrated after injection of OVA-antigen. The indexes of splenocyte proliferation (SP) to concanavalin A (Con A) and bacterial lipopolysaccharide (LPS) were measured in vitro by MTT assay. At low concentration BPA slightly increased splenocyte proliferation but at higher concentration it showed significant inhibitory effects on cell proliferation. Mitogen-stimulated SP was also determined with spleen cells from BPA treated mice. Con A-induced SP was slightly decreased and LPS-induced SP was especially inhibited at 1000 mg/kg and 2000 mg/kg of BPA. These results indicate that BPA is able to acutly evoke humoral and cell mediated immune suppression in mice.

  • A literature study on cancer therapy of warm-hot oriental medicine was done, and the results were as follows. 1. In oriental medicine, oncogens are six exopathogens, seven modes of emotion, overwork, pathogenic factors, and especially related with pathologic cold situation. 2. There are many capillaries in tuomr, and because temperature of inner space of tumor is higher than normal organization. Tumor cell has a character which is weak for high temperature. 3. Warm-hot herb drugs have effects of dissipating mass, warming kidney to reinforce yang and dispering, so it has a function of suppressing tumor as well as improving immunity in cancer therapy. 4. In traditional medical books, main prescriptions of cancer therapy are xinzhiyinyanggongjiwan(新製陰陽攻積丸), qianjinxiaoshiwan(千金硝石丸), feiqiwan(肥氣丸), xibenwan(息賁丸), fuliangwan(伏梁丸), beiqiwan, bentunwan(賁豚丸), zengsunwujiwan(增損五積丸), and these are composed of warm-hot herb drugs. 5. In current, the study of warm-hot drugs is progressed in immunological capacity, anti-tumor activity, stimulating bone marrow and regulating hormone secretion. It will be expected that advanced study of these must be accomplished in cancer patients.

우황이 생쥐의 면역반응에 미치는 영향 (Effects of the Administration of Bezoar Bovis on Immune Responses of Mice)

  • The effects of Bezoar Bovis on immune responses of ICR Mice were studied. In the present study, the Jerne hemolytic plaque assay (PFC) was used to evaluate the humoral immune response to sheep red blood cells, and macrophage regulatory function was measured by quantitating the production of molecules secreted by macrophages. Bezoar Bovis was given in single daily p.o doses for short term (1, 2 days) or long term (7, 14 days). PFC response for taken 3 days after short term exposure to Bezoar Bovis was increased. The production of nitirc oxide in macrophages was also stimulated. In contrast, long term exposure to Bezoar Bovis resulted in inhibitory effect of Bezoar Bovis on nitirc oxide, $TNF-{\alpha}$and IL-6 production in macrophages. These findings suggest that treatment with Bezoar Bovis may result in differential immunological effect depending on treatment schedule.

  • Cefodizime has originally been developed for treating infections as antibiotics. However. according to some of recent studies. cefodizime. a third generation cephalosporin. may potentially have the capability of stimulating chemotactic activity of neutrophils and monocytes as well as the strong immuno-modulator. In this study. we studied to learn about the expressive effect of dentritic cells and macrophage. With this background. We have studied to see if cefodizime can be a potential substance inducing an immunological function in dendritic cells and peritoneal macrophages. IL-12 activates NK cell and macrophage, and shows antiviral effect by excreting INF-${\gamma}$. In vitro. total RNAs were extracted from murine dentritic cell at 4, 8, 12, 24hr after the application of 10, 50, 100${\gamma}g$/ml of cefodizime wighout other stimulators. And we analyzed IL-12 mRNA using RT-PCR method. In conclusion. IL-12 mRNA was increased. and the results suggest that cefodizime activate TH1 cell induction, CTL differentiation as well as accelerating the increase of NK. LAK cell.

홍조류 유래 네오아가로올리고당의 면역 활성 증강 효과 (Immune Enhancing Activity of Neoagarooligosaccharides from Marine Red Algae)

  • Agar, a heterogeneous polymer of galactose, is the main component of the cell wall of marine red algae. It is well established as a safe, non-digestible carbohydrate in oriental countries. Neoagarooligosaccharides (NAOs) prepared by hydrolyzing agar by microbial β-agarase have been reported to show safety. However, their immunological effects have not been reported yet. Thus, the objective of this study was to investigate immune enhancing effects of neoagarooligosaccharides (NAOs) from marine red algae Gelidium elegans in mice by performing ex vivo experiments. Six-week-old mice were fed ad libitum. NAOs were orally administrated at three different concentrations (100, 500, and 2,500 mg/kg B.W./day) twice a week for four weeks. The group fed with NAOs at 2,500 mg/kg showed the highest proliferation of splenocytes and production levels of cytokines (IL-1β, IL-6, TNF-α) in the ex vivo experiment. In conclusion, NAOs can enhance immune function, increase proliferation of splenocytes, and increase cytokine production by activating macrophages in mice.

Ginsenosides are active ingredients in Panax ginseng with immunomodulatory properties from cellular to organismal levels

  • The immune system is one of the most important parts of the human body and immunomodulation is the major function of the immune system. In response to outside pathogens or high inflammation, the immune system is stimulated or suppressed. Thus, identifying effective and potent immunostimulants or immunosuppressants is critical. Ginsenosides are a type of steroid saponin derived from ginseng. Most are harmless to the body and even have tonic effects. In this review, we mainly focus on the immunostimulatory and immunosuppressive roles of two types ginsenosides: the protopanaxadiol (PPD)-type and protopanaxatriol (PPT)-type. PPT-type ginsenosides include Rg1, Rg2, Rh4, Re and notoginsenoside R1, and PPD-type ginsenosides include Rg3, Rh2, Rb1, Rb2, Rc, Rd, compound K (CK) and PPD, which activate the immune responses. In addition, Rg1 and Rg6 belong to PPT-type ginsenosides and together with Rg3, Rb1, Rd, CK show immunosuppressive properties. Current explorations of ginsenosides in immunological areas are in the preliminary stages. Therefore, this review may provide some novel ideas to researchers who study the immunoregulatory roles of ginsenosides.

류마티스 관절염에서 NLRP3 인플라마좀의 역할 (Role of NLRP3 Inflammasome in Rheumatoid Arthritis)

  • Objectives: Inflammasomes are molecular platforms that are generated inside cytoplasmic compartments. The objective is to mediate immunological responses of the host to cell damage and infection. Caspase-1 is triggered by inflammasome to generate interleukin-1𝛽 (IL-1𝛽), an inflammatory cytokine, and pyroptosis, an inflammatory form of apoptosis. Methods: In the past two decades, scientists have uncovered several inflammasomes. The most research has been conducted on NLRP3 inflamamsomes, whose activity can be stimulated by a variety of induction factors. However, the unregulated activation of NLRP3 inflammasomes is also a role in the etiology of several human disorders. Previous research has demonstrated that NLRP3 inflammasomes have a significant role in the innate and acquired immune systems, as well as in the prevalence of joint illnesses such rheumatoid arthritis. Conclusion: Within the scope of this review, we will present a brief overview of the biological features of NLRP3 inflamamsomes as well as a description of the underlying mechanisms governing activation and regulation. In particular, we explore the function of inflammasomes in the development of rheumatoid arthritis as well as the promise of recently identified medicines that target inflamasomes.

Immunomodulatory effects of six Acetobacter pasteurianus strains in RAW-Blue macrophage

  • In this study, we investigated the immunological properties of six strains of Acetobacter pasteurianus through nuclear factor-kappa B/activator protein-1 (NF-κB/AP-1) transcription factor activation and nitric oxide (NO) and cytokine production in macrophages. We found that the six A. pasteurianus strains had no significant inhibitory effect on the cell viability of RAW-BlueTM cells at the concentration of (25, 50, 100 CFU/macrophage). The production of NO and cytokines (TNF-α, IL-1β, and IL-6) showed different abilities of immune activation for each strain, and it was 0.7 to 0.9 times higher than that of the LPS (100 ng/mL, v/v) positive control and 7 to 8 times superior to that of the negative control group. To explore the underlying mechanism, we evaluated the mRNA expression of pro-inflammatory genes. Consequently, we found that inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 expression including genes expression of cytokines were elevated by the six A. pasteurianus treatment. These results suggested that the six strains of A. pasteurianus have an excellent industrial application value as a functional material for the purpose of enhancing immune function.

Effect of Lipopolysaccharide (LPS) Exposure on the Reproductive Organs of Immature Female Rats

  • Lipopolysaccharide (LPS), an endotoxin, elicits strong immune responses in mammals. Several lines of evidence demonstrate that LPS challenge profoundly affects female reproductive function. For example, LPS exposure affects steroidogenesis and folliculogenesis, resulting in delayed puberty onset. The present study was conducted to clarify the mechanism underlying the adverse effect of LPS on the delayed puberty in female rats. LPS was daily injected for 5 days ($50{\mu}g/kg$, PND 25-29) to treated animals and the date at VO was evaluated through daily visual examination. At PND 39, animals were sacrificed, and the tissues were immediately removed and weighed. Among the reproductive organs, the weights of the ovaries and oviduct from LPS-treated animals were significantly lower than those of control animals. There were no changes in the weights of uterus and vagina between the LPS-treated and their control animals. immunological challenge by LPS delayed VO. Multiple corpora lutea were found in the control ovaries, indicating ovulations were occurred. However, none of corpus luteum was present in the LPS-treated ovary. The transcription level of steroidogenic acute regulatory protein (StAR), CYP11A1, CYP17A1 and CYP19 were significantly increased by LPS treatment. On the other hand, the levels of $3{\beta}$-HSD, $17{\beta}$-HSD and LH receptor were not changed by LPS challenge. In conclusion, the present study demonstrated that the repeated LPS exposure during the prepubertal period could induce multiple alterations in the steroidogenic machinery in ovary, and in turn, delayed puberty onset. The prepubertal LPS challenge model used in our study is useful to understand the reciprocal regulation of immune (stress) - reproductive function in early life.

Expression of Immune-Related Genes during Loach (Misgurnus anguillicaudatus) Embryonic and Early Larval Development

  • Early life stage mortality in fish is one of the problems faced by loach aquaculture. However, our understanding of immune system in early life stage fish is still incomplete, and the information available is restricted to a few fish species. In the present work, we investigated the expression of immune-related transcripts in loach during early development. In fishes, recombination-activating gene 1 (RAG-1) and sacsin (SACS) have been considered as immunological function. In this study, the expression of the both genes was assessed throughout the early developmental stages of loach using real-time PCR method. maRAG-1 mRNA was first detected in 0 dph, observed the increased mostly until 40 dph. Significant expression of maRAG-1 was detected in 0 to 40 dph. These patterns of expression may suggest that the loach start to develop its function after hatching. On the other hand, maSACS was detected in unfertilized oocyte to molura stages and 0 to 40 dph. maSACS mRNA transcripts were detected in unfertilized oocytes, suggesting that they are maternally transferred.
