Longevity regulation by NMD-mediated mRNA quality control

  • Proper maintenance of biological components is crucial for longevity and healthy aging. Although the role of homeostatic maintenance systems for DNA and protein in longevity is established, it remains largely unknown for RNA. In our recent work, we show that nonsense-mediated mRNA decay (NMD) promotes longevity in the roundworm C. elegans by enhancing RNA quality control. We find that the activity of NMD decreases during aging, raising the possibility that RNA quality declines in old animals. We then show that key components of NMD complex are required for prolonged lifespan in C. elegans. In addition, animals with reduced insulin/insulin-like growth factor-1 (IGF-1) signaling (IIS), a representative longevity model, display increased NMD activity. Thus, up-regulation of NMD appears to play crucial roles in longevity conferred by reduced IIS via enhancing mRNA quality control. As both IIS and NMD pathways are evolutionarily conserved, mammals including humans may be equipped with similar RNA quality control systems to achieve longevity.

  • Carbofuran(CF) is one the most widely used carbamate pesticides in the world applied for insect and nematode control. Due to its widespred use in agriculture and households, contamination of food, water, and air has become serious, and consequently adverse health effects are inevitable in humans, animals, wildlife and fish, it has reported that CF alone or in combination with other carbamate insecticides influences the level of reproductive and metabolic hormones such as thyroxine and corticosterone, and results in impairment of endocrine. 동물 실험을 적용한 국내 간호 연구 동향 분석 (Use of Animals for Nursing Research in Korea)

  • Purpose: The purpose of this study was to analyze the trends in Korea of nursing research using animals. Methods: 45 articles were reviewed with descriptive statistics from 2000 to 2015. Results: Nearly 50% of the studies were supported by research funds. Most of the animal species in the studies were rats (81.1%) and mice (15.6%). The major animal models for human disease were cerebrovascular diseases (24.4%), skin problems (17.8%), and nerve injuries (15.5%). Immunohistochemistry (24.3%) was mainly used as an experimental technique. The main intervention and treatments used in the studies were chemical treatments, exercise and surgery. Conclusion: In this review, relevant results were described to identify the trends of nursing research using animal experimentation in Korea. Expansion of various nursing research using animal models can be required for gaining knowledge applicable to the same conditions in humans.

  • 인공생명 연구는 인공생명을 이용한 예술의 발전에도 영향을 주었다. 인공생명예술은 초기의 미시생물에 대한 탐구에서 출발하여 관람자의 선택을 통해 창발성을 일으키는 작업으로 발전해 왔다. 또한 실제 동물에 대한 관심은 사이버 펫과 같은 인공 동물의 제작으로 이어지고, 인간에 대한 관심은 아바타의 창조로 이어진다. 본고는 사이버 펫과 아바타를 활용한 나오꼬 토사 작품의 비가시적 감성에 대한 분석을 중심으로 인공생명 예술의 창발적 변화가 인터랙티브 아트의 발전에 어떠한 영향을 미쳤는지를 입증하고자 한다.

  • Phytoestrogens are naturally occurring compounds derived from plants. Structurally, some phytoestrogens resemble endogenous estrogen of humans and animals. Phytoestrogens exhibit estrogen agonist/antagonist properties and have many biological effects such as prevention of hormone-dependent breast cancer, anti-oxidative activity, inhibition of tyrosine kinase activities and inhibition of angiogenesis. In this study we investigated whether biochanin A, a phytoestrogen, and its metabolites such genistein, p-ethylphenol and phenolic aic affect IL-4 production in EL-4 thymoma cell-line and primary lymph node cells.   • Zinc plays a key role in genetic expression, cell division, and growth and is essential for the function of more than 200 enzymes; effects of zinc deficiency induce many syndromes, including abnormal visual adaptation. The pigment epithelium (EP) contains high concentrations of zinc in humans and in animals and it participates in threshold elevation, visual sensitivity increment, and acceleration of rhodopsin regeration during visual adaptation. The origin of c-wave of electroretinogram(ERG) is not only pigment epithelium as shown in present research, but also other cell layers, perhaps the photoreceptors. We propose zinc as a candidate for an internal messenger which participates in signal amplification.

Development of a Blocking ELISA for Measuring Rabies Virus-specific Antibodies in Animals

  • Rabies virus (RABV)-specific antibodies in animals and humans are measured using standard methods such as fluorescent antibody virus neutralization (FAVN) tests and rapid fluorescent focus inhibition tests, which are based on cell culture systems. An alternative assay that is safe and easy to perform is required for rapid sero-surveillance following mass vaccination of animals. Two purified monoclonal antibodies (4G36 and B2H17) against RABV were selected as capture and detection antibodies, respectively. A genetically modified RABV, the ERAGS strain, was propagated and concentrated by polyethylene glycol precipitation. Optimal conditions for the RABV antigen, antibodies, and serum dilution for a blocking enzymelinked immune sorbent assay (B-ELISA) were established. We evaluated the sensitivity, specificity, and accuracy of the B-ELISA using serum samples from 138 dogs, 71 raccoon dogs, and 25 cats. The B-ELISA showed a diagnostic sensitivity of 95.8-96.3%, specificity of 91.3-100%, and accuracy of 96.0-97.2% compared to the FAVN test. These results suggest that the B-ELISA is useful for sero-surveillance of RABV in dogs, raccoon dogs, and cats.

A Safety Evaluation of Genetically Modified Feedstuffs for Livestock Production; the Fate of Transgenic DNA and Proteins

  • Two genetic constructs used to confer improved agronomic characteristics, namely herbicide tolerance (HT) in maize and soyabean and insect resistance (Bt) in maize, are considered in respect of feeding to farm livestock, animal performance and the nutritional value and safety of animal products. A review of nucleic acid (DNA) and protein digestion in farm livestock concludes that the frequency of intact transgenic DNA and proteins of GM and non-GM crops being absorbed is minimal/non existent, although there is some evidence of the presence of short fragments of rubisco DNA of non-GM soya in animal tissues. It has been established that feed processing (especially heat) prior to feeding causes significant disruption of plant DNA. Studies with ruminant and non-ruminant farm livestock offered GM feeds demonstrated that animal performance and product composition are unaffected and that there is no evidence of transgenic DNA or proteins of current GM in the products of animals consuming such feeds. On this evidence, current HT and Bt constructs represent no threat to the health of animals, or humans consuming the products of such animals. However as new GM constructs become available it will be necessary to subject these to rigorous evaluation.

Environmental Sustainability and Social Desirability Issues in Pig Feeding

  • Feeding pigs used to be a means of managing domestic resources that may otherwise have been wasted into valuable animal protein. Feeding pigs thus was a form of husbandry. Following recent rapid industrial development, pig rearing has changed from extensive to intensive, but this transformation has been associated with major concerns. The concentration of large amounts of pig manure in small arrears is environmentally hazardous. Moreover, high densities of animals in intensive production systems also impose a health threat for both animals and humans. Furthermore, the use of growth promoters and preventive medicines for higher production efficiencies, such as in-feed antibiotics, also induces microbial resistance thus affects human therapeutics. In addition, consumers are questioning the ethics of treating animals in intensive production systems. Animal welfare, environmental and bio-safe issues are re-shaping the nature of pig production systems. Feeding pigs thus involves not only the consideration of economic traits, but also welfare traits and environmental traits. Thus, a focus on technological feasibility, environmental sustainability and social desirability is essential for successful feeding operations. Feeding pigs now involves multiple projects with different sustainability goals, but goal conflicts exist since no pattern or scenario can fulfill all sustainability goals and the disagreements are complicated by reduced or even no use of in-feed antibiotics. Thus it is difficult to feed pigs in a manner that meets all goals of high quality, safe product, eco- and bio-sustainability, animal welfare and profit. A sustainable pig production system thus requires a prioritization of goals based on understanding among consumers, society and producers and needs to view from both a local and global perspective.
