A Case of Pulmonary Paragonimiasis with Chronic Abdominal Pain and Erythematous Rash in a 6-year-old Girl (만성 복통과 발진을 주소로 내원한 6세 여아에서 진단된 폐흡충증 1예)

  • Pleural paragonimiasis is uncommon in the pediatric population and therefore can be challenging to diagnose. This is a case of a 6-year-old girl with pleural effusion who had been having intermittent persistent epigastric pain and erythematous rash on the face, hands, and arms for 6 months. Exudative pleural effusion with prominent eosinophils and serum eosinophilia were observed. As patient showed high immunoglobulin M (IgM) titers against Mycoplasma pneumoniae, she was treated with antibiotics; however, the pleural effusion did not improve during hospitalization. Despite showing negative stool ova and cyst results, patient's serum and pleural effusion were positive for Paragonimus westermani-specific IgGs on enzyme-linked immunosorbent assay. Respiratory symptoms, pleural effusion, and skin symptoms improved after praziquantel treatment.

Maternal Psychosocial Factors that Affect Breastfeeding Adaptation and Immune Substances in Human Milk (산모의 모유수유 적응과 모유 내 면역물질에 영향을 미치는 심리사회적 요인)

  • Purpose: This study was to identify relationships of maternal psychosocial factors including mother's mood state, childcare stress, social support and sleep satisfaction with breastfeeding adaptation and immune substances in breast milk, especially secretory immunoglobulin A (sIgA) and transforming growth factor-beta 2 (TGF-${\beta}2$). Methods: Data were collected from 84 mothers who delivered full-term infants by natural childbirth. Structured questionnaires and breast milk were collected at 2~4 days and 6 weeks postpartum. Data were analyzed using descriptive statistics, Pearson's correlation, multiple linear regression, and generalized estimating equation (GEE). Results: Scores for the breastfeeding adaptation scale were significantly related with child care stress, mood state and social support. Mother's anger was positively correlated with the level of sIgA in colostrum (p<.01). Immune substances of breastmilk was significantly influenced by time for milk collection (p<.001) and the type of breastfeeding (sIgA, p<.001, TGF-${\beta}2$, p=.003). Regression analysis showed that breastfeeding adaptation could be explained 59.1% by the type of breastfeeding, childcare stress, the Profile of Mood States, emotional support and sleep quality (F=16.67, p<.001). Conclusion: The findings from this study provide important concepts of breastfeeding adaptation program and explanation of psychosocial factors by immune substances in breast milk. Future research, specially, bio-maker research on breast milk should focus on the ways to improve breastfeeding adaptation.

Hu.4-1BB-Fc fusion protein inhibits allergic inflammation and airway hyperresponsiveness in a murine model of asthma

  • Purpose: 4-1BB (CD 137) is a costimulatory molecule expressed on activated T-cells. Repression by 4-1BB is thought to attenuate Th2-mediated allergic reactions. The aim of this study was to investigate the effect of 4-1BB on allergic airway inflammation in a murine asthma model. Methods: BALB/c mice were sensitized to and challenged with ovalbumin (OVA). Hu.4-1BB-Fc was administered 1 day before the first OVA sensitization or 1 day after the second OVA sensitization. Following antigen challenge, airway responsiveness to methacholine was assessed and bronchoalveolar lavage (BAL) fluid was analyzed. Total immunoglobulin (Ig) E, OVA-specific IgE, $IgG_1$, and $IgG_{2a}$ levels in sera were measured by enzyme-linked immunosorbent assay. Lung pathology was also evaluated. Results: In mice treated with Hu.4-1BB-Fc before the first OVA sensitization, there was a marked decrease in airway hyperresponsiveness, total cell count, and eosinophil count in the BAL fluid. In addition, Hu.4-1BB-Fc treatment decreased serum OVA-specific $IgG_1$ levels and increased serum $IgG_{2a}$ level significantly compared with the corresponding levels in mice sensitized to and challenged with OVA. Hu.4-1BB-Fc-treated mice also showed suppressed peribronchial and perivascular inflammatory cell infiltration. In contrast, treatment with Hu.4-1BB-Fc 1 day after sensitization had no effect on airway hyperresponsiveness and showed less suppression of inflammation in lung tissue. Conclusion: Administration of Hu.4-1BB-Fc can attenuate airway inflammation and hyperreactivity in a mouse model of allergic airway inflammation. In addition, administration before sensitization may be more effective. These findings suggest that 4-1BB may be a useful therapeutic molecule against asthma.

The Effects of Line Dance on Immune function and Inflammation for Elderly Women (라인댄스가 여성노인의 면역기능 및 염증에 미치는 영향)

  • This study was conducted to identify the characteristics of immune function and inflammatory changes for elderly women, by having them to participate in line dance program. For this purpose, 18 elderly women in their 60s to 70s, living in Y city were selected, and 9 of the participants were randomly assigned to participate in the line dance program for 12-weeks (line dance exercise group), while the other 9 participants were assigned to carry out their daily lives without participating in certain physical activity (control group) for the same period. The subjects' immune function and inflammation were measured using SPSS 24.0 for datafication, and the average and two-way ANOVA analysis were conducted. The result indicates the immunoglobulin, IgA, has increased, but not dramatically. The level of IgG(p<.05), and IgM(p<.05) increase were detected from the line dance exercise group, but not so much from the control group. Inflammation indicator, the CRP(p<.05), were significantly reduced from the line dance exercise group, but not much for the control group. According to the collected data, line dance for the elderly women can be considered as an effective exercise program, and the result shows that inflammation is reduced and immune function is improved. It also implies the needs of active physical activity arbitration to keep the elderly women's health and to prevent the chronic disease in the future.

Effect of Dietary Oligosaccharides on the Performance, Intestinal Microflora and Serum Immunoglobulin Contents in Laying Hens (Oligosaccharides 계열 생균제가 산란계의 생산성, 장내 미생물 및 혈청 면역 글로불린 함량에 미치는 영향)

  • This study was conducted to investigate the effects of dietary supplementation of mannan-oligosaccharides (MOS) and fructo-oligosaccharides (FOS) on the performance, immune response and small intestinal microflora in laying hens. A total of 960 Hy-Line $Brown^{(R)}$ laying hens of 27 wks old, housed in 2 bird cages, were assigned in a completely randomized block design into one of the following 6 dietary treatments: control, antibiotic (6 ppm avilamycine), 0.025% MOS, 0.05% MOS, 0.25% FOS, and 0.5% FOS. Each treatment had 4 replicates of 40 birds and was fed ad libitum for 6 wks under 16 h lighting regimen. There were significant differences among treatments in hen-day and hen-housed egg production. Hen-day egg production in 0.025% MOS was significantly higher than that of control. Hen-housed egg production in antibiotic-treated group was significantly higher compared with control. Egg weight, feed intake and feed conversion were not significantly different among treatments. Egg shell thickness was highest in 0.25% FOS, but was not significantly different among the rest of treatments. There were no significant differences among treatments in egg shell strength, egg shell color, egg yolk color and Haugh unit. IgG concentrations in serum were not significantly different among treatments. On the other hand, IgA concentrations of the treated birds tended to be increased compared with control. Dietary treatments tended to decrease Cl. perfringens and E. coli, and to increase Lactobacillus spp. The result of this experiment showed that dietary supplementation of MOS and FOS in laying hens tended to improve egg production comparable to the supplementation of antibiotics. The level of serum IgA and small intestinal microflora were also significantly affected by the treatments.

Effects of Various Chitosan Oligomer Molecular Weight Levels on the Disorders of Lipid Metabolism and Immune-related Factors in Rats Treated 2,3,7,8-Tetrachlorodibenzo-p-dioxin (다이옥신계 TCDD(2,3,7,8-Tetrachlorodibenzo-p-dioxin)에 노출된 흰쥐의 지질대사 및 면역관련 인자에 대하여 키토산 올리고머의 분자량별 섭취효과)

  • This study was conducted to investigate the effects of various levels of chitosan oligomer (CO) molecular weight on the disorders of lipid metabolism and immune-related factors induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), that is a endocrine disrupter, using adult male rats (SD) for 3 weeks. These 40 animals were divided into five groups. Three kinds of CO were used by molecular weight (MW) (less than 1000, 1000~3000, and 5000~10000) and added 4% to basal diets respectively. TCDD (40 ${\mu}g$/kg B.W) was intraperitoneally injected into rats at the beginning of the experiment. The relative weights of the livers were increased in all rats treated with TCDD, and the brain and testis weights were increased in all CO diet groups, compared to the control and TCDD groups. The levels of white blood cells (WBC) and red blood cells (RBC), hemoglobin, hematocrits (HCT), and platelets were significantly lowered by treating TCDD. By the way, RBC and HCT tended to recover by CO diets. The elevation of serum total and HDL cholesterol levels induced by TCDD treatment was significantly reduced by CO (5000~10000 MW) diets. The apparent increasing of the total lipid, cholesterol, and triglyceride levels of rat livers induced by TCDD was tended to be suppressed in those fed CO diets. Especially, diets with less than 1000 MW significantly diminished liver triglycerides. The levels of serum immunoglobulin (Ig) A, IgG1 and IgM were significantly high in rats fed CO (5000~10000 MW) diets. The decreasing levels of IgE by treatment with TCDD tended to recover all the CO diet groups to the level of control group. In histochemical observation, the fat droplets and apoptosis of liver due to TCDD treatment were markedly alleviated in all CO diet groups. These results indicated that CO, though not regular according to molecular weight, can exert improving effects on lipid accumulation, hepatocytic disorders, abnormal blood cells, and some immunoglobulins induced by TCDD.

Immunomodulatory effects of β-1,3/1,6-glucan and lactic acid bacteria in LP-BM5 murine leukemia viruses-induced murine acquired immune deficiency syndrome (면역결핍 모델에서 β-1,3/1,6-glucan과 유산균을 이용한 in vivo 면역 활성 조절 효과)

  • In this study, ${\beta}$-1,3/1,6-glucan, lactic acid bacteria, and ${\beta}$-1,3/1,6-glucan+lactic acid bacteria were tested for 10 weeks using an immunodeficient animal model infected with LP-BM5 murine AIDS virus On the immune activity. Cytokines production, plasma immunoglobulin concentration, T cell and B cell proliferation were measured. As a result, the T cell proliferative capacity which was weakened by immunization with LP-BM5 murine AIDS virus increased significantly T cell proliferative capacity compared with the red ginseng control group. B cell proliferative capacity was significantly higher than the infected control group. Increased B cell proliferation was reduced. In the cytokine production, IL-2, IL-12 and IL-15 in the Th1-type cytokine increased the secretion of IL-2, IL-12 and IL-15 compared to the infected control. The proliferative capacity of the treated group was higher than that of the mixed treatment group. TNF-${\alpha}$ was significantly decreased compared with the infected control group. The IL-4, IL-6 and IL-10 levels were significantly inhibited in the infected control group and the Th1/Th2 type cytokine expression was regulated by immunohistochemistry. IgE, IgA, and IgG levels were significantly lower in the immunoglobulin secretion assay than in the control. As a result, the immunomodulatory effect of ${\beta}$-1,3/1,6-glucan+lactic acid bacteria was confirmed by mixing with LP-BM5 murine AIDS virus-infected immunodeficient animal model.

Anti-Allergic Effect of Ulmus davidiana Cortex on Contact Dermatitis Induced by Dinitrofluoro-Benzene in Mice

  • Objective: The root bark of Ulmus davidiana var. Japonica (Ulmi Radicis cortex, URC) is a medicinal herb used for promoting diuresis and treating dampness. In Korea, URC has long been used as an efficacious therapy for inflammation, burns, frostbite and skin diseases such as eczema and psoriasis. Methods: In the present study, we used 1-fluoro-2,4-dinitrofluorobenzene (DNFB)-induced contact dermatitis (CD) mouse model to investigate the anti-allergic and the anti-inflammatory effects of URC on skin lesion, histopathological changes and specific antibody production. Results: URC treatment, 10 mg/mL, effectively inhibited skin lesions induced by repeated paintings with DNFB. In the histopathological observation, topical application of URC inhibited spongiosis. In addition, URC lowered the production levels of total immunoglobulin and IgG2a in serum. Conclusion: These data indicate that URC has an anti-inflammatory effect that produces an improvement of skin lesions in CD mice.

Activation-induced Cytidine Deaminase in B Cell Immunity and Cancers

  • Activation-induced cytidine deaminase (AID) is an enzyme that is predominantly expressed in germinal center B cells and plays a pivotal role in immunoglobulin class switch recombination and somatic hypermutation for antibody (Ab) maturation. These two genetic processes endow Abs with protective functions against a multitude of antigens (pathogens) during humoral immune responses. In B cells, AID expression is regulated at the level of either transcriptional activation on AID gene loci or post-transcriptional suppression of AID mRNA. Furthermore, AID stabilization and targeting are determined by post-translational modifications and interactions with other cellular/nuclear factors. On the other hand, aberrant expression of AID causes B cell leukemias and lymphomas, including Burkitt's lymphoma caused by c-myc/IgH translocation. AID is also ectopically expressed in T cells and non-immune cells, and triggers point mutations in relevant DNA loci, resulting in tumorigenesis. Here, I review the recent literatures on the function of AID, regulation of AID expression, stability and targeting in B cells, and AID-related tumor formation.

Increased Antitumor Immunity of Mouse GM-CSF in Mouse Colon Tumor (CT-26) Model

  • Oncolytic vaccinia virus is an engineered vaccinia virus that selectively destroys cancer cells and induces tumor immune response. Oncolytic vaccinia expressing mouse GM-CSF showed cytotoxic activity against various kinds of cancer cells when oncolytic vaccinia virus expressing human GM-CSF and mouse GM-CSF is intravenously administered in the mouse CT26 colon tumor model. Cancer cells treated with isolated immunoglobulin G from the serum with complement showed these cytotoxic activity and complement observed dose-dependent cytotoxic effect. These results suggest that oncolytic vaccinia virus expressing mouse GM-CSF can increase oncolytic vaccinia virus by inducing anticancer antibody in a mouse tumor model. Further studies are needed on antitumor immunity of GM-CSF.
