연구배경: IL-4는 기관지천식의 병인에 중요한 역할을 하는 cytokine으로서 B세포에서 IgE를 생성하게 하여 아토피의 발생에 중요한 역할을 한다. IL-4의 유전자 다형에 관한 연구로 promoter부위 (-589) 에 cytosine이 thymine으로 치환되는 polymorphism(-589 C$\rightarrow$T polymorphism) 이 존재한다는 것이 밝혀졌다. 그 후 IL-4 유전자 polymorphism이 천식 환자에 어떤 영향을 미치는 지에 대한 연구가 진행되어 왔다. 본 연구에서는 polymorphism이 IL-4 유전자의 발현을 조절하는 promoter에 위치하므로 혈액내 IgE level에 영향을 주어 증상의 정도에 관여할 것이라는 가정하에 천식 환자의 증상의 정도와 IL-4 유전자 다형과의 연관성을 조사하였다. 또한 한국의 천식환자에서의 이러한 유전자 다형의 유형과 빈도를 조사하였다. 방 법: 고려대학교 부속병원에 내원한 49명의 천식환자와 33명의 정상 대조군을 대상으로 하였다. 모든 천식 환자는 증상의 정도에 따라 경증, 중등증 및 중증의 두 대상군으로 나누었다. 모둔 천식 환자와 정상인의 혈액에서 DNA를 분리하였고 ARMS(Amplification Refractory Mutation System) 및 RFLP(Restriction Fragment Length Polymorphism)를 시행하여 polymorphism의 존재 및 유형을 검색하였다. 결 과: 천식 환자의 중상의 정도와 IL-4 유전자 다형의 유형과는 유의한 관계를 발견할 수 없었다 (P=0.709). 정상인이나 천식환자에서 polymorphism(C/T 및 T/T형)의 빈도가 각각 100% 와 95.9%로 서양보다 월등히 높았다. 그러나 두 군에서 polymorphism의 유형 및 그 빈도에 있어서 차이는 없었다. 33명의 정상 대조군에서는 C/C형을 발견할 수 없었고, 정상인과 천식환자에서 공히 T/T형이 C/T형보다 약간 많은 빈도를 보였다. 결 론: IL-4 유전자의 -589 C/T polymorphism과 천식 환자의 증상의 경중과는 유의한 연관성을 발견하지 못했다. 그러나 우리나라 천식 환자와 정상인에서 서양보다 월등히 많은 수에서 polymorphism을 관찰할 수 있었으며, 정상인에서는 C/C 형이 발견되지 않았다. 따라서 IL-4 유전자 다형성이 종족간에 현저한 차이를 보일 수 있다는 결론을 얻을 수 있었다.
Background : Recently, so many people are suffered from the allergic or auto-immune disease, and the representative disease is just Allergic Asthma. It is because human immune function has been decreased. Many treatments were done to treat this disease, and many methods were studied to increase immune function and to suppress the asthma. But, the effect of asthma-suppression and improvement of immune response of EH-HA(Ephedrae Herba Herbal Acupuncture) has not been studied in detail. To study the effects of EH-HA, we injected EH-HA at Joksamni(ST36) of C57BL/6 mice. Objectives : The purpose of this study is to investigate the effect of asthma-suppression and improvement of immune response of EH-HA(Ephedrae Herba Herbal Acupuncture). EH-HA was done at Joksamni(ST36) of the mice with ovalbumin-induced asthma. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks. Two experimental groups were treated with different concentrations(1%, 0.1%) of EH-HA at Joksamni(ST36) for the later 8 weeks(3times/week). Results : 1. The lung weight of the group treated with EH-HA decreased significantly compared with that of control group. 2. The total cells in lung, total leukocytes and eosinophils in BALF of the group treated with EH-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the group treated with EH-HA in photomicrographs decreased significantly compared with those of control group. 4. The concentrations of IL-13, IgE, IL-4 in serum and IL-4 in BALF of the group treated with EH-HA decreased significantly compared with those of control group. 5. The numbers of $Gr-1^+/CD11b^+\;cells,\;CD3e^-/CCR3^+\;cells,\;CD4^+\;cells,\;CD8^+\;cells,\;CD3e^+/CD69^+\;cells\;and\;IgE^+/B220^+\;cells$ in lung of the group treated with EH-HA decreased significantly compared with those of control group. 6. In RT-PCR, the mRNA expression of IL-4, IL-5 and IL-13 in the group treated with EH-HA decreased compared with those of control group. Conclusion : These results suggested that EH-HA at Joksamni(ST36) in C57BL/6mice may be effective to OVA-induced asthma of C57BL/6 mice.
We observed the efficacy of natural herbs and mixture in treating atopic dermatitis using anti-human IgE treated Human HMC-I cell and NCNga mice model. First, we selected three herbs, Cynonchum witfordii, Diospyros kaki, Ilite which were used to treat skin disease in Traditional Korea Medicine. Using Human HMC-I cell treated with anti-human IgE, we investigate in vitro whether each herb effects on IL-4, IL-13, $TNF-{\alpha}$ expression and $TNF-{\alpha}$, Histamine secretion value. Finally, we conducted study whether the mixture of the selected herbs is more effective than each herb which consist the mixture and control group. The results show that the mixture is better in improving atopic dermatitis condition.
Background: This study sought to determine whether the antioxidant effects of astaxanthin (AST) could have an anti-inflammatory effect to reduce inflammation caused by atopic dermatitis (AD). Methods: Using a mouse model of AD induced by phtalic acid (PA), the levels of inflammation, inflammatory agents, and evidence of antioxidant activity were examined in PA treated mice (n = 3), PA-AST treated mice (n = 3), and a control group of mice (n = 3). This included measurements of ear thickness, levels of mast cells, IgE, inflammatory cytokine, malondialdehyde (MDA), hydrogen peroxide, HO-1, and GPx-1. Results: AST treatment significantly prevented inflammation as measured by ear thickness (p < 0.05), mast cell count (p < 0.001), and IgE concentration in the blood (p < 0.001). Levels of TNF-α (p < 0.001), IL-1β (p < 0.001), IL-6 (p < 0.001), and MDA (p < 0.05) were also significantly lower. In addition, GSH levels increased significantly (p < 0.001), and the level of hydrogen peroxide significantly reduced (p < 0.01). The expression of HO-1, GPx-1 increased. Conclusion: In this small experimental study, AST acted on inflammatory mechanisms that induced AD, through anti-inflammatory and antioxidant mechanisms, and is a candidate of interest in the clinical treatment of AD.
Objectives : The aim of this study was to investigative the effects of Gagambojungikgi-tang (GBT), a Korean herbal medicine, on the immune mediators, T cell proliferation and wound healing in the recombinant Sj26 (rSj26) antigen induced atopic dermatitis(AD) model mice. Methods : GBT is the water extracts prepared from mixture of Ginseng Radix, Astragali Radix, Angelicae gigantis Radix, Atractylodes Rhizoma alba, Aurantii nobilis Pericarpium, Glycyrrhizae Radix, Artemisia iwayomogi Herba, Scutellaria Radix, Lonicera japonica Flos. This is a modified prescription of Bojungikgi-tang, which has been used for the treatment of indigestion, and immunological disease in east-asian countries. GBT was orally administered or externally applied at difference doses. The levels of immune mediators [(IgE, IgG1, prostaglandin E2 (PGE2), Th1/Th2 cytokines], T cell proliferation, and wound healing in the rSj26 or chemical antigen induced AD model BALB/c were investigated. Results : GBT dose-dependently suppressed the release of TNF-${\alpha}$, IL-$1{\beta}$ (Th1 cytokines), IL-4, IL-10 (Th2 cytokines), PGE2 (inflammatory mediators) and T cell proliferation. But GBT increased the production of IFN-${\gamma}$ (Th1 cytokine). Furthermore, A wound healing effect of GBT was similar to external application of dexamethasone. Conclusions : These results suggest that GBT suppresses the inflammatory mediators and regulates the Thl/Th2 cytokines, and promotes the wound healing. Therefore, these properties may contribute to the strong anti-AD effect of GBT.
Objectives & Methods : The purpose of this study is to observe the effects of Phellodendri Cortex Herbal-acupuncture solution (PC-HAS) at Joksamni (ST36) on collagen II induced arthritis in DBA-1J mice. The author performed several experimental items to analyze arthritis evaluation, change of weight, spleen size and adhesion rate, change of cytokine level, IgG, IgM and anti-collagen II, chang of immunocyte count, histological change of CIA mouse joint. Results : 1. In the PC-HA group, arthritis index, the incidence of arthritis and joint edema were significantly decreased. 2. In the PC-HA group, the change of spleen size, spleen adhesion rate and the knee joint were significantly decreased. 3. The levels of $IL-1{\beta}$, IL-6 and INF- in serum of the CIA mouse were significantly decreased by PC-HA. 4. The levels of IgG, IgM and anti-collagen II in serum of the CIA mouse were significantly decreased by PC-HA. 5. In the CIA mouse spleen cell culture, the levels of IFN- , IFN- / IL-4, IL-10 were significantly decreased by PC-HA, but the level of IL-4 was significantly increased by PC-HA. 6. In the PC-HA group, the ratios of $CD3e^+$ to $CD45R^+$ cell, $CD4^+$ to $CD8^+$ cell and $CD4^+/CD25^+$ cell were similarly maintained as normal group in the CIA mouse spleen cell. 7. In the PC-HA group, $CD4^+CD25^+$ and $CD45R^+/CD69^+$ cell were significantly decreased in the lymph nodes. 8. In the PC-HA group, $CD3^+/CD69^+$ and $CD11b^+/Gr-1^+$ cell were significantly decreased in knee joint. 9. In histology, the cartilage destruction and synovial cell proliferation in the PC-HA group were similar with that of the normal group and the collagen fiber expressions in the PC-HA group were similar with that of the normal group. Conclusions : Form the result above, the results suggest that the PC-HA at ST36 has significant effect on collagen-induced arthritis, and can be put to practical use in the future rheumatoid arthritis clinic.
Results: 1. In the AlPR-HA group, the incidence of arthritis and arthritis index were significantly decreased. 2. In the AlPR-HA group, the levels of $IL-6,\;IFN-{\gamma},\;TNF-{\alpha},\;IL-1{\beta},\;IgG,\;IgM,\;Anti-collagen\;II$ in serum of the CIA mouse were significantly decreased. 3. In the AlPR-HA group, the levels of $IFN-{\gamma:,\;IFN-{\gamma}\;/IL-4$ in spleen cell culture of the CIA mouse were significantly decreased. 4. In the Hematoxylin and eosin stain, the cartilage destruction and synovial cell proliferation were decreased in the AIPR-HA group. 5. In the Masson's trichrome stain, the collagen fiber expressions were similar with that of the Normal group. 6. In the AlPR-HA group, the expression ratio of $CD3e^+$ to $CD19^+$ cell and $CD4^+$ to $CD8^+$ cell were similarly maintained as Normal group in the CIA mouse lymph nodes. 7. In the AlPR-HA group, $CD3e^+/CD69^+$ cells in the CIA mouse joint and $CD11a^+/CD19^+$ cells and $CD11b^+/Gr-1^+$ cells in the CIA mouse lymph nodes were significantly decreased. 8. In the AIPR-HA group, $CD4^+/CD25^+$ cells were significantly decreased in the CIA mouse spleen cell and were similarly maintained as Normal group in the CIA mouse lymph nodes. Conclusions: These results suggest that AlPR-HA at 51'36 has an effect to control synovial cell proliferation and cartilage destruction in rheumatoid arthritis.
Objectives : To clarify the possible effect of Lycium chinese Mill (LC)., Morus alba L (MA)., and Lycium chinese Mill. +Morus alba L. (LC+MA), we have examined their influence on the development of pulmonary eosinophilic inflammation in the asthmatic murine model. Methods : Female Balb/c mice (5weeks) were immunized on two different days (21 days and 7 days before inhalational exposure) by intraperitonial injections of 0.2ml alum-precipitated Ag containing $100{\mu}g$ of OVA bound to 4 mg of aluminum hydroxide in PBS. Seven days after the second sensitization, mice were exposed to aerosolized ovalbumin for 30 minutes/day on 3 days/week for 8 weeks (at a flow rate of 250 L/min, 2.5% ovalbumin in normal saline) and, LC, MA, and LC+MA (500 mg/kg) were orally administered 3 times per a week for 8 weeks. Results : The suppressive effect of LC, MA, and LC+MA were demonstrated by the accumulation of eosinophills into airways, with the reduction of eosinophil, total lung leukocytes numbers. These were correlated with the marked reduction of IL-5, IL-13 and IL-4 levels in the BALF and serum. OVA-specific IgE levels were also decreased in serum and BAL from these mice. LC, MA, and LC+MA decreased eosinophil CCR3 expression and CD11b expression in lung cells. Conclusions : These results indicate that LC, MA, and LC+MA have high inhibitory effects on airway inflammation and hyper-responsiveness in the asthmatic murine model. The suppression of IL-5, IgE, eosinophil CCR3 expression and CD11b expression, and the increase of IFN-${\gamma}$ production in BALF seem to contribute to this effect. Hence, the results indicated that LC, MA, and LC+MA could act as a immuno-modulator which possesses anti-inflammatory and anti-asthmatic property by modulating the imbalance between Th1 and Th2 cytokines.
본 연구는 천식 생쥐 모델을 이용하여 사이프러스 오일(CS)이 알러지성 천식에 미치는 영향을 평가하기 위하여 수행되었다. Ovalbumin으로 천식을 유발시킨 천식 생쥐 모델을 사용하여 실험군에 0.3% CS를 3 주간, 1주 3회, 1회 30분간 분무기를 이용하여 흡입시켰다. 기도과민성, 백혈구 중 호산구 수, 폐 세포 내 면역세포와 Th2 싸이토카인의 변화를 관찰한 결과 CS를 처리한 실험군에서 기도과민성, 호산구 수 및 폐 세포 내 IL-5,IL-13 수치, 혈청내 IgE 분비량, 폐 세포 내 CCR3, CD3, CD4 세포의 수 등 모든 항목에서 현저한 감소효과를 보였다. 이러한 결과는 CS가 천식 반응의 주요인자인 Th2 싸이토카인과 호산구에 긍정적인 영향력이 있음을 시사한다. 따라서 CS는 천식 치료제로서 가능성이 있을 것으로 사료된다.
Objectives : To clarify the possible effect of JS (Juglans sinensis), PCF (Psoralea corylifolia L.), and J+P(JS+PCF), we examined their influence on the development of pulmonary eosinophilic inflammation in the asthmatic murine model. And also JS, PCF, and J+P decreased eosinophilic CCR3 and CD11b expression in lung tissue. Conclusions : These results indicate that JS, PCF, and J+P have deep inhibitory effects on airway inflammation and hyper-responsiveness in the asthmatic murine model. The suppression of IL-5, IgE, and eosinophilils and the increase of IFN-${\gamma}$ production in BALF seem to contribute to these effects. Specially, esosinophils and TNF-a in J+P combination group were significantly reduced in BALF and lung tissue. Hence, the results indicated that JS, PCF, and J+P could act as an immuno-modulator which possesses anti-inflammatory and anti-asthmatic property by modulating the imbalance between Th1 and Th2 cytokines.

제 19 조 (관할 법원)
