  • Urea is detected as an indicator of renal disease in the human body. For these reasons, many biosensors for urea have been developed based on the enzymatic reaction of urea hydrolysis catalyzed by urease. Potentiometric method is applied reversible reaction system. But urea hydrolysis reaction may not has a reversible reaction mechanism in electrode surface. Therefore we applied to voltammtricmethod to obtain a sensitivity curve. The sensitivity of sensors was 34 ${\mu}$A/decade.

Synergistic Effect of Glucoamylase and $\alpha$-Amylase in Enzymatic Hydrolysis of Raw Corn Starch in an Agitated Bead Reaction System (분쇄마찰매체 효소반응계에서 생전분 효소당화를 위한 Glucoamylase와 Alpha-Amylase의 보완작용)

  • The synergistic effect of glucoamylase and a -amylase on the hydrolysis of raw corn starch in an agitated bead reaction system was studied by investigating the changes of sugar profiles, the granular structure, particle size distribution, and X-ray diffraction pattern of residual raw corn starch. The enzymatic hydrolysis of raw corn starch was greatly enhanced by synergistic effect of glucoamylase and $\alpha$ -amylase. Even though the sugar profiles were mainly determined by the mixing ratio of glucoamylase and $\alpha$-amylase; raw starch was mainly converted to glucose directly without accumulation of any significant amount of oligosaccharides. The cavity formation and fragmentation phenomena of raw corn starch granule subjected to enzyme reaction were analyzed by means of SEM and the particle size distribution. The X-ray diffraction pattern of raw starch was not changed at the initial stage of reaction but slightly changed at the late stage of hydrolysis, which may be caused by the preferential degradation of amorphous region by enzymatic reaction, not by the destruction of microcrystalline structure of raw corn starch.

High Char-Yield in AN-AM Copolymer by Acidic Hydrolysis of Homopolyacrylonitrile

  • Acrylonitrile (AN)-acrylamide (AM) copolymers were prepared by nitric acidic hydrolysis of homopolyacrylonitrile. The acrylamino group increased as a function of hydrolysis time, while crystallinity decreased. Differential scanning calorimetry and a thermal gravimetric analysis indicated that the acylamino introduced by acidic hydrolysis effectively enhanced the cyclization reaction at low temperature due to the change of the cyclization reaction mechanism. Char-yield of AN-AM copolymers also gradually increased with increasing hydrolysis time. The maximum char-yield was 49.48% when hydrolized at $23^{\circ}C$ in 65% nitric acid solution for 18 h, which was 30% higher than that of non-acidic hydrolysis of homopolyacrylonitrile. Simulation of the practical process also showed an increase of char yields, where the char yields were 55.43% and 62.60% for homopolyacrylonitrile and copolyacrylonitrile, respectively, with a hydrolysis time of 13 h.

  • The hydrolysis-precipitation reaction of mixed aluminum salt solutions of aluminum sulfate, aluminum nitrate, and urea has been investigated to obtain narrow-sized and unagglomerated fine spherical precipitates of aluminum hydroxide required for coating core particles. The hydrolysis-precipitatin reaction could be controlled to be appropriate to coating processes by usign urea as a pH control-agent. As the concetration of total Al3+ ion and the molar ratio of SO42-/Al3+ in starting solutions became smaller and also as the vol. ratio of water/solution for hydrolyzing mixed aluminum salt solution became larger, the morphology of precipitates tended to be more unagglomerated and spherical, while their size(0.5longrightarrow0.05${\mu}{\textrm}{m}$) to be smaller. The optimum hydrolysis condition for coating processes was to hydrolyze the mixed aluminum salt solution, in which the molar ratio of SO42-/Al3+ was 0.75, while the amount of water corresponding to the vol. ratio of water/solution of 15. The precipitate was the aluminum hydroxide which sulfate ions were strongly adsorbed on and the maximum yield in the hydrolysis-precipitation reaction was about 20%.

Method of Using Acid Hydrolysis to Increase the Efficacy of Decreasing Alcohol Concentration from Hovenia dulcis Extract (헛개열매 추출물의 산 가수분해에 의한 알코올 분해 효능 증대)

  • This work was a method that used an acid hydrolysis for increasing the efficacy of decreasing alcohol concentration from Hovenia dulcis extract. For acid hydrolysis, the best pH was 2.0 to obtain a maximum alcohol dehydrogenase activity at fixed reaction temperature and time. At pH 2.0, reaction temperature $80^{\circ}C$ and reaction time 4 hr gave the highest activity which was $124\%$ of control. The bioactive compound, (+)-dihydromyricetin, content increased to $30\%$ after acid hydrolysis. This is very simple and efficient method to increase the efficacy of decreasing alcohol concentration from Hovenia dulcis extract.

  • Hydrolysis reactions of S-phenyl-S-vinyl-N-p-tosylsulfilimine (VSI) and its derivatives at various pH have been investigated kinetically. The hydrolysis reactions produced phenylvinylsulfoxide and p-toluene sulfonamide as the products. The reactions are first order and Hammett ρ values for pH 1.0, 6.0, and 11.0 are 0.82, 0.45, and 0.57, respectively. This reaction is not catalyzed by general base. The plot of k vs pH shows that there are three different regions of the rate constants $(k_t)$ in the profile.; At pH < 2 and pH > 10, the rate constants are directly proportional to the concentrations of hydronium and hydroxide ion catalyzed reactions, respectively. The rate constant remains nearly the same at 2 < pH < 10. On the bases of these results, the plausible hydrolysis mechanism and a rate equation have been proposed: At pH < 2.0, the reaction proceeds via the addition of water molecule to sulfur after protonation at the nitrogen atom of the sulfilimine, whereas at pH > 10.0, the reaction proceeds by the addition of hydroxide ion to sulfur directly. In the range of pH 2.0-10.0, the addition of water to sulfur of sulfilimine appears to be the rate controlling step.

Enzymatic Hydrolysis Optimization of a Snow Crab Processing By-product (홍게 가공부산물의 효소적 단백질 가수분해 최적화)

  • The objectives of this study were to evaluate a protease suitable for the enzymatic hydrolysis of a snow crab processing by-product (SPB) and to optimize the hydrolysis conditions using response surface methodology (RSM). The SPB was hydrolyzed at $50^{\circ}C$ and pH 7.0-7.2 to obtain various degree of hydrolysis (DH) using Flavourzyme at an enzyme/substrate (E/S) ratio of 3.0%. The reaction progress curve exhibited an initial fast reaction rate followed by a slowing of the rate. The DH was increased to 30% at 90 min with a final DH 32 to 36%. A central composite experimental design having three independent variables (reaction temperature, reaction time, and E/S ratio) with five levels was used to optimize the enzymatic hydrolysis conditions. Based on the DH data, the optimum reaction conditions for the enzymatic hydrolysis of the SPB were a temperature of $51.8^{\circ}C$, reaction time of 4 hr 45 min, and an E/S ratio of 3.8%. It was demonstrated that the enzymatic hydrolysate of SPB could be used as a flavoring agent or a source of precursors for the production of reaction flavors.

A STUDY ON AMIDI HYDROLYSIS CATALYZED BY MITAL COMPlEXES (금속착물로 아미드 가수분해 촉매화에 관한 연구)

  • This study is involved to develop new catalysts to decompose plastics, detergents and surfactants containing synthetic peptide bonds. As the first year research, the catalytic-hydrolysis of amide bond in copper complex was accomplished. The hydrolysis reaction in aqueous solution was monitored by UV/VIS spectroscopy. As the pH of the solution Is increased and the temperature is raised, the reaction rate increases. The reaction rate is observed as the first order kinetic behavior for the copper complex. The metal catalyzed hydrolysis mechanism is proposed cia metal-hydroxide in the pH region of 5.5 to 6.3. The results of characterization of the catalytic reaction mechanism can be applied to develop new catalysts for peptide bond degradation in further research.

  • The parameters of silanol formation reaction of organosilane including solvent type, solution concentration, pH and hydrolysis time influence the adhesion property of 2 layer flexible copper clad laminate (FCCL). Especially, the hydrolysis reaction time of silane coupling agent affects the formation of the silanol groups and their self-condensation to generate oilgomeric structure to enhance the surface treatment as an adhesive promoter. In our study, we prepared the binary silane coupling agents to control hydrolysis reaction rate and surface energy after treatment of silane coupling agents for increasing the adhesive property between a copper layer and a polyimide layer. The surface morphology of rolled copper foil, as a function of the contents of the coated binary silane coupling agent, was fully characterized. As fabricated 2-layer FCCL, we observed that adhesive properties were changed by hydrolysis rate and surface energy.

Catalytic Effect of $MO_4^{2-_4}$ (M=Cr, Mo and W) on Hydrolyses of Carbon and Phosphorus Esters

  • Second-order rate constants have been measured spectrophotometrically for the hydrolysis of p-nitrophenyl ac-etate (PNPA) and p-nitrophenyl diphenylphosphinate (PNPDPP) with MO42- (M = Cr, Mo and W) in phos-phate buffer (pH = 8.00) at 35.0 $^{\circ}C.$ Thes e MO42- species exhibit large catalytic effect in the hydrolysis of PNPA and PNPDPP except WO42- in the reaction with PNPA. The catalytic effect of these MO42- species has been observed to be much more significantin the hydrolysis of PNPDPP than in the hydrolysis of PNPA. Since the smallest CrO42-would be most highly solvated by H2O molecules, CrO42- is expected to exhibit the least catalytic effect, if solvation effect is the most important factor. However, in fact, CrO42- shows the highest cat-alytic effect toward PNPA, indicating that solvation effect is not solely responsible for the catalytic effect. The most basic CrO42- shows the highest catalytic effect, while the least basic WO42- is least reactive toward PNPA, indicating that the basicity of MO4 2- might bean important factor. However, in the hydrolysis of PNPDPP, no correlation is observed between the basicity and catalytic effect, suggesting thatbasicity alone can not be re-sponsible for the catalytic effect shown by the MO42- species. Formation of a chelate is suggested to be respon-sible for the high catalytic effect of MO42- in the hydrolysis reaction of PNPA and PNPDPP. The formation of chelate is considered to be more suitable for the reaction with PNPDPP than with PNPA based on the larger catalytic effect observed in the reaction with PNPDPP than with PNPA.
