Neuronal growth regulator 1 (NEGR1) is a GPI-anchored membrane protein that is involved in neural cell adhesion and communication. Multiple genome wide association studies have found that NEGR1 is a generic risk factor for multiple human diseases, including obesity, autism, and depression. Recently, we reported that Negr1-/- mice showed a highly increased fat mass and affective behavior. In the present study, we identified Na/K-ATPase, beta1-subunit (ATP1B1) as an NEGR1 binding partner by yeast two-hybrid screening. NEGR1 and ATP1B1 were found to form a relatively stable complex in cells, at least partially co-localizing in membrane lipid rafts. We found that NEGR1 binds with ATP1B1 at its C-terminus, away from the binding site for the alpha subunit, and may contribute to intercellular interactions. Collectively, we report ATP1B1 as a novel NEGR1-interacting protein, which may help deciphering molecular networks underlying NEGR1-associated human diseases.

Peptide nucleic acids (PNAs) that bind to complementary nucleic acid sequences with extraordinarily high affinity and sequence specificity can be used as antisense oligonucleotides against microRNAs, namely antagomir PNAs. However, methods for efficient cellular delivery must be developed for effective use of PNAs as therapeutic agents. Here, we demonstrate that antagomir PNAs can be delivered to hepatic cells by complementary DNA oligonucleotide and cationic liposomes containing galactosylated ceramide and a novel cationic lipid, DMKE (O,O'-dimyristyl-N-lysyl glutamate), through glycoprotein-mediated endocytosis. An antagomir PNA was designed to target miR-122, which is required for translation of the hepatitis C virus (HCV) genome in hepatocytes, and was hybridized to a DNA oligonucleotide for complexation with cationic liposome. The PNA-DNA hybrid molecules were efficiently internalized into hepatic cells by complexing with the galactosylated cationic liposome in vitro. Galactosylation of liposome significantly enhanced both lipoplex cell binding and PNA delivery to the hepatic cells. After 4-h incubation with galactosylated lipoplexes, PNAs were efficiently delivered into hepatic cells and HCV genome translation was suppressed more than 70% through sequestration of miR-122 in cytoplasm. PNAs were readily released from the PNA-DNA hybrid in the low pH environment of the endosome. The present study indicates that transfection of PNA-DNA hybrid molecules using galactosylated cationic liposomes can be used as an efficient non-viral carrier for antagomir PNAs targeted to hepatocytes.
This study investigated the effects of dietary monobasic potassium phosphate (MKP) on the growth of the far eastern catfish Silurus asotus and four leafy vegetables in a hybrid biofloc technology aquaponic system. To an experimental diet containing 45% protein and 7% lipid, 1, 2, 3 or 4% MKP was added and was designated as MKP1, MKP2, MKP3, and MKP4, respectively. The optimum MKP levels were determined for the growth of fish and four leafy vegetables over 10 weeks. After the 10-week feeding trial, weight gain, feed efficiency, specific growth rate and protein efficiency ratio were higher in the fish groups fed MKP2 and MKP3 than in the other groups (P<0.05). The growth of the four leafy vegetables was also higher in the fish groups fed MKP2 and MKP3. Water quality [dissolved oxygen, pH, water temperature, electrical conductivity, turbidity, total ammonia nitrogen (TAN), $NO_2-N$, $NO_3-N$ and $PO_4-P$] was measured six times a week using a portable water quality meter and reagent measurements. The TAN (4.58-20.40 mg/L), $NO_3-N$ (24.12-52.40 mg/L) and $PO_4-P$ (20.38-48.48 mg/L) levels increased with time, while the $NO_2-N$ level remained below 0.1 mg/L throughout the study.
The effects of dietary mannan oligosaccharides, Lactobacillus plantarum, Bacillus subtilis, and Bacillus licheniformis supplementation on hybrid grouper Epinephelus akaara ♀×Epinephelus lanceolatus ♂ were evaluated. The fish were fed a basal diet and five other diets consisting of 0.6% mannan oligosaccharides, L. plantarum, B. subtilis, and B. licheniformis and mixture of each 0.15% prebiotic and all the probiotics (designated as MOS, LP, BS, BL, and SYN) for 56 days. Growth performance and feed utilization showed no significant differences among all experimental groups. Lipid level of whole-body was significantly high in MOS and BL groups. Plasma aspartate aminotransferase was significantly low in BL and SYN groups. Nitro-blue tetrazolium, lysozyme and anti-protease, and glutathione peroxidase in BS, SYN, and all probiotic groups, respectively, were significantly high. Intestinal Vibrio bacteria was significantly low in all probiotic and SYN groups. Gene expression of interleukin-1β and interleukin-10 in SYN group; transforming growth factor β2 in MOS and BS groups, toll-like receptor 2-2 in BS and BL groups; and C-type lectin in MOS, LP and SYN groups were significantly upregulated. Our findings indicate that mannan oligosaccharides, L. plantarum, B. subtilis, and B. licheniformis could improve innate immunity, antioxidant capacity, anti-inflammation, and intestinal microbiota of hybrid grouper.

최근 고기능성 화장품이 출시되면서 기능성 원료가 빛, 열, 산소에 매우 불안정하여 다양한 방법으로 안정성을 높이려고 연구되어 지고 있다. 특히, 카테킨은 주름 개선에 탁월한 원료이지만 빛, 열, 산소에 매우 불안정하다. 본 연구에서는 카테킨을 3Dimension화 하여 안정성 및 피부 침투를 높였다. 1 dimension으로 sol-gel method로 실리카를 다공성으로 만들어서 다공성 부문에 카테킨을 흡착시킨다. 2 dimension으로 다공성 실리카에 흡착디어진 카테킨을 non-phospholipid 베지클을 이용하여 solid lipid nanoparticle(SLN)을 만든다. 마지막으로 3dimension은 SLN되어진 카테킨을 skin lipid matrix를 이용하여 lameller phase self organization시킨다. 3 Dimension-카테킨은 일반적인 리포좀에 비해 빛과 열에 대한 color 안정성을 chromameter로 측정한 결과 5-10배 더 안정하였으며, HPLC 분석 결과 카테킨의 생존율이 3-5배 더 개선되었다. 또한 penetration effect를 측정한 결과 일반 리포좀보다 더 깊게 침투되었다. Wrinkle reduction effect를 한달 후에 측정한 결과 일반 리포좀보다 주름이 현저하게 감소되었다. 이러한 여러 가지 실험을 위해서 Laser light scattering system, cryo-SEM, chroma meter, HPLC, image analyzer, microfludizer 등을 사용하였다.
We have cloned an LTP gene (PoLTP1) from poplar (Populus alba ${\times}$ P. tremula var. glandulosa) suspension cells and examined changes in its expression levels in response to various stresses and ABA treatment. The full-length PoLTP1 cDNA clone encodes a polypeptide of 116 amino acids with typical characteristics of LTPs, notably a conserved arrangement of cysteine residues. Southern blot analysis indicate that two or three copies of the PoLTP1 are present in the genome of the investigated hybrid poplar. In addition, northern analysis of samples from soil-grown plants indicate that PoLTP1 is tissue-specifically expressed in the leaves and flowers. The gene is significantly up-regulated by treatment with mannitol, NaCl and ABA, but not by either cold or wounding. These results indicate that PoLTP1 is involved in osmotic stress responses in poplar plants and suspension cells.
Objective: The aim of this study was to compare male and female geese of two contrasting genotypes in terms of fatty acid composition, indexes related to human health, lipid metabolism and oxidative stability of the meat. Methods: The experiment was carried out on total of 120 geese of two different genotypes; the native breed Czech goose (CG) and commercial hybrid Novohradska goose (NG). One-d-old goslings were divided into 4 groups according to genotype and sex, and 8 birds from each group were slaughtered at 8 weeks of age. Results: The effects of the interactions between genotype and sex were observed on growth performance and carcass traits. Final body weight (p<0.001), daily weight gain (p<0.001), daily feed intake (p<0.001), slaughter weight (p<0.001), and cold carcass weight (p<0.001) were highest in NG males and lowest in CG females. The meat fatty acid composition results showed effects of both genotype and sex on the total n-6 and the total polyunsaturated fatty acid (PUFA) content, as well as the PUFA n-6/PUFA n-3 ratio. Regarding genotype, the total n-6, the total PUFA content and the PUFA n-6/PUFA n-3 ratio were higher in CG, and higher values were found in females. In terms of the lipid metabolism, ${\Delta}^5-{\Delta}^6$ desaturase (p = 0.006) was higher in males. The meat oxidative stability results revealed an interaction between genotype, sex and storage time (p<0.001). The highest (13.85 mg/kg) malondialdehyde content was measured in the meat of CG females after 5 days of storage and was presumably related to a higher PUFA content. Conclusion: NG had a relatively higher growth rate and meat oxidative stability, whereas the advantage of CG meat is its favourable fatty acid profile characterized by a higher PUFA content.
본 연구는 자색옥수수 색소 1호 포엽과 속대 추출물의 항비만 활성을 검정하고자 지방분해효소 저해활성을 평가하고 3T3-L1 지방전구세포에서 지방분화억제 효과를 검정하고자 수행되었다. Pancreatic lipase 저해 활성 결과, 색소 1호 포엽 및 속대 추출물의 100, 500, $1,000{\mu}g/mL$ 농도처리구에서 양성대조군인 orlistat 보다 높은 저해 활성을 나타내었다. 3T3-L1 지방전구세포를 배양하여 색소 1호 포엽 및 속대 추출물의 세포독성 평가를 수행한 결과, 추출물은 모든 처리농도에서 세포 생존율에 영향을 미치지 않은 것으로 확인되었다. 분화된 3T3-L1 지방전구세포에서 색소 1호 포엽과 속대 추출물을 처리하지 않고 분화시킨 대조군은 lipid droplet의 형성이 활발하게 유발되었으나 색소 1호 포엽 및 속대 추출물의 처리에 의해 농도 의존적으로 lipid droplet의 형성이 억제되는 것으로 나타났다. Real-time PCR과 Western blot을 실시하여 $PPAR{\gamma}$와 $C/EBP{\alpha}$ 유전자 및 단백질 발현량을 측정한 결과, 추출물을 처리하지 않고 분화시킨 대조군에서는 $PPAR{\gamma}$와 $C/EBP{\alpha}$의 유전자 및 단백질 발현이 증가하였으며, 추출물 처리에 의해 $PPAR{\gamma}$와 $C/EBP{\alpha}$의 유전자 및 단백질 발현이 유의적으로 감소하였다. 본 연구 결과는 색소 1호 포엽 및 속대 추출물이 pancreatic lipase 활성 및 지방전구 세포의 분화를 억제시킴으로써 항비만 활성 기능성 물질로의 활용 가능성이 높음을 시사한다.

Objective: Reduced crude protein (CP) diets offer potential benefits such as optimized feed efficiency, reduced expenses, and lower environmental impact. The objective of this study was to evaluate black soldier fly larvae (BSFL) meal on a low-protein diet for duck performance, blood biochemical, intestinal morphology, gastrointestinal development, and litter. Methods: The experiment was conducted for 42 days. A total of 210-day-old male hybrid ducklings (5 replicate pens, 7 ducks per pen) were randomly assigned to 6 dietary treatments (3×2 factorial arrangements) in randomized design. The factors were CP level (18%, 16%, 14%) and protein source feed soybean meals (SBM), black soldier fly larvae meals (BSFLM). Results: Reduced dietary CP levels significantly decreased growth performance, feed intake, the percentage of nitrogen, pH (p<0.05), and tended to suppress ammonia in litter (p = 0.088); increased lipid concentration; and enhanced relative weight of gastrointestinal tracts (p<0.05). In addition, dietary BSFL as a source of protein feed significantly increased lipid concentration and impacted lowering villus height and crypt depth on jejunum (p<0.05). Conclusion: In conclusion, the use of BSFLM in a low-protein diet was found to have a detrimental effect on growth performance. However, the reduction of 2% CP levels in SBM did not have a significant impact on growth performance but decreased nitrogen and ammonia concentrations.
연구의 목적은 oleic acid로 지방생성이 유도된 HepG2 세포에서 자색옥수수 색소 1호 포엽 및 속대 추출물이 간세포 내 지방생성에 미치는 영향을 구명하는 것이다. 자색옥수수 색소 1호 포엽 및 속대 추출물에 의한 HepG2 세포 내 지방 축적의 변화를 확인하기 위하여 배양된 세포에 oleic acid로 지방 축적을 유도하고 추출물에 의한 중성지방생성 억제 효과를 측정하였으며 추출물을 처리하지 않은 대조군과 추출물을 처리한 실험군의 지방합성 및 축적에 관련된 유전자와 단백질 발현량을 RT-PCR과 Western blot을 통하여 측정하였다. Oil Red O와 Nile Red 염색을 통하여 추출물의 처리로 HepG2 세포 내 중성지방 축적이 억제된 것을 확인하였다. RT-PCR에 의하여 mRNA 발현량을 측정한 결과, oleic acid에 의하여 지방 생성이 유도된 대조군에 비하여 모든 추출물 처리군의 SREBP-1c와 SREBP-1a 유전자 발현량이 유의적으로 감소되었다. Western blot을 실시하여 p-AMPK, p-SREBP1, PPARα, FAS 단백질의 발현량을 측정한 결과, 간에서 지질대사에 관여하는 주요 인자인 SREBP1 단백질의 발현은 추출물의 처리 농도에 따라 유의하게 감소하였으며 지방산의 생합성 경로에 관여하는 주요 효소인 FAS의 단백질 발현향은 모든 처리 농도에서 현저하게 감소된 것이 확인되었다. 본 연구결과는 자색옥수수 색소 1호 포엽 및 속대 추출물이 간세포 내에서 중성지방의 축적을 억제시키고 지질 합성에 관련된 유전자 및 단백질의 발현을 억제시킴으로써 간 세포 내 지질 축적을 완화할 수 있는 기능성 소재로의 활용가치가 높다고 판단된다.
