사운덱스 알고리즘을 적용한 신경망라 뉴로-처지 기법의 호스트 이상 탐지 (Host Anomaly Detection of Neural Networks and Neural-fuzzy Techniques with Soundex Algorithm)

  • 본 논문에서는 시스템 호출을 이용하여 이상 침입 탐지 시스템의 성능을 향상시키기 위해, 특징 선택과 가변 길이 데이터를 고정 길이 학습 패턴으로 변환 생성하는 문제를 해결하기 위한 사운덱스 알고리즘을 적용한 신경망 학습을 통하여 이상 침입 탐지의 연구를 하고자 한다. 즉, 가변 길이의 순차적인 시스템 호출 데이터를 사운덱스 알고리즘에 의한 고정 길이의 행위 패턴을 생성하여 역전파 알고리즘과 퍼지 멤버쉽 함수에 의해 신경망 학습을 수행하였다. 역전파 신경망과 뉴로-퍼지 기법을 UNM의 Sendmail Data Set을 이용하여 시스템 호출의 이상침입 탐지에 적용하여 시간과 공간 복잡도 그리고 MDL 측면에서 성능을 검증하였다.

  • 이동 통신 기술의 급격한 발전으로, 이동 컴퓨팅 환경에서 데이터 서비스에 대한 수요가 점차 증가하고 있다. 이동 컴퓨팅 환경에서의 여러 단점을 극복하고자 캐쉬가 등장하게 되었다. 이동 호스트 내에 캐쉬가 존재하면, 통신 대역폭의 절약 및 질의에 대한 빠른 응답을 가져올 수 있지만, 이동 호스트가 서버 데이터와의 캐쉬 일관성(consistency)을 유지시켜야 한다는 문제가 생기게 된다. 본 논문에서는 기존의 기법들이 가지는 문제점들을 보완하기 위해 데이터의 주기성을 고려한 캐쉬 일관성 유지 기법을 제안한다. 제안한 기법은 데이터를 주기적, 비주기적 데이터로 분류하고 주기적 데이터에 만료시간을 삽입한다. 또한 무효화 메시지를 저장해서 단절 후에 선별적으로 캐쉬 데이터가 나누어질 수 있도록 해서, 캐쉬 내의 데이터가 접속 단절 후에 일방적으로 버려지는 것을 막을 수 있게 된다. 이렇게 함으로서 서버와 이동 호스트간의 통신에 사용하는 전체 대역폭의 낭비와 서버접속에 필요한 이동 호스트의 전력의 낭비를 줄일 수 있다. 마지막으로 기존의 기법과의 실험을 통해서 제안한 기법의 효율성을 알아보고자 한다.

Cooperation between Human DAF and CD59 in Protecting Cells from Human Complement-mediated Lysis

  • The complement (C) regulatory proteins decay accelerating factor (DAF, CD55) and CD59 could protect host cells using different mechanisms from C-mediated damage at two distinct levels within the C pathway. Co-expression of DAF and CD59 would be an effective strategy to help overcome host C-induced xenograft hyperacute rejection. In this study, we made a construct of recombinant expression vector containing DAF and CD59 cDNA and the stable cell lines were obtained by G418 selection. Extraneous genes integration and co-expression were identified by PCR, RT-PCR and Western blot analysis. Human c-mediated cytolysis assays showed that NIH/3T3 cells transfected stably with pcDNA3-CD59, pcDNA3-DAF, and pcDNA3-CD59DAF-DP were protected from C-mediated damage and that synchronously expressed human CD59 and DAF provided the most excellent protection for host cells as compared with either human CD59 or DAF expressed alone. Therefore, the construct represents an effective and efficacy strategy to overcome C-mediated damage in cells and, ultimately, in animals.

Fate of Genetically Engineered 2,4-D-Degrading Microorganisms in Natural Soils and Waters

  • To analyze the effects of host versus plasmid on survival of 2, 4-degrading bacteria in environmental samples, strains Pseudomonas cepacia/pJP4, Alcaligenes JMP228/pJP4, P. cepacia/p712, and Alcaligenes JMP228/p712 were separately inoculated into samples of field soil, paddy soil, lake water, and river water, and then the changes of their populations were measured. The strains used contained a 2, 4-D degradative plasmid, either pJP4 conferring fast-growing property to the host or p712 conferring slow-growing property, and were resistant to antibiotics such that the inoculated strains could be enumerated against the indigenous microbial populations. In sterile environmental samples, these strains were stably maintained at the levels used for inoculation, except in sterile paddy soil where Alcaligenes JMP228 strains died drapidly. In natural soil samples for four strains declined steadily with time, but in naturla water samples their polulations fell rapidly at the early phase and then remained almost constant. When the environmentla samples were treated with 2, 4-D, P. cepacia/pJP4 and P. cepacia/p712 maintained significant numbers, while Alcaligenes JMP228/pJP4 and Alcaligenes JMP228/p712 declined significantly in most of the samples. The results indicated that the survivability of genetically modified microorganisms could vary depending on the environments and that their abundance in the environments under s2, 4-D selection was markedly influenced by the nature of the 2, 4-D degradative plasmid as well as type of the host strain.

Chewing Lice of Swan Geese (Anser cygnoides): New Host-Parasite Associations

  • Chewing lice (Phthiraptera) that parasitize the globally threatened swan goose Anser cygnoides have been long recognized since the early 19th century, but those records were probably biased towards sampling of captive or domestic geese due to the small population size and limited distribution of its wild hosts. To better understand the lice species parasitizing swan geese that are endemic to East Asia, we collected chewing lice from 14 wild geese caught at 3 lakes in northeastern Mongolia. The lice were morphologically identified as 16 Trinoton anserinum (Fabricius, 1805), 11 Ornithobius domesticus Arnold, 2005, and 1 Anaticola anseris (Linnaeus, 1758). These species are known from other geese and swans, but all of them were new to the swan goose. This result also indicates no overlap in lice species between older records and our findings from wild birds. Thus, ectoparasites collected from domestic or captive animals may provide biased information on the occurrence, prevalence, host selection, and host-ectoparasite interactions from those on wild hosts.

The SL1 Stem-Loop Structure at the 5′-End of Potato virus X RNA Is Required for Efficient Binding to Host Proteins and forViral Infectivity

  • The 5′-region of Potato virus X (PVX) RNA, which contains an AC-rich, single-stranded region and stem-loop structure 1 (SL1), affects RNA replication and assembly. Using Systemic Evolution of Ligands by EXponential enrichment (SELEX) and the electrophoretic mobility shift assay, we demonstrate that SL1 interacts specifically with tobacco protoplast protein extracts (S100). The 36 nucleotides that correspond to the top region of SL1, which comprises stem C, loop C, stem D, and the tetra loop (TL), were randomized and bound to the S100. Remarkably, the wild-type (wt) sequence was selected in the second round, and the number of wt sequences increased as selection proceeded. All of the selected clones from the fifth round contained the wt sequence. Secondary structure predictions (mFOLD) of the recovered sequences revealed relatively stable stem-loop structures that resembled SL1, although the nucleotide sequences therein were different. Moreover, many of the clones selected in the fourth round conserved the TL and C-C mismatch, which suggests the importance of these elements in host protein binding. The SELEX clone that closely resembled the wt SL1 structure with the TL and C-C mismatch was able to replicate and cause systemic symptoms in plants, while most of the other winners replicated poorly only on inoculated leaves. The RNA replication level on protoplasts was also similarly affected. Taken together, these results indicate that the SL1 of PVX interacts with host protein(s) that play important roles related to virus replication.

  • 본 연구는 포천 이동갈비바비큐축제에 방문한 방문객을 대상으로 식생활 라이프스타일 유형에 따라서 포천 이동갈비바비큐축제를 선택할 때 집단별 추구하는 선택속성의 차이점과 고객반응에 미치는 영향을 알아보고자 라이프스타일에 따라 분류된 군집들의 인구통계학적 특성에 따라 교차분석을 실시하였고, 군집별로 포천 이동갈비바비큐축제 차이분석과 고객반응에 미치는 영향관계를 파악하였다. 연구결과, 축제를 주최자들은 향후 축제를 기획할 때는 가족이 함께 다양한 지역적인 전통문화체험과 함께 휴식을 취할 수 있는 프로그램이 필요하며, 현재 영업하고 있는 축제장의 음식점은 가격이 비싼 바비큐 음식보다는 가족이 쉽게 이용할 수 있는 적당한 가격의 바비큐메뉴를 개발해야 할 것이다. 축제 선택속성에 따른 고객반응에서는 청결은 기각되었다. 자연경간이 뛰어난 포천 이동갈비바베큐축제의 특성을 고려해볼 때 청결은 의미가 없는 것으로 판단된다. 따라서 타 지역의 축제와는 성격을 달리하는 자연과 함께 하는 체험프로그램을 개발해야할 것이다.

Complete Genomic Characterization of Two Beet Soil-Borne Virus Isolates from Turkey: Implications of Comparative Analysis of Genome Sequences

  • Sugar beet (Beta vulgaris L.) is known as a key product for agriculture in several countries across the world. Beet soil-borne virus (BSBV) triggers substantial economic damages to sugar beet by reducing the quantity of the yield and quality of the beet sugars. We conducted the present study to report the complete genome sequences of two BSBV isolates in Turkey for the first time. The genome organization was identical to those previously established BSBV isolates. The tripartite genome of BSBV-TR1 and -TR3 comprised a 5,835-nucleotide (nt) RNA1, a 3,454-nt RNA2, and a 3,005-nt RNA3 segment. According to sequence identity analyses, Turkish isolates were most closely related to the BSBV isolate reported from Iran (97.83-98.77% nt identity). The BSBV isolates worldwide (n = 9) were phylogenetically classified into five (RNA-coat protein read through gene [CPRT], TGB1, and TGB2 segments), four (RNA-rep), or three (TGB3) lineages. In genetic analysis, the TGB3 revealed more genetic variability (Pi = 0.034) compared with other regions. Population selection analysis revealed that most of the codons were generally under negative selection or neutral evolution in the BSBV isolates studied. However, positive selection was detected at codon 135 in the TGB1, which could be an adaptation in order to facilitate the movement and overcome the host plant resistance genes. We expect that the information on genome properties and genetic variability of BSBV, particularly in TGB3, TGB1, and CPRT genes, assist in developing effective control measures in order to prevent severe losses and make amendments in management strategies.

Exploring the molecular characteristics, detoxification functions, and immune responses of two glutathione S-transferases in redlip mullet (Liza haematochelia)

  • The mechanism for the elimination of xenobiotics undergoes three different phases of reactions in organisms. Among these, glutathione S-transferases (GSTs) are classified as phase II detoxification enzymes, catalyzing the conjugation of electrophilic substrates to glutathione or reduced hydroperoxides. This study aimed to investigate the molecular characteristics, detoxification functions, and immune responses of GST omega (LhGSTO1) and kappa (LhGSTK1) in redlip mullet. The open reading frames of LhGSTO1 (720 bp) and LhGSTK1 (687 bp) encoded proteins of 239 and 228 amino acids, respectively. Sequence analysis revealed that LhGSTO1 and LhGSTK1 possessed GSH-binding sites in their N-terminal domains. Substrate-binding sites in the C-terminal domain were exclusively identified in LhGSTO1. In the tissue-specific transcription profile analysis, both LhGSTO1 and LhGSTK1 were ubiquitously expressed in all tissues of healthy mullets. Temporal expression analysis of LhGSTO1 and LhGSTK1 in the blood showed that their expression was significantly modulated by polyinosinic:polycytidylic (poly I:C), lipopolysaccharide (LPS), and Lactococcus garvieae. Different chemical and cellular assays were performed to assess the detoxification and cellular protective abilities of the two proteins. A substrate specificity test using the recombinant proteins revealed that both proteins possessed specific activity toward 1-chloro-2,4-dinitrobenzene (CDNB). In the disk diffusion assay, the smallest clearance zones were observed for LhGSTO1 and LGSTK1 against CdCl2. In the cell protection assay, both LhGSTO1 and LhGSTK1 showed significant Cd detoxification ability compared to the control. Collectively, these results demonstrate that GST omega and kappa are involved in host defense against immune stimulants and xenobiotics in redlip mullet.

Effect of the different cover crop incorporation on glomalin-related soil protein and soybean and maize growth

  • The glycoprotein known as glomalin-related soil protein (GRSP) is abundantly produced on the hyphae and spores of arbuscular mycorrhizal fungi (AMF) in soil and roots. GRSP play a decisive role in the soil aggregation, but GRSP was also sensitive to agricultural managements. Thus, our objectives were to assess the effect of different cover crop incorporation on the GRSP content in soil and growth of subsequent soybean and maize. Pot experiments with the incorporation of four cover crops were set up. The same amount (666g) of aboveground plant parts of wheat (AMF host), hairy vetch (AMF host), mustard (non-host) and rapeseed (non-host) was separately incorporated into soils. The aboveground plant parts and roots of soybean and maize were grown in each incorporated pots and sampled at 6 and 9 weeks after sowing. Our results showed that the different cover crops incorporation affected soil biological and chemical properties such as EC, $NO_3-N$ content, ${\beta}-glucosidase$ activity, alkaline phosphatase (ALP) activity and GRSP content. The soil EC and $NO_3-N$ content in the hairy vetch, mustard and rapeseed was higher compared to the wheat. The ${\beta}-glucosidase$ activity in the wheat and hairy vetch was significantly higher than that in the mustard and rapeseed, and the ALP activity in the wheat was significantly higher than that in the hairy vetch, mustard, and rapeseed. The GRSP content in the mustard and rapeseed was significantly lower than that of the hairy vetch and wheat. Moreover, The top dry weight and leaf area of soybean and maize in the hairy vetch at 6 weeks were significantly higher compared to the other treatments. Our results indicated that the incorporation of mustard and rapeseed may cause indirectly the decrease of GRSP content and soil enzyme activity in soil. One possible explanation for the decrease of GRSP in non-AMF host crop treatments may be the decrease of AMF density in the soil. AMF are not able to form a symbiotic relationship with Brassicaceae roots due to the release of anti-fungal compounds. This means the AMF may not be able to produce GRSP in the soil. However, the differences in the benefit of cover crop incorporation were shown only by a pot experiment. Comparative investigations of crop residue managements would be applied to both pot experiment and field study to clarify a better selection of cover crops in rotation to encourage GRSP production.
