Purification and Characterization of Homoharringtonine from Cephalotaxus koreana

  • A new isolation and purification method was developed aiming at increasing yield and purity for homoharringtonine. This method was a simple andefficient procedures, for the isolation and purification of homoharringtonine from Cephalotaxus koreana, consisting of solvent extraction, adsorbent treatment, low-pressure chromatography, and high performance liquid chromatography (HPLC). The crude homoharringtonine was efficiently pre-purified adequately to perform HPLC through a combination withadsorbent treatment and low-pressure chromatogaphy. The homoharringtonine can be simply obtained with high purity and yield from crude homoharringtonine by HPLC. Purified homoharringtonine was characterized.

  • A novel purification method was developed for producing homoharringtonine from Cephalotaxus koreana, to guarantee high purity and yield. Our simple, efficient procedure for isolating and purifying homoharringtonine from C. koreanabiomass consisted of solvent extraction, synthetic adsorbent treatment, low-pressure chromatography, followed by high performance liquid chromatography (HPLC). The use of active clay treatment and silica gel low-pressure chromatography in the pre-purification process allowed for the rapid, efficient separation of homoharringtonine from interfering compounds and dramatically increased the yield and purity of crude homoharringtonine for high-performance liquid chromatography (HPLC) purification steps compared with alternative processes. Homoharringtonine could be obtained simply with high yield and purity from biomass using this purification method, while minimizing solvent use and the scale and complexity of HPLC operations for homoharringtonine purification. Purified homoharringtonine was identified and characterized.

  • Harringtonine and homoharringtonine known as anti-cancer agents were isolated from Korean native plumyew(Cephalotaxus koreana) using column chromatography(CHCl3:MeOH=19:1, Rf=0.28). The structure of the mixture of two compounds was characterized by 1H-NMR. Comparison of our spectra of harringtonine and homoharringtonine with previously reported ones indicated that the two are identical. The contents of harringtonine and homoharringtonine in the needles, stems, and roots of Korean native plumyew were determined by high performance liquid chromatography(HPLC). The contents of both compounds varied with the site of location and the part of plant. The content of harringtonine was higher in needles and roots than in stems, whereas the content of homoharringtonlne was lower than harringtonine. Homoharringtonine contents in needles at Mt. Palgong, Mt. Dukyu, Mt. Baekyang, Mt. Jiri, and Namhae were higher than in stems and roots. But homoharringtonine contents in needles al Mt. Jokye and Jindo were lower than in stems and roots.

  • The contents and composition of alkaloids homoharringtonine, harringtonine, and cephalotaxine were determined in Korean native plumyew tree (Cephalotaxus koreana) collected from 17 different sites in Korea. The alkaloid contents of C. koreana varied with locations and plant populations from 11.8 mg/g to 195.2 mg/g of dry weight while C. koreana in southern area had higher alkaloid contents. The alkaloid compositions in C. koreana were highest with homoharringtonine (40%) and followed harringtonine (32%) and cephalotaxine (28%). Total alkaloids and homoharringtonine contents in needles were higher than those of stem by 2 fold. Alkaloid contents of C. koreana were the highest in winter, and the lowest in summer. Comparison of alkaloid contents and composition with other species of Cephalotaxus revealed that C. koreana was different to C. harringtonia, and contents of homoharringtonine were higher than other species. C. koreana could be potential source for homoharringtonine and related alkaloids.

  • Homoharringtonine isolated from the plumyew tree (Cephalotaxus koreana) is currently considered as one of the most promising chemotherapeutic agents for anti-cancer. Variation of homoharringonine contents from eight natural populations of Korean native plumyew trees was determined by HPLC (high performance liquid chromatography), and compared with the variation among populations, the variation of the different parts(needle, root, stem and seed), and the variation according to their ages and growth features. Homoharringonine contents among populations were significantly different with Mt. South Dukyu ($1,048{\mu}g/g$) having the highest and Mt. Obong having the lowest. The analyses of plant pans showed that the homoharringtonine contents were highest in plant needles ($874{\mu}g/g$), and followed by roots, stems and seeds. Homoharringtonine contents of the plumyew trees decreased about 30% in increment with their ages and growth.

  • Cell proliferation inhibitory effects of homoharringtonine (HHT), an active drug substance in Cephalotaxus koreana, against blood cancer cell line K562 were evaluated. In addition, in vivo chronic toxicity test with mouse was carried out. When K562 cell line was treated everyday for 9, 6, 3 days, $IC_{50}$ values of HHT were determined as 0.27, 0.37, and 1.10 mM respectively. The anticancer activity of HHT was comparable to adriamycin, a known anticancer drug compound for blood cancer treatment. in vivo chronic toxicity test of the HHT, the number of red blood cell (RBC) showed no significant difference. From the analysis of the liver-functional enzymes in blood, all of liver damage related enzymes such as glutamate-oxalate-transferase (GOT), glutamate-pyruvate-transferase (GPT), cholesterol (Chol) and alkaline phosphatase (ALP) showed no significant change. However, from the histologic test, a neutrophil of the band type in liver tissue was observed.

A Novel Purification Process for Homoharringtonine from Celphalotaxus koreana

  • An effective purification method was developed for producing Homoharringtonine (HHT), to guarantee high purity and yield from Cephalotaxus koreana. This process was a simple and efficient procedure, for the isolation and purification of HHT form the biomass of Cephalotaxus koreana, consisting of extract, adsorbent treatment, precipitation and followed by a chromatography. The extraction, adsorbent treatment and precipitation in pre-purification process allows for rapid and efficient separation of HHT from many compound and dramatically increases the yield and purity of crude HHT for HPLC purification steps compared to alternative processes. This purification processes serves to minimize solvent usage, size, and complexity of the operations for HHT purification.

Biotic and Abiotic Factors Affecting Homoharringtonine Contents of Cephalotaxus koreana Nakai (개비자나무의 homoharringtonine 함량에 영향을 미치는 생물 및 무생물적 환경인자)

  • This study was carried out to investigate abiotic and biotic environmental factors affecting homoharringtonine (HHT) contents of Cephalotaxus koreana, whereby, to provide basic information of high value-added industry production of HHT as a promising anti-cancer agent. For correlation between abiotic environmental factors (soil moisture, soil pH, habitat density and temperature) and HHT contents, the contents were highly correlated with soil moisture (0.77) and soil pH (-0.68). For multiple regression analysis of relationship between abiotic environmental factors (soil moisture and soil pH) and HHT contents, soil moisture appeared to be strongly affecting the contents relatively due to being significant at only its regression coefficient ($26.48^{***}$). For the effect of biotic environmental factors (damage index) affecting HHT contents, the contents was quadratic with equation of $H=278.23+1242D-398.87D^2$, also, damage index had strong effect on the contents. Finally, for the result of the most influencing an environmental factor on HHT contents, both damage index and soil moisture were suitable in second polynomial regression, also, damage index ($R^2=0.73^{***}$) was turned out to be more influencing factor than soil moisture ($R^2=0.67^{**}$) on HHT contents relatively. Therefore, we predict that HHT contents in the trees of Cephalotaxus koreana is produced as a chemical defense mechanism triggered by a stress-related damage of fungi or insects.

  • This study was designed to investigate the cytotoxic effect of Cephaxus koreana on human breast cancer cell lines (SCC-1395 and SK-BR-3) us ins MTT(3- [4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide) assay. RPMII -1640 medium with $10\%$ FBS and $1\%$ of penicillin-streptomycin was used. The ethanol extrart of Cephaxus koreana after adsoption resin treatment showed higher anticancer activity against SCC- 1395 than the ethanol extract of Cephaxus koreana without adsoption resin treatment. SK-BR-3 showed higher sensitivity activity than SCC-1395 at the same concentration of 20000 ppm.
