Obesity Exacerbates Coxsackievirus Infection via Lipid-Induced Mitochondrial Reactive Oxygen Species Generation

  • Coxsackievirus B3 (CVB3) infection causes acute pancreatitis and myocarditis. However, its pathophysiological mechanism is unclear. Here, we investigated how lipid metabolism is associated with exacerbation of CVB3 pathology using high-fat diet (HFD)-induced obese mice. Mice were intraperitoneally inoculated with 1×106 pfu/mouse of CVB3 after being fed a control or HFD to induce obesity. Mice were treated with mitoquinone (MitoQ) to reduce the level of mitochondrial ROS (mtROS). In obese mice, lipotoxicity of white adipose tissue-induced inflammation caused increased replication of CVB3 and mortality. The coxsackievirus adenovirus receptor increased under obese conditions, facilitating CVB3 replication in vitro. However, lipid-treated cells with receptor-specific inhibitors did not reduce CVB3 replication. In addition, lipid treatment increased mitochondria-derived vesicle formation and the number of multivesicular bodies. Alternatively, we found that inhibition of lipid-induced mtROS decreased viral replication. Notably, HFD-fed mice were more susceptible to CVB3-induced mortality in association with increased levels of CVB3 replication in adipose tissue, which was ameliorated by administration of the mtROS inhibitor, MitoQ. These results suggest that mtROS inhibitors can be used as potential treatments for CVB3 infection.

Monascus pilosus 발효 뽕잎차가 고지방 식이 마우스의 체중과 간 조직 항산화계 효소 활성에 미치는 영향 (Effects of Mulberry Leaf Tea Fermented by Monascus pilosus on Body Weight and Hepatic Antioxidant Enzyme Activities in Mouse Fed High-Fat Diet)

  • 본 연구에서는 뽕잎차와 Monascus pilosus로 발효시킨 뽕잎차가 ICR mouse의 체중과 간 조직 항산화계 효소류의 활성에 미치는 영향을 조사하기 위하여 정상대조군(NC), 고지방 식이 대조군(HC), 뽕잎차 분말을 2% 혼합한 고지방 식이군(UM), 발효 뽕잎차 분말을 2% 혼합한 고지방 식이군(FM)으로 나누어 8주간 사육하였다. 체중증가량에 있어서는 HC군의 주당 체중증가량은 NC군에 비해 2.13배로 높았으나, UM군과 FM군은 NC군의 1.80 및 1.67배로 체중증가량이 크게 완화되었다. 장기 중량은 고지방 식이군에서 증가되었던 간의 중량은 UM과 FM군에서 회복되는 경향을 보였으며, 신장의 경우에도 NC군 수준으로 회복되는 경향을 나타내었다. 부고환 주변 지방의 함량은 UM과 FM군의 경우 모두 고지방 식이로 현저하게 증가된 지방함량을 유의적으로 감소시켰으며, 그 효과는 FM군에서 현저하였다. ALT 활성은 HC군이 NC군에 비하여 2.56배나 높은 수치를 나타내었으며, UM군과 HC 간의 유의차는 보이지 않으나 FM군은 HC군에 비하여 유의적으로 낮았다. TG, TC, LDL-cholesterol의 함량 및 동맥경화지수(AI)는 HC군은 NC군에 비하여 각각 18.42%, 39.59%, 212.56% 및 230.70%가 증가하였으며, HDL-cholesterol 함량은 17.84%가 감소하였다. UM군의 TG, TC, HDL- 및 LDL-cholesterol 함량은 HC군과 유의적인 차이가 없으나, FM군에서는 유의적인 차이를 보였으며, 동맥경화지수도 HC군 2.63, UM군 2.09, FM군 1.70으로 FM군에서 양호한 결과를 나타내었다. 간 조직 TG 함량은 UM 및 FM군이 HC군에 비하여 25.18~25.72%가 감소되었으며, 총 콜레스테롤의 함량에서도 TG와 유사한 경향을 보였다. Glutathione(GSH)의 함량은 FM군이 UM군에 비하여 유의적으로 증가하였으며, lipid peroxide(LPO)의 함량은 UM군과 FM군은 HC군에 비하여 각각 49.82% 및 44.52% 감소하였다. Xanthine oxidase(XO) 활성은 UM군과 FM군에서는 HC군에 비하여 각각 38.41% 및 53.62%가 감소하였다. SOD 활성은 UM과 FM군 모두 NC군과 유사한 활성을 나타내었다. Glutathione S-transferase(GST)활성은 UM군과 FM군은 고지방 식이에 의해 감소된 HC군의 활성을 14.81~23.46% 증가시켰다. Glutathione peroxidase(GPX) 활성(NADPH nmole/min/mg protein)은 고지방 식이 대조군인 HC군에서의 활성은 NC군의 65.59%에 불과하였으나, UM군과 FM군은 HC군에 비하여 각각 22.95% 및 38.52% 증가하였다. 간 조직을 검경한 결과, 고지방 식이 대조군인 HF군에서는 간 조직 세포가 지방 방울의 축적에 의해 확장되어 간굴모세혈관(hepatic sinusoids)이 폐쇄된 상태였다. 하지만, UM군과 FM군에서는 중심정맥 주위의 세포들이 정상적으로 잘 보존되어 있으며, 간 소엽의 간세포삭과 간 조직 세포 내에 지방방울의 축적이 보이긴 하나 구분이 가능하며, UM군에 비하여 FM군 쪽의 간 손상이 보다 경미한 것으로 나타났다. 이상의 실험결과, M. pilosus로 발효시킨 뽕잎차는 정상식이군에서는 간 조직 손상과 관련된 LPO의 함량을 경감시키는 효과와 함께 경미한 체중 감소 효과가 있는 것으로 사료되며, 고지방 식이에 의한 비정상 생리 상태에서는 비만 해소와 ROS 생성계효소의 저해와 소거계 항산화 효소의 활성화를 통한 간 기능 증진과 조직 손상을 예방 또는 경감시키는 효과가 있는 것으로 생각된다.

Lactobacillus acidophilus NS1 Reduces Phosphoenolpyruvate Carboxylase Expression by Regulating HNF4α Transcriptional Activity

  • Probiotics have been known to reduce high-fat diet (HFD)-induced metabolic diseases, such as obesity, insulin resistance, and type 2 diabetes. We recently observed that Lactobacillus acidophilus NS1 (LNS1), distinctly suppresses increase of blood glucose levels and insulin resistance in HFD-fed mice. In the present study, we demonstrated that oral administration of LNS1 with HFD feeding to mice significantly reduces hepatic expression of phosphoenolpyruvate carboxykinase (PEPCK), a key enzyme in gluconeogenesis which is highly increased by HFD feeding. This suppressive effect of LNS1 on hepatic expression of PEPCK was further confirmed in HepG2 cells by treatment of LNS1 conditioned media (LNS1-CM). LNS1-CM strongly and specifically inhibited $HNF4{\alpha}-induced$ PEPCK promoter activity in HepG2 cells without change of $HNF4{\alpha}$ mRNA levels. Together, these data demonstrate that LNS1 suppresses PEPCK expression in the liver by regulating $HNF4{\alpha}$ transcriptional activity, implicating its role as a preventive or therapeutic approach for metabolic diseases.

미숙감귤의 지질배설 촉진작용 (The Accelerating Action of Lipid Excretion of Immature Citrus Fruits)

  • In a series of investigations to develop a potential anti-obese agent, we prepared a immature Citrus fruits (IMF) and compared its lipid excretion effects to those of Citrus fruits (MF, CP, JP) in an ICR mouse model of obesity induced by a high fat diet. The body weights of IMF fed mice were found to be significantly lower from 2 weeks oral administration, despite the fact that their food intakes were similar to that of the HFD control mice. The fecal cholesterol content showed a significant increase in IMF (52.0 mg/g) at 7 weeks oral administration, and mature Citrus fruits (MF, 41.3 mg/g), immature Citrus peel (CP, 32.2 mg/g), mature Citrus peel (JP, 30.0 mg/g) respectively. And also, the triglyceride content in the feces showed a significant increase in MF (14.6 mg/g), and IMF (14.2 mg/g), CP (10.5 mg/g) and JP (10.0 mg/g), respectively. On the other hand, in the inhibition activity of pancreatic lipase, MF (0.78 nmol/min/ml) showed the greatest decrease in glycerol, CP (0.79 nmol/min/ml), JP, (0.93 nmol/min/ml) and IMF (1.42 nmol/min/ml).

Fermented Peel of Citrus sunki Hort. ex Tanaka Promotes Ethanol Metabolism and Suppresses Body Fat Accumulation

  • Citrus sunki Hort. ex Tanaka is one of several Jeju-native citrus fruits. A number of biological properties for citrus fruits have been suggested, however little is known about those of C. sunki. The present study tested whether fermented product of C. sunki peel (FSP) might affect the activities of enzymes regulating ethanol metabolism. Effects on body weight gain as well as on fatty liver formation were also investigated. The activities of alcohol dehydrogenase and aldehyde dehydrogenase were stimulated remarkably by FSP. Excessive ethanol-induced cytotoxicity was also prevented by FSP in HepG2 cells. FSP decreased the weight gain and fatty liver formation induced by a high-fat diet in mice. From these results, FSP might be a potent source of neutraceuticals useful for preventing ethanol-induced health problems.

사인과 백출 추출 혼합물의 항비만 효과 (Anti-Obesity Effects of Mixture of Atractylodes macrocephala and Amomum villosum Extracts)

  • In this study, we investigated the anti-obesity effects of various mixtures of Atractylodes macrocephala (AM) and Amomum villosum (AV) water extracts on high-fat diet (HFD) induced mouse model. We classified five groups as follows; control, HFD, HFD + AM extracts : AV extracts (100mg/kg) (1:1), HFD + AM extracts : AV extracts (100mg/kg) (2:1), HFD + AM extracts : AV extracts (100mg/kg) (3:1). Oral administration of various mixtures of AM and AV extracts for 6 weeks inhibited HFD-induced increases of body, liver and epididymal fat weights. Also, lipid profiles including LDL cholesterol were improved by various mixtures of AM and AV extracts treatment compared with HFD-fed group. Lipogenesis-related genes such as acetyl coA carboxylase (ACC) and fatty acid synthase (FAS) in liver changed in a favorable way for lipid biosynthesis by HFD compared to control, but various mixtures of AM and AV extracts-treated groups did not. Our results show that various mixtures of AM and AV extracts can prevent HFD-induced obesity in mice and suggests that the mechanisms are involved in expressions and modifications of lipogenesis-related genes such as ACC and FAS in liver.

Effects of Dyglomera® on leptin expression, pro-inflammatory cytokines, and adipocyte browning in 3T3-L1 cells

  • Dyglomera® is an aqueous ethanol extract derived from the fruit and pods of Dichrostachys glomerata. A previous study has revealed that Dyglomera regulates adipogenesis and lipolysis by modulating AMP-activated protein kinase (AMPK) phosphorylation and increased expression levels of lipolysis-related proteins in white adipose tissue of high fat diet-induced mice and 3T3-L1 adipocyte cells. To further investigate mechanisms of Dyglomera, additional studies were performed using 3T3-L1 cells. Results revealed that Dyglomera downregulated adipogenesis by inhibiting the protein kinase B/mammalian target of rapamycin signaling pathway and reconfirmed that it downregulated gene expression levels of proliferator-activated receptor (PPAR)-γ, CCAAT enhancer binding protein α, sterol-regulation element-binding protein-1c. Dyglomera also reduced adipokines such as tumor necrosis factor alpha, interleukin-1β, and interleukin 6 by regulating leptin expression. Moreover, Dyglomera promoted beige-and-brown adipocyte-related phenotypes and regulated metabolism by increasing mitochondrial number and expression levels of genes such as T-box protein 1, transmembrane protein 26, PR domain 16, and cluster of differentiation 40 as well as thermogenic factors such as uncoupling protein 1, proliferator-activated receptor-gamma co-activator-1α, Sirtuin 1, and PPARα through AMPK activation. Thus, Dyglomera not only can inhibit adipogenesis, but also can promote lipolysis and thermogenesis and regulate metabolism by affecting adipokine secretion from 3T3-L1 adipocytes.

LKB1/AMPK 신호 전달 경로의 활성화로 인한 새싹율무 열수 추출물의 항비만 효과 (Coix lacryma-jobi var. mayuen Stapf Sprout Extract Ameliorates High-Fat Diet-Induced Obesity by Upregulating LKB1/AMPK Signaling)

  • Objectives : AMP-activated protein kinase (AMPK) is a key metabolic regulator that reduces lipogenesis. AMPK is mainly activated via phosphorylation of liver kinase B (LKB) 1 under energy stress. Here, we highlighted the anti-obesity effect and underlying mechanism of Coix lacryma-jobi var. mayuen Stapf sprout water extract (CSW) sprout extract in connection with the LKB1/AMPK signaling pathway. Methods : C57BL/6 mice (20~25 g) fed HFD to induce obesity and at the same time administered CSW 100 mg/kg (CSWL; (CSWL; CSW low concentration) or CSW 200 mg/kg (CSWH; CSW high concentration) or Garcinia extract (Garcinia) 200 mg/kg orally for 6 weeks. Body weight and food intake were measured at the same time each day. After 6 weeks of CSW administration, liver tissue and serum were obtained through an autopsy. After the end of the experiment, biochemical analysis (triglycerides (TG), total cholesterol (TC), HDL-cholesterol, and LDL-cholesterol) was performed on the serum. And then, protein levels related to TG and TC synthesis were measured through western blot analysis in liver tissue. Results : As a result, serum TG, TC, and LDL-cholesterol levels were significantly increased in the control group and significantly decreased in the CSW administration group. On the other hand, the HDL-cholesterol level was increased in the CSW-administered group. And as a result of Western blot analysis, CSW significantly increased the phosphorylation of LKB1 & AMPK, and remarkably decreased the expression of factors related to TG and TC synthesis. Conclusions : Our findings suggest that CSW influences the TG and TC synthesis to positively affect HFD-induced obesity in C57BL/6 mice.
