Evaluation of Control Pollination Efficiency and Management Status in Control Pollinated Progeny Populations of Pinus densiflora using Pedigree Analysis based on Microsatellite Markers (소나무 인공교배 차대집단에서 Microsatellite marker 혈통분석을 이용한 인공교배 효율 및 관리상태 평가)

  • Controlled pollination (CP) is an important method in tree breeding programs because CP quickly generates desirable genotypes and rapidly maximizes genetic gains. However, few studies have evaluated the efficiency and success rate of CP in the breeding program of Pinus densiflora. To evaluate CP and the management of control pollinated progenies, we used 159 individuals in CB2 × KW40 or KW40 × CB2 populations that were established in 2015. After genotyping microsatellite loci, we estimated whether the number of primers was sufficient or not. Then, we performed pedigree analysis. The result showed that the number of primers was sufficient. By pedigree analysis, we found out that 60 of 159 individuals had been generated by the mating between CB2 and KW40. In the maternity analysis, there was evidence to indicate the possibility of management problems. Therefore, we excluded 54 individuals and repeated the pedigree analysis. In the second analysis, 47 of 105 individuals were generated by the mating between CB2 and KW40. To increase the efficiency of CP in tree breeding programs, several precautions are required. It is necessary to identify the exact clone names of the mother and father trees. In addition, CP processes should be performed properly, including deciding on the schedule of CP and the isolation of female strobili or flowers. Finally, the monitoring of hybrid progenies management after mating is important. Molecular markers should be used to identify the clone names of the mother and father trees and for monitoring post hoc management. This study provides a reference for tree breeding programs for the future control pollination of pine species.

Tracking lead contamination sources of sediments in Lake Andong using lead isotopes (납 동위원소를 이용한 안동호 퇴적물 중의 납 오염 기원)

  • The objective of this study was to identify Pb pollution sources of sediments in Lake Andong. We analysed Pb isotopes in sediments from Lake Andong, soils and mining tails from the watershed as well as sludges and wastewater from zinc smelting facilities which exists in upper stream of Lake Andong. The Pb isotope ratios ($^{207}Pb/^{206}Pb$ and $^{208}Pb/^{206}Pb$) for sediments are $0.827{\pm}0.004$ and $2.041{\pm}0.015$, which showed similar values with those of mining tails, $0.815{\pm}0.002$ and $2.016{\pm}0.006$, respectively. The isotopic ratio values of soils existed in the range of 0.756~0.881 and 1.872~2.187. In imported zinc ores, the isotopic ratio values existed in the range of 0.816~0.956 (mean 0.832) and 2.029~2.219 (mean 2.059). These values were similar to those in zinc and lead concentrate originated from Canada and South America. Additionally, isotopic ratio values for sludge and wastewater were $17.515{\pm}0.155$, $15.537{\pm}0.018$, and $37.357{\pm}0.173$, respectively. The Pb isotopic ratio of sediments showed binary combination patterns with soil and mining tails, which were similar to those for Korean Pb ore.

Sorption Efficiency of the Bamboo Charcoal to Remove the Cesium in the Contaminated Water System (오염수계 내 세슘 제거를 위한 대나무 활성탄의 흡착효율 규명)

  • The cesium (Cs) removal from the contaminated water system has been considered to be difficult because the cesium likes to exist as soluble phases such as ion and complexes than the solid in water system. Many researches have focused on developing the breakthrough adsorbent to increase the cesium removal efficiency in water. In this study, the laboratory scale experiments were performed to investigate the feasibility of the adsorption process using the bamboo charcoal for the Cs contaminated water system. The Cs removal efficiency of the bamboo charcoal were measured and the optimal adsorption conditions were determined by the adsorption batch experiments. Total 5 types of commercialized bamboo charcoals in Korea were used to identify their surface properties from SEM-EDS and XRD analyses and 3 types of bamboo charcoals having large specific surface areas were used for the adsorption batch experiment. The batch experiments to calculate the Cs removal efficiency were performed at conditions of various Cs concentration (0.01 - 10 mg/L), pH (3 - 11), temperature ($5-30^{\circ}C$), and adsorption time (10 - 120 min.). Experimental results were fitted to the Langmuir adsorption isotherm curve and their adsorption constants were determined to understand the adsorption properties of bamboo charcoal for Cs contaminated water system. From results of SEM-EDS analyses, the surfaces of bamboo charcoal particles were composed of typical fiber structures having various pores and dense lamella structures in supporting major adsorption spaces for Cs. From results of adsorption batch experiments, the Cs-133 removal efficiency of C type bamboo charcoal was the highest among those of 3 bamboo charcoal types and it was higher than 75 % (maximum of 82 %) even when the initial Cs concentration in water was lower than 1.0 mg/L, suggesting that the adsorption process using the bamboo charcoal has a great potential to remove Cs from the genuine Cs contaminated water, of which Cs concentration is low (< 1.0 mg/L) in general. The high Cs removal efficiency of bamboo charcoal was maintained in a relatively wide range of temperatures and pHs, supporting that the usage of the bamboo charcoal is feasible for various types of water. Experimental results were similar to the Langmuir adsorption model and the maximum amount of Cs adsorption (qm:mg/g) was 63.4 mg/g, which was higher than those of commercialized adsorbents used in previous studies. The surface coverage (${\theta}$) of bamboo charcoal was also maintained in low when the Cs concentration in water was < 1.0 mg/L, investigating that the Cs contaminated water can be remediated up with a small amount of bamboo charcoal.

Analysis of PAHs (polycyclic aromatic hydrocarbons) in Ground Coffee Using GC-tandem Mass Spectrometry and Estimation of Daily Dose (GC-tandem mass spectrometry를 이용한 분쇄원두커피 중 PAHs(polycyclic aromatic hydrocarbons) 분석법 연구 및 인체노출량 평가)

  • The purpose of this study was to develop an analytical method for determining 15 polycyclic aromatic hydrocarbons (PAHs) of EU priority using gas chromatography (GC)-tandem mass spectrometry (MS). The PAHs in ground coffee were analyzed after being extracted using methods such as saponification-liquid-liquid extraction, Soxhlet extraction, and solid-liquid extraction. The solid-liquid extraction method showed the greatest repeatability and most efficient reduction of the matrix effect. GC-tandem MS for the quantification of the 15 PAHs showed better resolution and lower limit of detections (LODs) than GC-MS-selected ion monitoring (SIM) and high performance liquid chromatography with fluorescence detector. LODs of this method for the ground coffee types were 0.002-0.1 ${\mu}g/kg$ and limit of quantifications (LOQs) were 0.006-0.2 ${\mu}g/kg$ The recoveries ranged from 52.6 to 93.3%. Forty-six commercial types of ground coffee were analyzed to determine their PAHs contamination levels. PAHs concentration ranged from ND to 5.988 ${\mu}g/kg$. This study was conducted with toxicity equivalence factors, the U.S. EPA recommendation to identify dietary risks for PAHs in different types of coffee. The estimated average daily dose of PAHs was $5.24{\times}10^{-8}$ mg/kg body weight/day.

Clinical Analysis of Pleuropneumonectomy for Chronic Inflammatory Lung Disease (만성염증성 폐질환에서 전폐절제술의 임상적 평가)

  • Background: Pneumonectomy for inflammatory lung disease has been of major concern because of its associated morbidity and mortality, particularly with respect to pleuropneumonectomy. The purpose of this study is to evaluate the surgical outcomes, and identify the risk factors contributing to postoperative complications in patients undergoing pleuropneumonectomy. Material and Method: Ninety-eight patients underwent pneumonectomy for benign inflammatory lung disease were retrospectively analyzed. Pleuropneumonectomy (Group A) was done in 48 patients and standard pneumonectomy (Group B) was done in 50 patients. Clinical characteristics, postoperative complications were examined and compared between 2 groups. In pleuropneumonectomy group, postoperative risk factors affecting morbidity were evaluated. Result: There was one in-hospital death. Twenty-three major postoperative complications occurred in 21 patients (21.4%). The common complications were empyema and bronchopieural fistula (BPF) in 8 (8.4%), re-exploration due to bleeding in 8. At least one postoperative complication occurred in 14 of 48 patients from Group A (29.2%) and in 7 of 50 patients from Group B (14%). In Group A, empyema and BPF encountered in 6 and re-exploration for bleeding in 6 were the most common complication. In univariate analysis, right pneumonectomy, completion pneumonectomy, large amount of blood loss (>1,000 mL), and intrapleural spillage were risk factors contributing to postoperative complications in Group A. In multivariate analysis, intrapleural contamination during operation was a risk factor of postoperative complication. Conclusion: The morbidity and mortality rates of pneumonectomy for chronic inflammatory lung disease are acceptably. However, we confirm that pleuropneumonectomy is a real technical challenge and a high-risk procedure and technically demanding. Meticulous surgical techniques are very important in preventing serious and potentially lethal complications.

Distribution and Characteristics of Heterotrophic Plate Count Bacteria in Water Samples from Drinking Water Dispensers (냉온수기에서 일반세균의 분포 및 분리한 세균의 특성)

  • To evaluate bacteriological water quality, samples were taken from drinking water dispensers placed at S company (S-C) and U highschool (U-H) in Ulsan. The medians of heterotrophic plate counts (HPCs) were 53 CFU/ml for the 74 water samples of S-C and 80 CFU/ml for the 36 cold water samples of U-H, and 38% of the S-C and 42% of the U-H samples showed HPC bacterial concentrations higher than 100 CFU/ml. Coliform bacteria were detected from one sample of S-C. To determine the major source of bacterial contamination, water samples were taken daily for $6\sim8$ days from the bottled water containers as well as the faucets of an experimental water dispenser. While the average HPCs in the bottled water containers were 33 CFU/ml for the first and 132 CFU/ml for the 2nd analysis, the HPC concentration in the cold water samples was 1,022 CFU/ml for the 2nd analysis. These results suggest that the majority of bacteria detected in the cold water samples were originated from the biofilms on the surface of water passages within the water dispensers. There was no significant increase in HPC bacterial concentrations within the bottled water container after installation on the water dispenser. We could isolate and tentatively identify 3 genera 6 species of Gram-positive and 7 genera 7 species of Gram-negative bacteria from the plate count agar plates of U-H samples. Among the isolates, 72% were observed as Gram-positive, and Micrococcus spp. was the most abundant with 54% of the total, followed by Sphingomonas paucimobilis with 16%. It appears that most of the HPC bacteria detected in water dispensers originate from indoor airborne bacteria, which may play important roles in the formation of biofilms on the surface of water passages within the water dispensers.

Distribution Characteristics of Environmental Contaminant at Soil in an Industrial Complex Area (공단지역 토양 중 환경오염물질 농도 분포 특성)

  • This study was performed to prevent the health damage of environmental contaminants in Industrial Complex Area. And, this study aimed to identify the concentration levels and distribution characteristics of environmental contaminants and Polycyclic Aromatic Hydrocarbons (PAHs) at soil in Industrial Complex Area and control area. The concentration of the soil pollution standard such as the heavy metals in the soil, VOCs, PAHs, and PCB were measured and analyzed using the soil specimens in the Industrial Complex Area and control area. Soil specimens from the Industrial Complex Area (the direct exposure area) and the control area were surveyed. Songdo-dong, Haedo-dong and Jechul-dong, which are in the direct exposure area and near the emission source, showed relatively high concentrations of contaminant materials when compared with Jangki-myeon, which is far off and in the control area. The concentration of zinc was 20.8-58.9% of the level of concern (300 mg/kg) in the 1st region, which is a relatively high concentration. The concentration of fluoride was under the standard in every region, but it was about 74% of the level of concern (400 mg/kg) in the 1st region. It is recommended that controlling fluoride emissions is necessary. Levels of organic phosphate, phenol, and VOCs like benzene, toluene, ethylbenzene and xylene were under the detection limit of the analysis instruments. The concentration of TPH was high in Songdo-dong. The concentration of contaminants in Jechul-dong was high. In addition, it was observed that the level of soil contamination changed depending on the distance from the emission source. The concentration of PAH compounds in the soil was 18.71-1744.59 ng/g, and the concentration of six potential cancer-causing PAH materials was 6.54-695.94 ng/g. The highest concentration was in Songdo-dong. The PAH concentration in the direct exposure area near the complex was relatively high compared to the indirect exposure area.

Comparison of Liver, Kidney, Bone Metal Concentration in OhJeokSan-Treated Rats (오적산(五積散)을 투여한 흰쥐의 간장, 신장, 골중 금속농도 비교에 관한 연구)

  • Traditional herbal medicine is used extensively among the Korean populations, and other Asian countries employ similar therapies as well In recent years, extensive focus was laid on adulteration of the herbal medicine with heavy metals. This may be mainly due to a soil contamination by an environmental pollution. The objective of this study is to identify the contents of various heavy metals in the blood from OhJeokSan-Decoction (OD) treated-rats. For this study, 13 kinds of metals including essential and heavy metals, i.e. Al, As, Cd, Co, Cr, Cu, Fe, Hg, Mn, Ni, Pb, Se and Zn were analyzed by a slight modification of EPA methods and the following results are obtained. ; 1. There is no significant difference between the OD-treated groups and control group in liver, kidney, bone, brain, weight, food intake. 2. The amount of each metal analyzed in the liver are as follows; Al ; $0.235{\sim}4.215mg/kg$, As ; $0.103{\sim}0.461mg/kg$, Cd ; $0.005{\sim}0.010mg/kg$, Co ; $0.017{\sim}0.046mg/kg$, Cr ; $0.137{\sim}0.403mg/kg$, Cu ; $1.736{\sim}4.827mg/kg$, Fe ; $54.472{\sim}381.447mg/kg$, Hg ; not detected, Mn ; $1.159{\sim}2.803mg/kg$, Ni ; $0.007{\sim}0.095mg/kg$, Pb ; not detected, Se ; $0.682{\sim}1.887mg/kg$, Zn ; $10.213{\sim}26.119mg/kg$, by groups, respectively. In control and other experimental group, several metal (Co, Cu, Mn, Zn, As, Cr) has a significant difference, but in experimental I and other experimental II, III, IV, V groups, there are no significant difference. 3. The amount of each metal analyzed in the kidney are as follows; Al ; $1.712{\sim}31.230mg/kg$, As ; $0.062{\sim}0.439mg/kg$, Cd ; $0.010{\sim}0.062mg/kg$, Co ; $0.000{\sim}0.101mg/kg$, Cr ; $0.125{\sim}0.636mg/kg$, Cu ; $3.385{\sim}12.502mg/kg$, Fe ; $41.148{\sim}99.709mg/kg$, Hg ; $0.000{\sim}0.270mg/kg$, Mn ; $0.433{\sim}2.301mg/kg$, Ni ; $0.000{\sim}0.221mg/kg$, Pb ; $0.000{\sim}0.584mg/kg$, Se ; $0.540{\sim}1.600mg/kg$, Zn ; $8.775{\sim}17.060mg/kg$, by groups, respectively. The amount of each metal analyzed in the bone(tibia and fibula) are as follows; Al ; $9.557{\sim}119.464mg/kg$, As ; $0.139{\sim}12.250mg/kg$, Cd ; $0.000{\sim}0.295mg/kg$, Co ; $0.022{\sim}0.243mg/kg$, Cr ; $0.239{\sim}1.999mg/kg$, Cu ; $0.000{\sim}2.291mg/kg$, Fe ; $240.249{\sim}841.956mg/kg$, Hg ; $0.000{\sim}0.983mg/kg$, Mn ; $0.214{\sim}7.353mg/kg$, Ni ; $5.473{\sim}11.453mg/kg$, Pb ; $0.000{\sim}8.502mg/kg$, Se ; $0.000{\sim}3.005mg/kg$, Zn ; $61.158{\sim}195.038mg/kg$, by groups, respectively. The concentration of Se, Cd are variated significantly in control and other experimental groups, and Se is variated significantly in experimental I and other experimental II, III, IV, V groups. 5. Exceptionally several metal concentration is increased or decreased. but there is no significant harmful difference of metal concentration in the liver, kidney and bone, from the OD-treated-rats compared to those of the control group even if higher dosage($1{\sim}8$ times dosage of person) of OD was administered. Thus, it is expected that the herbal decoction in the traditional herbal medicine would not lay any burden on the body and the heavy metal toxins would not affect our physiological system. However, other kinds of herbal treatment, such as i.v. and i.p. should be considered in terms of metal toxicity in the body since the level of certain metal.

The Behavior of Dissolved and Particulate Phases of Trace Elements within the Watershed of Juam Reservoir (주암호 집수유역 내 용존 및 입자상 미량원소의 거동 특성)

  • In order to investigate the amounts of trace elements flowing into reservoir, and to elucidate the relationship between trace element mobility and fraction size, the stream water and sediment samples were collected from thirty-two sites of the 3rd or 4th order stream within watershed surrounding the Juam reservoir. Chemical analyses of trace elements (As, Cd, Cr, Cu, Ni, Pb and Zn) for all samples were completed, and additionally cationi and anion for stream water samples. Considering the distribution of rocks and contamination sources in watershed, the eight stream sediments were selected from typical sites representing study areas, and we determined the concentrations of trace elements according to size fractions ($2\;mm{\sim}200\;{\mu}m$, $200{\sim}100\;{\mu}m$, $100{\sim}50\;{\mu}m$, $50{\sim}20\;{\mu}m$ and < $20\;{\mu}m$). The correlation relationships between concentrations and size fractions of stream sediments were important to identify the hydro-geochemical behavior of trace elements that flow into Juam reservoir. Stream waters showed four water types (Ca-Mg-$HCO_3$, Ca-Na-$HCO_3$-Cl, Ca-Na-$HCO_3-SO_4$, Ca-Na-$HCO_3$) depending on pollution sources such as coal mine, metal mine, farm-land and dwellings. Concentrations of trace elements increased clearly with the decrease in size fractions of stream sediments. Concentrations of Cu, Pb and Zn increased dramatically in silt size (< $20\;{\mu}m$) fraction, while As had high concentrations in sand size ($2\;mm{\sim}100\;{\mu}m$) fraction in downstream sediments of metal mines. These indicate that Cu, Zn, and Pb moved into Juam reservoir easily in the adsorbed form on silt size grain in sediments, and As was transported as As-bearing mineral facies, resulting in its less chance to reach into Juam reservoir.

Isolation and Morphological Identification of Fresh Water Green Algae from Organic Farming Habitats in Korea (유기농업 생태계로부터 담수 녹조류 분리 및 형태적 동정)

  • This study aimed to isolate and identify freshwater algae from the organic agricultural ecosystems and investigate its biological characteristics to study the possibility of utilizing a biomass freshwater algae in organic farming. In the survey area, average water temperature was $12.4{\sim}28.2^{\circ}C$ and the pH ranges were from 6.1 to 8.5. The solid culture method is more suitable than liquid culture method for isolation of freshwater algae with lower contamination level and higher isolation frequency. A total of 115 strains were isolated from six freshwater algae habitats in nine regions in Korea. BGMM (BG11 Modified Medium) amended with NaNO3 and $KNO_3$ as a nitrogen, and $Na_2CO_3$ as carbon source was designed to isolate and culture freshwater algae. Absorbance of freshwater algae culture has increased dramatically to four days and decreased after eight days after inoculation. CHK008 of the seven isolates showed the highest absorbance in seven days after culturing in BGMM. The optimal pH of BGMM for culturing freshwater algae was pH 6-7. As light intensity increased, growth of freshwater algae increased. Among the five kinds of carbon sources, glucose and galactose promoted good growth of freshwater algae in BGMM. The colony color of purified 16 green algae isolates showed a separation of green, dark and light green, and of them, eleven algae strains showed a strong fluorescent light under fluorescence microscopy. Cell size of the green algae showed a wide range of variation depending on the species. General morphology of the green algae strains was spherical. Chlamydomonas sp. was elliptical, and Chlorella sorokiniana was ellipsoidal and cylindrical. All strains of the green algae except for Chlamydomonas sp. did not have flagella. One isolate of Chlamydomonas sp. and five isolates of C. sorokiniana secreted mucus. Sixteen isolates of 16 green algae were identified as two family and six species, Chlorella vulgalis, C. sorokiniana, C. pyrenoidosa, C. kessleri, C. emersonii, and Chlamydomonas sp. based on their morphological characteristics.
