In vivo protein expression changes in mouse livers treated with dialyzed coffee extract as determined by IP-HPLC

  • Background: Coffee extract has been investigated by many authors, and many minor components of coffee are known, such as polyphenols, diterpenes (kahweol and cafestol), melanoidins, and trigonelline, to have anti-inflammatory, anti-oxidant, anti-angiogenic, anticancer, chemoprotective, and hepatoprotective effects. Therefore, it is necessary to know its pharmacological effect on hepatocytes which show the most active cellular regeneration in body. Methods: In order to determine whether coffee extract has a beneficial effect on the liver, 20 C57BL/6J mice were intraperitoneally injected once with dialyzed coffee extract (DCE)-2.5 (equivalent to 2.5 cups of coffee a day in man), DCE-5, or DCE-10, or normal saline (control), and then followed by histological observation and IP-HPLC (immunoprecipitation high performance liquid chromatography) over 24 h. Results: Mice treated with DCE-2.5 or DCE-5 showed markedly hypertrophic hepatocytes with eosinophilic cytoplasms, while those treated with DCE-10 showed slightly hypertrophic hepatocytes, which were well aligned in hepatic cords with increased sinusoidal spaces. DCE induced the upregulations of cellular proliferation, growth factor/RAS signaling, cellular protection, p53-mediated apoptosis, angiogenesis, and antioxidant and protection-related proteins, and the downregulations of NFkB signaling proteins, inflammatory proteins, and oncogenic proteins in mouse livers. These protein expression changes induced by DCE were usually limited to the range ± 10%, suggesting murine hepatocytes were safely reactive to DCE within the threshold of physiological homeostasis. DCE-2.5 and DCE-5 induced relatively mild dose-dependent changes in protein expressions for cellular regeneration and de novo angiogenesis as compared with non-treated controls, whereas DCE-10 induced fluctuations in protein expressions. Conclusion: These observations suggested that DCE-2.5 and DCE-5 were safer and more beneficial to murine hepatocytes than DCE-10. It was also found that murine hepatocytes treated with DCE showed mild p53-mediated apoptosis, followed by cellular proliferation and growth devoid of fibrosis signaling (as determined by IP-HPLC), and subsequently progressed to rapid cellular regeneration and wound healing in the absence of any inflammatory reaction based on histologic observations.

The Analysis of the Cytokine Expression in Musculoskeletal Tumors (근골격계 종양에서 사이토카인 발현의 분석)

  • The cytokines are the hormone-like proteins, which are produced in the mononuclear cells. They have many roles, such as immune mediators, cell differentiations, angiogenesis. The chemokines have chemotactic effects which control the host immune response. There were few reports about the cytokines associated with musculoskeletal tumors. From late 1980s, the cytokine studies of bone tumors such as osteosarcoma were started, but most studies for benign and malignant musculoskeletal tumors were left to be explored. To evaluate the characteristics of the cytokines in variable musculoskeletal tumors, tissues were obtained from the seven patients who visited the Yeungnam University hospital from February to July 2000. They were lipoma (1 case), parosteal osteoma (1 case), enchondroma (2 cases), pigmented villonodular synovitis (1 case), ganglion (1 case), and metastaic squamous cell carcinoma (1 case). The gene experession of the cytokines were analyzed by RNase protection assay (RPA) and reverse transcription-polymerase chain reaction (RT-PCR). The lipoma and parosteal osteoma expressed MIP-$1{\beta}$, and IP-10 genes. The two enchondromas showed different results, one expressed all of MIP-$1{\alpha}$, MIP-$1{\beta}$ and IP-10 genes but the other expressed none of above. The pigmented villonodular synovitis strongly expressed MIP-$1{\alpha}$ and IP-10 when compared with the other cases. The ganglion did not express all of the chemokines mentioned above. And the metastatic squamous cell carcinoma expressed all of the chemokines and especially IP-10 was highly expressed. Even though this study has only a few cases, these results provide a basis for the cytokine mediating network study in musculoskeletal tumors.

  • Our society is currently exposed to environment of various information that is exchanged real time through networks. Especially regarding medical policy, the government rushes to practice remote medical treatment to improve the quality of medical services for citizens. The remote medical practice requires establishment of medical information based on big data for customized treatment regardless of where patients are. This study suggests establishment of regional medical cluster along with defense and protection cooperation models that in case service availability is harmed, and attacks occur, the attacks can be detected, and proper measures can be taken. For this, the study suggested forming networks with nationwide local government hospitals as regional virtual medical cluster bases by the same medical information system. The study also designed a mutual cooperation security model that can real time cope with IP Spoofing attack that can occur in the medical cluster and DDoS attacks accordingly, so that the limit that sole system and sole security policy have can be overcome.

  • This study compares and analyzes the national competitiveness in IP finance using the diamond model. The main findings of our analysis are as follows. First, assuming Korea's competitiveness in IP finance to be 1, USA with 1.63 has the highest competitiveness of all countries compared, and Israel is 1.49 followed by Japan with 1.37. Second, Korea and other countries compared show large differences in the areas of demand conditions, strategic structure, and competition. Third, Korea is inferior compared to other countries considered in terms of the factors including the protection of investors and IPR, and government policies. Since most of the factors in which Korea shows inferiority can be managed by public policies, to strengthen the competitiveness of IP finance, the government's active support to build industrial foundation are required.

  • Various researches are actively studied to satisfy exponentially increasing the usage of the Internet as well as the diverse service demands. Especially the Optical Internet that delivers several Tbps through a single optical fiber requires the intelligence to satisfy the various types of survivability requirements. In the paper, a novel recovery scheme that satisfies the various restoration demands in IP over WDM networks is proposed. The scheme classifies the restoration services into three classes and applies dedicated protection and shared restoration scheme with different priorities for each class. Also, a configuration scheme for information database to support the scheme is proposed. This scheme satisfies the different degree of restoration demands in terms of restoration time, blocking rate and resource usage. With the scheme, medical data can be transmitted without loss.

  • IPSec(IP Security)은 데이터가 공개적으로 전송되는 네트워크 데이터에 암호화와 인증, 무결성을 제공하기 위해 사용되는 프로토콜이다. IPSec 안에는 여러 프로토콜이 있는데, 그 중에 실제 패킷에 암호화와 인증, 무결성을 추가해 전달하기 위해서는 ESP(Encapsulation Security Payload)라는 프로토콜이 사용된다. 이 ESP(Encapsulation Security Payload)라는 프로토콜이 사용된다. 이 ESP는 패킷을 암호화하기 위해 DES-CBC 모드를 사용하는데, 여기에서 IV(Initialization Vector) 값이 쓰인다. 이 값은 패킷 복호화를 하기 위해 공개적으로 전달이 되기 때문에 중간에 공격자에 의해 공격 당할 위험이 많다. 본 논문에서는 IV 공격을 방지하기 위해 IV의 값을 해쉬 함수를 통해 한번 해슁을 한 다음에, IV 값을 안전하게 전달하는 방법을 제시하고자 한다.

  • 최근 인터넷의 트래픽이 다양해지고, 서비스 질 보장에 대한 사용자의 요구가 증가하며, 가상 사설망(VPN, Virtual Private Network)이나 트래픽 엔지니어링 등 향상된 IP 서비스가 요구되고 있다. 이러한 요구 사항을 충족시켜주기 위하여 IETF에서는 MPLS (Multiprotocol Label Switching) 기술을 표준화하고 있다. 다양한 서비스와 QoS 보장을 가능하게 하는 MPLS 망에서 신뢰성(Reliable) 보장은 또 하나의 중요한 문제이다. 본 논문에서는 이러한 MPLS 망의 신뢰성을 보장하기 위한 기존의 방법들을 설명하고 기존 방법의 문제를 해결하기 위해 선택적인 보호 연결(Protection Path) 설정방법을 제안한다. 본 논문에서 제안하는 보호 연결 설정방법은 과거의 특정 장애 정보를 이용하여 선택적으로 보호 연결을 미리 설정함으로써 빠른 연결 복구를 수행하게 하고 보호 연결 설정으로 인한 자원의 낭비를 최소화 하고자 하는 방법이다.

Molecular Characterization and Variation of the Broad bean wilt virus 2 Isolates Based on Analyses of Complete Genome Sequences

  • The full-genome sequences of fourteen isolates of Broad bean wilt virus 2 (BBWV2), collected from broad bean, pea, spinach, bell pepper and paprika plants in Korea during the years 2006-2012, were determined and analyzed comparatively along with fifteen previously reported BBWV2 genome sequences. Sequence analyses showed that RNA-1 and RNA-2 sequences of BBWV2 Korean isolates consisted of 5950-5956 and 3568-3604 nucleotides, respectively. Full-length genome sequence-based phylogenetic analyses revealed that the BBWV2 Korean isolates could be divided into three major groups comprising GS-I (isolates BB2 and RP7) along with isolate IP, GS-II (isolates BB5, P2, P3 and RP3) along with isolate B935, and GS-III including 16 BBWV2 Korean isolates. Interestingly, GS-III appears to be newly emerged and predominant in Korea. Recombination analyses identified two recombination events in the analyzed BBWV2 population: one in the RNA-1 of isolate K and another one in the RNA-2 of isolate XJ14-3. However, no recombination events were detected in the other 21 Korean isolates. On the other hand, out of 29 BBWV2 isolates, 16 isolates were found to be re-assortants, of which each RNA segment (i.e. RNA1 and RNA2) was originated from different parental isolates. Our findings suggested that reassortment rather than recombination is a major evolutionary force in the genetic diversification of BBWV population in Korea.

  • This paper is aimed at intensively examining the scope of legal protection for fashion designs under the Design Protection Act of Korea. For this purpose, this looked into how the Act defines the concept of design, its requirements, and the prerequisites for acquiring design rights. The study also reviewed statutory interpretations over the judgment of the identity and similarity of designs. For more practical and substantial discussions, this research utilized cases and precedents, which had relevant legal principles. This study also figured out how both the requirements for a design definition - such as merchantability, configuration, visibility, and aesthetics - and the prerequisites for acquiring design rights - like industrial applicability, novelty, and creativity - are interpreted and utilized in actual circumstances. The authors expressed their opinions regarding the criteria of judging the identity and similarity of designs, based on a study of previous cases. Previous rulings show that aesthetics of the exterior design is used as the criteria for determining whether a design is same or similar. So, two designs, which have different specific details, are deemed same or similar, if both designs show similarity in the dominant elements. This is because both designs will produce similar aesthetic qualities. However, if the dominant elements of a design are part of the public domain, and the specific details characterize the design, the latter has to be evaluated in the process. This paper examined scope of legal protection for fashion designs using relevant precedents. The study may serve as academic materials that lead to the establishment of rightful ownership in creative activities.

  • The international government committee is progressing their agreements about intellectual property protection of traditional knowledge(TK), gene resource(GR), folklore(FL) in WIPO. It is in the course of selection with precedence of TK, GR, FL in WlPO, focused on discussions about listing of TK documents, standardization of DB construction, sharing and profit distribution of GR. Their are disagreements between developed countries and underdeveloped countries about intellectual property protection agreements of TK, GR. The developed countries insist on using, the existing intellectual property protections, but the underdeveloped countries ask new ones on character of TK, GR, It causes intangible assets to be valuable trade properties in future world trade. We have to make national plans to face agreements about IP of TK, GR in WIPO with our own TK, GR. We must have a basic attitude about laying down these plans such as raising national economy, considering our TK, GR to be relics of mankind culture. In addition to these, it needs accurate analysis about agreements in WIPO, our TK, GR, scientific mechanical level and capital power.
