어성초(魚腥草)가 자궁근종 세포의 성장억제와 세포자멸사에 미치는 영향 (The Effect of Houttuynia cordata thunberg(魚腥草) on the inhibition of growth of leiomyomas and apoptosis)

  • Purpose: Uterine leiomyomas (fibroids) are common estrogen-dependent uterine tumors. Houttuynia cordata thunberg has cancer-preventing properties and often used in Chinese medicine. In the present study we used Houttuynia cordata thunberg to determine its effect on cell proliferation and apoptosis in human uterine leiomyoma cells. Methods: Primary cultured human uterine leiomyoma cells were treated with Houttuynia cordata thunberg. Cell viability analysis was analyzed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTS) assay and Flow cytometry was performed to ascertain the effects Houttuynia cordata thunberg. Expression of cell cycle related proteins and apoptosis related proteins were evaluated by Western blot analysis. Results: Cell viability was significantly influenced by Houttuynia cordata thunberg treatment in a dose-dependent manner in leiomyoma cells compare to normal myometrial cells. Flow cytometric analysis showed that Houttuynia cordata thunberg induced Sub G1 arrest. DNA fragmentation assay was carried out and apoptosis was detected. Activation of caspase-3, down-regulation of Bcl-2, with concomitant increase in p21 was observed. Houttuynia cordata thunberg treatment of uterine leiomyoma cellsresulted in a concentration-dependent cell death induced via the caspase dependent mechanism. Conclusion: These results suggest that Houttuynia cordata thunberg treatment in uterine leiomyoma cells leads to growth inhibition and induced apoptosis. These results suggest that Houttuynia cordata thunberg will be a promising agent for use in therapeutics agents against human uterine endometrial cancer.

어성초 추출물의 세포독성과 향균효과 (IV) (Studies on the cytotoxicity and Antimicrobial Effects of the Extract of Houttuynia cordata (IV))

  • This study was carried out to evaluate cytotoxic effects of Houttuynia cordata Thunberg extracts on murine leukemia tumor cell lines. Disruptions in cell organelles were determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazol iumbromide (MMT) assay. The comparison of $IC_{50}$ values of Houttuynia cordata Thunberg extracts on $L1210,\;P388D_1$ and Vero cell lines showed that the methanol extract of Houttuynia cordata Thunberg indicated the most antitumor activity in the MTT assay. In order to develop a antimicrobial agent, dried Houttuynia cordata Thunberg was extracted with several solvents, and then antimicrobial activity was investigated. The minimal inhibitory concentration (MIC) of the extracted substance against microorganisms were also examined. Antimicrobial activity of amocla and ketoconazole as references was compared to those of other solvent extracts such as $H_2O$, n-hexane, chloroform, ethyl acetate ethanol and methanol. The antimicrobial activity of all extracts from the sample had growth inhibition activity against gram-negative bacteria, yam-positive bacteria and fungi $(MIC,\;>\;200\;{\mu} g/ml)$. These results suggest that the methanol soluble extract of Houttuynia cordata Thunberg may be a valuable choice for the studies on the treaeent of murine leukemia tumor cell lines and antimicrobial agents.

괴화, 어성초, 익모초에서 효소 분해에 의한 항산화 활성 변화 연구 (Study on the Change of Antioxidant Activity by Enzymatic Hydrolysis in Sophora japonica Linne, Houttuynia cordata Thunberg, Leonurus japonicus Houttuyn)

  • Objectives: The enzymatic hydrolysis is one of the processing methods that improve its effectiveness on medicinal herbs. In this research, changes in ingredients and activity by enzymatic hydrolysis were studied. Methods: For this study, a carbohydrate hydrolase such as viscozyme, which converts glycosides to aglycone, was applied to induce constituent changes in Sophora japonica Linne, Houttuynia cordata Thunberg and Leonurus japonicus Houttuyn. Changes in antioxidant activity were measured using the 1,1-diphenyl-2-picrylhydrazl (DPPH) method, and changes in ingredients were analyzed by high performance liquid chromatography. Results: As a result of enzymatic hydrolysis, the content of quercetin was increased from 1.26 mg/g to 29.66 mg/g in Sophora japonica Linne, from 0 mg/g to 0.66 mg/g in Houttuynia cordata Thunberg and from 0.43 mg/g to 0.71 mg/g in Leonurus japonicus Houttuyn. As a result of the antioxidant experimentation, the IC50 of Sophora japonica Linne decreased from 5 ug/ml (MeOH extract) and 9.1 ug/ml (EtOAc fraction) to 3.0 ug/ml, Houttuynia cordata Thunberg decreased from 15.6 ug/ml (MeOH extract) and 13.6 ug/ml (EtOAc fraction) to 11.2 ug/ml, and Leonurus japonicus Houttuyn decreased from 14.4 ug/ml (MeOH extract) and 12.6 ug/ml (EtOAc fraction) to 10.2 ug/ml. Conclusion: In conclusion, it was confirmed that glycoside rutin contained in the three medicinal herbs was changed to quercetin which is the aglycone, by the enzymatic hydrolysis using viscozyme. In terms of antioxidant activity, Sophora japonica Linne showed a significant antioxidant activity value that closes to the control group butylated hydroxyanisole. Houttuynia cordata Thunberg and Leonurus japonicus Houttuyn showed a minor increase in antioxidant activity.

전립선 비대증에 대한 Houttuynia cordata Thunberg 에탄올 추출물의 최적용량 결정 (Determination of Optimal Dosage of Ethanol Extract of Houttuynia cordata Thunberg Against Benign Prostatic Hyperplasia)

  • Houttuynia cordata Thunberg는 전통의학에서 다양한 약리학적 작용에 대해 연구되어 왔다. 본 연구에서, 거세 및 테스토스테론 프로피오네이트(TP) 주입에 의해 유도된 양성 전립선 비대증(BPH) 모델에 대한 Houttuynia cordata 에탄올 추출물(HCE)의 영향을 조사했다. 총 30마리의 쥐를 6개의 그룹으로 나누었다. 한 그룹은 정상 대조군으로 사용되었고 다른 그룹은 거세 후 14일 동안 복강내 TP를 주입하여 BPH를 유도하였다. 양성 대조군은 BPH 모델에 매일 피나스테라이드(5 mg/kg)를 투여했다. 피나스테라이드 대신 HCE (0.5, 1, 2mg/kg)를 투여한 군은 실험 군으로서 대조군과 비교하였다. 체중 100 g 기준으로 전립선 무게에 대한 통계적 유의성은 없었다. ELISA 검사로 5-α 환원효소 및 디하이드록시 테스토스테론(DHT) 농도를 측정하였으며, 모든 실험군에서 유의미한 감소가 있었다. 5-α 환원효소의 경우 HCE (0.5 mg/kg) 군이 가장 낮았고, DHT는 HCE (2 mg/kg) 군에서 가장 낮은 수치를 보였다. 전립선에 대한 조직병리학적 관찰에서 대조군과 HCE (2 mg/kg) 투여 군은 정상 세포 형태를 보였고 세포팽창이 없었다. 그러나 음성 대조군과 HCE (1 mg/kg) 군에서는 세포가 부풀어 오르고 세포 사이의 간격이 좁아졌다. 특히 HCE (0.5 mg/kg) 군에서는 일부 세포가 터지는 현상이 나타났다. 따라서 2 mg/kg 이상의 HCE를 투여하는 것이 BPH의 보호 효과에 적합할 것이다.

Houttuynia cordata Thunberg exhibits anti-tumorigenic activity in human gastric cancer cells

  • Objectives : Gastric cancer is a leading cause of cancer-related deaths, worldwide. Houttuynia cordata Thunberg (H. cordata) has been used as a medicinal plants and it has an anti-cancer activity in human colorectal cancer and leukemic cancer. However, the potential anti-cancer activity and mechanisms of H. cordata for human gastric cancer cells have not been tested so far. Thus, this study examined the biological effects of H. cordata on the human gastric cancer cell line SNU-1 and AGS. Methods : Inhibition of cell proliferation and cell cycle by H. cordata was carried out by MTT assay and Muse cell cycle analysis and the expressions of protein associated with apoptosis and cell cycle regulation were investigated with Western blot analysis. Results : In MTT assay, the proliferation of SNU-1 and AGS cells was significantly inhibited by H. cordata in a time and dose dependent manner, Inhibition of cell proliferation by H. cordata was in part associated with apoptotic cell death, as shown by changes in the expression ratio of Bax to Bcl-2 by H. cordata. Also, H. cordata regulated the expression of cell cycle regulatory proteins such as pRb, cyclin D1, cyclin E, CDK4, CDK2, p21 and p15. Conclusion : The antiproliferative effect of H. cordata on SNU-1 and AGS gastric cancer cells revealed in this study suggests that H. cordata has intriguing potential as a chemopreventive or chemotherapeutic agent.

Ovalbumin으로 유발된 아토피피부염 모델 마우스에 대한 복합한약추출물 경구투여의 효과 (Oral Administration Effects of Herbal Extracts on Atopic Dermatitis in Balb/c Mice Sensitized by Ovalbumin)

  • Objective: This study was to evaluate whether herbal mixture (HM-A : Houttuynia cordata Thunberg, Rubus coreanus, Rehmannia glutinosa, Prunus yedoensis, HM-B : Houttuynia cordata Thunberg, Rubus coreanus, Rehmannia glutinosa, Angelica gigas nakai) supresses the development of atopic dermatitis in Balb/c mice sensitized by ovalbumin. Methods: Mice were sensitized by intraperitoneal injection of ovalbumin plus aluminum hydroxide hydrate, followed by epicutaneous sensitization for 6 weeks. After induced atopic dermatitis, HM-A and HM-B were orally administrated for two weeks(once a two days) as a 50 mg/kg concentration. After all mice were sacrificed at the end of the experiment, skin and blood were harvested. Results: Oral administration group was reduced the infiltration of eosinophils, mast cells and total T cells on the skin areas as well as blood analysis. Also, cutaneous expression of IL-4,13,17 decreased. Blood IgE level was decreased. Conclusion: These drugs could be potential candidates for the atopic dermatitis.

익모초, 어성초, 진피 복합 추출물의 항염증 및 항산화 평가 (Evaluation of Anti-inflammatory and Antioxidant Abilities of Complex Extracts Produced from Leonurus japonicus Houtt., Houttuynia cordata Thunberg, and Citrus unshiu Markovich)

  • 본 연구는 복합추출물(익모초, 어성초, 진피)의 생리활성을 측정하여 식·의약품, 화장품 등의 소재로서의 가능성을 확인하고자 하였다. 복합추출물은 열수 추출하였으며, 세포독성, 항산화, 항염증, MAPKs 등의 평가는 ELISA 및 western blot을 통해 확인하였다. 그 결과, 총 폴리페놀 함량은 35.53±0.48 mg GAE/g, 플라보노이드 함량은 8.18±0.16 mg QE/g으로 나타났으며, DPPH 와 ABTS radical 소거능은 농도 의존적으로 증가하였다. 대조군과 비교하여 세포독성은 400 ㎍/㎖ 농도까지 100% 이상의 생존율이 나타났으며, NO 및 염증성 사이토카인을 유의하게 감소시켰다. 또한, 복합추출물은 MAPKs(ERK, JNK, p38) 단백질 발현을 유의하게 억제시켰다. 이러한 결과는 복합추출물이 산화적 손상 및 염증에 사용될 수 있음을 나타낸다. 따라서 본 연구 결과는 식·의약품, 화장품용 복합소재의의 기초자료로 활용될 수 있을 것이다.

  • This study was carried out to evaluate antitoxic effects of Houttuynia cordata extract on cadmium by colorimetric methods. The antitoxic activity ofc in NIH 3T3 fibroblasts was evaluated by MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide) and SRB (sulforhodamine B protein) assays. The light microscopic study was carried out to observe morphological changes of the treated cells. The concentration of 10-2 mg/ml of Houttuynia cordata extract was shown significant antitoxic activity. The number of NIH 3T3 fibroblasts were antitoxic and tend to regenerate. These results suggest that the ethyl acetate extract of Houttuynia cordata retains a potential antitoxic activity.

감마선조사 생약재 (어성초, 구기자)의 안전성에 관한 유전독학성적 평가 (Genotoxicological Safety of the Two gamma-Irradiated Herbs ; Houttuynia cordata Thunberg and Kycium Chinense Miller)

  • These experiments were performed to investigate the safety of two herbs-Houttuynia cordata Thunberg and Lycium chinese Miller-irradiated with gamma-rays in respect of genotoxicity. Water extracts from the 10 kGy gamma-irradiated herbs were examined in two short -term in vitro tests ; (1) Salmonella typhimurium reversion assay (Ames test) in strain TA 98 and Ta100 and (2) Micronuclues test on clutured Chinese hamster ovary(CHO) cells. No mutagenicity was detected in the two assays with or without metabolic activation . From these results , the safety of the herbs irradiated with gamma-rays at practical doses could be revealed in further tests of genotoxicity in vivo, chronic and reproductive toxicity.

어성초(魚腥草)의 줄기배양에 의한 식물체(植物體) 재분화(再分化) (Plant Regeneration by the Stem Culture in Houttuynia cordata Thunberg)

  • 약용식물(藥用植物)로써 이용 가능성 큰 어성초(魚腥草)의 이차(二次) 대사산물(代謝産物)의 생산에 기초가 되는 조직배양체계(組織培養體系)를 확립하고자 어성초(魚腥草)의 절간(節間), 절(節) 및 엽(葉)의 절편체(切片體)를 이용(利用)하여 캘러스를 형성(形成)시키고, 형성(形成)된 캘러스로부터 식물체(植物體) 재분화(再分化)를 시도하였다. 1. Embryogenic 캘러스 형성(形成)은 모든 절편체(切片體)에서 2, 4-D 1 mg/L와 kinetin 0. 5 mg/L을 첨가(添加)하였을 때 가장 효과적이었다. 2. 절간(節間)에서의 캘러스 형성(形成)이 엽(葉)보다 더 효과적이었다. 3. 어성초(魚腥草)의 절간(節間)으로부터 embryogenic 캘러스 형성(形成)은 ${\frac{1}{2}}\;MS$ 배지(培地)가 효과적이었다. 4. 배지내(培地內)에 1%의 활성탄(活性炭)을 첨가(添加)하였을 경우 캘러스를 형성(形成)시키지 않고 배양(培養)된 절(節)로 부터 직접 식물체(植物體)가 분화(分化)되었다. 5. Embryogenic 캘러스로부터 식물체(植物體) 재분화(再分化)는 0. 2 mg/L NAA와 1 mg/L BA를 첨가(添加)한 배지(培地)가 다른 조합(組合)의 배지(培地)보다 분화율(分化率)이 높았다.
