Effects of feeding corn naturally contaminated with aflatoxin on growth performance, apparent ileal digestibility, serum hormones levels and gene expression of Na+, K+-ATPase in ducklings

  • Objective: A 14-d trial was conducted to determine the effects of feeding corn naturally contaminated with aflatoxin $B_1$ ($AFB_1$) on growth performance, apparent ileal digestibility, serum hormones levels and gene expression of $Na^+$, $K^+-ATPase$ in ducklings. Methods: A total of 704 ducklings were blocked on the basis of sex and body weight (BW), and then allocated randomly to one of the following two treatments: i) CON, basal diet and ii) $AFB_1$, diets with 100% of normal corn replaced with $AFB_1$ contaminated corn. There were 22 pens per treatment and 16 birds per pen. The concentration of $AFB_1$ was 195.4 and $124.35{\mu}g/kg$ in the contaminated corn and $AFB_1$ diet, respectively. Results: The $AFB_1$ decreased average daily gain, average daily feed intake, d 7 BW, final BW in the whole trial, and feed conversion ratio (FCR) during d 8 to 14 and d 1 to 14 by 10% to 47% (p<0.05), while FCR during d 1 to 7 was increased (p<0.05). $AFB_1$ did not affect mortality to 7 d of age, and then increased to 5.8% from 8 to 14 d of age (p<0.01). Apparent ileal gross energy digestibility was reduced by $AFB_1$, whereas apparent ileal digestibility of dry matter, nitrogen, and amino acid was improved (p<0.01). Feeding $AFB_1$ diets increased serum concentration of leptin and insulin-like growth factors-1 (IGF-1) (p<0.05), but had no effect on neuropeptide Y, ghrelin, cholecystokinin-8 or insulin (p>0.05). Dietary treatments did not influence relative expression of jejunal $Na^+$, $K^+-ATPase$ gene (p>0.05). Conclusion: Taken together, feeding corn naturally contaminated with $AFB_1$ reduced growth performance, improved apparent ileal digestibility, and affected serum leptin and IGF-1 in ducklings from d 1 to 14.

Evaluation of energy and amino acids of brown rice and its effects on laying performance and egg quality of layers

  • Two experiments were conducted to determine apparent metabolizable energy (AME), nitrogen-corrected AME (AMEn), and ileal digestible amino acid (AA) content of brown rice (BR) and to investigate the effect of dietary supplementation of BR on laying performance and egg quality of laying hens. In Exp. 1, 72 Hy-line Brown layers (49-week-old) were allocated to two treatments using a completely randomized block design, and each treatment included six cages per treatment and six hens per cage. A semi-purified diet was formulated to include BR as the sole source of AA and energy and an N-free diet was used to determine basal endogenous loss of AA. The hens were fed a commercial layer diet for adaptation to the experimental environment and diet for 7 days from d 0, and then fed experimental diets for 5 days from d 7. Excreta were collected from d 10 to 11 and ileal digesta were collected on d 12. On a dry matter (DM) basis, the AME and AMEn of BR was determined at 3,773 and 3,729 kcal/kg, respectively. The apparent ileal digestibility (AID) of BR ranged from 32.7% for Thr to 73.7% for Arg. The range of the standardized ileal digestibility (SID) value was between 79.4% for Met and 96.6% for Lys. In Exp. 2, 252 Hy-line Brown layers (44-week-old) were divided into four groups, comprising seven replicates of nine birds each and assigned to four experimental diets containing 0 (Control), 5%, 10%, or 15% BR for 5 weeks. The BR-containing diets were formulated to be equal in the content of AMEn and digestible AA to those of the diet without BR. No significant differences were observed in laying performances. Egg quality and blood profiles were not linearly or quadratically affected by dietary treatments. These results suggest that up to 15% BR can be included into layer feed without any adverse effects on laying performance and egg quality, if its energy and digestible AA values are well evaluated.

Nutrient ileal digestibility evaluation of dried mealworm (Tenebrio molitor) larvae compared to three animal protein by-products in growing pigs

  • Objective: This study was to investigate the nutrient ileal digestibility of dried mealworm (Tenebrio molitor) larvae and compare with those of three animal protein by-products in growing pigs. Methods: A total of 12 crossbred ($[Landrace{\times}Yorkshire]{\times}Duroc$) growing pigs with average body weights of $24.12{\pm}0.68kg$ were surgically equipped with simple T-cannulas after being deprived of feed for 24 h according to published surgical procedures. These pigs had a recovery period of two weeks. A total of 12 pigs were assigned to individual metabolic crates and allotted to one of four treatments with 3 replicates in a fully randomized design. Dietary treatments included the following: i) Fish meal, corn-vegetable by-product basal diet+9.95% fish meal; ii) Meat meal, corn-vegetable by-product basal diet+9.95% meat meal; iii) Poultry meal, cornvegetable by-product basal diet+9.95% poultry meal; iv) Tenebrio molitor, corn-vegetable by-product basal diet+9.95% dried Tenebrio molitor larvae. Results: Results showed that the apparent ileal digestibility (AID) of Lys was higher (p<0.05) in pigs fed Tenebrio molitor diet than that in pigs fed fish meal diet. Pigs fed Tenebrio molitor diet showed increased (p<0.05) AID of His and Arg compared to pigs fed Fish meal or Meat meal diet. The AID of Cys was increased (p<0.05) in pigs fed poultry meal and Tenebrio molitor diets compared to that in pigs fish meal diet. Pigs fed meat meal, poultry meal, and Tenebrio molitor diets showed higher (p<0.05) standardized ileal digestibility (SID) of total energy compared to pigs fed fish meal diet. The SID of Arg was higher (p<0.05) in pigs fed Tenebrio molitor diet than that in pigs fed fish meal or meat meal diet. Furthermore, pigs fed poultry meal or Tenebrio molitor diets showed increased (p<0.05) SID of Cys compared to pigs fed fish meal diet. Conclusion: In conclusion, providing pigs with diets that contained Tenebrio molitor larvae meal improved AID and SID of nutrients as well as essential and non-essential amino acids. The digestibility of dried mealworm larvae protein and its utilization in vivo are also good. Therefore, dried mealworm larvae protein can be used as protein source at 10% level in growing pigs.

Pea starch increases the dry matter flow at the distal ileum and reduces the amino acids digestibility in ileal digesta collected after 4 hours postprandial of pigs fed low-protein diets

  • Objective: The study was aimed to investigate the rules of postprandial changes in intestine digesta dry matter (DM) flow and amino acid digestibility of growing pigs fed low-protein (LP) diets made of different starch. Methods: Eight barrows (28.8±2.1 kg) with a T-cannula at the distal ileum were randomly allotted to an 8×3 Youden square design. Treatments included: waxy corn starch LP (WLP); corn starch LP (CLP) and pea starch LP (PLP). Diets were given at 08:00 and 20:00. Digesta samples were collected in six 2-h stages from 08:00 to 20:00. Results: The Cr concentrations of ileal digesta increased and then decreased in WLP and CLP, while increased continuously in PLP as time passed after postprandial (p<0.05). Higher average Cr concentrations (0.78% and 0.84% vs 0.70%; p<0.05) and lower average DM flow (181.1 g/kg and 166.3 g/kg vs 240.3 g/kg; p<0.001) were observed in WLP and CLP, compared with PLP. The apparent ileal digestibility coefficient of most amino acids in WLP and CLP increased compared with that in PLP. No difference in lysine or methionine digestibility was observed. When digesta were collected in 2-h periods, the apparent ileal digestibility coefficient of amino acids did not change over time. When digesta was collected in 4-h periods from 16:00 to 20:00 and 6-h periods from 14:00 to 20:00 (p<0.05), WLP and CLP showed markedly higher amino acid digestibility than PLP Conclusion: High-amylose slowly digested starch can increase the DM flow at the distal ileum and reduce the apparent ileal digestibility coefficient of amino acids of pigs fed LP diets. Compared with waxy corn starch and corn starch, pea starch reduced the digestibility of amino acids in digesta collected after 4 h postprandial.

회장 상피세포에서 비브리오균(Vibrio vulnificus)의 염증 유도 기작 연구: protein kinase C와 nuclear factor kappa-B의 관련성 (Vibrio Vulnificus Induces the Inflammation of Mouse Ileal Epithelium: Involvement of Protein Kinase C and Nuclear Factor-Kappa B)

  • 비브리오균(Vibrio vulnificus)은 심각하고 치명적인 감염을 일으킬 수 있는 호염성의 식중독균이지만, 숙주세포 내에서 염증반응을 일으키는 분자적 기작은 아직 잘 알려지지 않았다. 본 연구에서는 오염된 식품 섭취를 통해 유입되는 비브리오균의 소장 특이적 염증 반응 위치와 기작을 알아보기 위해, 7주령의 수컷 마우스에 비브리오균($1{\times}10^9CFU$)을 16시간 동안 경구 투여하였다. 그 결과 비브리오균은 주로 회장(ileum) 부위에서 비브리오균(WT) 수가 가장 많이 증가하였고, 공장(Jejunum), 근위부대장(proximal colon), 원위부 대장(distal colon)에도 유의적으로 군집현성이 이루어짐을 알 수 있었지만, 십이지장(duodenum)과 비장(spleen), 그리고 간(liver) 조직들에서는 관찰되지 않았다. 특히 비브리오균의 표적기관인 회장상피조직에서는 비브리오균(WT)이 침입 시 융모 안으로 다량의 염증세포들이 유입되었고, 융포의 폭이 넓어지고 길이가 짧아지는 전형적인 조직학적 염증 반응을 보여주었다. 비브리오균이 유도한 조직 특이적 염증반응기작을 알아보기 위해, 비브리오에 감염된 회장상피조직으로부터 단백질과 mRNA를 분리하였다. 비브리오균은 숙주세포의 중추적 신호전달 단백질인 protein kinase C (PKC)의 인산화 및 $PKC{\alpha}$의 세포막이동을 촉진시켰고, mitogen-activated protein kinase (MAPK) 중 extracellular signal-regulated kinases (ERK)와 c-Jun N-terminal kinases (JNK)의 인산화를 유도하였지만, p38 MAPK 인산화에는 영향을 미치지 않았다. 특히 비브리오균은 inhibitory factor-kappa B (I-${\kappa}B$)의 활성을 촉진시킴으로써 nuclear factor-kappa B (NF-${\kappa}B$)의 인산화를 유도하였다. 마지막으로 비브리오균(WT)에 감염된 회장의 경우, 정상마우스에 비해 염증성 cytokine인 interleukin (IL)-6, IL-8, tumor necrosis factor (TNF)-${\alpha}$의 mRNA 수준이 유의적으로 증가되었다. 염증매개 수용체인 toll like receptor (TLR)-4, TLR-5, TLR-9의 mRNA의 발현 또한 비브리오균 처리에 의해 증가되어 있음이 관찰되었다. 종합적으로 오염된 식품 섭취를 통해 유입되는 비브리오균은 회장상피세포를 표적으로 염증반응을 일으키며, 그 기작은 PKC, ERK1/2, 그리고 JNK의 인산화를 통한 NF-${\kappa}B$ 활성의 촉진이며, 이로 인한 다양한 염증 매개 단백질 발현의 증가를 통해 이루어진다고 할 수 있다.

외상성 횡경막 Hernia: 2례 보고 (Traumatic Diaphragmatic Hernia: A Report of Two Cases)

  • Two cases of traumatic diaphragmatic hernia are reported, who were operated on in this department during the last 12 months` period. The first case, a 34 year old male, fell from 6 meters` height while he was working on electric pole. He sustained rib fractures, left 8th, 9th and 10th rib, left diaphragmatic rupture and ileal perforation. In the pleural cavity, there were stomach, omentum, left lobe of liver, transverse colon and ileum, which were reduced into the peritoneal cavity, and the diaphragmatic aperture was closed through anterolateral thoracotomy. After closure of the thoracic incision, median abdominal incision was made and closed the ileal perforation by primary suture. The second case was a 19 year old tyre repairman, who felt abrupt severe abdominal pain during lifting a heavy lyre. A barium study revealed a marked displacement of the stomach into the left pleural cavity. Immediately, thoracotomy was performed and closed the ruptured diaphragm after reduction of the herniated stomach, omentum, transverse colon, spleen and small intestine. The size of the diaphragmatic aperture were measured 17cm. in first case and 12cm. in the other respectively. Both cases discharged after uneventful recovery.

소아에서 복부둔상 후 발견된 특발성 소장 궤양의 천공 1예 (Perforation of an Idiopathic Small Bowel Ulceration after Blunt Abdominal Trauma in a Child)

  • Idiopathic small bowel ulceration distal to the duodenum is rare. Less than 5 % of the reported cases were in children. In the majority of the patients, a single ulcer of unknown cause is found in the jejunum or ileum. The diagnosis is difficult and usually made at the time of surgical exploration for complications, such as perforation, hemorrhage or obstruction. We treated a pediatric patient with perforation of an idiopathic ileal ulceration. The child was an 11-year-old boy who sustained blunt abdominal trauma. The involved ileal segment was resected. Pathologic findings were compatible with idiopathic small bowel ulceration. The clinical and pathological aspects of idiopathic ulcerations are discussed, and the literature reviewed.

장 평활근의 수축성에 대한 berberine의 효과 (Effect of berberine on intestinal contractility)

  • Berberine $(10^{-7}-10^5M)$ increased the contractility dose-dependently in isolated rabbit ileal and jejunal segments. Atropine and hemicholinium abolished this response but not mecamylamine. Berberine$(10^{-8}-10^5M)$ enhanced the transmurally-stimulated(80 V, 0.5 ms, 0.05 Hz) twitch response in the isolated guinea-pig ileal segments. Atropine and hemicholinium also abolished this response but not mecamylamine. Effect of KCI, carbachol and histamine were not affected by pretreatment with berberine$(10^{-5}M)$. The results of our study suggest that berberine increases the intestinal contractility by increasing a small amount of acetylcholine release from the postganglionic parasympathetic nerve terminal but not by a direct activation of muscarinic receptors.

Measurement of ileal permeability with different-sized polyethylene glycols (PEG 400, 600 and 1000)

  • Polyethylene glycols (PEGs; 400, 600, and 1000) were used to study the molecular weight (MW) permeability dependence in the rat ileal mucosa. Absorption of the PEGs was measured by following their recirculation perfusion over a 3 hr collection period. HPLC methods were used to separate and quantitate the individual oligomers present in the solution of PEGs mixtures (MW range 330 to 1 1 22 D). In the range studied, a distinct molecular weight cutoff was not identified. Corrected for the length of ileum used in the study, over the molecular weight range 330 to 1122 D, the apparent permeability $(P_{app)$ of PEG ranged from $3.2\pm0.06\times10_{-5} cm/sec(mean\pmSEM, n=7)\; to\; 0.1\pm0.02\times10^{-5} cm/sec.$ Also, it was observed that the apparent permeability was inversely proportional to approximately $MW^{2.4}$.

  • H$_2$O$_2$ has been shown to act as a signaling molecule involved in many cellular functions such as oxidant-induced stress, apoptosis, proliferation. In this study, we investigated the action mechanisms of H$_2$O$_2$ on activation of Extracellular Signal-Regulated Protein Kinase(ERK) in cultured feline ileal smooth muscle cells(ISMC). Western blot analysis done with phospho-specific MAP kinases antibodies demonstrated that potent activation of ERK and moderate activation of SAPK/JNK occurred within 30 min of H$_2$O$_2$ treatment.
