간질감염(肝蛭感染) 및 구충제(驅蟲劑) DS-6 부작용(副作用)에 관(關)한 조사연구(調査硏究) (Studies on Bovine Fascioliasis with Special Reference to Infestation Rate and Side Action of DS-6 in Cattle)

  • A servey on bovine fascioliasis in 52,876 cattle of Euiseong, Andong, Yecheon, Wolseong areas in Gyeongbug Province was carried out by the intradermal test, and the clinical side action of the positive cattle administered with DS-6 intramuscularly was observed. In addition pathological examination of 2 cattle died with side action of DS-6 was also conducted. The results of the studies are summarized as follows; 1. The average infestation rate of bovine fascioliasis was 31.1% and regional infestation rates were 35.1% in Eiseong, 34.5% in Yecheon, 32.2% in Andong, 26.9% in Wolseong, respectively. 2. The side actions were occurred in 1.5%(115/7,667) cf cattle administered with DS-6 and the clinical observations were salivation, dyspnea, hyperamia of conjunctiva, lacrimation, perspiration, urticaria, tremor, diarrhea, staggering, convulsion, edema and abortion. 3. Within 1 hour after the injection, side actions were occurred in 61.7% of the 115 cattle while they were observed in 37.4% of the 115 catte from 1 hour to 2 hours post injection. 4. The duration of the side action was less than 2 hours in 29.5%, 2 to 10 hours in 47.0% and more than 10 hours in 23.5% of the 115 cattle. 5. Autopsy findings and histopathological observations indicate that the death of cattle is caused by anaphylactic shock.

  • Avian chlamydiosis is caused by Chlamydophila psittaci and considered as one of an important zoonotic disease throughout the world. Among more than 400 avian species including poultry and pet birds susceptible to the disease, psittacine birds were known to be mostly susceptible hosts. In Korea, no outbreak of the disease and genetic analysis of the agent in poultry and pet birds have been reported. With histopathological findings and genetic identification of a causative agent, avian chlamydiosis was identified in parrots submitted from the same pet bird farm in 2006 and 2009 for the diagnosis. Based on genetic sequences and phylogenetic analysis of ompA gene, the two isolates of Chlamydophila psittaci showed 100% of genetic similarity and belonged to genotype A, suggesting that the same agent might be continuously circulated in the farm. This result indicates that serological survey of the disease in pet bird farms and impact of the disease on significance in public health may be further studied.

  • Carbon tetrachloride ($CCl_4$) is well known hepatotoxicant. Its overdose cause severe centrilobular hepatic necrosis in human and experimental animals. We administered $CCl_{4}$ at low (0.2 mL/kg p.o.) and high (2 mL/kg p.o.) doses to mice. Mice were sacrificed at 24 h after administration. We evaluated liver toxicity by serum AST and ALT level and by microscopic observation. Using cDNA chip, we conducted gene expression analysis in liver. Mean serum activities of the hepatocellular leakage enzymes, ALT and AST, were significantly increased compare to control, respectively, in the low and high dose groups. H&E evaluation of stained liver sections revealed $CCl_{4}-related$ histopathological findings in mice. Moderate centrilobular hepatocellular necrosis was present in all $CCl_{4}$ treated mice. We found that gene expression pattern was very similar between low and high dose group. However, some stress related genes were differently expressed. These results could be a molecular signature for the degree of liver injury. Our data suggest that the degree of severity could be figure out by gene expression profiling.

사염화탄소로 유발된 간손상에 대한 구기자 추출물의 치료효과 (Therapeutic Effect of Lycii fructus Extract in Hepatotoxicity Induced by Carbon Tetrachloride)

  • The hepatotherapeutic effect of the extract of Lycii fructus has been studied in rats against $CCl_4$ induced liver toxicity. The rats were orally treated with $CCl_4$ (corn oil/$CCl_4$ 1:1, $1m{\ell}/kg$) and then $CCl_4$ ($0.5m{\ell}/kg$) administered four times for 2 weeks. The extracts of L. fructus have been administered every day for 2 weeks after the last $CCl_4$ injection. The experimental groups consisted of negative control (G1), positive control ($CCl_4$ alone; G2), extract of L. fructus (50 mg/kg; G3, 100 mg/kg; G4, 200 mg/kg; G5), respectively. There was a significant decrement to G2 on the serum level of aspartate aminotransferase, alanine aminotransferase, and alkaline phosphatase in G5. Also, the content of thiobarbituric acid reactive substance, and phosphatidylcholine hydroperxidase, a marker of lipid peroxidation, in the liver were decreased significantly G5 and G4 compared with G2. Although, catalase or superoxide dismutase, antioxidant enzyme, in the liver were decreased significantly too, it would not be a good sign for the liver. In histopathological findings, such a hepatocellular vacuolar degeneration, lobular restructure, cellular infiltration, necrosis, and so on were shown severely in G2. However, G4 and G5 was shown a mild cytoplasmic vacuolation and inflammatory cell. In conclusion, as a protection against cell damage, lipid peroxidation and serum level, it suggested that the extract of Lycii fructus would have been a therapeutic effect of liver injury directly.

Is Surgical Treatment Necessary after Non-curative Endoscopic Resection for Early Gastric Cancer?

  • Purpose: Additional surgery is commonly recommended in gastric cancer patients who have a high risk of lymph node metastasis or a positive resection margin after endoscopic resection. We conducted this study to determine factors related to residual cancer and to determine the appropriate treatment strategy. Materials and Methods: A total of 28 patients who underwent curative gastrectomy due to non-curative endoscopic resection for early gastric cancer between January 2006 and June 2009 were enrolled in this study. Their clinicopathological findings were reviewed retrospectively and analyzed for residual cancer. Results: Of the 28 patients, surgical specimens showed residual cancers in eight cases (28.6%) and lymph node metastasis in one case (3.8%). Based on results of the endoscopic resection method, the rate of residual cancer was significantly different between the en-bloc resection group (17.4%) and the piecemeal resection group (80.0%). The rate of residual cancer was significantly different between the diffuse type group (100%) and the intestinal type group (20%). The rate of residual cancer in the positive lateral margin group (25.0%) was significantly lower than that in the positive vertical margin group (33.3%) or in the positive lateral and vertical margin group (66.7%). Conclusions: We recommended that patients who were lateral and vertical margin positive, had a diffuse type, or underwent piecemeal endoscopic resection, should be treated by surgery. Minimal invasive procedures can be considered for patients who were lateral margin positive and intestinal type through histopathological examination after en-bloc endoscopic resection.

Effects of Callophyllis japonica powder on carbon tetrachloride-induced liver injury in rats

  • A limited number of studies have been conducted on the bioactivity of Callophyllis japonica (C. japonica), which is a red seaweed that is traditional in the oriental diet. In this report, the hepatoprotective effect of C. japonica was studied in a carbon tetrachloride $(CCl_4)-induced$ hepatotoxicity model in rats. A single intraperitoneal injection of 1.25 ml/kg of 20% $CCl_4$ in olive oil produced an elevated level of serum glutamic pyruvic transaminase (SGPT) and increased enzyme activity of serum glutamic oxaloacetic transaminase (SGOT). Pre-treatment with C. japonica (150 mg/kg/d) for 3 days in $CCl_4-injected$ rats reduced the levels of SGPT and SGOT compared with control levels (P < 0.05), while higher doses (300 and 600 mg/kg) were ineffective. The administration of C. japonica (150 mg/kg/d) for 3 days after the $CCl_4$ injection in rats was ineffective at reducing SGPT and SGOT. The histopathological findings in each group largely agreed with the biochemical data. The results of this study support the suggestion that C. japonica has a hepatoprotective effect on chemical-induced liver injury.

Demonstration of the Usefulness of Optical Coherence Tomography in Imaging a Mouse Tail Model of Lymphedema

  • To investigate the usefulness of optical coherence tomography (OCT) for imaging lymphedema, we directly compared it to other histological methods in a mouse model of lymphedema. We performed detailed imaging of the lymphedema lesion on a mouse tail. We imaged the mouse tail in vivo with OCT and created histopathological samples. We constructed a spectrometer-based OCT system using a fiber-optic Michelson interferometer. The light was directed to 50:50 couplers that split the light into reference and sample arms. Backscattered light from a reference mirror and the sample produced an interference fringe. An OCT image of the lymphedema model revealed an inflammatory reaction of the skin that was accompanied by edema, leading to an increase in the light attenuation in the dermal and subcutaneous layers. Similar to OCT image findings, histological biopsy showed an inflammatory response that involved edema, increased neutrophils in epidermis and subdermis, and lymphatic microvascular dilatation. Furthermore, the lymphedema model showed an increase in thickness of the dermis in both diagnostic studies. In the mouse tail model of lymphedema, OCT imaging showed very similar results to other histological examinations. OCT provides a quick and useful diagnostic imaging technique for lymphedema and is a valuable addition or complement to other noninvasive imaging tools.

꼬마하마(Choeropsis liberiensis)에서 확인된 다낭신장병 (Polycystic Kidney Disease in the Adult Female Pygmy Hippopotamus (Choeropsis liberiensis))

  • 다낭신장병은 신장실질에 다수의 낭포가 형성되는 것을 특징으로 하는 사람, 개, 고양이에서 흔히 있는 유전성 질환으로서, 사육상태의 꼬마하마(pygmy hippopotamus)에서도 몇 몇 증례가 보고되고 있다. 2013년 1월 15일 체중 198킬로그램, 33년령 암컷 꼬마하마의 부검과정 중에 양쪽 신장에서 다낭신장병이 관찰되었다. 한 쪽 신장은 약간 종대된 반면 다른 쪽 신장의 아랫부분은 옅은 황색의 수양성 액체로 채워진 한 개의 큰 낭포가 있었다. 양측 신장 모두 직경 2 mm에서 20 mm의 다양한 크기의 액체가 함유된 다수의 낭포들이 관찰되었다. 상당한 부분의 신장 피질과 수질부가 낭포들로 대체되어 있었다. 현미경 검사에서 낭포들의 안쪽은 낮은 입방세포에서부터 편평상피세포들로 구성되어 있었다. 육안적인 소견과 조직병리학적인 검사로 다낭신장병으로 진단하였다. 본 증례보고는 한국에서 최초로 꼬마하마에서 다낭신장병이 확인된 것이다.

푸들에서 발생한 melanotrichia: 임상적 그리고 조직병리학적 특징 (Melanotrichia in Poodles : Clinical and Histopathological Findings)

  • 5살과 2살의 푸들 견이 미용 직후, 등 부위의 털의 변색과 전신적 탈모증세로 건국대학교 부속 동물 병원에 내원하였다 병변 부위에서는 육안적으로 약간의 피부 발적과 소농포가 발견되었지만 총 혈구 계산, 혈액 화학 검사에서는 특이한 소견이 발견되지 않았다. 아울러 최근 약물투여 병력도 없었다. 병변부의 trichogram에서는 모근 쪽과 모간에 멜라닌 과립이 착색되어 있었고, 병변 이외의 부위에서는 멜라닌 착색이 관찰되지 않았다. 그리고 병변부 피부 생검에서는 약한 부전 각화증이 관찰되었으며 아주 미약한 진피층의 호중구 침윤이 관찰되었다. 본 케이스에서는 melanotrichia가 미용 직후에 피부에 가해지는 각종 자극에 의해 발생할 수 있음을 보여주고 있다. 그러나, 미용 후 멜라닌 신생의 정도와 델라닌 세포의 증식 기전은 본 증례로서는 현재까지 명확하게 밝혀지지는 않았다.

개에서 발생한 이상성 활막육종 증례 (Imaging Diagnosis: Biphasic Synovial Sarcoma in a Dog)

  • 우측 후지가 점진적인 파행을 보이는 11년령의 수컷 시츄견이 내원하였다. 신체검사 결과 부정형의 종괴가 촉진 되었다. 방사선 촬영에서는 균질한 연부 조직성 밀도를 보였으며, 초음파 검사에서는 혼합성 에코를 보이는 경계가 명료한 특징을 갖고 있었다. MRI 검사에서는 주변 근육과 비교한 결과 유사 또는 강한 비균질성 신호강도를 보였다. 종괴는 주로 대퇴부 주변에 형성되어 있었으며, 일부는 골반뼈 폐쇄구멍을 통해 골반강내에서 직장의 변위를 일으키고 있음이 확인할 수 있었다. 병리조직 검사 결과 중간엽 세포와 상피세포로 구성된 활막육종으로 진단되었다. 종괴는 외과적으로 적출하였으나, 지속적인 근위축으로 있었으며 인해 파행은 개선되지 않았다. 적출 후 6개월에 종괴가 형성됨이 확인되었으며 점진적으로 그 크기 비대해져 10개월이 되는 시점에서 견주의 요청으로 안락사 하였다. MRI는 활막육종 형성됨이 확인되었으며 이에 대한 특징적인 소견은 내포하고 있지 않지만 종괴의 수술적 적출과 종괴의 범위를 확인하는데 있어 유용한 정보를 제공함을 알 수 있었다.
